염색폐수의 수질독성시험을 이용한 한국의 수질배출허용기준 평가연구 (Assessment of Korean Water Quality Standards for Effluent Discharged from the Dye Industry Based on Acute Aquatic Toxicity Tests Using Microbes and Macroinvertebrates)

  • Acute aquatic toxicity of effluents discharged from five dyeing plants in Gyeong-gi province were evaluated to assess whether the current Korean water quality standards(KWQS) could protect aquatic life. Chemical analyses of all parameters regulated under KWQS, except for E-coli, were also carried out to determine regulation compliance of the samples. All the effluent samples were satisfied with KWQS except for the color in only one sample. In acute Daphnia magna toxicity tests, significant mortality was observed in one of five samples and EC50 was 12.1%(95% confidence interval 9.1-16.2), which was in compliance with KWQS. The result of the Microtox assay indicated that acute microbial toxicity existed in effluents from three out of five plants, two of which were in compliance with KWQS. The agreement between regulation compliance of chemical concentrations of effluent and observed toxicity from various biological toxicity tests was very poor to fair (kappa = 0.194~0.250). The data presented suggest that exposure to dyeing wastewater which were in compliance with Korean water quality standards may not be safe to aquatic biota, and multiple tropical levels should be considered in aquatic toxicity monitoring of dyeing industry.

  • Toxicity identification and quantification are important factors to evaluate the effect of industrial effluent on the aquatic environment. In order to measure the potential and real toxicity of mixed chemicals in the effluents, the biological method (i.e., WET test) should be used as well as chemical analysis method. In this study, we conducted WET test for various kinds of industrial effluents using aquatic organisms such as water flea (Daphnia magna), algae (Pseudokirchneriella subcapitata), fish (Oryzias latipes, Danio rerio), and microorganism (Vibrio fisheri). In addition, we carried out chemical analysis and TIE (Toxicity Identification Evaluation) for effluents in order to identify the substances causing toxicity. Among the 30 kinds of wastewater, S13 showed the highest eco-toxicity and $Ca^{2+}$ and $Cl^-$ ion were suspected as major compounds causing toxicity for aquatic organisms. In order to confirm these suspected compounds, various confirmation procedures need to be carried out.

  • Stink bug, Canthecona furcellatta (Hemiptera: Pentatomidae), is an important predator of silkworm larvae. Nymphs and adult attack the early stage silkworm larvae and causes about 10-15 per cent loss to silk industry. Synthetic organic pesticides has tremendous impact on minimizing the pest population but repeated and frequent use has created problems of residual toxicity, development of resistance to insecticides, pest resurgence and out break, phyto-toxicity and hazards to non target species and beneficial organism. Silkworms are very sensitive to pesticides; therefore, attempt has made to control the bug population through introduction of its native natural enemies in the silkworm-rearing field. Biological control has tremendous scope in sericulture because it is eco-friendly in nature and non-harmful farmers. Native natural enemies have been screened. Psix striaticeps, Trissolcus spp. and Telenomus spp. have been recorded as the most potential parasitoid against pentatomid bug. Life cycle, sex ratio and other various attributes of the par-asitoids have been recorded. The parasitization potential of the parasitoid is very high and they have the ability to discriminate between parasitized and unpar-asitized host. Mass propopagation technique under laboratory condition has been standaydized.

수생 지렁이를 이용한 생태 독성 평가 연구에 대한 고찰 (Ecotoxicological Studies Using Aquatic Oligochaetes: Review)

  • Oligochaetes distribute widely in freshwater ecosystem, and some species are used as bioindicators for water quality assessment because they are tolerant to organic enrichment. They are acknowledged for potential for environmental health recovery of organic polluted environment. There are a lot of studies on ecology and toxicity assessment using oligochaetes in aquatic environment. In this study, we reviewed literature on ecotoxicology of aquatic oligochaetes. We searched literature from a database 'google scholar' by using keywords such as aquatic, oligochaete, and toxicity. The literature were summarized according to publication years, species, test methods, and chemicals. We obtained 133 articles published from 1953 to 2015 from the database. Among them, 58 papers(43.6% of total) have been published in 1990s. Three species(Lumnbriculus variegatus, Tubifex tubifex, and Limnodrilus hoffmeisteri) have been used most frequently in the study. Different species displayed different toxicological responses to different toxic chemicals. The results on the ecotoxicological study with aquatic oligochaetes revealed the possibility of the development for early warning system using aquatic oligochaetes to monitor aquatic ecosystem disturbance.

Acute Oral Toxicity of Salicornia herbacea L. Extract in Mice

  • Salicornia herbacea L. (S. herbacea) is an annual herbaceous plant of Chenopodiaceae. It grows in groups on the coast or mud flat of Korea is known to be rich in minerals. S. herbacea has potent anti-cancer, antioxidant, anti-obesity, bowel function improvement. However, pharmacological mechanisms of S. herbacea extract (SHE) remain poorly understood. The aim of this study was to investigate the potential acute toxicity of SHE in ICR mice administered a single oral dose of 0, 500, 1,000, and 2,000 mg/kg by gavage. After administration of the extract, signs of toxicity were observed every day for 14 days. No mortality, abnormal clinical signs, body weight, organ weight or pathological changes were observed compared to a control group, and there were no differences in the body weights of the control and treatment groups. Biological serum activities and histological tests were not significantly changed in the treatment group compared to the control group. Especially, treatment of SHE was significantly decreased of total cholesterol and triglyceride levels. These results indicated that a single oral administration of SHE does not exerts any toxic effects at a dose of 2,000 mg/kg and that the $LD_{50}$ of SHE is greater than 2,000 mg/kg. Accordingly, SHE appears to have potential in various functional agents of foods, without toxicity.

  • In 1990, Tolerable Daily Intake (TDI) of 10 pg TCDD/kg/day for dioxins based on carcinogenicity and reproductive toxicity was determined by WHO/EURO, that resulted in the establishment of TDIs in other countries. In Japan, Ministry of Health and Welfare and Environment Agency, respectively established the TDI of 10 pg TCDD/kg/day and Health Risk Assessment Index of 5 pg TCDD/kg/day in 1996. Accumulation of new scientific data, especially by molecular toxicology since 1990, resulted in the reevaluation of TDI by WHO-ECEH and IPCS in May, 1998. At this meeting, it was stressed that \circled1 toxic effects of dioxin is mediated through Ah-receptor in both animals and humans, \circled2 use of ebody burdeni concept is better than the use of traditional NOAEL/UF approach, \circled3 inclusion of coplanar PCBs in the TDI by the use of new WHO-TEF. LOAELs (0.16~200 ng TCDD/kg/day) obtained from reproductive toxicity and immunotoxicity in rats, and neurobehavioral toxicity and induction of endometriosis in rhesus monkeys are calculated to be the body burden of 10~50 ng TCDD/kg that is 14~37 pg TEQ/kg/day as human daily intake. Finally TDI of 1~4 pg TEQ/kg/day was established by applying the UF of 10. In Japan, reproductive toxicity and immunotoxicity in rats were used to obtain LOAELs (100~200 ng TCDD/kg/day). Finally TDI of 4 pg TEQ/kg/day was established in June 1999 by applying the UF of 10 to human daily intake of 43.6 pg TEQ/kg/day which corresponds to the body burden of 86 ng TCDD/kg.

In vitro cytotoxicity and in vivo acute toxicity of selected polysaccharide hydrogels as pharmaceutical excipients

  • Polysaccharide hydrogels constitute a structurally diverse class of biological macromolecules with a wide range of physicochemical properties. They also constitute important members of the family of industrial water-soluble polymers. They find application in Pharmacy as binders, disintegrants, suspending, emulsifying and sustaining agents. According to the International Pharmaceutical Excipients Council (IPEC), an excipient must have an established safety profile. Hence, in the present study, in vitro cytotoxicity on Vero and HEp-2 cell lines, and in vivo acute toxicity in rats were carried out to establish the safety of polysaccharide hydrogels from the seeds of Plantago ovata and Ocimum basilicum. The in vitro cytotoxicity was determined by MTT and SRB assays. In the in vivo acute toxicity, the effects of three different doses of hydrogels (100, 200 and 400 mg/kg body weight) on food and water intake, body weight, biochemical and hematological parameters were studied. The results of in vitro did not show any cytotoxicity on both the cell lines used. In the in vivo acute toxicity, the hydrogels did not show any toxic symptoms in all three dose levels. This establishes the safety of the selected hydrogels. Hence, they can be used as excipients in pharmaceutical dosage forms.

Identification and toxigenic potential of a Nostoc sp.

  • Cyanobacteria are well known for their production of a multitude of highly toxic and / or allelopathic compounds. Among the photosynthetic microorganisms, cyanobacteria, belonging to the genus Nostoc are regarded as good candidate for producing biologically active secondary metabolites which are highly toxic to humans and other animals. Since so many reports have been published on the poisoning of different animals from drinking water contaminated with cyanobacteria toxins, it might be assumed that bioactive compounds are found only in aquatic species causes toxicity. However, the discovery of several dead dogs, mice, ducks, and fish around paddy fields, prompted us to study the toxic compounds in a strain of Nostoc which is most abundant in the paddy fields of Iran, using polymerase chain reaction and liquid chromatography coupled with a diode array detector and mass spectrophotometer. Results of molecular analysis demonstrated that the ASN_M strain contains the nosF gene. Also, the result of ion chromatograms and $MS^2$ fragmentation patterns showed that while there were three different peptidic compound classes (anabaenopeptin, cryptophycin, and nostocyclopeptides), there were no signs of the presence of anatoxin-a, homoanatoxin-a, hassallidin or microcystins. Moreover, a remarkable antifungal activity was identified in the methanolic extracts. Based on the results, this study suggests that three diverse groups of potentially bioactive compounds might account for the death of these animals. This case is the first documented incident of toxicity from aquatic cyanobacteria related intoxication in dogs, mice, and aquatic organisms in Iran.

Syntheses of Biologically Non-Toxic ZnS:Mn Nanocrystals by Surface Capping with O-(2-aminoethyl)polyethylene Glycol and O-(2-carboxyethyl)polyethylene Glycol Molecules

  • Water-dispersible ZnS:Mn nanocrystals were synthesized by capping the surface of the nanocrystal with O-(2-Aminoethyl)polyethylene glycol (PEG-$NH_2$, Mw = 10,000 g/mol) and O-(2-Carboxyethyl)polyethylene glycol (PEG-COOH, Mw = 10,000 g/mol) molecules. The modified PEG capped ZnS:Mn nanocrystal powders were thoroughly characterized by XRD, HR-TEM, EDXS, ICP-AES and FT-IR spectroscopy. The optical properties were also measured by UV/Vis and photoluminescence (PL) spectroscopies. The PL spectra showed broad emission peaks at 600 nm with similar PL efficiencies of 7.68% (ZnS:Mn-PEG-NH2) and 9.18% (ZnS:Mn-PEG-COOH) respectively. The measured average particle sizes for the modified PEG capped ZnS:Mn nanocrystals by HR-TEM images were 5.6 nm (ZnS:Mn-PEG-NH2) and 6.4 nm (ZnS:Mn-PEG-COOH), which were also supported by Debye-Scherrer calculations. In addition, biological toxicity effects of the nanocrystals over the growth of wild type E. coli were investigated. They showed no biological toxicity to E. coli until very high concentration dosage of 1 mg/mL of the both nanocrystal samples.
