Synthesis and Biological Activity of Benzoxazole Containing Thiazolidinedione Derivatives

  • The peroxisome proliferator-activated receptors (PPARs) are a primary regulator of lipid metabolism. Potency for activation of PPAR$\gamma$, one of a subfamily of PPARs, particularly mirrors glucose lowering activity. We prepared thiazolidinediones featuring benzoxazole moiety for subtype selective PPAR$\gamma$ activators. 5-[4-[2-(Benzoxazol-2-yl-alkylamino)ethoxy]benzyl]thiazolidine-2,4-diones have been prepared by Mitsunobu reaction of benzoxazolylalkylaminoethanol 8 and hydroxybenzylthiazolidinedione 6 and their activities were evaluated. Most compounds tested were identified as potent PPAR$\gamma$ agonists.

  • Ginsenoside, major components of ginseng have been reported to show various biological activities including an increase of cholesterol metabolism. stimulation of serum protein synthesis, immunomodulatory effects. To explain these pharmacological actions, it is thought that ginseng saponins should be metabolized by human intestinal bacteria after they are orally administered. (omitted)

  • Carbamates are generally used as insecticide, thus 8-hydroxy-5-chloroquinolinyl-N-ethylcarbamate was newly synthesized. Its physical properties were determined and chemical structure was identified by means of IR, nmr in addition to elemental analysis. The effect of the compound on rat's ileum, and antibacterial activity against Staphylococcus aureus, Salmonella typhi, E. coli, and Pseudomonas aeruginosa were examined. It was observed that the dosage over 100 $\mu$g/ml of the compound relaxed rat's ileum and the same dosage of the compound inhibited growth of the above strains of bacteria.

  • The roots of Scutellaria baicalensis Georgi have been used in the chinese medicine for the treatment of inflammation, fever and headache. Recent studies had shown that baicalin and baicalein showed a detoxifying effect and could inhibit peripheral capillary permeability, and Wogonin was found to inhibit the proliferation of tumor cells. Wogonin has been synthesized as shown in schem. Compound 2 was prepared from 2,6-dimethoxy-1,4-benzoquinone(1) followed by acylation to give compound 3. The base treatment of 3 gave cyclized product which was carried by deprotection of methyl group chemoselectively. Overall step included relatively simple ten steps.
