  • In this study, Banhasasim-tang extracts (BSTE) have an inhibitory effects on cisplatin-induced nephrotoxicity in mouse model. Cisplatin is the most widely used anticancer drug for treatment of various cancer. However, cisplatin treatment to cancer patients leads to many side effects such as nephrotoxicity and body weight decrease. We hypothesize that BSTE improve the cisplatin-induced side effects in mouse model. We found that BSTE administration protected tubular injury by cisplatin in mouse model. BSTE also inhibited increase of creatinine and BUN induced by cisplatin injection in serum. Collectively, our data suggest that BSTE could be a therapeutic agent for reducing kidney injury induced by cisplatin treatment in cancer patients.

  • This study was performed to investigate the effects of garlic on uncoupling protein 2 (UCP2) transcriptional regulation of UCP2-luciferase transgenic mice fed on a high fat diet to induce obesity. To examine the transcriptional regulation of UCP2, we generated transgenic mice with a UCP2 promoter (-1,830/+30 bp) containing luciferase as a reporter gene. UCP2-luciferase transgenic mice were fed a 45% high-fat diet for 8 weeks to induce obesity. Subsequently, mice were maintained on either a high-fat control diet (TG-CON), or high-fat diets supplemented with 2% (TG-GL2) or 5% (TG-GL5) garlic for a further 8 weeks. Dietary garlic reduced body weight and energy efficiency ratio in the TG-GL5 group, compared to the TG-CON group. Furthermore, garlic supplementation significantly decreased white adipose tissue fat mass and plasma levels of triglycerides, total cholesterol, and leptin in the TG-GL2 and TG-GL5 groups, compared to the TG-CON group. Specifically, UCP2 promoter activity in metabolic tissues such as liver, white adipose tissue, brown adipose tissue, and skeletal muscle was increased by garlic supplementation. These results suggest that dietary garlic was partially associated with an increase of UCP2 transcriptional activity in metabolic tissues for decreasing obesity.

  • Purpose: The purpose of the study was to develop an emotional self-regulation program for obese children and to examine the program's effectiveness on depression, eating habits, and body mass index. Methods: The emotional self-regulation program was developed from theory-based strategies and a pre-post pilot study was conducted with 46 children (19: intervention, 27: control). The program included 6 weekly group sessions over 2 months. The embedded experimental model of concurrent mixed methods was used to better understand research outcomes by converging both quantitative and qualitative data. Anthropometric measurements and surveys were performed before and after the intervention. Three focus group interviews were conducted with all participants after intervention to explore their experiences. Results: The intervention group displayed decreased depression levels in the quantitative outcomes. The participants also expressed positive outcomes such as improvements in confidence, impulse management, intimacy, happiness, and losing weight. Conclusion: These findings suggest that emotional self-regulation skills are crucial factors for obese children not only to manage their emotional problems but also to control their weight. Further research is needed with a larger sample size and randomized controlled trials.

  • Leptin gene, an obesity gene, has been known to involve in the regulation of food intake and body weight. It is also thought to be related to the glucose metabolism, insulin secretion and type II diabetes mellitus. Recently, the production of recombinant leptin protein has been attempted for the application in the treatment of obesity and the correction of hereditary obesity and type II diabetes. In the present study, leptin cDNA was cloned from mouse fat cells by RT-PCR and prokaryotic expression of leptin was attempted in order ot prepare a leptin-specific antigen. Immunization of a rabbit with the leptin-specific antigen into a rabbit resulted in the generation of leptin-specific antiserum that could be useful in the detection of leption expressed in various tissues. The sequence of leptin cDNA prepared in the present study wa identical to the previously reported one. Transformation of E. coli(DH5a) cells with the leptin cDNA-inserted translation vector, pGEX-4T-3-leptin followed by treatment with IPTG (0.1mM) resulted in the expression of a large amount of GST-leptin fusion protein with a molecular weight of 44 KDa as an inclusion body. Denaturation of the insoluble fusion protein by 8M urea, 6M guanidium-HCI or 0.1% 2-mercaptoethanol followed by a slow oxidation could not solubilize the inclusion body. The cell extract was subjected to SDS-PAGE and GST-leptin protein electroeluted from the gel was then injected into a rabbit subcutaneously for the immunization. Anti-GST-leptin rabbit antiserum which had a cross reactivity to the GST-leptin protein was generated. Leptin protein expressed in mouse brain and fat tissues was detected by Western blot immunodetection system using the antiserum generated in the present study.

  • Recently, research investment in the improvement of food safety as a food source and specializing of nutritional source of edible insects is being actively conducted. Cricket especially has been attracting considerable interest in entomophagy; however, research on the safety assessment of cricket is limited. This study investigated the effects of cricket ethanol extract when orally administrated in Sprague-Dawley rats. Here, we performed a 4 wk repeated oral dose toxicity test in Sprague-Dawley rats following the Organization for Economic Cooperation and Development test guidelines 407 under Good Laboratory Practice regulation. Rats were randomly allocated 4 groups: vehicle control, 250, 500, 1,000 mg/kg test groups and administrated based on body weight for 28 d. The animals were observed for mortalities and clinical signs, body weight changes, food and water consumption. At the end of treatment period, blood and urine were collected and analyzed. Subsequently, the animals were sacrificed and subjected to gross pathological examination and organ weight measurement. The organs were preserved for histopathological examination. The results showed that there were no systemic toxicological effects related with the cricket ethanol extract in the 4 wk oral repeated dose toxicity study. It is considered that NOAEL of cricket ethanol extract is greater than 1,000 mg/kg/d and there was no target organ detected.

  • Objectives: The herb of Agastache rugosa (AR) is a traditional herbal medicine used for colds, vomiting and furuncles. However, there are few reports to investigate the inhibitory effects of AR on obesity. In this study, the effects of AR on high fat diet (HFD)-induced obesity and its mechanism of actions were investigated in experimental animals. Methods: The mice were fed HFD for 4 weeks to induce obesity. After randomly divided into normal fat diet, HFD and AR groups, 200 mg/kg of AR was administrated for 4 weeks with continuous HFD feeding while vehicle was orally treated to HFD group. Food intake and body weight were recorded weekly. Results: Increased body weight by HFD was improved by AR treatment. AR administration inhibited an increase of visceral fat weight as well as adipocyte hypertrophy. Hepatic steatosis was ameliorated in AR-treated mice. In addition, treatment of AR attenuated the expression of adipogenic transcription factor, peroxisome proliferator-activated receptor (PPAR)-gamma in the epididymal adipose tissue. Also the increased serum leptin level by HFD was maintained in AR group, leading to inhibition of food intake. Conclusions: AR treatment showed inhibitory effects on HFD-induced obesity by inhibition of PPAR-gamma and reduction of food intake. AR could be an alternative treatment for obesity.

  • The biochemical characteristics of immunorecative atrial natriuretic peptide (irAUPP and the changes in the levels of irANP in the heart were investigated during the metamorphosis of frog tadpoles. Immunohistochemical localization of pro-ANP in cardiocfes and the presence of irAW in the peritoneal fluid of metamorphosing tadpoles were also examined. The major form of irANP in the cardiocvtes of tadpoles (Rana omurensisp was high molecular weight on gel filtration chromatographv and reverse-phase HPLC. The levels of irANP in the atrium of tadpoles were five to seven times higher than those in the ventricle. In metamorphosing tadpoles the levels of irANP in the atrium increased at stage XX, the climax of metamorphosis, and decreased at stage XXV (P < 0.051, the completion of metamorphosis. When the levels of irANP was expressed as a function of body weight of tadpoles, a continuous increase in the levels of irANP was observed from pre- to postmetamorphosis (P < 0.051. The levels of irAUP in the ventricle were found to be higher in the adult frogs than in tadpoles (R. omurensisl (P < 0.01). Pro-ANP (31-67) immunoreactivity was detected in the ventricle as well as in the atrium of tadpoles fR. nigromaculotal. The peritoneal fluid was also found to contain low molecular weight of irAUP and the levmils of irANP were 55.4 $\pm$ 9.1 pg/ml. Changes of the level of irANP at different stases of the life cycle suggest that ANP may play a role in the regulation of body fluid homeostasis of frog tadpoles during the metamorphosis.

  • In this study, we examined the hepatic anti-steatosis activity of carnosic acid (CA), a phenolic compound of rosemary (Rosmarinus officinalis) leaves, as well as its possible mechanism of action, in a high-fat diet (HFD)-fed mice model. Mice were fed a HFD, or a HFD supplemented with 0.01% (w/w) CA or 0.02% (w/w) CA, for a period of 12 weeks, after which changes in body weight, blood lipid profiles, and fatty acid mechanism markers were evaluated. The 0.02% (w/w) CA diet resulted in a marked decline in steatosis grade, as well as in homeostasis model assessment of insulin resistance (HOMA-IR) index values, intraperitoneal glucose tolerance test (IGTT) results, body weight gain, liver weight, and blood lipid levels (P < 0.05). The expression level of hepatic lipogenic genes, such as sterol regulating element binding protein-1c (SREBP-1c), liver-fatty acid binding protein (L-FABP), stearoyl-CoA desaturase 1 (SCD1), and fatty acid synthase (FAS), was significantly lower in mice fed 0.01% (w/w) CA and 0.02% (w/w) CA diets than that in the HFD group; on the other hand, the expression level of ${\beta}$-oxidation-related genes, such as peroxisome proliferator-activated receptor ${\alpha}$ (PPAR-${\alpha}$), carnitine palmitoyltransferase 1 (CPT-1), and acyl-CoA oxidase (ACO), was higher in mice fed a 0.02% (w/w) CA diet, than that in the HFD group (P < 0.05). In addition, the hepatic content of palmitic acid (C16:0), palmitoleic acid (C16:1), and oleic acid (C18:1) was significantly lower in mice fed the 0.02% (w/w) CA diet than that in the HFD group (P < 0.05). These results suggest that orally administered CA suppressed HFD-induced hepatic steatosis and fatty liver-related metabolic disorders through decrease of de novo lipogenesis and fatty acid elongation and increase of fatty acid ${\beta}$-oxidation in mice.
