Effects of Cheonghyul-San on the Generation of Redox Status and on the Expression of NF-${\kappa}$B Dependent Proteins (청혈산(淸血散)이 Redox Status 및 NF-${\kappa}$B 의존성 단백질에 미치는 영향)

  • The aim of this study was to investigate the effects of Cheonghyul-san on the generation of peroxynitrite ($ONOO^-$), nitric oxide (NO) and superoxide anion radical ( ${\cdot}\;O_2^-$), and on the expression of NF-${\kappa}$B-dependent proinflammatory proteins in ob/ob mice. Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57BL/6J black mice) and control obese (ob/ob mice) groups have received the standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein-2 (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blot was performed using anti-IKK-${\alpha}$, anti-phospho I${\kappa}$B-${\alpha}$, anti-NF-${\kappa}$B (p50, p65), anti-COX-2, anti-iNOS, anti-VCAM-1 antibodies, respectively. Cheonghyul-san prevented $H_2O_2$-induced cell death. Cheonghyul-san inhibited the generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ in the $H_2O_2$-treated LLC-$PK_1$ cells. The generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ were inhibited in the Cheonghyul-san-administered ob/ob mice groups. The GSH/GSSG ratio was decreased in the ob/ob mice, whereas the ratio was improved in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the protein expression levels of phospho-I${\kappa}$B-${\alpha}$, IKK-${\alpha}$, NF-${\kappa}$B (p50, p65), COX-2, iNOS and VCAM-1 genes. These results suggest that Cheonghyul-san is an effective scavenger of $ONOO^-$, ${\cdot}\;O_2^-$ and NO, and has an inhibitory effect on the expression of NF-${\kappa}$B-dependent inflammatory genes in ob/ob mice. Therefore, Cheonghyul-san might be used as a potential therapeutic drug against the diabetes- and obesity-related proinflammatory diseases.

  • Winterkill can be defined as any injury including freeze stress kill, winter desiccation, and low temperature disease to turfgrass plants that occurs during the winter period. The major damages from winterkill were low temperature kill, crown hydration, and winter desiccation. Low temperature kill is caused by air and soil temperature. Soil temperature affect more severe to turfgrass than air temperature because low soil temperature cause fetal damage to turfgrass crown. Crown hydration is a form of winter injury in which intercellular water within the plant freezes and causes physical injury to the cell membrane and wall. This is eventually resulted in dehydration of cell. Winter desiccation is the death of leaves or whole plants due to drought during the winter period. To reduce winterkill damage, cultivar selection is very important. If changing cultivar is not allowed, cold temperature hardiness needs to be increased by providing nutrients especially phosphorus and potassium in the late fall. Turf cover is effective way to reduce winterkill damage. Remaining snow is positive process to reduce winterkill damage by insulating soil temperature. The previous researches reported many materials as turf cover such as straw, polypropylene, polyester, and wood mat. Aeration and topdressing is one of the process against winterkill. Both methods are mainly conducted to reduce thickness of thatch layer. In recent, relatively new materials called black or winter topdressing sand are used to protect soil temperature from low air temperature and thaw ice crystal that may remain in soil.

Resistance Evaluation of Several Turfgrass Species and Graminious Crop Species against Rhizoctonia cerealis and Typhula incarnata under Controlled Conditions (주요 잔디류와 화본과 식량 밭작물의 황색마름병원균 및 설부소립균핵병원균에 대한 저항성 평가)

  • During 2008~2009 winter season, yellow patch and gray snow mold occurred on turfgrass plants in golf courses in Kangwon and Jeonbuk provinces, respectively. The fungi associated with the diseases were identified as Rhizoctonia cerealis Van der Hoeven and Typhlua incarnata Lasch ex Fr., based on the morphological characteristics of hyphae and sclerotia. R. cerealis and T. incarnata were pathogenic to most turfgrass and crop species tested. R. cerealis infected crown, stem and leaf tissue of the host plants, and the symptom was light yellow circular patch. Individual infected leaf near the margin of patch developed red color first and finally turn brown. The symptoms caused by gray snow mold pathogen are water-soaked spots, and became a watery soft rot. Infection parts became yellow and then turned brown followed by death of the whole plant. White mycelia were developed on higher petioles, leaves, and on soil where these plant parts lay, and black sclerotia of variable size and shape formed in the mycelial mass. All isolates tested were pathogenic on most turfgrass and crop plants, and significantly different in aggressiveness. Disease severity increased with longer snow cover days on target plants, suggesting that disease severity was expressed over snow cover days. There were significant differences in disease severity among the graminious species, and among cultivars within each species, indicating varying levels of susceptibility to R. cerealis and T. incarnata.

Contamination level of commercialized pepper and sterilization effect by intense pulsed light in batch system (시중 판매 후추의 오염도 및 회분식 광펄스 처리에 의한 살균 효과)

  • Twenty-nine pepper products commercially available in the market were collected and investigate for contamination levels. Pepper products purchased from traditional markets had a degree of contamination of $10^6-10^7CFU/g$ aerobic bacteria, $10^4-10^5CFU/g$ Bacillus sp., and less than $10^2CFU/g$ yeast and molds. Organic pepper showed a degree of contamination of $10^4$ aerobic bacteria, $10^2-10^3$ Bacillus sp., and less than $10^1$ yeast and molds. Intense pulsed light (IPL) treatment of 10 min (1,000 V, 5 pps and 4 cm sample-to-lamp distance) showed a bacterial death rate of 1.45-1.55 log for whole peppers, and of 0.8-0.85 log for black and white pepper powder. The sterilization rate using IPL was higher than that using other non-thermal sterilization methods, such as ozone treatment or low-pressure discharge plasma sterilization, indicating that the IPL sterilization method may find potential application in the industry. However, further studies may need to be conducted to enhance the effect of sterilization.

  • 2020 was a year in which the world spent in disorder due to the pandemic of Coronavirus infection-19(COVID-19). The pandemic was at the beginning of a turning point in history. For examples, the Black Death(Pest) that destroyed the feudal system of medieval Europe in the 14th century, smallpox that led to the destruction of the Inca Empire by Spain in the 17th century, and the Spanish flu that ended World War I early. The great transformation that will come after COVID-19 is presented from various fields and perspectives, but the understanding and direction of the transformation is ambiguous. This study attempts to derive and to analyze core terms based on a network of the future of globalization after COVID-19. Four Networks related to globalization, anti-globalization, and globalization and digitalization after COVID-19 were established respectively. A network integrating four networks was also constructed. The core terms were extracted from the hub nodes, the stress centrality, and the simplified network to which the K-core algorithm was applied. After COVID-19, the changes in globalization were analyzed from the extracted core terms. This study is thought to be meaningful to propose a method of deriving and analyzing core terms based on a network in understanding social changes after COVID-19.

Fundamental Studies for the Breeding of Marine Algae 3. Effects of Plant Hormones on the Growth of Porphyra-fronds (해조의 육종을 위한 기초적 연구 3. 김 엽체에 미치는 식물호르몬의 생장효과)

  • As one of the fundamental studies for the breeding of marine algae, the effects of several plant hormones (IAA, Gibberellin, 2.4-D, NAA, Kinetin) on the growth of Porphyra-fronds, P. tenera Kjell. form tamatsuensis Miura, were investigated from January 21 to February 21 1981. The fronds used for the experiment were dissected out at $25mm^2$ size, and cultured in modified Provasoli's ESP medium supplemented with various concentrations of each plant growth regulators. The culture was kept under constant water temperature of $5^{\circ}C$ in 14 hrs. photoperiod and illuminated with 2,400 lux by fluorescent light. Based on the results of first experiment, the culture of fronds for the secondary experiment was carried out at $5^{\circ}C\;and\;10^{\circ}C$ in medium containing various levels of Kinetin from April 6 to 24, and compared the growth of two groups at each concentrations with each other, The results obtained are summarized as follows : (1) The best growth efficiencies were observed at 5.0mg/1 of each plant hormones except Gibberellin. Among them, the highest growth-rate was $312.5\%\;(345.3\%\;in\;frond\;size)$ in contrast with control at 5.0mg/1 of Kinetin, and was followed by $257.5\%\;(236.1\%)$ in 2.4-D,$166.7\%(147.6\%)$ in IAA and $141.7\%\;(167.7\%)$ in NAA, but that in Gibberellin was $247.9\%(241.9\%)$ at 10.0mg/l. (2) Especially, the fronds cultured at 5.0mg/1 of Kinetin were deep black-brown in colour, and had vivid, healthy chloroplasts in their all cells. On the contrary, the fronds cultured in other media were discoloured to light black-brown or green-drown, and almost all cells were vacuolated or shrunk gradually into death.(3) There was an obvious difference between the best growth-rates of the fronds cultured at 5.0mg/l of Kinetin at $5^{\circ}C$ and those at $10^{\circ}C$. The former was $366.7\%$, the latter $318.8\%$ but the difference was little at lower concentrations. (4) Many abnormal cells grown up to $25.0-27.5\mu$ in diameter were found among the marginal cells of fronds which showed the best growth in Kinetin, and the fronds wire $41.0-42.0\mu$ in thickness which was thicker by $10.0\mu$ or so than the others. (5) In two fronds at 1.0mg/1 of Kinetin cell-divisions were observed, which might developed into antheridium, but it was doubtful whether due to the efficiency of Kinetin.

Effects of N-Methylacetamide on the Viability, Fertility and Hatchability of Cryopreserved Ogye (Korean Native Black Fowl) Semen (N-Methylacetamide 동결보호제가 오계 동결정액의 생존성, 수정 및 부화율에 미치는 영향)

  • The use of methylacetamide (MA) as a cryoprotective agent for freezing Korean Native Black rooster Ogye semen was examined with artificial insemination. The diluted Ogye semen with HS-1 was subjected for 2 step dilution method of cryopreservation in which the final concentration of MA was adjusted to 7.5%. The sperm viability after thawing was reduced from $95.17{\pm}0.93%$ to $55.93{\pm}1.38%$ which was confirmed by live-death analysis based on Fluorescence-Activated Cell Sorting (FACS). The rates of fertilized eggs with fresh or frozen-thawed semen were reduced from $94.98{\pm}3.93%$ to $66.36{\pm}8.43%$ at day 7 with significant difference. However, the hatching rates of experiments at day 21 did not shown difference between $92.64{\pm}2.33%$ and $90.45{\pm}8.05%$ (P<0.05). With these results, the utilization of MA for freezing of Ogye spermatozoa could affect on viability of frozen-thawed semen but not on the fertility of lain eggs and hatchability of fertilized eggs and also provide possible tools of freezing for poultry genetic resource conservation.

A case study of verifying a suicide by carbon monoxide intoxication committed by burning an ignition charcoal briquette (착화탄 연소에 의한 일산화탄소 중독사에서 자살입증에 관한 사례연구)

  • Carbon monoxide (CO) intoxication, arising from CO from an ignited charcoal briquette (ICB), is a popular means of committing suicide in Korea. Most CO intoxications are related to suicide attempts; however, the possibility of a homicide disguised as a suicide cannot be ruled out. Therefore, forensic investigation of the deceased and the crime scene is crucial to confirm that the deceased committed suicide. Detection of the components of an ICB on the objects suspected of being contacted by the deceased, such as the hands, nostrils, and doorknobs, is essential for linking the crime scene to the victim in the case of suicides by ignited ICBs. The traces from an ICB were analyzed by investigating the morphological characteristics and obtaining elemental compositions. The ICBs were completely different from blackened wood, as detected by discriminant analysis with the elements of carbon and oxygen. We analyzed one case of CO intoxication to demonstrate an excellent procedure for verifying whether a suicide occurred with an ICB. We employed SEM-EDX for the analysis of an ICB, microscope-FT/IR and pyrolysis-GC/MS for a partly burnt resin-type substance, GC/MS for diphenhydramine (a sleeping drug), and GC/TCD for the CO-Hb level. We detected traces of an ICB on the hands, nostrils, and doorknobs, which were all discriminated into an ICB group. Detection of ICB traces from the nostrils could indicate that the deceased started the fire themselves to commit suicide. The partially burnt black material was analyzed as an acrylronitrilestyrene polymer, which is normally used to make bags for carrying or wrapping and could be assumed to have been used to transport the ICB. Diphenhydramine, a sleeping drug, was detected at a level of 2.3 mg/L in the blood, which was lower than that in fatal cases (8-31 mg/L; mean 16 mg/L). A CO-Hb level of 79% was found in the blood, which means that the cause of death was CO intoxication. The steps shown here could represent an ideal method for reaching a verdict of suicide by CO intoxication produced by burning an ICB in a sealed room or a car.
