Analytical trend of perfluorinated compounds in environmental and biota samples (환경 및 생체시료 중 과불화 화합물의 분석 동향)

  • Perfluorinated compounds have characteristics of resistance to heat, acidic, basic conditions and also resist water, oil, grease, pollutant. Futhermore they are used by various industrial material, nowadays, they produced in large scale for indutrial and commercial areas. However, they also resist metabolizing and degrading in environmental system (plant, animal, even human body). Moreover, in animal's bodies, PFCs can be accumulated in organ (eg; liver) and lead to liver cell necrosis even oncogenesis. Perfluorinated compounds are newly registered as new persistent organic pollutants (POPs) on Stockholm convention in 2009. Therefore necessity for analytical methodology for determination of PFCs in various environmental samples is even more increased. This study discussed sample preparation and instrumental conditions for the analysis of PFCs in environmental and biota samples.

Effects of Garlic and Thyme Extracts on Growth Performance and Carcass Characteristics of Broiler Chicks

  • A total of 800 one-day-old chickens (Cobb 500) were obtained from a local hatchery and allocated randomly to five dietary treatments. This study conducted for 42 d. Treatment groups included 1) CON (control), 2) T0.3 (0.3% of thyme extract), 3) T0.6 (0.6% of thyme extract), 4) G0.3 (0.3% of garlic extract) and 5) G0.6 (0.6% of garlic extract). Thyme extract included $90{\mu}g/ml$ Carvacrol and 400 mg/ml thymol while garlic extract included 100 mg/ml elastin. There were no significant effects among the treatments on weight gain, feed intake and feed conversion ratio over the entire trial. Also, no significant differences were observed between CON group and other treatments in plasma parameters (cholesterol, triglyceride and HDL) and hematocrit. Sex of chickens had no effect on blood parameters. Supplementation of herb extracts did not affect carcass characteristics including carcass yield, breast, thigh and abdominal fat. There were no significant differences observed in carcass characteristics between males and females fed with different levels of herb extracts. In conclusion, supplementation of garlic and thyme extracts as feed additives at two levels of 3 and 6% in the present study did not improve performance and carcass traits of broiler chicks.

Bioequivalence of Senital Tablet to Airtal Tablet (Aceclofenac 100 mg) (에어탈 정(아세클로페낙 100 mg)에 대한 세니탈 정의 생물학적 동등성)

  • Bioequivalence of two aceclofenac tablets, the $Airtal^{TM}$ (Daewoong Pharmaceutical Co., Ltd.) and the $Senital^{TM}$ (Hana Pharmaceutical Co., Ltd.), was evaluated according to the guideline of KFDA. Fourteen normal male volunteers (age $20{\sim}29$ years old) were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After one tablet containing 100 mg of aceclofenac was orally administered, blood was taken at predetermined time intervals and the concentration of aceclofenac in plasma was determined with an HPLC method using UV detector. The pharmacokinetic parameters ($C_{max}$, $T_{max}$ and $AUC_t$) were calculated and ANOVA was utilized for the statistical analysis of parameters. The results showed that the differences in $C_{max}$, $T_{max}$ and $AUC_t$ between two tablets were 3.69%, 2.44% and 0.51%, respectively. The powers $(1-{\beta})$ for $C_{max}$, $T_{max}$ and $AUC_t$ were 87.85%, 98.70% and more than 99%, respectively. Detectable differences $({\Delta})$ and confidence intervals were all less than ${\pm}20%$. All of these parameters met the criteria of KFDA for bioequivalence, indicating that $Senital^{TM}$ tablet is bioequivalent to $Airtal^{TM}$ tablet.

Sperm Fertility of Transgenic Boar Harboring hEPO Gene is Decreased

  • This study was conducted to compare the reproduction ability of the wild type boar and recombinant human erythropoietin (hEPO) transgenic boar semen. Ejaculated boar semen was analyzed by flow cytometry, Elisa and IVF methods. In experiment 1, flow cytometric analysis showed that the live sperm ratio of transgenic boar sperm significantly lower (P<0.05) than that of wild type boar after incubation at 20, 22, 24 and 26 hr. In experiment 2, the presence and levels of various cytokines (IL-6, IL-10 and $TNF-{\alpha}$) to related animal reproduction in the seminal and blood plasma were examined using specific enzyme immunoassay. There was no significant difference between both groups. In experiment 3, the fertilizing capacity and developmental ability of both boar sperm were compared. The transgenic boar sperm had a significantly low capacity of penetration, sperm-zona binding, embryo development, and blastocyst formation compared to wild type sperm (P<0.05). These results suggest that transgenic boar sperm harboring hEPO gene has low sperm viability than wild type boar, and it is a reason to decrease of fertility and litter size.

Antidiabetic Activity of Polysaccharide Extract from Tetragonia tetragonoides in Streptozotocin-induced Diabetic Mice (스트렙토조토신으로 유도한 당뇨 마우스에서 번행초 다당 추출물의 항당뇨 효과)

  • Tetragonia tetragonioides seems to be a promising antiulcer medicinal plant due to the presence of water-soluble polysaccharide and cerebroside as its major constituents. There have been no previous studies using T. tetragonioides polysaccharide extract (TPE) to assess its antidiabetic effect in streptozotocin (STZ)-induced diabetes in mice. This study was designed to evaluate the antidiabetic effect of TPE in diabetic mice, which was established by one-week intraperitoneal injection of 65 mg/kg STZ. After three weeks of TPE treatment at a dose of 100 mg/kg, a maintenance of body weight, a decrement in plasma glucose, and low levels of triglyceride, lactate dehydrogenase, alkaline phosphatase, and glutamic pyruvic transaminase were observed in diabetic mice. Furthermore, the ingestion of TPE lowered the blood glucose levels during the oral glucose tolerance test (OGTT) and restored most of the tested parameters to their normal levels. Therefore, the antidiabetic potential of T. tetragonioides has been demonstrated for the first time by our research.

Concentration of Heavy Metals in Octopus minor in Seosan, Chungnam and Food Safety Assessment (충남 서산에서 어획된 낙지(Octopus minor)의 중금속 함량과 인체 위해성 평가)

  • This study estimated the heavy metal concentrations in octopus (Octopus minor) and conducted a food safety assessment of octopus. Octopus, a benthic cephalopod, was collected from the Seosan intertidal zone on the west coast of Korea. The samples were digested with acids, and then the cadmium (Cd), copper (Cu), and zinc (Zn) contents were analyzed using inductively coupled plasma mass spectrometry (ICP-MS). The Cd, Cu, and Zn range of concentrations in octopus were 0.06-19 (mean 5.8), 44-1,463 (mean 354.8), and 76-929 (mean 247.9) mg/kg on a dry weight basis, respectively. The concentrations of heavy metals were higher in the internal organs than in the mantle. Of the three heavy metals, copper had the highest concentrations in the internal organs because of the existence of hemocyanin bound with copper in octopus blood, whereas zinc had the highest concentrations in the mantle. No relationship between the concentration of heavy metals and biological parameters (length, weight, and sex) was found. The ratios (I/M) of the heavy metal concentrations in internal organs and mantle were highest for cadmium, although cadmium had much lower concentrations in the internal organs compared with copper and zinc. Considering the provisional tolerable weekly intake (PTWI) of the three heavy metals and the average intake of octopus per day, all three elements should have no adverse effects on humans.

Diol-ginsenosides from Korean Red Ginseng delay the development of type 1 diabetes in diabetes-prone biobreeding rats

  • Background: The effects of diol-ginsenoside fraction (Diol-GF) and triol-ginsenoside fraction (Triol-GF) from Korean Red Ginseng on the development of type 1 diabetes (T1D) were examined in diabetes-prone biobreeding (DP-BB) rats that spontaneously develop T1D through an autoimmune process. Methods: DP-BB female rats were treated with Diol-GF or Triol-GF daily from the age of 3-4 weeks up to 11-12 weeks (1 mg/g body weight). Results: Diol-GF delayed the onset, and reduced the incidence, of T1D. Islets of Diol-GF-treated DP-BB rats showed significantly lower insulitis and preserved higher plasma and pancreatic insulin levels. Diol-GF failed to change the proportion of lymphocyte subsets such as T cells, natural killer cells, and macrophages in the spleen and blood. Diol-GF had no effect on the ability of DP-BB rat splenocytes to induce diabetes in recipients. Diol-GF and diol-ginsenoside Rb1 significantly decreased tumor necrosis factor α production, whereas diol-ginsenosides Rb1 and Rd decreased interleukin 1β production in RAW264.7 cells. Furthermore, mixed cytokine- and chemical-induced β-cell cytotoxicity was greatly inhibited by Diol-GF and diol-ginsenosides Rc and Rd in RIN5mF cells. However, nitric oxide production in RAW264.7 cells was unaffected by diol-ginsenosides. Conclusion: Diol-GF, but not Triol-GF, significantly delayed the development of insulitis and T1D in DP-BB rats. The antidiabetogenic action of Diol-GF may result from the decrease in cytokine production and increase in β-cell resistance to cytokine/free radical-induced cytotoxicity.

Anti-obesity Effect of Fermented Whey Beverage using Lactic Acid Bacteria in Diet-induced Obese Rats

  • High-protein fermented whey beverage (FWB) was manufactured using whey protein concentrate (WPC) and Lactobacillus plantarum DK211 isolated from kimchi. This study was designed to evaluate the anti-obesity activity of FWB in male rats fed a high-fat diet. Male Sprague-Dawley rats were randomly assigned to three groups (n=8 per group). The three groups differed in their diet; one group received a normal diet (ND), another, a high-fat diet (HD), and the third, a HD plus fermented whey beverage (HDFWB), for 4 wk. Supplementation with FWB (the HDFWB group) prevented weight gain and body fat accumulation. The food intake in the HDWFB group was significantly lower (p<0.05) than that of the HD group. The HDWFB group also showed a significant decrease in organ weights (p<0.05), except for the weight of the testis. There was a significant decrease in total cholesterol, LDL-cholesterol, and triglycerides in the HDFWB group compared with the HD group (p<0.05), but there was no significant difference in serum HDL-cholesterol levels among the experimental groups. Rats ingesting FWB (the HDFWB group) also showed a significant decrease in blood glucose levels, and plasma levels of insulin, leptin, and ghrelin compared to HD group (p<0.05). These results indicate that FWB has beneficial effects on dietary control, weight control, and reduction in fat composition and serum lipid level; consequently, it may provide antiobesity and hypolipidemic activity against high fat diet-induced obesity in rats.

  • GABA tea which was increased of GABA by anaerobic conditions in green tea leaves) was fed in the diet to spontaneously hypertensive rats(SHR). The mean systolic blood pressure of SHR fed a diet containing 1.5% GABA tea after 3 weeks was not different with zero day as 177.3mmHg. However, 1.5% green tea and control after 3 weeks were more increased 4.2 percents and 10 percents than in zero day. The plasma levels of aldosterone of SHR fed a diat containing 1.5% GABA green tea after 3 weeks was more decreased 2.5 times than in water treatment.

  • 1. Introduction : The conventional grafting polymerization technique requires chemically reactive groups on the surface as well as on the polymer chains. For this reason, a series of prefunctionalization steps are necessary for covalent grafting. The surface prefunctionalizational technique for grafting can be used to ionization radiation, UV, plasma, ion beam or chemical initiators. Of these techniques, radiation method is one of the useful methods because of uniform and rapid creation of active radical sites without catalytic contamination in grafted samples. If the diffusion of monomer into polymer is large enough to come to the inside of polymer substrate, a homogeneous and uniform grafting reaction can be carried out throughout the whole polymer substrate. Radiation-induced grafting method may attach specific functional moieties to a polymeric substrate, such as preirradiation and simultaneous irradiation. The former is irradiated at backbone polymer in vacuum or nitrogen gas and air, and then subsequent monomer grafting by trapped or peroxy radicals, while the latter is irradiated at backbone polymer in the presence of the monomer. Therefore, radiation-induced polymerization can be used to modification of the chemical and physical properties of the polymeric materials and has attracted considerable interest because it imparts desirable properties such as blood compatibility. membrane quality, ion excahnge, dyeability, protein adsorption, and immobilization of bioactive materials. Synthesizing biocompatible materials by radiation method such as preirradiation or simultaneous irradiation has often used $\gamma$-rays to graft hydrophilic monomers onto hydrophobic polymer substrates. In this work, in attempt to produce surfaces that show low levels of anti-fouling of bovine serum albumin(BSA) solutions, hydroxyethyl methacrylate(HEMA) was grafted polypropylene membrane surfaces by preirradiation technique. The anti-fouling effect of the polypropylene membrane after grafting was examined by permeation BSA solution.
