말초혈액영상에서 신경망 모델을 이용한 적혈구의 형태학적 변이 분류 (Morphological Variation Classification of Red Blood Cells using Neural Network Model in the Peripheral Blood Images)

  • Recently, there have been researches to automate processing and analysing images in the medical field using image processing technique, a fast communication network, and high performance hardware. In this paper, we propose a system to be able to analyze morphological abnormality of red-blood cells for peripheral blood image using image processing techniques. To do this, we segment red-blood cells in the blood image acquired from microscope with CCD camera and then extract UNL fourier features to classify them into 15 classes. We reduce the number of multi-variate features using PCA to construct a more efficient classifier. Our system has the best performance in recognition rate, compared with two other algorithms, LVQ3 and k-NN. So, we show that it can be applied to a pathological guided system.

The Role of CD4 T Cell Help in CD8 T Cell Differentiation and Function During Chronic Infection and Cancer

  • CD4 and CD8 T cells are key players in the immune response against both pathogenic infections and cancer. CD4 T cells provide help to CD8 T cells via multiple mechanisms, including licensing dendritic cells (DCs), co-stimulation, and cytokine production. During acute infection and vaccination, CD4 T cell help is important for the development of CD8 T cell memory. However, during chronic viral infection and cancer, CD4 helper T cells are critical for the sustained effector CD8 T cell response, through a variety of mechanisms. In this review, we focus on T cell responses in conditions of chronic Ag stimulation, such as chronic viral infection and cancer. In particular, we address the significant role of CD4 T cell help in promoting effector CD8 T cell responses, emerging techniques that can be utilized to further our understanding of how these interactions may take place in the context of tertiary lymphoid structures, and how this key information can be harnessed for therapeutic utility against cancer.

Zinc Increases Chemotactic Activity of Porcine Peripheral Blood Polymorphonuclear Cells

  • Zinc is necessary for normal functions in the immune system. The objective of the study is to examine the effect of zinc on the chemotactic activity of porcine peripheral blood polymorphonuclear cells (PMNs). A modified Boyden chamber was used to determine the directional migration distance of PMNs. Various concentrations of zinc showed no chemotactic activity to PMNs. However, culture supernatant from peripheral blood mononuclear cells (PBMCs) treated with zinc remarkably increased the chemotactic activity of PMNs when compared with culture supernatant from PBMCs treated without zinc. Culture supernatant from PBMCs treated without zinc also increased the migration distance of PMNs relative to vehicle control (medium alone). Increasing effect in chemotactic activity of PMNs by culture supernatant from PBMCs treated with zinc was inhibited by treatment of porcine anti-interleukin (IL)-8 polyclonal antibody (pAb). This effect was not affected by heat treatment ($4-85^{\circ}C$). This corresponded with heat stable physical characteristics of IL-8. These results suggest that zinc can upregulate the chemotaxis of PMNs, which is primary mediated by IL-8 chemotactic factor released from PBMCs treated with zinc.

Tea Flavonoids Induced Differentiation of Peripheral Blood-derived Mononuclear Cells into Peripheral Blood-derived Endothelial Progenitor Cells and Suppressed Intracellular Reactive Oxygen Species Level of Peripheral Blood-derived Endothelial Progenitor Cells

  • Endothelial dysfunction in atherosclerosis is associated with increasing oxidative stress that could be reversed by antioxidant. Therefore epigallocatechin gallate (EGCG), epicatechin gallate (ECG), epigallocatechin (EGC) and catechin (C) of tea flavonoids were investigated for their roles in regenerating endothelial cell. Peripheral blood mononuclear cells (PB-MNCs) were isolated, plated and cultured in medium with/without treatment of EGCG, ECG, EGC and C. Results showed that among all EGCG, ECG, EGC and C concentrations tested, $12.5{\mu}mol/L$ was not cytotoxic for peripheral blood-derived endothelial progenitor cells (PB-EPCs). Treatment of EGCG, ECG, EGC or C increased the percentages of CD34, CD133, VEGFR-2 expressions and suppressed hydrogen peroxide-induced percentages of reactive oxygen species (ROS) level in PB-EPCs. Taken together, our current results showed that EGCG, ECG, EGC or C of tea flavonoids could induce differentiation of PB-MNCs into PB-EPCs as well as protect PB-EPCs from oxidative damage by suppresing the intracellular ROS levels.

Aflatoxin-mediated Sperm and Blood Cell Abnormalities in Mice Fed with Contaminated Corn

  • The effect of aflatoxin-contaminated corn on albino mice was investigated using the sperm morphology assay. Blood parameter levels including; total white blood cells (WBC), total red blood cells (RBC), packed cell volume (PCV), serum bilirubin (SB) and fasting blood sugar (FBS) were also determined in the tested mice. Test mice were exposed to aflatoxin-contaminated corn (contamination level of 100 ppb) for $1{\sim}4$ weeks while aflatoxin-free corn and cyclophosphamide were used as negative and positive controls, respectively. Sperm cells showed varieties of morphological abnormality when assessed after 5 weeks. The percentage frequencies of the negative and positive controls were 18.8% and 48.87%, respectively, while the percentage abnormalities for the 1, 2, 3 and 4 weeks exposures were 41.38%, 48.17%, 57.13% and 61.67%, respectively. PCV, WBC, total bilirubin and glucose level values of mice in all concentrations were higher and statistically significant as compared to the negative control values using Dunnett's test. Therefore, abnormal sperm cell induction is concentrationdependent such that continuous consumption of aflatoxin-contaminated corn is capable of negatively affecting spermatogenesis by inducing or increasing the frequency of morphologically abnormal sperm cells produced.

한우 무손상 적혈구의 superoxide 및 과산화수소 제거능력 (Scavenge of superoxide and hydrogen peroxide by bovine intact red blood cells)

  • The ability of bovine intact red blood cells to scavenge superoxide and hydrogen peroxide by superoxide dismutase, catalase and glutathione peroxidase was investigated. Intact red cells(up to 0.4%) suspensions did not inhibit ferricytochrome c reduction by superoxide in the superoxide generating system. On the other hand, intact red cell(0.4%) suspensions almost completely inhibit ferrocytochrome c oxidation by hydrogen peroxide. The ability of intact red cells to scavenge hydrogen peroxide was mainly attributed to either membrane bound catalase or glutathione peroxidase. The scavenge of hydrogen peroxide by 0.1~0.2% intact red cells showed a trend of dependence on mainly glutathione peroxidase. However, at blood cell concentration higher than 0.3%, the process depended upon peroxidase-independent scavengers like catalase. Enhancement of ferrocytochrome c oxidation by red cells treated with aminotriazole proved that the protection against hydrogen peroxide was due to catalase, while the protection in the presence of glutathione indicated scavenging effect of glutathione peroxidase against hydrogen peroxide.

대식세포의 Fc 수용체를 통한 탐식에 미치는 Inositol-phosphatase의 영향 (Effect of Inositol-phosphatase on Fc Receptor-mediated Phagocytosis of Macrophages)

  • Background: Fc receptor-mediated phagocytosis is a complex process involving the activation of kinases and phosphatases. FcgammaRIIB has been known to transduces inhibitory signals through an immunoreceptor tyrosine-based inhibitory motif (ITIM) in cytoplasmic domains. In this study, we examined the involvement of inositol-phosphatase in the Fc receptor-mediated phagocytosis. Methods: J774 cells were infected using vaccinia viral vector containing SH2 domain-containing inositol-phosphatase (SHIP) cDNA and stimulated with the sensitized sheep red blood cells. Results: Stimulation of J774 cells induced the tyrosine phosphorylation of SHIP which was maximal at 5 minutes. Phosphatidylinositol-3 (PI-3) kinase inhibitor (wortmannin) inhibits J774 cell phagocytosis of sensitized sheep red blood cells in a dose-dependent manner. Heterologious expression of SHIP in J774 cells inhibits phagocytosis of sensitized sheep red blood cells in a dose-dependency manner, but catalytically dead mutants of SHIP has no effect on phagocytosis. Conclusion: These results strongly suggest that the active signals mediated by PI-3 kinase are opposed by inhibitory signals through SHIP in the regulation of Fc receptor-mediated phagocytosis.

Histology and morphometry of the skin of the trident goby Tridentiger brevispinis (Perciformes, Gobiidae)

  • The Korean trident goby, Tridentiger brevispinis, lives in adverse habitats that can easily become hypoxic due to low precipitation, regional dry periods, and high amounts of solar radiation. Histological and morphometric studies revealed the goby's specialized skin (35.4-150.0㎛ in thickness), consisting of an epidermis and dermis. The thicker epidermis comprises an outermost surface layer (having taste buds, stratified fattened cells, mucous cells, pigment cells, and stratified polygonal cells), middle layer (having stratified polygonal cells), and stratum germinativum (stratified columnar cells). In particular, the dermis has scales, well-developed vascularization, and a few blood capillaries just above the basement membrane, and a reduced diffusion distance was present in the lateral body. Consequently, adaptations such as thicker epidermis, well-developed vascularization, few blood capillaries, and a reduced diffusion distance may provide cutaneous respiration for survival in poorly oxygenated water during the periodic dry season.

  • The purpose of this study was to investigate effects of inner skin of chestnut on the activation of a living body's function (regional cerebral blood flow and mean arterial blood pressure in Sprague-Dawley rats, proliferation of thymocytes in normal mice and L1210 cells transplanted mice) . We used inner skin of chestnut extract(Sample A : inner skin of chestnut-panbroiled after driedextract (100$\^{C}$ ), Sample B , inner skin of chestnut-panbroiled-extract(100$\^{C}$ ) , Sample C : inner skin of chestnut -panbroiled after dried-extract(80$\^{C}$ ), Sample D : inner skin of chestnut-panbroiled-extract(80$\^{C}$)} Regional cerebral blood flow(rCBF) and Mean arterial blood pressure(MABP) were tested using Leser -Doppler Flowmetry(LDF), and the proliferation of thymcytes was tested using a colorimetric tetrazoliun assay ( MTT assay) The experimental results as follows 1. rCBF was significantly increased by Sample C in a dose-dependent manner. 2. MABP was not changed by Sample C in a 0.1mg/kg∼10.0mg/kg treated group. 3. Proliferation of thymocytes was not changed by Sample C in normal mice. 4. Proliferation of thymocytes was significantly accelerated by Sample C in L1210 cells transplanted mice.

Secreted decoy of insulin receptor is required for blood-brain and blood-retina barrier integrity in Drosophila

  • Glial cells play important roles during neurogenesis and in maintaining complex functions of the nervous system. Here, we report the characterization of a gene, Sdr, which contains a putative insulin-like growth factor receptor domain and is required to maintain critical nervous system functions in Drosophila. Sdr is expressed in glial cells during embryonic and larval stages of development, but its role in adult flies is poorly understood. As insulin signaling is important throughout the lifespan in human, we investigated the Sdr's role in adult flies. Our results demonstrate that Sdr is expressed on surface glial cells that surround the nervous system. Mutation of Sdr did not affect development but caused defects in locomotion and lifespan. Sdr mutants also showed increasingly severe defects in the blood-brain- and blood-retina-barriers as they aged. Therefore, we suggest a novel role of Sdr in maintaining the integrity of the blood-brain- and blood-retina-barriers in adult flies.
