고상반응법에 의한 아커마나이트 분말의 합성 및 생체활성도 평가 (Synthesis of akermanite bioceramics by solid-state reaction and evaluation of its bioactivity)

  • 치과용 임플란트 재료로 주로 사용되는 지르코니아 및 티타늄 합금은 생체불활성 특징으로 인하여 골유착 및 골형성 능력이 떨어진다. 이러한 문제를 쉽고 간단하게 해결하기 위한 방법으로는 생체활성 물질을 표면에 코팅하여 생체 활성을 높이는 방법이 있다. 본 연구에서는 우수한 골결합 능력을 가진 실리케이트계 세라믹인 아커마나이트(Ca2MgSi2O7)를 고상반응법으로 합성하고, SBF 용액 내 침적실험을 통하여 합성 아커마나이트 분말의 생체활성을 분석하였다. 고상반응 출발원료로는 탄산칼슘(CaCO3), 탄산마그네슘(MgCO3), 이산화규소(SiO2) 분말을 사용하였다. 분말을 혼합 및 건조한 후, 가압 성형하여 디스크 형태로 만든 후, 고상반응 온도를 변화시키며 아커마나이트 상의 합성을 유도하였다. 합성된 아커마나이트 펠릿의 용해 및 생체활성 분석을 위하여 SBF 용액 내 침적 시키고, 침적시간에 따라 아커마나이트의 표면 용해 및 하이드록시아파타이트 석출을 분석하였다. 합성반응 온도가 높아질수록 아커마나이트 상이 뚜렷하게 나타난 반면에, SBF 용액 내 용해는 천천히 진행되었다. 합성된 아커마나이트 분말의 생체활성도는 대체적으로 우수하였으나, 그 중에서도 1100℃에서 고상반응 하여 합성한 분말에서 적절한 용해 및 하이드록시아파타이트 입자의 석출이 잘 일어나는 것으로 분석되었다.

Comparison of removal torques between laser-etched and modified sandblasted acid-etched Ti implant surfaces in rabbit tibias

  • PURPOSE. The purpose of this study was to analyze the effects of two different implant surface treatments on initial bone connection by comparing the Removal Torque Values (RTQs) at 7 and 10 days after chemically modified, sandblasted, large-grit and acid-etched (modSLA), and Laser-etched (LE) Ti implant placements. MATERIALS AND METHODS. Twenty modSLA and 20 LE implants were installed on the left and right tibias of 20 adult rabbits. RTQs were measured after 7 and 10 days in 10 rabbits each. Scanning electron microscope (SEM) photographs of the two implants were observed by using Quanta FEG 650 from the FEI company (Hillsboro, OR, USA). Analyses of surface elements and components were conducted using energy dispersive spectroscopy (EDS, Horiba, Kyoto, Japan). RESULTS. The mean RTQs were $12.29{\pm}0.830$ and $12.19{\pm}0.713$ Ncm after 7 days (P=.928) and $16.47{\pm}1.324$ and $16.17{\pm}1.165$ Ncm after 10 days (P=.867) for LE and modSLA, respectively, indicating no significant inter-group differences. Pore sizes in the LE were $40{\mu}m$ and consisted of numerous small pores, whereas pore sizes in the modSLA were $5{\mu}m$. In the EDS analysis, Ti, O, and C were the only three elements found in the LE surfaces. Na, Ca, Cl, and K were also observed in modSLA, in addition to Ti, O, and C. CONCLUSION. The implants showed no significant difference in biomechanical bond strength to bone in early-stage osseointegration. LE implant can be considered an excellent surface treatment method in addition to the modSLA implant and can be applied to the early loading of the prosthesis clinically.

산호 유래 실리콘 함유 다공성 수산화아파타이트의 합성 및 특성 분석 (Preparation and Characterization of Porous Hydroxyapatite Containing Silicon Derived from Natural Coral)

  • 생체친화성 인공골 개발을 위하여 해면골과 매우 유사한 기공구조를 갖는 천연 산호(CaCO$_3$, aragonite phase)를 수열처리하여 수산화아파타이트 다공체를 제조하였다. 생체 친화성을 증진시키기 위하여 산호를 수열 처리하는 과정에서 실리콘을 치환시키는 실험을 하였다. 수열합성과 solvothermal 방법을 반복적으로 사용하여 실리콘이 치환된 수산화아파타이트를 합성하였으며 이들의 각 온도에 따른 상변화 및 실리콘 농도를 XRD, ICP, EDS 등을 통하여 분석하였다. 분석결과로부터 실리콘이 수산화아파타이트 골격 내에 균일하게 분산되어 있음을 확인하였다.

In-vitro and In-vivo Biocompatibility Evaluation of Silica Based Bio-active Glass Prepared by Hydrothermal Method

  • Bioactive glass powders were synthesized by hydrothermal chemical route by the use of ultrasonic energy irradiation. We used sodalime, calcium nitrate tetra hydrate and di ammonium hydrogen phosphate as the precursor material to synthesize $SiO_2$ rich bio-active glass materials. The $SiO_2$ content was varied in the precursor mixture to 60, 52 and 45 mole%. Dense compacts were obtained by microwave sintering at $1,100^{\circ}C$. Mechanical properties were characterized for the fabricated dense bioactive glasses and were found to be comparable with conventional CaO-$SiO_2$-$Na_2O$-$P_2O_5$ bioactive glass. Detailed biocompatibility evaluation of the glass composition was investigated by in-vitro culture of MG-63 cell and mesenchyme stem cell. Cell adhesion behavior was investigated for both of the cell by one cell morphology for 30, 60 and 90 minutes. Cell proliferation behavior was investigated by culturing both of the cells for 1, 3 and 7 days and was found to be excellent. Both SEM and confocal laser scanning microscopy were used for the investigation. Western blot analysis was performed to evaluate the bimolecular level interaction and extent and rate of specific protein expression. The ability to form biological apatite in physiological condition was observed with simulated body fluid (SBF). In-vivo bone formation behavior was investigated after implanting the materials inside rabbit femur for 1 and 3 month. The bone formation behavior was excellent in all the bioglass compositions, specially the composition with 60% $SiO_2$ content showed most promising trend.

골감소증을 동반한 지속성 복막투석환자의 다빈도섭취 음식조사 및 골밀도 개선을 위한 레시피 개발 (Frequently Consumed Dishes and Development of Recipes to Improve Bone Mineral densities in Continuous Ambulatory Peritoneal Dialysis Patients with Osteopenia)

  • The purpose of this study was to assess the frequently consumed dish consumption frequencies of continuous ambulatory peritoneal dialysis(CAPD) patients with osteopenia and develop recipes to improve bone mineral density of CAPD. The subjects were 96 CAPD patients with osteopenia(male 39, female 57) (osteopenia group) and 45 CAPD patients with normal BMD(male 24, female 21), matched with key variables(normal group). Fifty dishes(foods) that most frequently consumed were determined and food consumption frequency for each dish(food) for two groups were compared. Osteopenia group showed lower consumption frequency for ice-cream but higher frequency in apple. Of the 50 most frequently consumed dishes(foods), 20 dishes assessed as safe and recommendable for CAPD patients with osteopenia based on the contents of protein and mineral were selected : white boiled rice, white gruel, beef soup, steamed cabbage, roasted dried laver, fried egg, roasted bean-curd, cooked and seasoned bean sprouts, corn-starch jelly, cheese, ice-cream, orange juice, apple, grape, peach, peanut, raw lettuce, raw cucumber, and injulmi rice cake. wenty eight new dishes with modified recipes were developed for CAPD patients. Protein and mineral contents were analyzed for frequently consumed 17 dishes, assessed as modification of recipes are needed. The recipes were modified to decrease P, Na and K contents and to increase protein and Ca contents. Twenty dishes(foods) selected as having reasonable protein and mineral contents ratio or 28 newly developed dishes modified with protein and mineral contents or ratio would be helpful for nutrition education or counseling for CAPD patients with osteopenia. Dishes(foods) suggested in this study would also be useful for all CAPD patients for preventing osteoporosis.

난소 적출 마우스 모델에서 Oryza sativa L.의 뿌리 추출물에서 분리된 화합물의 골다공증 개선 효과에 관한 예비연구 (Preliminary Study on the Osteoporosis Improvement Effect of Compounds Isolated from Oryza sativa L. root Extract in the Ovariectomized Mouse Model)

  • Bisphosphonates, estrogen, and calcium supplements are commonly used medications for postmenopausal osteoporosis, but they are associated with various side effects such as vaginal bleeding, deep vein thromembolism, and breast cancer. In this study, we aimed to investigate the potential of a compound isolated from the roots of Oryza sativa L. to improve osteoporosis using an ovariectomized mouse model. We isolated and identified oryzativol A, a lignan compound, through chemical analysis of an ethanol extract using a bioassay-guided fractionation protocol. We also examined the metabolism, clearance, and CYP enzyme activity of oryzativol A, and found that it showed plasma stability of over 80% at all analysis times, and indicating a low likelihood of inactivation or excretion by the CYP3A4 enzyme. Our results showed that the high-dose group of oryzativol A exhibited a significant increase in bone mineral density compared to the control group. Although the ALP concentration did not differ significantly compared to the control group, it showed a tendency to increase in the high-dose group of oryzativol A. Furthermore, the abnormal ratio of serum Ca/P, caused by osteoporosis, was improved to a level closer to that of the normal group as the dosage of oryzativol A increased. Taken together, these findings suggest that oryzativol A is stable in vivo and has potential as a therapeutic agent for osteoporosis, particularly when administered in high doses.

Aflatoxin $B_1$과 Vitamin $D_3$ 결여가 Broiler병아리의 중족골 무기물함량에 미치는 영향 (Effect of Feeding Aflatoxin $B_1$ and Vitamin $D_3$ on Metatarsus Bone Minerals of Broiler Chicks)

  • Aflatoxin $B_1$($AFB_1$ 0 또는 1ppm)과 Vitamin $D_3$($VD_3$0, 500, 1,000 또는 1,500IU/kg)의 교호작용을 조사코자, Hubbard계통 broiler 병아리 336수를 이용, 2$\times$4 요인시험이 실시되었으며, 전체8처리가 3반복으로 설계되어 탈지건조 중족골의 회분 및 무기물함량과 중량 등을 측정하였다. 얻어진 결과를 요약하면 다음과 같다. L 중족골의 신선중량에 있어서 $AFB_1$ 급여에 의한 차이는 나타내지 않았으며 $VD_3$의 수준이 증가됨에 따라 감소되는 경향을 보였다(P<.01). 2. 탈지건조 중족골의 회분함양을 보면 $AFB_1$의 급여로 인하여 감소되었으며(P<.01), $VD_3$수준이 증가될 때 반대로 증가되었다(P<.01). 교호효과는 인정되지 않았으나 $AFB_1$ 1ppm 급여로 인한 회분감소를 극복하기 위하여 $VD_3$ 1,500IU/kg의 급여가 필요하다는 것이 증명되었다. 3. Ca의 함양은 AFB$_1$급여의 영향을 받지 않았으나 $VD_3$에 의하여 증가되는 경향을 보였다(P<0.05). 4. P의 함양은 $AFB_1$급여로 감소되었으나(P<.01) $VD_3$의 영향은 없었다. 5. Cu의 함양은 AFB$_1$$AFB_1$에 의하여 감소되었다(P<.05). 6. Na Mg, K, Zn, Fe 및 Mn의 함량은 $AFB_1$이나 $VD_3$에 의하여 영향을 받지 않았다.

강화 석모도 지역 온천수와 지하수의 수리지구화학 및 동위원소 연구 (Hydrogeochemical, Stable and Noble Gas Isotopic Studies of Hot Spring Waters and Cold Groundwaters in the Seokmodo Hot Spring Area of the Ganghwa Province, South Korea)

  • 강화 석모도 지역 석모도 온천수의 영족기체와 온천수의 지화학적 진화와 기원을 해석하고 온천수의 지화학적 특성을 규명하기 위해 온천수, 지하수, 지표수의 수리화학, 안정 동위원소, 영족기체 동위원소 분석이 이루어졌다. 온천수와 지하수의 pH는 각각 $6.42{\sim}6.77,\;6.01{\sim}7.71$로 약산성을 보이고 있다. 석모도 온천 지역의 온천수의 유출수온은 $43.3{\sim}68.6^{\circ}C$이다. 온천수의 전기전도도는 $60,200{\sim}84,300{\mu}S/cm$으로 비교적 높은 값이며 석모도 온천수가 해수와 혼합되어졌음을 암시하고 있다. 석모도 온천수의 화학 조성은 Na-Ca-Cl형이다. 반면, 지하수와 지표수는 각각 Na(Ca)-$HCO_3$, Na(Ca)-$SO_4$형과 Ca-$HCO_3$ 형으로 구분된다. 석모도 온천수의 산소와 수소 동위원소비는 각각 $-4.41{\sim}-4.47%o$$-32.0{\sim}-33.5%o$로 순환수 기원이다. 지하수에서의 산수 수소 동위원소비는 각각 $-7.07{\sim}-8.55%o,\;-50.24{\sim}-59.6%o$이다. 석모도 온천수에 $^{18}O$$^2H$가 부화된 특성은 온천수가 담수와 해수의 혼합대에서 유래되었음을 암시하고 있다. 석모도 온천수 중의 황산염이온의 황 동위원소비는 $23.1{\sim}23.5%o$로 이 지역 해수의 황 동위원소비(20.2%o)와 유사하다. 이는 온천수의 황이 해수의 황산염으로부터 유래되었음을 의미한다. 석모도 온천수의 $^3He/^4He$ 비는 $1.243{\times}10^{-6}{\sim}1.299{\times}10^{-6}cm^3STP/g$로 온천수 중의 He 가스가 부분적으로 맨틀에서 유래되었음을 보여준다. 온천수에서의 아르곤 동위원소비$(^{40}Ar/^{36}Ar=298{\times}10^{-6})cm^3STP/g$는 대기기원의 값을 보인다.

Microstructure and Mechanical Properties of Ti-35Nb-7Zr-XCPP Biomaterials Fabricated by Rapid Sintering

  • Ti-6Al-4V ELI (Extra Low Interstitial) alloy have been widely used as alternative to bone due to its excellent biocompatibility, although it still has many problems such as high elastic modulus and toxicity. Therefore, biomaterials with low elastic modulus and non toxic characteristics have to be developed. A novel ${\beta}$ Ti-35wt%Nb-7wt%Zr-Calcium pyrophosphate (CPP) composite that is a biocompatible alloy without elemental Al or V was fabricated by spark plasma sintering (SPS) at $1000^{\circ}C$ under 70 MPa using high energy mechanical milled (HEMM) powder. The microstructure and phases of the milled powders and the sintered specimens were studied using SEM, TEM, and XRD. Ti-35wt%Nb-7wt%Zr alloy was transformed from ${\alpha}$ phase to ${\beta}$ phase in the 4h-milled powder by sintering. The sintered specimen using the 4h-milled powder showed that all the elements were distributed very homogeneously and had higher density and hardness. ${\beta}$ Ti alloy-CPP composite, which has nanometer particles, was fabricated by SPS using HEMMed powder. During the sintering process, $CaTiO_3$, TixOy, and CaO were formed because of the reaction between Ti and CPP. The Vickers hardness of the composites increases with the increase of the milling time and the addition of CPP. The biocompatibility of the Ti-Nb-Zr alloys was improved by addition of CPP.

난소절제 흰쥐에서 대두 이소플라본의 섭취수준에 따른 Ca과 P의 평형 연구 (Calcium and Phosphorus Balance Study by Soy Isoflavone Intake in Ovariectomized Rats)

  • This study was performed to determine the effect of three different concentrations of soy-isoflavones on calcium and phosphorus balance in either sham-operated or ovariectomized female rats. Seventy-two 16-week old Sprague-Dawley rats underwent sham operation or bilateral ovariectomy. They were provided diets containing different levels of soy iso-flavones for 6 weeks: 50 ppm (Jow isoflavone intake; LI) , 250 ppm (medium isoflavone intake; MI) and 500 ppm (high isoflavone intake; HI). The subsequent fecal and urinary excretions of calcium and phosphorus were then measured. In the sham-operated rats, body weight gains and food efficiency ratio of the MI and HI groups were significantly lower than the control group while food intake was not different. However, there was no significant difference in the ovariectomized rats. The fecal excretion of calcium was significantly lower in the LI, MI and HI groups than the control group in sham operated rats, and significantly lower in the HI group than the control group in ovariectomized rats. Also, apparent ab-sorption rate of calcium and phosphorus did not show any significant difference among groups. Urinary excretion of calcium and phosphorus was significantly lower in the HI group than the LI group in the sham-operated rats. Urinary excretion of calcium was significantly higher in the control ovariectomized rats than in the control sham-operated rats. Retention of calcium and phosphorus did not show any significant difference in both groups. From the above result, we see that isoflavone intake increases calcium retention through an increase in calcium absorption and also suppresses the increase of calcium excretion in urine in ovariectomy. Therefore, it is suggested that isoflavone intake is recommended for menopausal women who experience sharp bone loss due to the decrease in estrogen honnone.
