Effect of Organic Selenium (Sel-Plex) on Thermometabolism, Blood Chemical Composition and Weight Gain in Holstein Suckling Calves

  • The objective of this study was to determine the effects of milk supplemented with Sel-Plex on thyroid hormones, rectal temperature, plasma glucose and cholesterol concentrations, and body weight in suckling calves. Ten Holstein suckling male calves of approximately 1 month of age were selected and randomly allocated to one of two groups and fed either unsupplemented milk (control) or milk supplemented with 0.3 mg/kg DM Se from Sel-Plex (treated). The animals received treated milk and free choice starter for two months. Dry matter intake for each individual animal was recorded daily throughout the study. The calves were weighed and blood samples were taken weekly. Plasma concentrations of triiodothyronine ($T_{3}$), thyroxin ($T_{4}$) and $T_{3}$-Uptake were determined using radioimmunoassay kits. Plasma cholesterol and glucose concentrations were determined by enzymatic-colorimetric methods. Rectal temperature was recorded at blood sampling time. During the experiment, ambient temperature and relative humidity were recorded at the time of blood sampling and every four hours. Treatment significantly (p<0.01) affected the concentration of $T_{3}$, free $T_{3}$ index ($FT_{3}I$) and ratio of $T_{3}$:$T_{4}$ in plasma. The plasma concentrations of $T_{3}$ in treated calves were 33% higher than in the control. The plasma concentrations of $T_{4}$ (p<0.05), glucose (p<0.01) and cholesterol (p<0.01) were decreased in the treated group. Calves fed Sel-Plex had higher rectal temperature (p<0.01). Plasma free $T_{4}$ index (FT4I) did not differ significantly between the two groups. An increase in body weight was observed (p<0.09) in the treated group. The results indicated that milk fortified with Sel-Plex could increase tissue conversion of $T_{4}$ to $T_{3}$ and therefore improve thermometabolism in suckling calves.

  • The nature of feed gas is essential for the active species formed in the nonthermal plasma jets, which would induce various biological phenomena. We investigated the different physiological effects of atmospheric pressure soft-plasma jets on Esherichia coli and blood cells according to the feed gas. Cell death rate, growth curve, membrane molecular changes and induced genes were examined. The relationship between cellular reactions and active species generated by discharge will be discussed.

Effects of Multi-Extracts of Mori Folium and of Exercise on Plasma Insulin and Glucose Levels in Streptozotocin-Induced Diabetic Rats

  • This research was conducted to study the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on plasma insulin and glucose levels in streptozotocin (STZ)-induced diabetic rats. Eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for 10 weeks. The animal groups consisted of a normal-control group, a STZ-control group, 3 STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 along with exercise. In the normal-control group, glucose tolerance tests resulted in the peak blood glucose level being achieved in 15 minutes and a fasting blood glucose level being achieved in 60 minutes. In the STZ-control group, the peak blood glucose level was reached after 60 minutes and, even after 90 minutes, blood glucose shown at a significantly higher level compared to the fasting levels. In the groups supplemented with MF extracts, the blood glucose level peaked after 30 minutes of glucose challenge, and returned to the fasting level after 90 minutes; the MF-360 and MF-360+exercise groups showed the best levels of glucose tolerance. Blood glucose levels in the STZ-induced diabetic groups were significantly higher compared to the normal-control group. However, after 7 weeks of supplementation with MF extracts, a significant lowering of blood glucose levels was observed in all groups supplemented with the MF extract. The best effect was observed in the group given MF extract combined with exercise. Compared to the normal-control group, blood insulin levels were significantly lower in all STZ-induced diabetic groups; however, a significantly higher level of insulin was observed in the groups given MF extracts compared to the STZ-control group. This study shows that the supplementation of MF extracts in STZ-induced diabetic rats resulted in increased blood insulin levels and lower blood glucose levels.

  • To evaluate effects of Ga Gam Sam Kee Tang(GGSKT) water extract on the plasma cortisol concentration and arterial blood $PCO_2,\;PO_2$ in the rabbit. The results obtained were as follows : 1. Intravenous administration of GGSKT water at the dose of $0.2m{\ell}/kg$ remarkably increased plasma cortisol concentration from 1 to 2 hours. 2. Oral administration of GGSKT water extract at the dose of $10m{\ell}/kg$ significantly decreased plasma cortisol concentration on 3 hours. 3. Intravenous administration of GGSKT water extract at the dose of $0.4m{\ell}/kg$ remarkably decreased arterial blood $PO_2$ on 2 hours. 4. Oral administration of GGSKT water extract at the dose of $1.0m{\ell}/kg$ significantly decreased arterial blood $PO_ 2$ on 2 hours and remarkably increased on 4 hours. 5. Intravenous administration of GGSKT water extract at the dose of $0.2m{\ell}/kg$ remarkably increased arterial blood $PCO_2$ on 4 hours. 6. Oral administration of GGSKT water extract at the dose of $2.0m{\ell}/kg$ significantly decreased arterial blood $PCO_2$ from 1 to 2 hours. These results suggest that therapeutic action of GGSKT water extract for athma may be related with the increment of plasma cortisol concentration and the decrease of arterial blood $PCO_2$.

  • Eight Korean native black goats were used, $10m{\ell}$ of blood was collected from the Jugular vein into heparinized tubes a week interval. The heparinized blood was centrifuged for separation to blood plasma and corpuscles. The hematocrit, per centage of blood that is red blood cells, was reschuffuled of 10 %, 20 %, 30 %, 40 % and 50 % using blood plasma, 0.9 % NaCl solution and 5.4 % dextrose solution. The sedimentation rates of red blood cell obtained at $7^{\circ}C{\pm}1^{\circ}C$ and $27^{\circ}C{\pm}1^{\circ}C$ are summarized as follows. 1) The sedimentation rates of red blood cell were more increased by lower PCV%, i.e. there was a reverse relationship between the sedimentation rates and PCV% at any condition of these experiments. 2) The RBC were sedimented the most quickly in the NaCl solution and slower in the plasma compare with the dextrose solution at the same PCV%. 3) There was no temperature effect on the sedimentation rates between the two groups of $7^{\circ}C{\pm}1^{\circ}C$ and $27^{\circ}C{\pm}1^{\circ}C$(at PCV 20% and 10%), even though the temperature difference is $20^{\circ}C$.

  • The concentrations of some blood constituents are known to be influenced by the postural change. The blood glucose and insulin concentrations were measured, first, in the supine, and then (30 minutes later) in the erect positions under the Lasting state. The effects of a duretic, furosemide, were also studied under the same condition for 5 consecutive days. The materials were 5 healthy volunteers aging 20-29 years old with out any diabetic past, or family histories. The blood glucose was measured by the Nelson's method, and plasma insulin by the radioimmunoassay method. Following are the results; 1. The plasma insulin concentration in the erect position is slightly higher than in the supine position, however, the increase is statistically insignificant because of the notable individual variations in the values of the supine position. 2. Four cut of 5 cases show the increase of about 80% of plasma insulin in the erect position, which is statistically significant if analyzed on the basis of frequency distribution. 3. The blood glucose concentration showed no postural changes. 4. The increase of the plasma insulin concentration in the erect position seems to be the result of limited extra vasation of insulin in the lower extremities.

  • The aim of the experiments was to investigate the effect of Semen Cuscutae aqua-acupuncture at the meridian point BL 20(脾兪) and BL 23(賢兪) on the blood pressure, plasma renin activity, plasma levels of aldosterone and atrial natriuretic peptide (ANP) in spontaneously hypertensive rats (SHR). The results of this study were as follows: 1. Systolic blood pressure was decreased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 20, BL 23. 2. Plasma renin activity was increased significantly after Semen Cuscutae aqua-acupuncture at meridian point BL 23, BL 20. 3. Plasma levels of aldosterone was increased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 20. 4. Plasma levels of atrial natriuretic peptide (ANP) was increased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 23, but it was decreased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 20. These results suggest that the changes of the depressor response after Semen Cuscutae aqua-acupuncture at the meridian point BL 20, BL 23 are related to the changes of the plasma renin activity, plasma levels of atrial natriuretic peptide (ANP) and aldosterone.

Correlation between the Plasma Insulin and Glucose Concentration in Normal Korean Adults (한국인 혈장 Insulin과 혈당량의 상호관계에 관한 고찰)

  • The correlation between the plasma insulin, and glucose concentration was studied in healthy Korean adults consisting of 20 males and 22 females of 16 to 38 years of age. The blood samples of above subjects were obtained through cubital vein at arbitrary times, during their usual working hours. Plasma insulin was assayed by means of double antibody system of radioimnmunoassy technics, and blood glucose was determined by means of Van Slyke-Folch method. Results were as follows: 1. There were no differences in the blood sugar levels in relation to the plasma insulin concentration either by sex or age. 2. In the case, when the plasma insulin concentration was within $200m{\mu}D/ml$, the correlation between the insulin, and glucose concentration existed, the ratio of which was expressed as; Plasma glucose concentration(mg/dI)=$91.9+0.08{\times}Insulin$ concentration r=0.62 3. Insulinogenic index was 12.4%, which was somewhat higher than other reports. 4. It is suggested that the correlation between plasma insulin and glucose concentration could be determined at arbitrary times instead of fasting times.

  • The stress of consecutive blood collectings resulted in evident elevation of plasma glucose level and significant lowering of chemiluminescent response of peripheral blood phagocytes in sea bass (Lateolabrax japonicus). Fish responded to the consecutive stressors in cumulative manners. The plasma glucose level in response to consecutive stressors depended on the stressor intervals. When the plasma glucose level of individual fish was compared with the chemiluminescent response, statistically significant (P<0.05) negative correlations existed.

BLOOD PLASMA MINERALS AND FERTILITY OF DAIRY COWS IN CENTRAL THAILAND

  • Blood plasma minerals and their effects towards the fertility in 136 cows randomly selected from 26 dairy farms in central Thailand were studied. An average of 8.60 mg% Ca, 5.97 mg% P, 2.45 mg% Mg, $85.9{\mu}g%$ Cu, and $160.9{\mu}g%$ Zn in plasma was observed. Compared to the critical deficient values, 24.3, 10.3, 11.8, 28.7 and 0.0% of the surveyed cows obtained the respective elements in plasma below the standard levels. No significant difference (p >.05) in plasma concentrations of Ca, P, Mg and Zn for the low fertile (conception rate > 3) and fertile (conception rate < 3) cows was found. However, plasma Cu of the low fertile cows (averaged $77.2{\mu}g%$) was lower (p <.01) than that of the fertile ones (averaged $91.12{\mu}g%$). Additionally, it is observed that 47.1% of the cows with the low fertility problem obtained plasma Cu below the 65.0 pg% critical deficient value.
