Chilli anthracnose (Colletotrichum spp.) disease and its management approach

  • Chilli is a widely consumed crop throughout the world. However, chilli anthracnose is a major constraint in chilli production leading to huge economic losses worldwide. Colletotrichum is a large genus of Ascomycete fungi, containing species that cause anthracnose diseases on a wide range of crops of economic value. This review is aimed at critically and accurately examining the taxonomic identification of Colletotrichum species by morphological and molecular approaches as well as assessing their management options. The use of appropriate integrated management practices, such as cultural, mechanical, chemical, and biological control, are important in chilli anthracnose disease prevention and control. Emphasis is laid on the use of biological control because it is cost effective and eco-friendly, and is an appropriate approach for disease management. The use of resistant cultivars is the cheapest, easiest, safest, and most effective means of controlling crop diseases. But, since no resistant cultivars of chilli have been developed and commercialized, it is very important to develop biological management strategies. Further studies leading to integrated disease management strategies need to be carried out.

Automatic Addition Control of the External Carbon Source by the Measurement of ORP in Biological Nitrogen Removal Process (생물학적 질소 제거공정에서 ORP 측정을 통한 외부탄소원의 자동 주입 제어)

  • For the cost-effective biological nitrogen removal (BNR) process whose characteristics of influent have low COD/N ratios, the automatic control system for the addition of external carbon based on oxidation-reduction potential (ORP) data in an anoxic reactor has been developed. In this study, it was carried out with a pilot-scale Bardenpho process which was consisted of anoxic 1, aerobic 1, aerobic 2, anoxic 2, aerobic 3 tank and clarifier. Firstly, the correlation coefficient ($R^2$) of the dosage of external carbon source and ORP value was about 0.97. Consequently, the automatic control system using ORP showed that the dosage of external carbon source was decreased by about 20% compared with a stable dosage of 75 mg/L based on the COD/N ratio of the anoxic influent.

Complete Genome Sequence of Myxococcus stipitatus KYC2006, a Myxobacterium That Affects the Growth of Photosynthetic Microorganisms

  • Here, we report the whole-genome sequence of Myxococcus stipitatus KYC2006, a bacterium whose conditioned media affect the growth of photosynthetic microorganisms such as cyanobacteria and microalgae. The genome of M. stipitatus KYC2006 was assembled into a 10,311,252 bp circular genome with 68.5% of GC content, containing 7,949 protein-coding genes, 12 rRNA genes, and 79 tRNA genes. Further analysis revealed that there are 29 secondary metabolite biosynthetic gene clusters in M. stipitatus KYC2006. These results suggest that M. stipitatus KYC2006 holds a significant potential as a resource for research on the development of biocontrol agents and value-added products from photosynthetic microorganisms.

Biological control of Lettuce Sclerotinia rot using Bacillus mojanvinensis Pro-EB 15 strain.

  • This studies were investigated the occurrence of Sclerotinia rot by Sclerotinia Sclerotiorum at the lettuce field in Uiryeong-Gun, Gyeongsangnam-Do and were isolated the most effective microorganism for the biological control to the pathogen, S. sclerotiorum YR-1 strain from diseased soil and lettuce leaves. For the pathogenicity test, the most suitable inoculmn density of YR-1 strain was selected as the mycelial suspension of 40m1 showing disease incidence of 80%, and the symptom showed as same as at the fields, the leaves and stem had rotten and developed white downy mycelial at the diseased lesion on the leaves and stems, and produced black and irregular sclerotinia. On the PDA dual test, about 300 isolates were examined the antifungal activity to the pathogen, YR-1 strain, and among them, A-2, A-7, and RH-4 strain were selected most effective antagonistic bacteria. At pots test, the control value of A-7 strain showed the highest value as 85% which was more effective than that of others in a growth chamber. For the promotion of control effect, the selected 3 isolates were spayed on the lettuce leaves as a sole and/or mixed treatments in a growth chamber, the mixed treatment of A-7 and RH-4 strain showing the control value of 90% was most effective than that of sole treatment with A-7 or RH-4 strain showing the control value of 80%, respectively and mixed treatment with A-2 and A-7 strain and A-2, A-7 and RH-4 strain. In addition, 3 bacteria re-isolated from diseased soils, and all of the selected 6 isolates investigated the control effect at pots in a growth chamber, According to the results, A-7 and Pro-EB 15 strain showed the control value of 91.0% and 90.1% respectively, and they were selected most effectual antagonistic bacteria to control lettuce sclerotinia rot and identified as the Bacillus mojanuinensis by 16s RNA analysis. This is the first report on the biological control using by B. mojanvinensis to the lettuce Sclerotinia rot.

Combined Effects of High Pressure and Heat on Shear Value and Histological Characteristics of Bovine Skeletal Muscle

  • Changes in shear force value, transverse sections, myofibrils and intramuscular connective tissue of bovine skeletal muscle exposed to the combination of high-pressure up to 400 MPa and heat (30 and $60^{\circ}C$) were studied. The shear force value decreased by pressure-heat treatment up to 200 MPa at 30 and $60^{\circ}C$, and then slightly increased over 200 MPa at $30^{\circ}C$. Shear force values of treated muscles were lower than those of untreated ones. Gaps between muscle fibers in the untreated muscle were a little clear, and then they became very clear in the treated muscles up to 200 MPa at 30 and $60^{\circ}C$. However, the gaps reduced significantly over 200 MPa at $30^{\circ}C$. The remarkable rupture of I-band and loss of M-line materials progressed in the myofibrils with increasing pressure applied. However, degradation and loss of the Z-line in myofibrils observed in the muscle treated at $60^{\circ}C$ was not apparent in the muscle treated at $30^{\circ}C$. The length of the sarcomere initially contracted by pressure-heat treatment of 100 MPa at $30^{\circ}C$ seemed to have recovered with increase of the pressure up to 400 MPa. In the muscle treated at $60^{\circ}C$, the length of sarcomere gradually decreased with increase of the pressure up to 400 MPa. In the treated muscles, changes in the honeycomb-like structure of endomysium were observed and accelerated with increase of the pressure. A wavy appearance clearly observed at the inside surface of endomysium in the untreated muscles gradually decreased in the treated muscles with increase of the pressure. Tearing of the membrane was observed in the muscles treated over 150 MPa at $30^{\circ}C$, as observed in the sample pressurized at 100 MPa at $60^{\circ}C$. The roughening, disruption and fraying of the membrane were observed over 200 MPa at $60^{\circ}C$. From the results obtained, the combination of high-pressure and heat treatments seems to be effective to tenderize tough meat. The shear force value may have some relationship with deformation of intramuscular connective tissue and myofibrils.

The Study of the Integrated Technology of the Dual Fluidized-bed Gasification for Producing Medium Heating Value Syngas (중발열량 합성가스 생산을 위한 일체형 이중유동층 가스화 기술 연구)

  • A 5 kg/hr scale integral dual fluidized-bed gasifier for producing medium heating value syngas from biomass or combustible wastes was manufactured. The effect of operating variables including gasification temperature, rate of feeding, and weight ratio of steam/feed on the behavior of the gasifier was investigated. The contents of $H_2$ and CO in syngas, flow rate of feeding, cold gas efficiency increased with the increased gasification temperature or rate of feeding, but decreased with the increased weight ratio of steam/feed within the experimental range. With wood powder as the feed, the concentrations of $H_2$ and CO in the syngas were as high as 41% and 32%, and the cold gas efficiency and lower heating value of the syngas were as high as 70.1% and $3,428kcal/Nm^3$. With food wastes as the feed, the concentrations of $H_2$ and CO in the syngas were as high as 37% and 23.9%, and the cold gas efficiency and lower heating value of the syngas were as high as 66.7% and $3,670kcal/Nm^3$.

Assessment of absorption ability of air pollutant on forest in Gongju-city

  • Background: Some researchers have attempted to evaluate the ecological function of various additional services, away from the main point of view on the timber production of Korean forests. However, basic data, evaluation models, or studies on the absorption of air pollutants related to major plant communities in Korea are very rare. Therefore, we evaluated the functional value of the forest ecosystem in Gongju-city. Plantation manual for air purification, supplied from the Ministry of Environment in Japan, was referred to process and method for assessment of air pollutant absorption. Results: Gross primary production was calculated about average 18.2 t/ha/year. It was a relatively low value in forests mixed with deciduous broad and evergreen coniferous compared to pure coniferous forest. Net primary production was the highest value in deciduous coniferous and was the lowest value in mixed forest with deciduous broad and evergreen broad. And the mean sequestration amount of each air pollutant per unit area per year assessed from gross primary production and concentration of gas was the highest with 75.81 kg/ha/year in $O_3$ and was 16.87 and 6.04 kg/ha/year in $NO_2$ and $SO_2$, respectively. In addition, total amounts of $CO_2$ absorption and $O_2$ production were 716,045 t $CO_2$/year and 520,760 t $O_2$/year in all forest vegetation in Gongju-city. Conclusions: In this study, we evaluated the absorption ability of air pollutant in 2014 on forest in Gongju-city area. Gongju-city has the broad mountain area about 70.3%, and area of deciduous broad leaves forest was established the broadest with 47.4% of genus Quercus. Pg was calculated about average 18.2 t/ha/year. The mean sequestration amount of each air pollutant per unit area per year assessed from Pg and $C_{gas}$ was the highest with 75.81 kg/ha/year in $O_3$ and were 16.87 and 6.04 kg/ha/year in $NO_2$ and $SO_2$, respectively. Absorption rates of $O_3$, $NO_2$, and $SO_2$ were the highest in evergreen coniferous forest about $14.87kgO_3/ha/year$, $3.30kgNO_2/ha/year$, $1.18kgSO_2/ha/year$, and the lowest were $5.95kgO_3/ha/year$, $1.32kgNO_2/ha/year$, and $0.47kgSO_2/ha/year$ in deciduous broad forest. In conclusion, it was evaluated that Japanese model is suitable for estimating air pollutants in Japan to Korean vegetation. However, in Korea, there is a very limited basic data needed to assess the ability of forests to absorption of air pollutants. In this study, the accuracy of a calculated value is not high because the basic data of trees with similar life form are used in evaluation.

Role of oxygen in plasma induced chemical reactions in solution

  • Many researchers have paid attention to the studies on the interaction between non-thermal plasma and aqueous solutions for biomedical applications. The gas composition in the plasma is very important. Oxygen and nitrogen are the main gases of interest in biological applications. Especially, we focus on the oxygen concentration. In this experiment, we studied the role of oxygen concentration in plasma induced chemical reactions in solution. At first, the amount of ions are measured according to changing the oxygen concentration. And we checked the relationship between these ions and pH value. Secondly, when the oxygen concentration is changed, it identified the type and amount of radical generated by the plasma. In order to confirm the effect of these chemical property change to biological material, hemoglobin and RBCs are chosen. RBCs are one of the common basic biological cells. Thirdly, when plasma treated according to oxygen concentration in nitrogen feeding gas, oxidation of hemoglobin and RBC is checked. Finally, membrane oxidation of RBC is measured to examine the relation between hemoglobin oxidation and membrane damage through relative hemolysis and Young's modulus. Our results suggest that reactive species generated by the plasma differsdepending on the oxygen concentration changes. The pH values are decreased when oxygen concentration increased. OH decrease and NO increase are also observed. These reactive species makes change of chemical properties of solution. We also able to confirm that the difference in these reactive species to affect the oxidation of the Hb and RBCs. The Hb and RBCs are more oxidized with the high oxygen concentration conditions. But membrane is damaged more by plasma treatment with only nitrogen gas. It is shown that red blood cells membrane damage and oxidation of hemoglobin are not directly related.

Biological Control of Bacterial Fruit Blotch of Watermelon Pathogen (Acidovorax citrulli) with Rhizosphere Associated Bacteria

  • Bacterial fruit blotch (BFB), which is caused by Acidovorax citrulli, is a serious threat to watermelon growers around the world. The present study was conducted to screen effective rhizobacterial isolates against 35 different A. citrulli isolates and determine their efficacy on BFB and growth parameters of watermelon. Two rhizobacterial isolates viz. Paenibacillus polymyxa (SN-22), Sinomonas atrocyanea (NSB-27) showed high inhibitory activity in the preliminary screening and were further evaluated for their effect on BFB and growth parameters of three different watermelon varieties under greenhouse conditions. The greenhouse experiment result revealed that SN-22 and NSB-27 significantly reduced BFB and had significant stimulatory effect on total chlorophyll content, plant height, total fresh weight and total dry weight compared to uninoculated plants across the tested three watermelon varieties. Analysis of the 16S ribosomal RNA (rRNA) sequences revealed that strains SN-22 belong to P. polymyxa and NSB-27 to S. atrocyanea with the bootstrap value of 99% and 98%, respectively. The isolates SN-22 and NSB-27 were tested for antagonistic and PGP traits. The result showed that the tested isolates produced siderophore, hydrolytic enzymes (protease and cellulose), chitinase, starch hydrolytic enzymes and they showed phosphate as well as zinc solubilizing capacity. This is the first report of P. polymyxa (SN-22) and S. atrocyanea (NSB-27) as biocontrol-plant growth promoting rhizobacteria on watermelon.

Understanding and Knowledge of the Eye in Biological Science Education of Middle and High School Students in Busan (부산광역시 중고등학생의 눈에 대한 생물교육학적 이해도와 지식도)

  • Purpose: This study was to investigate and analyze understanding and knowledge of the eye in biological science education for middle and high school students in Busan. Methods: A total of 821 middle and high school students from four different schools participated by self-administered questionnaire test. The results were compared and analyzed by grade and gender. Results: In the survey of the understanding about the eye in the biological science curriculum, the results indicated that the more learning of the eye in biological science lecture led to the more understanding of the role of the eye. The difference of understanding the eye between learned and unlearned students in biological sciences lecture was significant (p<0.01). In the survey of the knowledge of the eye, students did not know about myopia and astigmatism (average value <50). Conclusions: As biological science education is one of the major factors to understand the role of the eye, this result suggests that more information about the eye in the curriculum of biology is included.
