The Practical Application of Aqueous Two-Phase Processes for the Recovery of Biological Products

  • Although the generic implementation of aqueous two-phase systems (ATPS) processes for the recovery of biological products has been exploited for several years, this has not resulted in a wide adoption of the technique. The main reasons involve the poor understanding of the mechanism governing phase formation and the behavior of solute partitioning in ATPS processes, the cost of phase forming polymers, and the necessary extended time to optimize the technique. In this review paper, some of the practical disadvantages attributed to ATPS are addressed. The practical approach exploited to design ATPS processes, the application to achieve process integration, the extended use for the recovery of high-value products, and the recent development of new low-cost ATPS, are discussed. It is proposed that the trend of the practical application of ATPS processes for the recovery of biological products will involve the purification of new high-value bioparticulate products with medical applications. Such a trend will give new impetus to the technique, and will draw attention from industries needing to develop new, and improve existing, commercial processes.

Optimization of compression ratio in closed-loop CO2 liquefaction process

  • We suggest a systematic method for obtaining the optimal compression ratio in the multi-stage closed-loop compression process of carbon dioxide. Instead of adopting the compression ratio of 3 to 4 by convention, we propose a novel approach based on mathematical analysis and simulation. The mathematical analysis prescribes that the geometric mean is a better initial value than the existing empirical value in identifying the optimal compression ratio. In addition, the optimization problem considers the initial installation cost as well as the energy required for the operation. We find that it is best to use the fifth stage in the general closed-loop type carbon dioxide multi-stage compression process.

  • 1. A-value in the soils studied in this experiment were found to be about 40-50kg/10a (as $P_2O_5$) 2. Percentage of utilized phosphorus in Soybeans was 10%. 3. The utilization and A-value of phosphorus were very different among the varieties of soybean plants, and the range was about 30%. 4. A/M-value (A means maximum value of total amount of available phosphorus and M means minimum value of absorbed phosphorus by plants from soils) indicated the critical range of available phosphorus to the crops.

The Estimation of Physical/Biological Parameters of Greenhouse Soil by Image Processing (컬러 영상처리에 의한 시설재배지 토양의 생물 물리적 환경변수 추정)

  • This study was conducted to find out the coefficient relationships between intensity values of image processing and biological/physical parameters of soil in greenhouses. Soil images were obtained by an image processing system consisting of a personal computer and a CCD earners. A software written in Visual C$\^$++/ systematically integrated the functions of image capture, image processing, and image analysis. Image processing data of the soil samples were analyzed by the method of regression analysis. The results are as follows. For detecting soil density of unbroken soil samples, the highest correlation coefficients of 0.82 and 0.84, respectively were obtained fur R-value and S-value among image processing data while it was 0.97 for G-value. Considering the relationship between biological characteristics and image processing data of soil in greenhouse, the correlation was found generally low. For pH of unbroken soil sample, the correlation coefficients were found 0.87, 0.85, and 0.94, respectively with G, I, and H values of image processing data. In the case of bacteria, any correlation was not found with the image processing data For Actinomyctes, they were 0.86 and 0.85, respectively with G-value and B-value of image processing data showing high correlation coefficient compared to the other variables. The correlation coefficient between Fungi and H-value was shown 0.88, the highest among the variables higher than 0.8 while the other variables showed low correlation. For broken soil samples from greenhouse, the relation between biological parameter and image processing data were rarely shown in this study. The results of this study indicated that most of correlation coefficient between the variables were usually lower than 0.01. Accordingly, it was assumed that the soil should be used without broken to fairly estimate biological characteristics using CCD camera.

Effects of Dietary Supplemental Phosphate from Different Sources on Performance of Young Broiler Chicks and It's Biological Availability (서로 다른 인 공급원들의 생물학적 이용율과 어린 육계의 능력에 미치는 영향)

  • An experiment was conducted with male broiler chicks to determine the effect of different dietary phosphorus sources and evaluate the biological availability of phosphorus sources. The biological availability of phosphorus from dicalcium phosphate (DCP) was used as a reference standard (100%) compared to defluorinate phosphate (DFP). DCP and DFP was supplemented to a corn-soy basal diet at levels of 0.05, 0.15, 0.25, 0.35%. Each of 24 pens of 10 male broiler chicks with three replications was used for three weeks. The results indicated that weight gain, feed in-take, feed efficiency, nonphytic phosphorus (NPP) and tibia ash were significantly different among treatments. Dietary supplemental phosphorus of DFP improved weight gain, NPP intake and feed efficiency consistently, whereas supplements of DCP did not show consistent increase. Regression equations was used for the availability of DPP compared with DCP when percent bone ash was a function of total phosphorus in the diet. The percent (%) bone ash of DFP groups compared to that of DCP groups showed a value of 59.98% as a slope ratio. DFP indicated lower biological availability compared to DCP, but it's dietary supplementation tended to increase bone ash and maximize the growth of young broiler chicks.

Evaluation of Biological Kinetic Parameters in the Granular Sludge (입상슬러지의 동력학적 인자 산정)

  • Design approach of upflow Anaerobic Sludge Blanket(UASB) process based on the biological kinetic parameters are known to be very difficult since the characteristics of the granular slut비e depends on the type of wastewater and size distribution of the granular sludge also depends on the upflow velocity in the UASB reactors. Furthermore, industrial wastewater containing toxic substances has been treated by UASB process without the clear knowledge of toxic effects on the granular slut형e. Hence, the present research was aimed on the intensive evaluation of biological kinetic parameters of the granular sludge in UASB reactor with and without toxic substance of 2, 4-dichlorophenol in order to present the basic design measures for UASB process design. The results could be summarized as follows. The biological kinetic parameters(k and Ks) considerably varied with the granular size of the sludge. Generally, 연e k and ks values of the granular sludge increased with the particle size of the granule. The biological kinetic parameters(k and Ks) of the granular sludge obtained from batch test were not applicable to design purpose of UASB process due to substrate diffusional limitation into the granular sludge in the completely mixed UASB reactors. The toxic effects on k and Ks greatly varied with the granular sixte. And as the toxicant concentration increased, the k value decreased while the Ks value increased. Inhibition constant(hi) for k with the toxicant of 2, 4-dichlorophenol varied from 0.5 to 2.3 depending on the granular sizes while the inhibition constant(Ki) for Ks varied from 20.7 to 80.1, showing the mixed inhibition.

  • This study gives the optimal reaction conditions, reaction mechanisms, reaction rates leaded from the oxidation of phenol by electron beam accelerator and ozone used for recent water treatment. It gives the new possibility of water treatment process to effectively manage industrial sewage containing toxic organic compounds and biological refractory materials. The high decomposition of phenol was observed at the low dose rate, but at this low dose rate, the reaction time was lengthened. So we must find out the optimal dose rate to promote high oxidation of reactants. The reason why the TOC value of aqueous solution wasn't decreased at the low dose was that there were a lot of low molecular organic acids as an intermediates such as formic acid or glyoxalic acid. In order to use both electron beam accelerator and biological treatment for high concentration refractory organic compounds, biological treatment is needed when low molecular organic compounds exist abundantly in sewage. In this experiment, the condition of making a lot of organic acids is from 5 kGy into 20 kGy dose. Decomposition rate of phenol by electron beam accelerator was first order reaction up to 300ppm phenol solution on the basic of TOC value and also showed first order reaction by using both air and ozone as an oxidants.

Optimization of Culture Medium for Lactosucrose ($^4G-{\beta}$-D-Galactosylsucrose) Production by Sterigmatomyces elviae Mutant Using Statistical Analysis

  • In this study, the optimization of culture medium using a Sterigmatomyces elviae mutant was investigated using statistical analysis to increase the cell mass and lactosucrose ($^4G-{\beta}$-D-galactosylsucrose) production. In basal medium, the cell mass and lactosucrose production were 4.12 g/l and 140.91 g/l, respectively. However, because of the low cell mass and lactosucrose production, optimization of culture medium was carried out to increase the cell mass and lactosucrose production. Culture media were optimized by the S. elviae mutant using analysis of variance (ANOVA) and response surface methodology (RSM). Central composite designs using RSM were utilized in this investigation. Quadratic models were obtained for cell mass and lactosucrose production. In the case of cell mass, optimal components of the medium were as follows: sucrose 1.13%, yeast extract 0.99%, bactopeptone 2.96%, and ammonium sulfate 0.40%. The predicted maximum value of cell mass was about 5.20 g/l and its experimental value was 5.08 g/l. In the case of lactosucrose production, optimal components of the medium were as follows: sucrose 0.96%, yeast extract 1.2%, bactopeptone 3.0%, and ammonium sulfate 0.48%. Then, the predicted maximum value of lactosucrose production was about 194.12 g/l and the corresponding experimental value was about 183.78 g/l. Therefore, by culturing using predicted conditions, the real cell mass and lactosucrose production increased to 23.3% and 30.42%, respectively.

Studies on the Amino Acid Composition of Korean Fermented Soybean Meju Products and the Evaluation of the Protein Quality (장류제품의 아미노산 조성과 그 단백질 품질평가에 관한 연구)

  • This study analyses and compares the amino acid composition and available lysine content between Korean fermented soybean Meju and its products as well as home-made and commercially made products. The protein quality of the products was evaluated by the result, and the biological value of the proteins was estimated by using the regression equation for chemical score to biological value as calculated by B.O. Eggum. The amino-N content of soybean is found to be 85% of the total nitrogen content and is reduced to approximately 75% in the fermented products except home-made soysauce, where as the content of ammonia-N and other N-compounds is increased. The difference in protein quality between home-made and commercially made products is not found to be significant. The protein quality of soybean is not damaged greatly in the making of Meju but is seriously damaged during the long periods of ripening. After the ripening the chemical score of the products' protein is reduced to approximately one half of the original soybean protein and the available lysine content to $1/3{\sim}1/2$.
