Usefulness of Automated PCR Test for Detection of Mycobacterium tuberculosis in Clinical Samples (임상검체별 결핵균 검출을 위한 자동화 중합효소연쇄반응 검사의 유용성)

  • The purpose of this study was to evaluate the usefulness of the automated TB-PCR assay for the detection of Mycobacterium tuberculosis. The 807 cases were analyzed with their TB-PCR, AFB smear and culture in bronchial washing fluids, sputum and body fluids samples. The TB-PCR positive of the bronchial washing fluid, sputum and body fluids were 11.3%, 7.3% and 3.6%, respectively, in cases of AFB smear-negative and culture-negative. The sensitivity values of the bronchial washing fluid, sputum and body fluids were 93.3%, 100% and 50%, respectively, according to the culture result. The sensitivity of body fluids was lower than that of the bronchial washing fluid and sputum. The specificity values of the bronchial washing fluid, sputum and body fluids were 83.3%, 89.0% and 95.7%, respectively, according to the culture result. In conclusion, the automated TB-PCR assay proved to be a useful method for the detection of Mycobacterium tuberculosis in the bronchial washing fluid and sputum. But we think that there is still a need for us to study body fluids further.

Influences of Human Body Fluids and Gonadotropins Supplemented in the Maturation Medium on the Nuclear Maturation and Fertilizability of Mouse Immature Oocytes (성숙배양액에 첨가하는 인간체액 (Human Body Fluids) 및 성선자극호르몬이 생쥐 미성숙난자의 핵성숙과 수정능력에 미치는 영향)

  • Purpose of the present study was to find the optimal culture conditions for the maturation and fertilization of immature oocytes by the use human body fluids and gonadotropins (Gn) in the mouse model. Cumulus-enclosed mouse immature oocytes were incubated in the medium containing various human body fluids with or without Gn in vitro, and examined to confirm nuclear maturation (NM) and fertilization. Female ICR mice were stimulated with 7.5 IU pregnant mares' serum gonadotropin (PMSG). Cumulus-enclosed immature oocytes were isolated at 48-52 hr post PMSG injection and cultured in TCM 199 supplemented with various concentrations (20, 50, and 70%) of human body fluids such as fetal cord serum (hCS), follicular fluid (hFF), peritoneal fluid (hPF) and amniotic fluid (hAF) in the presence or absence of 10 IU/ml PMSG and 10 IU/ml human chorionic gonadotropin (hCG) for 18 hr. Fetal calf serum (FCS) was used as a control for the supplements. Matured oocytes were fertilized with sperm collected from the epididymis of male mice. Fertilization was conducted in T6 medium containing 15 mgl ml bovine serum albumin, and confirmed at 6 hr post-insemination. Evaluation of nucler maturation and fertilization was carried out by rapid staining using fuchin. There was no significant difference between the effects of human body fluids and FCS supplements on nuclear maturation of cumulus enclosed mouse immature oocytes. When maturation medium was supplemented with 20% hPF or 20% hAF, fertilization rates were significantly (P<0.01) lower than that of 20% FCS, hCS and hFF groups. However, higher concentrations of body fluids during IVM were not more beneficial on fertilizability of oocytes. The addition of Gn significantly increased the fertilization rates in hPF and hAF groups (hPF without Gn; 51.5%, compared with 85.1% for addition of Gn, and hAF without Gn; 30.1% compared with 85.8% for addition of Gn) at 20% concentration. These results suggest that human body fluids at 20% concentration and gonadotropins can be used as supplements for the maturation of mouse immature oocytes in vitro. When gonadotropins supplemented with the human body fluids in the maturation medium, fertilizability of mouse immature oocytes was increased in hPF and hAF groups. These results can be applied to maturation of human immature oocytes in vitro.

Volatile Free Acid Profiling of Body Fluids by Gas Chromatography (기체 Chromatograph를 이용한 체액내 Volatile Free Acid의 분석연구)

  • A gas chromatographic method is described for the determination of volatile free acids(VFAs) in body fluids. VFAs were trace enriched from body fluids by liquid-solid extraction using Chromosorb P as the solid sorbent and ether as the eluant. The enriched VFAs were injected in splitless injection mode onto a HP-20M fused silica capillary column. The flame ionization detector was used as the detector. The present method was applied to the profiling of VFAs in body fluids from patients suffering from the infectious disease, hepatitis. The VFAs concentrations were high in saliva of hepatitis patients and isobutyric acid detected in sera of hepatitis patients compared to healthy subjects.

Salty taste: the paradoxical taste

  • Taste is a basic sensation to get attracted toward nutritious foods or avoid possible harmful substances. The basic taste qualities in humans consist of sweet, bitter, umami, salty, and sour. Basically, sweet and umami tastes make food attractive, whereas bitter and sour tastes make it avoidable. Salty taste comprises basic salty and high salt taste. The basic salty taste is known as amiloride-sensitive salty taste, which is inhibited by amiloride, but the high salt taste is not sensitive to amiloride. Moreover, high salt taste can also cause avoidance behavior in human beings. Sodium, one of the most important cations in the body fluids of vertebrates, controls the volume of total body fluids and is a risk factor for cardiovascular diseases, such as hypertension. The concentration of sodium in body fluids must be under delicate control. A distinction between the salty taste and high salt taste would be a contributing mechanism to control the volume and/or osmolarity of body fluids.

  • The oriental medicine based on the traditional Chinese medicine has developed characteristically according to the history and racial character respectively; China, Korea and Japan. Japan, among these nations, has accepted western medicine earlier than other nations and has tried to compare western and oriental medicine and combine them. In Japanese traditional medicine, it is characteristic that the old medical classics focusing on Sanghannon (傷寒論) and Geumgyeyoryak(金?要略) has developed The recent tendencies of clinical medicine and researches in Korean oriental medicine are mostly about the study of oriental medicine in view of western medicine and the combination of western and oriental medical treatment like Japan. But the study on the Japanese oriental medicine hasn't so far been tried before in Korea. From now on, we should not overlook that a more interest on Japanese oriental medicine will be very useful. Therefore we have surveyed the background of its origin and the process of development of the theory of ${\ulcorner}$Qi, Blood and Body Fluids${\lrcorner}$. What we wish to show in this paper is to provide a source for the basic understanding by explaining a fundamental theory of physiology and pathology of Japanese oriental medicine. Concepts of ${\ulcorner}$Qi, Blood and Body Fluids${\lrcorner}$ suggested by Nangai Yoshimashi in 1792 is the way of thinking that the circulation of 3 factors- ${\ulcorner}$Qi, Blood and Body Fluids${\lrcorner}$ nourish human body. Among these 3 factors, if Qi does not function smoothly, it causes the condition of a disease like Qi-deficiency, imbalance of Qi-distribution or Qi-depression and stasis; in Blood's case, deficiency of Blood and Blood stasis; and as for Body Fluids, stasis of Body Fluids. In the recent trend of study, there's a try to combining the western and oriental medicine, Qi is considered as psychoneurotic system, Blood as circulatory and endocrinologic system and Body Fluids as immunologic system.

Exposure to Blood and Body Fluids during the Clinical Practicum of Paramedic Students

  • This study is a descriptive research of paramedic students who could be exposed to blood and body fluids during the clinical practicum to analyze the risk factors. From June 1 to June 30, 2018, 172 paramedic students who attended the University in Jeolla-do region were analyzed. The collected data were analyzed by using the SPSS WIN 21.0 program. The general characteristics of subjects, the exposure level of blood and body fluids, and etc were obtained for frequency and percentage analysis. The study found that 72.7 percent of students had experience to blood or body fluids exposure during the clinical practicum. Except for any needle injury, 70.4 percent of student were exposed to blood or body fluids. 28.8 percent of them which was the highest percentage of injection injury were exposed during the venous blood draw. 36.5 percent of exposure were related to wound dressings which was the highest percentage related to clinical procedures. 71.2 percent of students mentioned that they did not report this exposure because 68.5 percent students thought that it has no danger. According to the survey on hepatitis B, 50.6% of students had antibodies, but 31.8 percent of students did not confirm that the antibodies were formed. Even though paramedic students do practical training in a hazardous environment with repeated exposure to blood and body fluids due to the nature of job characteristics, the systemic infection control education program is insufficient. In order to prevent exposure and to protect paramedic students who do practical training with patients, it is necessary to make more systematic and active efforts in the continuous monitoring and the preventive education.

Forensic Body Fluid Identification by Analysis of Multiple RNA Markers Using NanoString Technology

  • RNA analysis has become a reliable method of body fluid identification for forensic use. Previously, we developed a combination of four multiplex quantitative PCR (qRT-PCR) probes to discriminate four different body fluids (blood, semen, saliva, and vaginal secretion). While those makers successfully identified most body fluid samples, there were some cases of false positive and negative identification. To improve the accuracy of the identification further, we tried to use multiple markers per body fluid and adopted the NanoString nCounter system instead of a multiplex qRT-PCR system. After measuring tens of RNA markers, we evaluated the accuracy of each marker for body fluid identification. For body fluids, such as blood and semen, each body fluid-specific marker was accurate enough for perfect identification. However, for saliva and vaginal secretion, no single marker was perfect. Thus, we designed a logistic regression model with multiple markers for saliva and vaginal secretion and achieved almost perfect identification. In conclusion, the NanoString nCounter is an efficient platform for measuring multiple RNA markers per body fluid and will be useful for forensic RNA analysis.

Corrosion Behavior of Titanium for Implant in Simulated Body Fluids (인공 체액 조건에서 임플랜트용 티타늄 소재의 부식 특성)

  • The corrosion of pure titanium (CP- Ti Grade 2) and titanium alloy (Ti6Al4V ELI) were studied under various conditions of simulated body fluids. The static immersion test and the electrochemical test were performed in accordance with ISO 10271 : 2001. For the electrochemical test, the open circuit potential was monitored as a function of time, and the cyclic polarization curve was recorded. The corrosion resistance was evaluated from the values of corrosion potential, passivation current density, breakdown potential, and the shape of hysteresis etc. The effects of alloy type, surface condition, temperature, oxygen, and constituents in the fluids such as acid, chloride were estimated. Both specimens had extremely low dissolution rate in the static immersion test. They showed strong passivation characteristics in the electrochemical test. They maintained negligible current density throughout the wide anodic potential range. The passive layer was not broken up to 2.0 V (vs. SCE). The hysteresis and the shift of passivation potential toward the anodic direction was observed during the reversed scan. The passivation process appeared to be accelerated by oxygen in air or that dissolved in the fluids. The passivation also proceeded without oxygen by the reaction of constituents in the fluids. Acid or chloride in the fluids, specially later weakened the passive layer, and then induced higher passivation current density and less shift of passivation potential in the reversed scan. CP-Ti Grade 2 was more reactive than Ti6Al4V ELI in the fluids containing acid or chloride, but thicker layer produced on its surface provided higher corrosion resistance.

DNA methylation-based age prediction from various tissues and body fluids

  • Aging is a natural and gradual process in human life. It is influenced by heredity, environment, lifestyle, and disease. DNA methylation varies with age, and the ability to predict the age of donor using DNA from evidence materials at a crime scene is of considerable value in forensic investigations. Recently, many studies have reported age prediction models based on DNA methylation from various tissues and body fluids. Those models seem to be very promising because of their high prediction accuracies. In this review, the changes of age-associated DNA methylation and the age prediction models for various tissues and body fluids were examined, and then the applicability of the DNA methylation-based age prediction method to the forensic investigations was discussed. This will improve the understandings about DNA methylation markers and their potential to be used as biomarkers in the forensic field, as well as the clinical field.

Numerical Analysis of Hydrodynamic Forces on a Floating Body in Two-layer Fluids (밀도가 상이한 두 유체층에서 부유체 동유체력 특성의 수치적 해석)

  • In this study, a radiation and a diffraction problems of a floating body in two-layer fluids were solved by the Numerical Wave Tank(NWT) technique in the frequency domain. In two-layer fluids, two different wave modes exist and the hydrodynamic coefficients can be obtained separately for each mode. The two-domain Boundary Element Method(BEM) in the potential fluid using the whole-domain matrix scheme was used to investigate the characteristics of wave forces, added mass and damping coefficients. The effects of the ratio of density and water depth in the lower domain were also evaluated and compared with given references.
