  • The purpose of this present study evaluated the osseous response around Ca-P coated xenogenic bone and compared osteogenic potential of Ca-P coated xenogenic bone to that of combination with type I collagen derived from bovine tendon as a biocompatible binder to prevent migration of bone particle on the repair of calvarial defects in rabbits. To study the effects of Ca-P coated xenogenic bone and collagen on bone healing, four 5-mm-diameter skull defect were made in calvaria with trephine filled with an autogenous bone chip or Ca-P coated xenogenic bone or Ca-P coated xenogenic bone and type I collagen (1:1 mixture by volume) or left empty. The defects were evaluated histologically at 1, 2, 4 and 8 weeks following implantation. Ca-P coated xenogenic bone at the calvarial defects of rabbits showed osteoconductivity at the margin of defect in the early stage of bony healing, but no direct contact with new bone was observed. With time passed by, it was resorbed slowly and showed consistent inflammatory reaction. An additional use of type I collagen derived from bovine tendon improved clinical handling, but no new bone formation was observed histologically. Above all, autogenous bone graft showed most prominent healing in quantity and density of new bone formation. According to this study, the use of Ca-P coated xenogenic bone alone and combination with type I collagen did not showed effective healing in quantity and density of new bone formation.

  • This study was undertaken to investigate the effects of age and of the dietary protein sources & levels on urinary calcium excretion and bone metabolism in the rats. Two experiments were conducted . In experiment U, 6& 20 weeks old rats were fed 8 & 36%, casein & ffish protein diet for 5 weeks . In experiment II, 16& 52 weeks old rats were fed 40 % protein diets, protein sources were casein, fish and gluten. High-protein diet group excreted more caldium in urine than low-protein group. Urinary calcium excretion was affected by the sources of protein ; gluten group excreted more Ca, followed by fish & casein group. Total bone Ca & Ca proportion in ash were higher in 20 weeks old rats than 6 weeks old rats, but 16 & 52 weeks old rats showed no differences. bone composition showed that water proportion was high and ash proportion was low in 6 weeks old rats than in 20 weeks old rats. However, these tendencies were not observed between 16 weeks and 52 weeks old rats. And bone composition was affected by protein sources ; higher ash proportions were noted in one order of casein, fish, and gluten water proportion were lower in one same order.

  • Purpose: This study was aimed to evaluate the effect of the deproteinated bovine bone powder (DBBP) coated with calcium phosphate (Ca-P) on osseous regeneration in the calvarial bone defect of rat. Materials and Methods : The DBBP (Control group, n=6) and the Ca-P coated DBBP (Experimental group, n=6) were grafted in the critical sized calvarial bone defect (8 mm) of rat weighing 250 g. The animals were sacrificed at 1, 4 week. The biopsy specimens were decalcified with 5%formaldehyde and embedded in paraffin. The rats were sacrificed at 8 week received tetracycline (1 week), calcein blue (4 week), and alizarin red (7 week), and the biopsy specimens were taken. The specimens were embedded in methylmethacrylate and ground to 10 ${\mu}m$ thin sections were made. All of the specimens were stained with H & E and Masson's trichrome and examined under light microscope. The specimens at 8 week were examined under fluorescent microscope. Results : In the Control group, the grafted DBBP was surrounded with connective tissue, and osteoblasts were observed partially around the grafted particles at 1 week. At 4 week, some osteoid was observed and, new bone formation was observed at the periphery of grafted materials at 8 week, In the Experimental group, some osteoid was seen at the periphery of the grafted Ca-P coated DBBP at 1 week, and osteoblast and newly formed bone were observed around the grafted materials. At 8 week, newly formed bone was observed at the periphery of the grafted materials. Conclusion: These results suggest that Ca-P coated DBBP group was more and faster than DBBP group in new bone formation and Ca-P could contribute to enhance bone formation in the critical sized calvarial bone defect of rat.

  • The purpose of present study was to assess the change of bone mineral density (BMD) by age and the dietary factors influcencing on BMD in Korean women in Taegu. The subjects were 242 healthy female in the range of 7-67 years old, and were divided into 4 age groups. BMD of lumbar of lumbar spine, femur(neck, ward's triangle, trochanter)and total body was measured by dual energy X-ray absorptionmetry. The nutrient intake measured by convenient method was similar to or more than the level of RDA. The significant relationship between nutrient intake and BMD was observed. Particularly for lumbar spine and total body in group 4 (50-60yr), such as energy, carbohyrate, protein, fat, Ca, animal Ca, meat Ca, Fe, thiamin and niacin were found significantly positive correlations. By analysis of multiple regression, significant relationships were shown between protein and lumbar spine and total body BMD in group 4, between ascorbic acid and total body BMD in group 2(17-34 yr). Energy expenditure showed better corrlations with BMD rather than nutrient intake. BMD was significantly greater in subjects consuming a mean dietary Ca intake more than 125% of RDA compared with less than 75% of RDA. In high Ca intake group ($\geq$125% of RDA), there were the significantly negative correlations between animal (meat) Ca and BMD in each bone of 7-9 years. The excessive intake of animal protein in this age group was found. And the correlations between BMD and past milk consumptin were significant in all skeletal sites of group 1(7-16 yr). This study confirms that the most effective way of preventing osteoporosis and the fractures is to maximize peak bone mass in early life and to minimize bone loss through the balanced adequate intake of Ca and other nutrients and regular physical activity.

  • Effects of liquefied calcium supplement on bone mineral density (BMD) and bone health index (osteocalcin, deoxypyridinoline) were investigated in 12 middle aged women. Middle aged women were arranged into 3 groups such as control, Ca500 and Ca750 by liquefied calcium supplement level. BMD was measured at the spine (vertebrae L2-L4). After 6 month, BMD of control group dropped but that of Ca750 were increased from 0.850 to 0.865. The increase in serum calcium and osteocalcin level, and the decrease in urine deoxypyridinoline level were observed in by calcium supplement groups. The bone health index of serum was changed as much as level of calcium supplement. The serum osteocalcin level of Ca750 significantly increased to 7.93 ng/mL after 6 months of calcium supplement. Although we didn't get any significant difference in BMD, we found that the liquefied calcium had no side effect and led effective change in bone health index. Hereafter, we suppose that the liquefied calcium will be available to develop healthy products for preventing osteoporosis.

  • The aim of this investigation was to examine the effects of osteocalcin, bone-specific alkaline phophatase, estrogen, insulin-like growth factor-I (IGF-I), Ca, P and bone mineral density on osteoporosis induced by ovariectomy in rats. Female Sprague-Dawley 30 rats of three-forth's birth, weighing $215{\pm}10g$, were divided into two groups including the sham operation group(5 heads) and ovariectomy group(25 heads). They were fed normal diets for 2 weeks before the experimental operation and for 8 more weeks after operation. The level of osteocalcin, TALP, BALP, estrogen, bone mineral density and IGF-I were increased in experimental group, but a little increased in sham operation group at same period. The change of rates of osteocalcin, TALP, BALP, estrogen, bone mineral density and IGF-I were significantly higher in experimental group than sham operation group. $Ca^{2+}$ was not changed between two groups and P was significantly decreased in experimental group and Ca/P ratio was higher in experimental group than sham operation group. Body weights were increased in all two groups and growth rate per day was higher in experimental group than sham operation group. However, femur weight I body weight ratio was lower in experimental group than sham operation group.

  • This study was carried out to investigate the free amino acid content, the free amino-nitrogen content and minerals such as Ca, P and Fe of chicken bone stock. The results were as follows. 1. In chicken bone stock, glutamic acid was the most abundant amino acid, Iysine, alanine, cysteine and glycine were followed oderly and these free amino acids seemed to be the most intensive flavor compounds. 2. The free amino acid-nitrogen content of chicken bone stock was increased after 3 hours boiling and began to decrease after 4 hours boiling. 3. After 3 hours boiling, Ca, P were 5.0 mg/100 g chicken bone and 42.5 mg/100 g chicken bone, respectively and began to decrease after 4 hours boiling. The ratio of Ca:P was about 7.8. Fe was increased after 4 hours boiling and began to decrease after 5 hours boiling. These results revealed the low effectiveness of Ca, P and Fe utilization of chicken bone stock, because of the very low extraction of minerals. But these results revealed the significant correlation of free amino acid, free amino-nitrogen contents and brothy flavor. So chicken bone stock on heating at 90-95$^{\circ}C$ for 3∼4 hours seemed to be the most intensive brothy flavor.

  • Using the favorable resorption behavior of amorphous Calcium phosphate (CaP) we fabricated a gelatin basednano fibrous mat by electrospinning for using as a fast healing patch for minorbone defects. Bone is predominantly formed by an inorganic phase of nano-crystalline HAp materials and nano fibrous protein material of collagen. The osteoblast cells, which are the bone formation cells and are key to the new bone formation, receive these materials to form new bone. Taking these considerations we make a new nano fibrous mat of amorphous CaP and gelatin, which is derived from collagen itself. A polymer carrier of poly caprolactone(PCL) was used in the system to stabilize the materials in biological condition. The electrospinning conditions were optimized for smooth mat without any droplet formation. The fabricated mat was characterized for its morphologyby SEM. Mechanical properties like tensile strength was evaluated. To investigate the bio-compatibility we performed the MTT assay and investigated its resorption behavior and apatite formation behavior by SBF immersion.
