Anti-Thrombotic Effects of Egg Yolk Lipids In Vivo

  • In this study, we investigated the effect of egg yolk lipids (EYL) on collagen ($10\;{\mu}g/ml$)-stimulated platelet aggregation in vivo. Dietary EYL significantly inhibited collagen-induced platelet aggregation, in addition, increased the formation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), intracellular $Ca^{2+}$-antagonist as aggregation-inhibiting molecules, in collagen-stimulated platelets. These results suggest that EYL inhibits the collagen-induced platelet aggregation by up-regulating the cAMP and cGMP production. On the other hands, prothrombin time (PT) on extrinsic pathway of blood coagulation was potently prolonged by dietary EYL in vivo. These findings suggest that EYL prolongs the internal time between the conversion of fibrinogen to fibrin. Accordingly, our data demonstrate that EYL may be a crucial tool for a negative regulator during platelet activation and blood coagulation on thrombotic diseases.

새로운 퀴놀론계 항생제 LB20304a의 일반약리작용 (General Pharmacology of LB20304a, a New Quinolone Antibiotic)

  • General pharmacological studies of LB20304a (a mesylate salt form of a new quinolone antibiotic LB20304 following oral administration of 300 mg/kg and 1000 mg/kg, almost maximum tolerance dose in mice and rat, respectively, were performed in terms of effects on general behaviour, central nervous system, gastrointestinal system, and blood coagulation system in mice and rats. With regards to general behaviour of mice, at oral dose of 300 mg/kg, LB20304a reduced muscle tone and locomotor activity. In terms of CNS, at oral treatment of 300 mg/kg, LB20304a showed some analgesic effects in mice, and oral dose of 1000 mg/kg caused drop in normal body temperature of rat, while it enhanced the pentylenetetrazole-induced clonic convulsion to tonic convulsion and/or death in mice at the doses of unto 300 mg/kg. In addition, LB20304a increased hexobarbital-induced sleeping time two and three times in mice at oral doses of 20 mg/kg and 300 mg/kg, respectively. Rota-rod and traction test in mice were not influenced by the dose of 300 mg/kg and 200 mg/kg, respectively. LB20304a reduced gastric secretion of rat at dose of 1000 mg/kg, and increased intestinal motility of mice at dose of 300 mg/kg. In rats, blood coagulation index, such as PT (prothrombin time) and aPTT (activated partial thromboplastin time) were not affected by the treatment of upto 1000 mg/kg of LB 20304a.

신체냉각이 혈액세포성분 및 응혈기전에 미치는 영향에 관한 실험적 연구 (Hematologic and Coagulation Changes in Hypothermic Dogs)

  • This experiment was carried out to study the responses of cellular component of blood and bone marrow to cold and also the changes of coagulation during cooling. Forty-two mongrel dogs were subjected to hypothermia by ice-water surface cooling technique. Lowest body temperature ranged from 21-23 degree. Dogs were divided into 3 groups,Group I, 12 dogs: pentothal anesthesia for 3 hours, Group II, 20 dogs;hypothermic group and Group III,10 dogs;postsplenectomy hypothermic group. Results were summarized as follows: 1. Hemoglobin, hematocrit and red blood cell count significantly increased when animals were cooled, and increase was noted in similar magnitude among the animals of Group I. 2. White blood cell count extremely decreased after cooling and effect of splenectomy on white blood cell count was not apparent. No significant changes were seen among Group I. 3. Differential count of white blood cell when cooled showed relative increase of polymorphonuclear neutrophil and decrease of lymphocyte. 4. There was marked decrease of platelets when body temperature reached to 21-23degree and essentially. no changes was noted in Group I. 5. Clotting time, bleeding time, plasma prothrombin time, recalcification time, and fibrinolysis showed no significant changes when dogs were cooled. Clot retration and prothrombin consumption during hypothermia appeared to be poor. In Group II, bleeding time decreased after splenctomy and when body temperature was lowered, plasma prothrombin time, clot retraction, and prothrombin consumption decreased. Decreased bleeding time and poor clot retraction were noted in Group I. 6. It was found that megacaryocyte count decreased even though platelet count of peripheral blood markedly diminsished when animals were cooled. There was some tendency of erythroid hyperplasia noted during hypothermia.

The activity of factor VIII and IX of cord blood at mid-trimester in fetuses without hemophilia

  • Purpose: Molecular genetic analysis is the main approach used for prenatal diagnosis of hemophilia A and B. However, in certain cases, such analysis is uninformative. In such situations, direct measurement of fetal coagulation factor levels is still the best option, and it may be the only option in some cases. This study was conducted to determine the normal ranges of mid-trimester cord blood factor VIII (FVIII) and IX (FIX) in a Korean population. Materials and Methods: Twenty-six FVIII samples and 29 FIX samples were assayed in fetal cord blood acquired by ultrasound-guided cordocentesis. Sampling was conducted during gestational ages of 19-24 weeks. Results: The mean and standard deviations for FVIII and FIX activity were $45.5{\pm}30.5%$ and $19.9{\pm}12.2%$, respectively. Ranges for FVIII and FIX were 1.5-125.0% and 6.0-52.0%, respectively. Conclusion: Our study revealed the normal ranges and lowest level of factor VIII and factor IX in non-affected normal fetus by fetal cord blood sampling during the mid-trimester in a Korea population. The factor assay of the fetal cord blood is invasive but feasible and provides important basic data related to hemophilia.

Endotoxin에 의한 혈전증에 미치는 Propolis의 효과 (The effect of Propolis on Endotoxin-induced thrombosis)

  • Propolis, a natural resinous compound collected from honey bees, contains many biochemical constituents and has been used for traditional medicines as early as 300 B .C. Recently, it has been reported to possess many biological activities such as antibacterial, antiviral, fungicidal, local anaesthetic, immunostimulating, antiinflammatory and free radical scavenging properties. To investigate activities of chrysin, one of propolis effective compounds for blood coagulation system was injected endotoxin (4000 EU/kg, i.v.) in rats at 1 hr after administered chrysin (20 mg/kg, p.o.). This study was resulted that chrysin has antiplatelet aggregation activity in vitro, delay of blood clotting time and prothrombin time, and reduction of fibrinogen and FDP in vivo. Chrysin has increased SOD activity, GSH content and GST activity, and decreased MDA content in liver. The result suggests that the antithrombosis effect of chrysin is suppressive activity for a blood coagulation system and antioxidative activity.

Effects of Ulinastatin on Postoperative Blood Loss and Hemostasis in Atrioventricular Valve Surgery with Cardiopulmonary Bypass

  • Background: Cardiopulmonary bypass (CPB) induces variable systemic inflammatory reactions associated with major organ dysfunction via polymorphonuclear neutrophils (PMNs). Ulinastatin, a urinary trypsin inhibitor, inhibits PMN activity and reduces systemic inflammatory responses. The aim of this study is to evaluate the effect of ulinastatin on postoperative blood loss and laboratory changes in patients undergoing open heart surgery. Materials and Methods: Between January 2008 and February 2009, 110 patients who underwent atrioventricular valve surgery through right thoracotomy were divided into two groups. Patients received either 5,000 U/kg ulinastatin (ulinastatin group, n=41) or the equivalent volume of normal saline (control group, n=69) before aortic cross clamping. The primary end points were early coagulation profile changes, postoperative blood loss, transfusion requirements, and duration of intubation and intensive care unit stay. Results: There were no statistically significant differences between the two groups in early coagulation profile, other perioperative laboratory data, and postoperative blood loss with transfusion requirements. Conclusion: Administration of ulinastatin during operation did not improve the early coagulation profile, postoperative blood loss, or transfusion requirements of patients undergoing open heart surgery. In addition, no significant effect of ulinastatin was observed in major organs dysfunction, systemic inflammatory reactions, or other postoperative profiles.

구름버섯 기원 항응고성 다당류의 혈액응고 저해기작 (Inhibitory Mechanism of Blood Coagulation by the Anticoagulant Polysaccharide from Coriolus versicolor)

  • 구름버섯 자실체에서 분리된 항응고성 다당류의 혈액응고 저해기작을 검토하였다. 항응고성 다당(CV-40-Va-1)은 vWF의 활성을 감소시킴으로써 혈소판응집을 억제시키는 것으로 나타났으며, 혈액응고인자중 thrombin뿐만이 아니라 내인성경로의 factor VII, IX, XI, XII의 활성 또한 억제 하였다. 그러나 CV-40-Va-1의 항응고 활성은 thromin에 직접 작용하는 것이 아니라 antithrombin III 의존적 활성을 보였다. Sulfation에 의하여 CV-40-Va-1의 항응고활성의 증가와 다당의 desulfation시 상반된 결과에서 CV-40-Va-1의 황산기가 항응고활성의 중요한 인자임을 알 수 있었다. CV-40-Va-1을 TFA로 부분가수분해하여 얻은 획분들(Fr.I, Fr.II)에서는 항응고 활성이 감소하였으나 분자량 1,000정도로 추정되는 획분(Fr.II)은 오히려 혈소판응집을 강력하게 억제하였다.

Stability Evaluation of National Reference Standards for Blood Products in Korea

  • National reference standards (NRSs) for biologics are established through potency estimation by a multi-center joint study of standard materials used in the approval process for national lot release and quality control of vaccines, blood products, and other biologics. In this study, a stability evaluation was conducted to determine whether the potency of NRSs for six blood products was being maintained at a consistent level in Korea. The present study conducted real-time stability tests via in-vivo/in-vitro bioassay on NRSs for blood coagulation factor VIII concentrate (2nd standard), antithrombin concentrate, prekallikrein activator, anti-hepatitis B immunoglobulin, blood coagulation factor IX concentrate, and anti-tetanus human immunoglobulin, as well as a trend analysis using cumulative annual results. The real-time stability test results showed that the mean potency of six NRSs was all within the control limit. In the trend analysis, the potency of NRS for blood coagulation factor VIII concentrate (2nd standard) showed a decreasing trend, while the potency of all other products had been stably maintained. The present study confirmed that the mean potency of NRSs for six blood products had been stably maintained in Korea. The findings of the present study establish a foundation that can ensure the quality of NRSs for biologics in Korea, and it is expected to make a major contribution to the supply of high-quality biologics.

Characterization of Binding Mode for Human Coagulation Factor XI (FXI) Inhibitors

  • The human coagulation factor XI (FXI) is a serine protease that plays a significant role in blocking of the blood coagulation cascade as an attractive antithrombotic target. Selective inhibition of FXIa (an activated form of factor XI) disrupts the intrinsic coagulation pathway without affecting the extrinsic pathway or other coagulation factors such as FXa, FIIa, FVIIa. Furthermore, targeting the FXIa might significantly reduce the bleeding side effects and improve the safety index. This paper reports on a docking-based three dimensional quantitative structure activity relationship (3D-QSAR) study of the potent FXIa inhibitors, the chloro-phenyl tetrazole scaffold series, using comparative molecular field analysis (CoMFA) and comparative molecular similarity analysis (CoMSIA) methods. Due to the characterization of FXIa binding site, we classified the alignment of the known FXIa inhibitors into two groups according to the docked pose: S1-S2-S4 and S1-S1'-S2'. Consequently, highly predictive 3D-QSAR models of our result will provide insight for designing new potent FXIa inhibitors.
