HPTLC와 GC/MS를 이용한 의약품의 데이타베이스화 및 생체시료에의 응용 (Constructing Database for Drugs and its Application to Biological Sample by HPTLC and GC/MS)

  • 생체시료에서 미지 의약품의 검출을 위하여, 민감하고 선택적인 박층크로마토그래피인 자동화 점적기 및 자외선 스펙트럼 검출기를 장착한 고성능박층크로마토그라피(HPTLC)를 시도하였다. 205종의 의약품에 대한 Rf값과 자외선흡수스펙트럼(scan 200-360 nm)을 이용하여 HPTLC 데이터베이스를 구축하였으며, 또한 96종의 의약품에 대하여는 리도카인에 대한 상대적 머무름시간(relative retention time; RRT) 및 질량스펙트럼을 이용하여 GC/MS 데이터베이스를 구축하였다. 생체시료중 의약품성분은 고상추출법(Clean Screen ZSDAU020)으로 추출한 후, 이들 시료들을 HPTLC 및 GC/MS 데이터베이스에 적용시켜 검색한 결과 90% 이상의 매우 양호한 일치율로 검색할 수 있었으며, 이들 의약품들을 GC/MS로 확인하였다. 결론적으로, 우리가 구축한 HPTLC 및 GC/MS 데이터베이스체계는 생체시료에서 미지의 의약품성분을 검색하는데 매우 유용한 방법으로 사료된다.

Double clad fiber probe for fluorescence spectroscopy

  • We report a probe using a single double clad fiber (DCF) for fluorescence spectroscopy. Bidirectional separate transmission for excitation and fluorescence light in a single fiber was implemented. A DCF coupler made by side-polished method could extract none but the collected fluorescence signals propagating in inner cladding mode, thereby diminishing the interference of silica background generated by the excitation in core mode. The experimental results show that the fluorescence spectra of biological tissues obtained using the DCF probes have much less silica background than using a standard multiple-mode fiber.

  • A series of copper complexes of some amino acid derivatives of 6-methoxy 3-methyl-coumarilic acid were prepared. The infrared, visible spectra and magnetic susceptibility of these compounds were reported. All copper complexes were foundt to have antimicrobial activity against gram-positive bacteria only.

Isolation and Partial Characterization of Two Ferredoxins from the Photosynthetic Bacterium Heliobacillus mobilis

  • Two ferredoxin (Fd) fractions, namely, Fd-A and Fd-B were isolated from Heliobacillus mobilis cells, and purified by ammonium sulfate fractionation, DEAE, gel-permeation and Phenyl-Superose column chromatographies under anaerobic conditions. Their absorption spectra were typical of 2[4Fe-4S] cluster type Fds with peaks at about 385 and 280 nm and a shoulder at about 305 nm. Their N-terminal amino acid sequences were determined, which showed that both of them contain a [4Fe-4S] cluster binding motif. Fd-B was sensitive to oxygen, and itsA$_{385}$ value decreased by about 50% in 2 h at 4$^{\circ}C$ under aerobic conditions. In contrast, $A_{385}$ of Fd-A was essentially unchanged up to 24 h under the same conditions.

  • An alkaloid, 3-methylcanthin-5, 6-dione, was isolated from the stem of Picrasma quassioides (D. Don) Benn. and characterized by comprehensive analyses of its 1D and 2D NMR spectra. It was also evaluated for its cytotoxic activity in vitro against three human cancer cell lines (MDA-MB-231, HT-29 and NCI-N87), using MTT assays. We found that 3-methylcanthin-5, 6-dione exhibited significant anti-inflammatory activity via inhibiting NO production induced in LPS-stimulated murine macrophage RAW264.7 cells. The antioxidant activity of 3-methylcanthin-5, 6-dione was measured by DPPH free radical scavenging assays, hydroxyl radical scavenging assays and reducing power assays. Our results showed that 3-methylcanthin-5, 6-dione has significant biological activities.

The release of cellular constituents of Lactobacillus lactis by freezing and freeze-drying

  • Lactobacillus lactis cell was markedly damaged when shocked by freezing and freeze-drying. The supernatant of shocked cells in 1% phosphate buffer (pH7.3) showed a maximum u.v. absorption spectra at 260nm after further incubation for 90min at $37-^{\circ}C.$ The leakage of cellular constituents by shocking could be prevented by dimethyl sulfoxide, glycerol, dextrose and $MgCl_2.$ Among them, $MgCl_2$ showed the preventing effect before and after freezing wh-ereas the rests, only before the treatment. This leakage was proved not to be related to the cellular repairing mechanism.

  • Compositions of methionine-metal chelates have been investigated by FT-IR and XRD studies to elucidate their molecular structures. It was concluded that Copamin and Zincamin contain a high percentage of crystalline products, presumably 2:1 Methionine-Cu or Zn complexes. On the contrary, FT-IR and XRD spectra of Ferramin didn't show any characteristics of the chelate and it was concluded to contain major components of starting $FeSO_4$ and methionine without chelation.

Conformation and Biological Activity of Mastoparan B and Its Analogs I

  • The mode of action of mastoparan B, an antimicrobial cationic tetradecapeptide amide isolated from the hornet Vespa basalis, toward phospholipid bilayers was studied with synthetic mastoparan B and its analogs with individual Ala instead of hydrophobic amino acids (1-Ile, 3-Leu, 6-Leu, 7-Val, 9-Trp, 13-Val, 14-Leu) in mastoparan B. Mastoparan B and its analogs were synthesized by the solid-phase method. Circular dichroism spectra showed that mastoparan B and its analogs adopted an unordered structure in buffer solution. In the presence of neutral and acidic liposomes, most of the peptides took an α-helical structure. The calcein leakage experiment indicated that mastoparan B interacted strongly with neutral and acidic lipid bilayers than its analogs. Mastoparan B also showed a more or less highly antimicrobial activity and hemolytic activity for human erythrocytes than its analogs. These results indicate that the hydrophobic face in the amphipathic α-helix of mastoparan B critically affect biological activity and helical contents.

MEASUREMENT OF PESTICIDES RESIDUES USING SPECTROSCOPY ON AGRICULTURAL PRODUCTS

  • A new spectroscopic method for pesticide residues detection on agricultural products was developed. The general determination methods are high performance liquid chromatography (HPLC), gas chromatography (GC) or GC-mass spectrometry. They have provided relatively good detection limit and accuracy with complicated and time-consuming (5hrs above) procedures. In addition freshness is very important for evaluating qualities of agricultural products. This requires a simple and fast method for detection of pesticides. Reflectance, transmittance and fluorescence spectrometry of pesticides were tested using UV range because most of pesticides contain conjugation band in the molecular structures. Fluorescence spectrometry showed better sensitive to detect pesticide residues than did reflectance and transmittance spectrometry. Intensity and shape of fluorescence spectra showed different patterns with different structures of pesticides. Detection limit for fluorescence spectrometry was 0.1 ppm to 10 ppm depending on the structures of pesticides. Application of fluorescence spectrometry appears to be an easy method for detection of pesticide residues on agricultural products.

InhA-Like Protease Secreted by Bacillus sp. S17110 Inhabited in Turban Shell

  • A strain producing a potent protease was isolated from turban shell. The strain was identified as Bacillus sp. S17110 based on phylogenetic analysis. The enzyme was purified from culture supernatant of Bacillus sp. S17110 to homogeneity by ammonium sulfate precipitation, SP-Sepharose, and DEAE-Sepharose anion exchange chromatography. Protease activity of the purified protein against casein was found to be stable at pH 7 to pH 10 and around $50^{\circ}C$. Approximately 70% of proteolytic activity of the enzyme was detected either in the presence of 100 mM SDS or Tween 20. The enzyme activity was enhanced in the presence of $Ca^{2+},\;Zn^{2+},\;Mg^{2+}$, but was inhibited by EDTA, indicating that it requires metal for its activity. The purified enzyme was found to be a monomeric protein with a molecular mass of 75 kDa, as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and gel filtration chromatography. The purified enzyme was analyzed through peptide fingerprint mass spectra generated from matrix-assisted laser desorption ionization-time-of-flight-mass spectrometry (MALDI-TOF-MS) and a BLAST search, and identified as immune inhibitor A (inhA) deduced from nucleotide sequence of B. cereus G9241. Since InhA was identified as protease that cleave antibacterial proteins found in insect, inhA-like protease purified from Bacillus sp. S17110 might be pathogenic to sea invertebrates.
