Synergic Effects of Bitter Melon and ${\beta}$-Glucan Composition on STZ-Induced Rat Diabetes and Its Complications

  • ${\beta}$-Glucan purified from oats (OG) and bitter melon, Momordica charantia Linn (MC), water extracts have shown favorable effects on diabetes and its complications. We investigated to find out the optimal composition showing hypoglycemic and antidiabetic complication effects in variable compositions (OG:MC = 1:1, 1:2, 1:4, 1:6, 1:8, 1:10, 2:1, 4:1, 6:1, 8:1, 10:1). Extracts were administered orally once a day for 28 days following 7 days post streptozotocin (STZ) dosing. Five rats per group (total 15 groups; Intact, STZ, OG, MC, and the variable composition groups) were selected according to the blood glucose and body weight at 6 days after STZ dosing. After 28 days of extracts dosing, the changes on the body weight, liver and kidney weight, blood glucose, blood urea nitrogen (BUN), creatinine, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), low-density lipoprotein (LDL), and total-cholesterol levels were observed. As the result of STZ-induced diabetes, decreases of body weight, increases of the liver and kidney weights, blood glucose, BUN, creatinine, AST, ALT, LDL, and total-cholesterol levels in STZ control were detected compared with intact control. However, these changes of hyperglycemia, diabetic nephropathy, hepatopathy, and hyperlipemia were dramatically decreased in the OG and MC single-dosing group, and all composition groups. In addition, there were more favorable effects in all composition groups compared with the OG and MC single-dosing groups. Among variable compositions, the OG:MC 1:2 mixed group showed the most synergic effects in this study.

Effects of Selenium and Vitamin I Administration on Blood Components in Hanwoo Young Bulls (한우 후보종모우에 Selenium과 Vitamin E 투여가 혈액성분에 미치는 영향)

  • To identify an antioxidant system, Se and Vit. E were administered into Hanwoo young sire and the effects of administration on blood components(Se, Vit. E, chemical values, estradiol-17 $\beta$, testosterone) were examined. The 16 heads ranging from twenty to thirty two months of age were randomly assigned to control group, Se-administered group(Se-group), Vit. E-administered group(Vit. E-group) and Se and Vit. E administered group(Se and Vit. E-group). Each reagent (Se : 0.1 mg, Vit. E . 1,500IU, Se+vit. E : 0.1 mg+1,500IU per kg of body weight, respectively) administered 3 times every 30days by intramuscular injection. Se concentration in serum was higher in Se-group and Se and Vit. E-group than in control group and Se and Vit. E-group also was higher than Vit. E-group(p<0.05). Although all Se-, Vit. E-administered groups were a little higher than control group, the injection of Se and Vit. E were not significant effect on Vit. E concentration in serum(p>0.05). All groups showed significant variance by periods, but there were not significantly different among groups in blood chemical values. The estradiol-17 $\beta$ concentrations of all Se-, Vit. E-administered groups were a little higher than those of control group, but there were not significant(p>0.05). There have no significant difference among groups in testosterone concentration. These results indicate that the administration of Se, Se + Vit. E increase Se concentration in Hanwoo young bull.

Effects of honeybee (Apis mellifera L.) venom by water supply on the performance of broiler chicken (봉독의 급수투여가 육계의 생산성에 미치는 영향)

  • This study was conducted to investigate the antibiotic effects of bee venom (Apis mellifera) on the growth performance and blood characteristics in broiler chicken. 1-day-old broiler chicks were randomly divided into 3 groups with 3 replicates of 5,000 birds each. The treatments were control without antibiotic and bee venom, 0.5ppm or 1ppm bee venom. The final body weight and body weight gain were significantly higher in bee venom than control (P<0.05). The feed conversion ratio in all treated groups were significantly improved as compared to that of control (P<0.05). No significant differences among the groups were observed in the contents of total cholesterol, aspartate aminotransferase (AST), alanine aminotransferase (ALT), total protein, albumin and globulin in blood serum. The white blood cell, red blood cell, heterophil, lymphocyte, and stress indicator (heterophil:lymphocyte rate) were not significantly different among the groups. The superoxide dismutase-like activities in the groups that were water containing bee venom were significantly increased compared to those in the control group (P<0.05). It has been suggested that bee venom promotes the growth of broilers without any negative effect when added in broiler water.

Comparison of Dietary Behaviors and Blood Clinical Indices in Underweight, Normal Weight, Normal Weight Obese and Obese Female College Students (저체중, 정상, 마른 비만 및 비만 여대생의 식행동 및 혈액 임상지표 비교)

  • Objectives: Normal weight obesity (NWO) is defined as excessive body fat in the context of a normal body mass index (BMI). This condition carries a greater risk of developing noncommunicable chronic disease and has been associated with early inflammation. This study was conducted to compare the anthropometric measurements, eating behaviors, and blood clinical indices among four groups: underweight, normal, normal weight obesity and obesity. Methods: The subjects included 215 female college students. A questionnaire was administered regarding general characteristics, dietary behaviors, food consumption frequency. Anthropometric measurements and blood clinical indices were also investigated. Results: The average BMI, body fat percentage, waist circumference, fat-free mass, and muscle mass were highest in the obesity group (p<0.05). Most subjects had tried to lose weight and perceived that their health was worse than before they became college students. The ratio of students in the NWO group who thought their health was very poor was significantly higher than in the other three groups (p<0.05). The obesity and NWO groups seemed to eat more and their eating speed was significantly faster than the other groups (p<0.001). The consumption frequency of caffeinated beverages was significantly higher in the NWO group than in the other three groups (p<0.01). WBC was significantly higher in the obesity group (p<0.05). Serum levels of TG and total cholesterol were also significantly higher in the obesity group (p<0.05). Serum GPT was significantly higher in the obesity group (p<0.05) while BUN level was highest in the NWO group (p<0.05). Conclusions: The obesity group showed the most health problems while the NWO group seemed relatively healthy. However, NWO can lead to problems such as metabolic syndrome and cardiovascular disease in later life if poor dietary habits are maintained. Therefore, education in appropriate eating habits is needed for these subjects.

Effects of cultivated wild ginseng pharmacopuncture at CV12 and ST25 on blood cell composition and cytokine levels in rats (中脘(중완)(CV12) 및 천추(天樞)(ST25) 산양산삼약침 처치가 흰쥐의 혈액성상 및 싸이토카인에 미치는 영향)

  • Objectives : To investigate whether cultivated wild ginseng pharmacopuncture at CV12 and/or ST25 has any immune-enhancing effect in normal rats. Methods : Effects of cultivated wild ginseng pharmacopuncture at CV12 and/or ST25 on blood cell counts, cell composition, cytokine and plasma protein levels were investigated in normal rats. Results : Red blood cell count, hematocrit and hemoglobin levels significantly increased in cultivated wild ginseng pharmacopuncture groups compared with those of the normal group. There was, however, no significant difference in white blood cell (WBC) count and WBC differential count between the normal group and the pharmacopuncture groups. Cultivated wild ginseng pharmacopuncture groups had significantly higher levels of tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ and interleukin-6 (IL-6) than the normal group while no significant difference between groups was found in interleukin-$1{\beta}\;(IL-1{\beta})$ level. Total protein and albumin levels were not different between groups. Conclusions : The results suggest that cultivated wild ginseng pharmacopuncture at CV12 and/or ST25 may have no immune-enhancing effect in normal rats.

Nutrient Intakes and Blood Lipids According to Obesity Degree by Body Fat Percentage among Middle-Aged Women in Gunsan City (군산시 중년여성의 체지방률에 의한 비만도 분류에 따른 영양소 섭취와 혈중지질에 관한 연구)

  • The purpose of this study was to compare nutrient intakes and blood lipids of middle-aged women according to the obesity index by %Fat. Subjects were assigned to one of the following groups based on percentage of body fat (%Fat): normal weight (18% ~ < 28%), overweight (28% ~ < 33%) and obesity (over 33%). Nutrient intakes were evaluated based on questionnaires of 24 hours recall method and blood lipids were analyzed by blood analyzer. The results were as follows. 1) Nutrient intakes were that carbohydrates and fat intakes in obesity group were higher than normal and overweight group but the difference was not significant, and protein intake rate was similar all groups. The intake rate of calcium was higher in normal group than obesity group (p < 0.05), and obesity group ingested under dietary reference intakes. The intake rate of iron was higher in obesity group than normal group (p < 0.05). 2) TG, TC, VLDL and TC/HDL in obesity group were higher than normal group (p < 0.001). The attack rate of coronary heart disease in obesity group is higher than normal group (p < 0.01), LDL, blood glucose and blood pressure in obesity group were higher than normal group (p < 0.01), and HDL was a little higher in normal group than obesity group, but the difference was not significant. 3) The correlation of anthropometric measurements, blood glucose, blood lipid, and blood pressure had significant results. Weight was associated positive level with blood glucose, blood lipid and blood pressure. Waist and WHR were associated positive level with blood glucose and blood lipid but weren't associated with blood pressure. BMI was associated positive level with blood glucose, blood lipid and blood pressure, %Fat was associated positive level with blood lipid and blood pressure, but wasn't associated with blood glucose. These results suggest that the decrease of waist on blood glucose control is better than decrease of % Fat and BMI, the decrease of %Fat and BMI on blood pressure control is better than decrease of waist, and the decrease of %Fat on blood lipid control is better than waist and BMI. The nutritional education for obesity treatment must perform to analyze the blood and assess the obesity degree by %Fat, waist and BMI before nutritional education, so the obesity treatment will be effectively.

The Antiulcer Effects of Alove vera on the Stomach ulcer Induced by Stress in rats (흰쥐의 스트레스성 위궤양에 대한 Alove vera의 항궤양작용)

  • This was performed to investigate the antiulcer effects of Aloe vera on the stomach ulcer induced by restraint and water-immersion stress in rats. For this experiment, 60 male Sprague-Dawley strain were used. The experimental groups were divided into five: a control(CA) and 4 aloe treatment groups. Each dose of aloe was 50 mg/kg BW(AA), 100mg/kg BW(AB), 200 mg/kg BW(AC), and 400 mg/kg BW(AD). The rats were allocated to each group by 12 and observed for 4 weeks. The results are as following. 1. The stomach surface pH in each group showed no significant difference, byt the values of aloe treatment groups were higher than the value of the control group. 2. The gastric wall mucus was significantly increased in all aloe treatment groups(p<0.05) compared with the control group. Especially in AC and AD group the differences were higher(p<0.01). 3. At shear rate rate 11.25, 45.0, 90.0, 225.0 sec-1, whole blood viscosity and plasma viscosity were measured. Most of the values of aloe treatment groups were significantly low compared with those of the control group(P<0.05). 4. The ulcer index of aloe treatment groups were significantly low campared with control group(p<0.05). Especially in AC and AD group the differences were more significant(p<0.01). 5. Less severe ulcers were observed in AA and AB group than in the control group. Tissues of AC and AD group had only slight ulcers and necrosis of tissue was not observed in these groups. Especially in AD group, there was more mucus than other groups and it seemed that alove vera stimulated the epithelial regeneration. From the results of this study, it can be concluded that the oral administration of Alove vera results in protection of stomach ulcer by stimulating the secretion of gastric mucus and the circulation of blood.

The Relationship of Nutritional Satus to the Degree of Hypertension in Residents of Kangbuk-gu, Seoul (서울 강북구 일부 주민들의 고혈압 분류에 따른 영양상태 비교)

  • The purpose of this study was to relate the degree of hypertension to nutritional status, in order to prepare basic data for a nutrition program. The study using the WHO standard divided the residents in Kangbuk-gu into a normal group (NG) of 254 adults with systolic blood pressure (SBP) < 130 or diastolic blood pressure (DBP) < 85; a borderline hypertensive group (BG) of 81 adults with 130 $\leq$ SBP $\leq$ 140 or 85 $\leq$ DBP $\leq$ 90; and a hypertensive group (HG) of 143 adults with SBP > 140 or DBP > 90. The mean age of HG was older than the other groups (p < 0.001). The dietary intake was investigated by the 24-hour recall method. The Ca intake as the percentage of the Korean Recommended Dietary Allowances (RDA) for the HG were higher than that of the other groups (p < 0.01). The Nutrient Adequacy Ratio (NAR) of Ca and Vit. A were below 0.75 for all three groups. The Mean Adequacy Ratio (MAR) was above 0.75 (p > 0.05) for all three groups. The Dietary Variety Score (DVS) of the NG was 22.7, and higher than those of the other groups (p > 0.05) The fasting blood sugar (p < 0.001), total cholesterol (p < 0.001), protein (p < 0.001), albumin (p < 0.01), hemoglobin (Hb) (p < 0.001), alkaline phosphatase (ALP) (p < 0.001), and creatinine (p <0.05) values of the HG were higher than those of the other groups. The Body Mass Index (BMI) and the waist-hip ratio (WHR) of the HG were higher than those of the other groups (p < 0.001) . In conclusion, these results showed that there tended to be differences among the three groups. For effective nutritional education and counseling of each group, we should furtherstudy the differences and understand their characteristics of each group in order to provide nutritional education for controling and preventing hypertension.

The Effects of Antioxidant Vitamins Via Comet and Mitogenic Assay in Mice Exposed to $AFB_1$ ($AFB_1$에 노출된 마우스에서 Comet Assay와 Mitogenic Assay에 의한 항산화 비타민의 효과)

  • The objective of this study was to examine the effects of antioxidant vitamins on the cellular oxidant damage by observing the mitogenicity in the mouse spleen and the strand breaks of DNA in mouse blood induced by $AFB_2$. Intraperitoneal(i.p.) injections of vitamin C(VC) of 10 mg/kg and vitamin E(VE) of 63.8 mg/kg were repeatedly administered to male ICR mice of 6 weeks old at intervals of 4 times every 2 days. After one hour vitamin treatments, $AFB_1$ of 0.4 mg/kg was injected into the $AFB_2$ plus vitamin treated groups in the same way. On the other hands, into the $AFB_2$ only treated group, only $AFB_2$ was injected without vitamins in the same method as above. The results of the experiment are as follows ; as regard to comet assay, DNA strand breaks were clearly present and they formatted a typical comet tail in the mice blood of the $AFB_2$ only treated groups. However, comet tails apparently disappeared in $AFB_2$ plus antioxidant vitamins treated groups since oxidant damage was controlled in an almost similar level to the control group. Mitogenicity of the spleen also showed a similar tendency as before, and these differences were more remarkably observed in the reaction against Con-A, which is a T-cell mitogen. In these data, the statistical significance was p<0.01. The LDL and VLDL levels were 408.72, 504.47 mg/dl respectively in the $AFB_2$ only treated groups. Compared with the $AFB_1$ only treated groups, those of $AFB_2$ plus antioxidant vitamin treated groups decreased to 272.06(VC), 305.28 mg/dl(VE), respectively. On the other hand, HDL levels were diminished to 32.60, 29.60 mg/dl in $AFB_2$ only treated groups, compared to 42.23, 41.14 mg/dl in the $AFB_2$ plus antioxidant vitamins treated groups. But, blood glucose levels were not statistically significant.

The antidiabetic properties of fractions of Lycopus lucidic Turcz in streptozotocin diabetic rats (택란 분획물이 당뇨유발 흰쥐에서의 혈당강하에 미치는 영향)

  • This study was carried out to evaluate the effects of fractions of methanol(MeOH) extracts of Lycopus lucidic Turcz on hyperglycemia and energy metabolites in streptozotocin(STZ) diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of STZ dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight, and the rats were divided into 7 groups, that is, one normal group and 6 diabetic groups: STZ-control, hexane, chloroform(CHCl$\sub$3/). ethylacetate(EtOAc), butanol(BuOH) and H$\sub$2/O fraction-fed groups. All groups were fed an AIN-93 diet and the fractions of Lycopus lucidic Turcz were administered orally with 2 % Tween 80 for 14 days after the STZ injection. Body weight, diet intake and organ weights were monitored. The plasma levels of blood glucose, insulin and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglycerides and free fatty acid were assayed. The plasma activities of aspartate aminotransferase (AST) and alanine aminotransferase(ALT) were also measured. Body weight losses were observed by feeding the fractions of Lycopus lucidic Turcz in STZ experimental groups, and the kidney weight was increased. The extent of blood glucose decrement was significantly greater in the hexane and BuOH fraction-fed groups than STZ-control group. The plasma protein level was significantly lower in the H$\sub$2/O fraction-fed group. The plasma cholesterol level was decreased in BuOH and H$\sub$2/O fraction-fed groups compared with the STZ-control group. The levels of free fatty acids in the CHC1$\sub$3/ and H$\sub$2/O fraction-fed groups were significantly decreased(p<0.05). ALT activitiy of BuOH fraction-fed group was lower than control but it was not significantly different. These results suggest that the fractions of Lycopus lucidic Turcz are capable of lowering blood glucose and fat metabolites concentrations when administered to STZ-treated rats, and AST/ALT activity and insulin levels show the possibility of therapeutic use to diabetes mellitus.
