Using a 50-1 airlift bioreactor, for the effective production of tylosin from Streptomyces fradiae TM-224 using raw cornmeal as the energy source, various environmental factors were studied in flask cultures. The maximum tylosin concentration was obtained at $32^{\circ}C$ and pH between 7.0 and 7.5. When seed was inoculated after 24 h of culture, the maximum tylosin concentration, 5.7 g/l, was obtained after 4 days of culture. Various concentrations of raw cornmeal were tested to investigate the optimum initial concentration for the tylosin production. An initial raw cornmeal concentration of 80 g/l gave the highest tylosin concentration, 5.8 g/l, after 5 days of culture. Of the various nitrogen sources, soybean meal and fish meal were found to be the most effective for the production of tylosin. In particular, with the optimal mixing ratio, 12 g/l of soybean meal to 14 g/l of fish meal, 7.2 g/l of tylosin was obtained after 5 days of culture. To compare raw cornmeal and glucose for the production of tylosin in the 50-1 airlift bioreactor for 10 days, fed-batch cultures were carried out under the optimum culture conditions. When raw com meal was used as the energy source, the tylosin production increased with increasing culture time. The maximum tylosin concentration after 10 days of culture was 13.5 g/l, with a product yield from raw cornmeal of 0.123 g/g of consumed carbon source, which was about 7.2 times higher than that obtained when glucose was used as the carbon source.
Recently, developments of large scale and high density cell culture methods have been the objects of many researches, because the demand of various pharmaceutical products produced by animal cell culture has been rapidly increasing. The cell culture equipment should have the requirements such as sufficient oxygen transfer and mixing, low shear stress and surface tension, and small foaming. In order to develop a proper bioreactor meeting these requirements simultaneously, a perfluorocarbon having high solubility of oxygen was sprayed into the medium as an oxygen carrier instead of air. Also, a new impeller was developed and combined together with the perfluorocarbon spraying system so as to design a new bioreartor for cell cultivation. The new impeller had better characteristics of mixing and oxygen transfer than the paddle and cell-lift impellers based on the same, shear rate. But, it was observed that the volumetric oxygen transfer coefficient of the new bioreactor decreased with increasing cell density during E. coli fermentation.
Oxygen is a key substrate in animal cell metabolism and its consumption is thus a parameter of great interest for monitoring and control in animal cell culture bioreactor. The use of a gas-permeable membrane offered the possibility to provide the required quantity of oxygen into the culture. while avoiding problems of foaming or shear damage generally linked to sparging. For determining the optimum DO control strategy of this gas-permeable membrane aeration bioreactor, the oxygen transfer rate coefficient was measured with varying $N_2$ ratio in inlet air. The results showed that an increasing mass flow rate of nitrogen reduced the $K_La$ value. and 5% nitrogen in air did not result in any oxygen limitation.
The optimal conditions for operating a moving-aeration bioreactor were determined as 30rpm and 150 (ml/min) of air flow rate, which can yield ca. 7.3 (l/h)of maximum mass transfer coefficient. It was also found that the agitation speed played much much important role than air input rate in oxgen transfer into the medium. $2.6{\times}10^6$ (cells/ml) and 0.6 (ml/l) of maximum cell denisty and IL-2 production were observed in batch cultivation of IL-2 producing BHK cell line. 0.53 (mM/l/h) of oxygen uptake rate was also estimated. The performance of a moving-aeration bioreactor (specific growth rate and oxygen uptake rate, etc.) was superior to other culture systems, such as cell-life and static membrane aeration bioreactors. Ii must be useful to apply this reactor to many culture processes by improving structural limitations in scaling-up the system.
본 연구에서는 음식폐기물 효소당화액을 BC생산 배지원으로 이용하여, flask정치배양과 50 L로 규모확대한 공기순환배양기에서 BC생산성을 검토하고, 생산된 BC의 제반물성을 조사하였다. 전단력을 낮추기 위해 당화액배지에 agar를 첨가하여 10 L, 50 L 공기순환배양기에서 배양한 결과, 50 L 규모로 scale-up한 반응기에서 5.64 g/L로 10 L 공기순환배양기 (5.84 g/L)에서와 동등한 BC 농도가 얻어져 BC 대량생산에 효과적인 배양방법임이 입증되었다. 50 L 규모로 확대된 공기순환배양기를 이용하는 본 배양 방법은 BC의 저비용 대량생산을 위한 효율적인 생산 방법 중 하나가 되리라 사료된다. 또한, 당화액을 배지로 하여 공기순환 방법으로 제조한 BC의 물성을 정치배양 방법으로 제조한 BC의 물성과 비교 검토한 결과, 공기순환법으로 제조하였을 때 중합도 및 역학적 성질이 약간 저하하였으나 결정성은 비슷하였으며, 배양기 용량의 대소에 따른 물성의 변화는 거의 없었다.
Optimal criterion for the scale-up production of schizophyllan, a fungal polysaccharide secreted by Schizophyllum commune, was investigated. For the production of the polysaccharide in a 150-l bioreactor, the culture conditions optimized in a 15-l bioreactor were applied to a 150-l bioreactor with scale-up process, by changing impeller speed and airflow rate. The optimized impeller speed in the 15-l bioreactor was 50 rpm in a technical medium based on barley. For establishment of the scale-up process, 3 kinds of criteria were used while the gas throughput number was kept constant, as follows; constant volume-related power input, constant tip speed of stirrer, and constant Reynolds number. In the 150-l bioreactor, the highest values for the maximum specific growth rate (1.17/day) and productivity (0.63 g/L${\cdot}$day) were achieved in the culture condition from constant volumerelated power input criterion.
Biodegradation of toluene in liquid effluent stream was carried out using biofilms of Pseudomonas putida formed on celite particles in the bubble column bioreactor. Silicon rubber tubing was installed at the bottom of the bioreactor and liquid toluene was circulated within the tubing. Toluene diffused out of the tube wall and was transferred into the culture broth where degradation by biofilms occurred. The operating variables affecting the formation of biofihns on celite particles were investigated in the bubble column bioreactor, and it was found that formation of bifilm is favored by high dilution rate and supply rate of carbon source which stimulate the growth of initially attached cells. Continuous biodegradation of toluene using biofilms was stablely conducted in the bioreactor for more than one month without any significant fluctuation, showing a removal efficiency higher than 95% at the toluene transfer rate of 1.2 g/L/h.

For the purpose of establishing an efficient propagation through airlift bioreactor system of Zingiber of officinale Rosc. Korean native Seosanjong, the effect of different factors and bioreactor on cultured plantlets were investigated. The highest number of plantlets, fresh weight per plant was obtained from explants when cultured in MS liquid medium including 0.3 mg/L NAA and 2.0 mg/L kinetin for 40 days. A 10 L bottle type bubble bioreactor, compared with 250 mL Erlenmeyer flask, was more efficient, producing 4.7 plantlets or from 1.5 to 1.6 times more than did the Erlenmeyer flask. The results demonstrate the rapid mass propagation of airlift bioreactor to produce normal ginger.

In this study, the relationship between morphology and viscosity of the main culture broth of Cephalosporium acremonium M25 was investigated in a 2.5 L bioreactor. The differentiation of C. acremonium M25 showed a complex pattern during the main culture. The morphological changes of C. acremonium M25 were related to the rheological properties of the culture broth and it was well agreed with the power law model. As a result of rheology study, it was found that rheological properties of the main culture broths of C. acremonium M25 in bioreactor were closely related to morphological changes. Also, fractal dimension fairly predicted morphological and rheological changes in the main culture broth.
공기부양 생물반응기에서 천연 청색 색소원인 쪽 (Poly,앵wm tinctorium) 세포를 대량 배양하기 위한 배양조건을 찾기 위해 영양성분 빛 색소 전 구체를 포함한 배지 성분의 생육에 미치는 영향 과 여러 배양조건에서 세포의 생육 특성에 대하 여 설험하였다. 최적 생육조건과 생육 특성을 규 명하기 위 해 external 1oop와 internal loop 두 가지의 arr-lift bioreactor 종류를 사용하여 14일간 배양하였다. 초기 접종량을 달리한 결과 각 reactor에서 공히 접종량이 0.5-2.5% packed cell volume 보다 많을수록 생육에 좋은 결과를 나타내였다. 초기 당농도의 영향을 살펴보기 위하여 2-4%로 그 조건을 달리하여 14일간 배양한 결과 4% 초기 당농도가 생육에 가장 좋은 조건임을 알 수 있었다. Sucrose는 대수기 초기단계에서 모두 소비되었다. Tryptophan(lmM)을 첨가한 경우 14일 배양 후에 internal loop reactor에서 3.8g/l, external loop reactorr에서 3.5g/l 의 cell mass를 얻었다. Indole을 첨가했을 때에는 세포 생육 저해효과 를 나타내였는데 external과 internal Ioop reactor에서 각각 2.5g/l와 2.2g/l를 얻었다. 무기질소원 으로는 potassium nitrate가 선발되었고, contrl이에 비해 110%d의 생육도 증가가 있였다. 최적 조건 하에셔의 14일간 internal loop bioreactor 배양에서 세포 건조 중량 16.34 g/l를 얻었다.
