누에, 맥문동복합물의 흉선 림프절내 T세포 활성 유도 효능 연구 (Mixture Bombyx mori L. and Liriopis seu Ophiopogonis Tuber effects on T cells in Thymus, Lymph Nodes)

  • Objectives : The purpose of this study is to investigate effects on the immune system of Bombyx mori L. and Liriopis seu Ophiopogonis Tuber mixture (BL) in Thymus, Lymph Nodes. Methods : Eight-week-old male Balb/c mice were divided into five groups : Group one included the normal mice (Nor). Positive control group two administrated with red ginseng (RG) 100 mg/kg. Group three administrated with Bombyx mori L. (BX) 300 mg/kg. Group four administrated with Liriopis seu Ophiopogonis Tuber (LP) 300 mg/kg. Group five administrated with the mixture of Bombyx mori L. and Liriopis seu Ophiopogonis Tuber (BL) 300 mg/kg. After 2 weeks administration, mice were sacrified and antigen receptor in Thymus, Lymph Nodes was analyzed by using Fluorescence Activated Cellorter Sorting (FACS). we counted the total of Thymus and Lymph Nodes cells. GOT (glutamlc oxaloacetic transaminase), GPT (glutamlc pyruvic transamlnase) in serum were analyzed after experiment. Results : In Effects of Nor, RG, BX, LP, BL on the ratio of CD4+CD8+, CD4+CD69+ and CD4+CD25+ T cell in Thymus and Lymphnode, BL is higher than other groups except Nor in CD4+, CD4+CD69+, CD4+CD25+ T cell. The number of Thymus and Lymph Nodes increased in BL. In the level of GOT and GPT, BL decreased comparing to others group except Nor. Conclusions : BL may have effect on T cells in Thymus, Lymph Nodes. In addition, Bombyx mori could be immune functional material with others herb materials.

  • Experiments were held in Egypt to test a new Package of practices for commercial cocoon production, including adding secondary macro and micronutrients to the mulberry garden, disease free rearing regime, low temperature rearing in young instars and natural mounting fur silkworms. This package for seed silkworm rearing lead to increase in fecundity by 67-121eggs (15.12-26.22%) and yield of standard boxes per 1 parent egg boxes by 57-58 egg boxes(48.33-51.66%) respectively in comparison th the traditional cocoon production technology.

  • Ecophysiologically diapause represents a syndrome of physiological and biochemical characteristics, all of which ensure survival during a long period of dormancy. Since, silkworm enters diapause as embryo at the early embryonic stage, the duration of egg life depends on the duration of embryonic diapause. The nature of diapause in silkworm, Bombyx mori, is primarily determined by genetic characters and endocrinologicnl mechanisms, mediated by environmental factors such as temperature and photoperiod. Hibernating potency value besides nucleic acid and carbohydrate metabolism, production and utilization of sorbitol are also equally responsible for induction, initiation, determination, maintenance and termination of diapause. Embryonic diapause in Bombyx moir, induced by active secretion of sub-oesophageal ganglion is attributed to hormonal system and metabolic adjustment, which serves to bring about a new physiological state. Metabolic conversion of trehalose to glycogen at induction, glycogen to sorbitol at initiation and sorbitol to glycogen at termination of diapause is correlated and in each metabolic shift a key enzyme becomes active in response to hormonal and environmental stimulation. An attempt has been made in this review article to discuss briefly the nature of embryonic diapause, influence of various factors on diapause nature, hormonal mechanism of diapause besides biochemical composition of egg, nucleic acid and carbohydrate metabolism, production and utilization of sorbitol in relation to induction, determination, maintenance, initiation and termination of diapause in the silkworm, Bombyx mori.

The Physical Methods for Induction of Anti-Bacterial Substances in the Silkworm Larva, Bombyx mori

  • To understand the physical method for induction of anti-bacterial substances from the silkworm larvae, Bombyx mori, three physical methods, i.e., infrared radiation, ultraviolet radiation and ultrasonic wave, have been used in this study. The results have shown that ultrasonic wave can induce anti-bacterial sub-stances effectively than radiations in the B. mori larva. The induction of anti-bacterial substances was different from silkworm race to race. Summer-autumn silkworm race (Qiufeng${\times}$Baiyu) was easy to induce antibacterial substances. It is suggested that the ultrasonic wave is a simple and easy method for induction.

  • A cDNA encoding the luciferase of firefly Luciola lateralis was cloned downstream from the polyhedrin gene promoter of Bombyx mori nucleopolyhedrovirus and expressed in B. mori cells (BmN-4). The coding soquence for luciferase was inserted into pBmKSK2 rectors) which was reconstructed from the polyhedrin-based transfer vector pBmKSKl by modifying cloning sites. Recombinant virus, BmK2-LUCDF, containing the luciferase gene was selected and purified in BmN-4 cells. The emission of luminescence by luciferase was only detected in BmK2-LUCDF-infected cell extracts. This result indicates that the cloned new luciferase gene of firefly L. lateralis can be expressed efficiently in baculovirus expression system and used as a useful reporter gene.

Resistance to Bombyx mori Densonucleosis Virus Type 1 and Its Inheritance in Silkworm, Bombyx mori L.

  • Bombyx mori densonucleosis virus type 1 (BmDNV1)- a non occluded virus causes flacherie disease in the susceptible stocks of the silkworm, Bombyx mori. However, some stocks are non-susceptible. Non-susceptibility to BmDNV1 in B. mori is a unique case where the virus infection is completely inhibited by a single gene of the host. A survey conducted by this institute in some parts of Karnataka state has revealed that, 43.05% of the total incidence of flacherie disease caused by non-occluded viruses, are due to the synergistic infection of B. mori densonucleosis and infectious flacherie virus. Earlier study indicated that rearing of BmDNV1 resistant silkworm stock is effective in protecting silkworm against BmIFV also. In the present study the response of 78 silkworm stocks which include 42 of non-diapausing and 36 of diapausing groups, to BmDNV1 is investigated. Newly ecdysed third instar larvae were inoculated per-os with 10% inoculum of BmDNV1 extracted from the mid-gut of infected silkworm. One non-diapausing and three diapausing silkworm stocks were found to be resistant to BmDNV1. Eleven silkworm stocks were found to possess moderate resistance whereas rest sixty three were found to be susceptible to BmDNV1. Genetic analysis has shown that the resistance to BmDNV1 is autosomally inherited and controlled by a major dominant or a major recessive gene in different silkworm stocks. These resistant stocks can be utilized as the resource material to develop BmDNV1 resistant commercial hybrids. The selection strategies, depending upon the mode of inheritance of resistance in the resource material chosen, are discussed.

Accumulation of Dietary Conjugated Linoleic Acid (CLA) in Silkworm, Bombyx mori

  • Conjugated linoleic acid (CLA) exhibits potent anti-carcinogenic and other biological activities in several animal models. We report here that dietary CLA, chemically synthesized from corn oil, accumulates I silkworm, Bombyx mori, which is used as therapeutic agent for diabetes in Korea and Japan. Mulberry leaves treated with 0.1 or 10% CLA in ethanol were supplied to silkworms from the end of 3rd instar to the 3rd day of the 5th instar. Fresh mulberry leaves or leaves treated with 10% corn oil in ethanol were fed as a check treatment. the amount of total lipids in the larval body ranged from 17.4 to 19.1mg/g of body tissue, which was not significantly affected by the source of the diets. No CLA was found in the control silkworms. But the level of CLA significantly increased to 83.5 mg/g of fat, when fed with mulberry leaves treated with 10% CLA. Only trace amounts of CLA were detected in the larvae reared with check leaves and 0.1% CLA-treated leaves. Mulberry leaves treatd with corn oil or CLA were not palatable to the larvae, resulting in a reduction of larval weight. These results suggest that silkworms containing CLA in body lipids could be producted by dietary CLA.

  • The silkworm Bombyx mori L. was studied as the potential “host” of popular in Uzbekistan biocontrol ectoparasite, entomophagous Bracon hebetor Say. Being one of representatives of economic-beneficial insects, the silkworm (larvae, pupae and imago) can be used as highly sensitive test organism for revealing of neuro toxic effects of insects venom as well as of their purified components in screening assays. In comparative aspect, except a mulberry silkworm, representatives of Uzbekistan pests cotton-boll worm Helycoverpa armigera Hbn., lesser mulberry pyralid Glyphodes pyloalis Wlk., codling moth Corpocapsa pomonella L., malaria mosquito Anopheles claviger and Colorado potato beetle Leptinotarsa decemlineata Say have been subjected to insect toxic test of bracon venom gland extract (VGE) and its fractions which were obtained by gel-chromatography on Sephadex G-100. The paralyzing effect of the VGE and its fractions was shown in a various degree on the pests.

  • This study was designed to investigate the effects of silkworm (Bombyx mori L.) powder (SWP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley(SD) male rats(160$\pm$10g) were fed basic diet (control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a consistent decreases (4.0% and 7.2%, 5.0% and 14.1%, respectively) in liver mitochondria and microsomes of SWP-200 SWP-400 groups compared with control group, and O2 radical level was significantly decreased about 12% in liver cytosol of SWP-400 group compared with control group. Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

Implications of High Temperature and Low Humidity on the Hatching in the Silkworm, Bombyx mori L. Under LD 12 : 12 Condition

  • The implications of temperature (25, 30 and $35^{\circ}C$) and relative humidity (RH; 60, 70 and $80\%$) on the hatching rhythmicity and hatching parameters (percentage and duration) were studied in the silkworm, Bombyx mori L. under natural photoperiod (LD 12 : 12). Disease free layings (DFLs) of two pure silkworm breeds, Pure Mysore (PM, a multivoltine breed) and $NB_4D_2$ (a bivoltine breed), and their hybrid, $PM{\times}NB_4D_2$ were introduced into the experimental conditions on the $3^{rd}$ day of oviposition till completion of hatching. The hatching rhythm was predominantly diurnal under all temperature and humidity conditions, with peaks just after 'lights-on' phase (6 hrs). Extreme temperature and humidity conditions did not alter the hatching rhythmicity, but prolonged the hatching durations, extending it to the next day, coupled with reduced hatching percentage in PM and $PM{\times}NB_4D_2{\cdot}In\;NB_4D_2$, on the other hand, hatching did not extend to the next day. Hatching percentage in this breed, however, reduced below the economic level under high temperature and low humidity conditions. The high temperature and low humidity together, though did not alter the rhythmicity, seems to exert synergetic effect on the hatching percentage and its duration in the silkworm, B. mori.
