항종양 면역반응 유도를 위한 수지상세포의 최적 활성화 조건 (The Optimal Activation State of Dendritic Cells for the Induction of Antitumor Immunity)

  • 수지상세포는 종양면역에서 필수적인 강력한 CTL 반응을 개시할 수 있는 유일한 세포이다 . 특히 외인성 종양항원에 대한 CTL 반응 유도는 활성화된 수지상세포의 IL-12 분비를 통한 CD4+ helper T세포의 cross-priming을 필요로 한다. 그러나 최근에 활성화된 수지상세포는 $Th_1$ 면역반응을 유도하지만 활성화 시간이 경과함에 따라 오히려 $Th_2$반응을 유도 할 수 있다. 따라서 본 연구에서는 OVA를 종양항원 모델로 설정하여 종양특이적인 CTL 반응을 형성하기 위한 최적의 수지상세포 활성화 조건을 조사하였다. 마우스 골수세포에 서 수지상세포로의 분화는 항원제시 기능을 위한 표면분자의 발현 측면에서 볼 때 배양 6일-7일 정도가 적합하였다. 수지상세포의 IL-12 생성능은 배양 6일 이상, OVA 항원 탑재 8시간 이상의 경우에 연이은 LPS 성숙자극으로 오히려 감소하는 경향을 보였다. 즉 배양 6일의 수지상세포에 OVA 항원 탑재를 8시간 수행한 경우(8-h DC)가 in vitro에서의 IL-12생성능, ex vivo에서의 세포내 $IFN-{\gamma}$를 발현하는 CD8+ T세포의 증가 및 OVA 특이적인 세포독성효과 등에서 가장 좋은 결과를 보였다. 또한 in vivo에서 종양 치료 및 예방효과에서도 8-h DC로 면역한 경우에 가장 우수한 종양형성 억제 효과와 생존기간 연장효과를 보였다. 현재 대부분의 수지상 세포를 이용한 항종양 백신에서 항원 탑재반응을 24시간 동안 수행하고 있으나, 본 실험의 결과로 볼 때, 8시간의 in vitro 항원 탑재가 보다 효과적인 종양특이적 CTL 반응과 항종양 면역반응을 유도함을 알 수 있다. 결론적으로 본 연구를 통하여 8시간 이상의 항원접촉은 수지상세포의 기능적 활성능력을 오히려 고갈시킬 수 있음을 제시한다.

백서 삼차신경절내 신경세포체의 치아이동에 따른 CGRP 면역염색성의 변화 (Changes of CGRP immunoreactivity in rat trigeminal ganglion neurons during tooth movement)

  • 이 연구의 목적은 중추신경계에서는 동통의 전달을 조절하며 말초조직에서는 혈관 이완, 면역체계의 조절, 탐식기능 조절 등의 염증에 관여되며 골 생성에도 관여하여 치아이동시 중요한 기능을 할 것으로 추측되고 있는 CGRP의 생성 장소인 삼차신경절내의 세포체의 크기에 따른 CGRP면역 반응성의 변화를 관찰하여 삼차 신경절내 CGRP면역양성을 띄는 각 크기의 세포들의 치아이동의 시간 경과에 따른 반응을 밝혀보고자 하는 것이다. 생후 9 주령의 Sprague-Dawley계 백서 30 마리를 정상 대조군 6 마리, 3 시간 교정력 적용군 5 마리, 12 시간군 4마리, 1 일군 5마리, 3 일군 5 마리, 7 일군 5 마리로 나누어 실험군은 각 해당 시간동안 상악 우측 제 1대구치에30gm내외의 교정력을 가한 후 희생시켰다. 희생시킨 백서의 삼차 신경절을 적출 하여 냉동 절편을 형성한 후 토끼의 항체를 이용하여 면역화학 염색을 시행하였다. 삼차 신경절의 CGRP 면역양성 신경세포체를 크기에 따라 소형 ($20{\mu}m$ 미만), 중형($20-35{\mu}m$), 대형($35{\mu}m$ 초과)으로 나누어 교정력 적용시간에 따른 변화를 관찰하였다. 1. 삼차 신경절내 전 신경세포체중 CGRP면역양성 신경세포체가 차지하는 비율은 정상 대조군이 33.0%였으며 교정력 적용후 1 일 후에는 24.5%로 감소하였으나 7 일 후에는 41.8%로 증가하였다. 2. 정상 대조군에서 CGRP 면역양성 신경세포체중 소형, 중형, 대형은 각각 51.4%, 44.0%, 4.7%였다. 3. 소형의 CCRP 면역양성 신경세포체는 3시간, 12시간 군에서 높게 나타났다. 4. 중형의 CGRP면역양성 신경세포체는 3일, 7일군에서 대형의 CGRP면역양성 신경세포체는 7 일군에 높게 나타났다. 위를 종합하여 볼 때 치아이동 초기에는 CGRP 면역양성 신경세포체중 소형의 세포가, 후기에는 중형, 대형의 세포가 반응한다는 것을 알 수 있었다.

미역취(Solidago virga-aurea var. gigantea Miq.) 뿌리 추출물이 MC3T3-E1 조골세포의 활성과 분화에 미치는 영향 (Effects of Solidago virga-aurea var. gigantea Miq. Root Extracts on the Activity and Differentiation of MC3T3- E1 Osteoblastic Cell)

  • 골조직은 골아세포, 파골세포, 골세포 등으로 구성되며, 골개조시 여러 인자가 세포증식, 분화, 활성화 및 골대사 조절에 관여한다. 이 때 조골세포의 활성은 골형 성 에 중요하므로, 본 연구에서는 MC3T3-E1 조골세포주를 이용하여 식용자원인 미역취뿌리의 조골세포의 증식과 분화활성에 미치는 영향을 조사하였다. 미역취뿌리 메탄을 추출물 및 분획물이 조골세포의 성장에 미치는 영향을 MTT검색법으로 조사한 결과, 미 역 취 뿌리 메탄을 추출물 10, 100${\mu}g/mL$ 처리시 대조군과 비교하여 각각 140, $120\%$ 증가하여 조골세포의 높은 성장률을 보였다. 미역취 뿌리 추출물 및 순차 분획물 시료가 ALP 효소 활성에 미치는 영향을 3일 간격으로 배지교환 및 시료처 리를 하면서 27일 동안 배양시간에 따른 변화를 측정하여 조사하였다 그 결과, 농도 1, 10, 100${\mu}g/mL$ 처리시 n-hexane과 chloroform분획을 제외한 나머지 분획물들이 시간이 지남에 따라 약간의 ALP활성을 증가시켰고, 메탄올 추출물은 27일째에 대조군에 비해 약 4.4배 이상, 양성 대조군에 비해 약 2배 이상 ALP 활성을 증가시켰다. 미 역취뿌리 메탄올추출물은 다시 ALP효소 염색법과 alizarin red 염색으로 조골세포의 ALP활성유도, 분화와 석회화 형성능을 재확인하였다. 따라서 미역취뿌리 추출물중 골세포의 활성을 촉진시키는 물질은 극성 에 따른 분획물보다 mothanol추출물에서 활성이 높게 나타나는 결과로 미뤄볼 때, 단일성 분에 의한 것보다 복합적 성분들의 상승 작용에 의하여 조골세포의 증식과 분화를 증진시킬 수 있다고 할 수 있다. 아울러 종래의 골질환에 좋다고 알려 진 식품과 양성 대조군에 비해 빠르게 유도하고 있어 앞으로 미역취뿌리에 대한 분자생물학 수준 등의 구체적인 연구들과 기작연구가 지속적으로 이루어져야 할 것이다.

방사선에 대한 생약복합조성물(HemoHIM)의 재생조직 및 면역계 방호 · 회복촉진 효과 (Protective Effect of a Herbal Preparation (HemoHIM) on the Self-Renewal Tissues and Immune System against γ-Irradiation)

  • 본 연구팀에서는 방사선으로부터 위장관과 면역 조혈계를 보호하기 위하여 당귀, 천궁, 백작약으로부터 새로운 생약복합물 HIM-I을 개발한 바 있다. 본 연구에서는 방사선 방호뿐 아니라 다양한 생체질환의 예방에 있어 면역조혈기능이 중추적인 역할을 한다는 관점에서, HIM-I로부터 면역조혈 기능을 더욱 증진시킨 생약복합조성물 HemoHIM을 제조하여 그 효능을 검증하였다. HIM-I를 에탄을 침지하여 에탄을 분획(HIM-I-E)과 조다당 분획 (HIM-I-P)을 얻은 후, HIM-I 에 조다당 분획을 첨가하여 HemoHIM을 제조하였다. 이렇게 얻은 HemoHIM, HIM-I 및 각 분획에 대하여 재생조직 및 면역계 방호와 회복촉진 효과를 비교 검증하였다. HemoHIM 과 HIM-I는 시험 관내에서 방사선에 의한 DNA 손상을 유의 적으로 억제하고 수산화 라디칼을 소거하는 효과가 있음이 관찰되었으며, HemoHIM과 HIM-I는 거의 비슷한 활성을 나타내었다. 시험 관내 면역 세포 활성화와 골수세포 성장촉 진 실험에서는 HemoHIM이 HIM-I 비하여 높은 활성을 보였으며, 이는 HIM-I에 비해 높은 조다당 함량에 기인한 것으로 보인다. 감마선 조사 마우스를 이용한 생체 보호효과를 살펴본 결과, HemoHIM은 HIM-I와 비슷한 정도의 소장 움 생존율 증가효과를 보였으나, 내재성 비장 조혈세포집락 형성 시험에서는 HemoHIM은 HIM-I보다 높은 효과를 나타내었다. 또한 방사선조사 마우스에서 HemoHTM의 투여는 방사선 조사후 급격히 감소된 말초 혈액내 백혈구 및 림프구수의 회복을 촉진시키고, 생존율을 증대시키는 효과가 관찰되었다. 이상의 결과들은 생약복합물 HIM-I 조다당 분획을 첨가하여 개발한 새로운 생약복합조성물 HemoHIM이 방사선에 의해 유발된 위 장관 및 면역계 조직의 손상을 감소 시켜 생존을 증가시키는 효과가 있음을 제시하였다. 특히 HemoHIM은 HIM-I와 비교하여 재생조직의 산화적 손상억제 효과는 비슷하게 유지되면서도 면역 조혈세포 방호 및 회복촉진 효과가 높은 것으로 관찰되어 방사선 방호제로서 뿐만 아니라 면역조혈기능 증진제로서 유용하게 활용될 수 있을 것으로 사료된다.

골관절염 실험모델에서 꾸지뽕나무 추출물의 골관절염 억제효과 연구 (Therapeutic Effects of Curdrania tricuspidata Leaf Extract on Osteoarthritis)

  • 본 실험에서는 primary culture된 연골세포 in vitro 실험모델과 MIA로 유발한 골관절염 in vivo 실험모델을 이용하여 꾸지뽕나무 잎 추출물의 관절염 예방 효과를 확인하였다. 먼저 MTT 시험법을 통해 세포 사용 적정농도를 $500{\mu}g/mL$ 이하로 결정하여 연골세포사멸 억제를 확인하고, 이를 근간으로 골관절염 동물실험 모델에서 골관절염 예방효과를 확인하였다. $H_2O_2$ 처리에 따른 산화적 독성으로 연골세포 사멸을 유도한 실험에서 꾸지뽕 잎 추출물은 정상세포 수준으로 사멸을 억제하였으며, 이러한 효과는 CTL80의 $200{\mu}g/mL$, CTL10의 $300{\mu}g/mL$ 농도에서 비교적 높게 나타났다. 교원질 합성을 억제하고 분해를 촉진시키는 MMPs(MMP-7, MMP-13)의 발현을 실시간 정량 PCR로 측정하여 발현변화를 살펴보았다. 그 결과 앞선 세포실험 결과와 마찬가지로 CTL80과 CTL10 처리군에서 발현이 유의적으로 낮아졌음을 살펴볼 수 있었다. 특히 CTL80에서 MMP-7과 MMP-13의 발현이 농도 유의적으로 감소하였으며, CTL10의 경우 200, $300{\mu}g/mL$ 농도에서 유의적으로 발현이 감소하는 것을 확인하였다. 세포실험 결과를 바탕으로 동물실험에서의 적정농도를 결정하였으며, 동물독성실험 결과 이상이 없음을 확인하고 실험을 진행하였다. 이때 세포실험결과 선정된 두농도(200, $300{\mu}g/mL$) 간의 차이가 미미하여 동물실험에 적용할 경우 비슷한 실험결과가 나타날 것으로 사료되어 두 실험군 간의 결과를 정확히 구분 짓기 위해 200, $500{\mu}g/mL$ 농도를 선정하여 사용하였다. 골관절염 유발 동물모델을 만들기 위해 SD rat의 관절강에 MIA를 injection 하였으며, 꾸지뽕 잎 에탄올 추출물 투여에 따른 관절염 예방 효과를 관찰하기 위해 관절염 유발 2주일 전부터 1일 1회 경구투여를 실시하고, 유발 후 3주간 지속적으로 투여하고 관찰하였다. 동물 관절의 병리학적 변화를 관찰하기 위하여 Micro-CT 촬영 및 분석을 실시한 결과 Control 군은 골의 강도와 밀도가 감소한 반면, 양성대조군인 MTX 투여군에서 정상군과 비슷한 수준으로 회복된 것을 확인하였고, CTL80-200군과 CTL10-500군에서 Control 군에 비해 유의적으로 수치가 감소하여 골관절염에 따른 손상이 감소한 것을 확인하였다. 동물의 관절조직의 H&E 염색을 통한 조직학적인 변화에서는 골관절염 유발로 연골세포의 손상과 뼈조직의 손상을 관찰하였으며 관절형태를 알아볼 수 없을 정도로 손상된 것을 확인하였다. 반면 CTL80과 CTL10에서는 관절강 세포의 형태가 정상군과 비슷한 둥근모양을 띤 양상을 보였으며 연골조직의 형태가 잘 유지되어 Control 군에 비해 꾸지뽕잎의 투여효과가 나타났음을 관찰하였다. 이상의 결과를 통하여 꾸지뽕 잎 에탄올 추출물은 높은 항관절염 효과가 있을 것으로 사료되며, 항관절염 효능을 지니는 기능성 소재로써 개발 가능성을 확인하였다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. And the surface charge density (or the number of ionizable functional groups) of the microspheres synthesized by "two stage shot growth method" was 6~13 times higher than that of the microspheres synthesized by conventional seeded batch copolymerization. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.
