Tripterygium wilfordii (Celastraceae): A new generic and species record for the flora of Vietnam

  • Tripterygium wilfordii Hook.f. (Celastraceae), collected from Phia Oac-Phia Den National Park of Vietnam, is reported here as a new generic and species record for the flora of Vietnam. The genus Tripterygium differs from other genera of Celastraceae by its samara fruits, which become three-winged at maturity, and a scandent shrub. Taxonomic notes, descriptions, line drawing and photographs are provided, together with short notes on the distribution, ecology and phenology of Tripterygium wilfordii Hook.f. (Celastraceae).

Hepatoprotective Effects of the Extracts of Alnus japonica Leaf on Alcohol-Induced Liver Damage in HepG2/2E1 Cells (알코올로 유도된 간손상 모델 HepG2/2E1 세포에서 오리나무 잎 추출물의 간보호효과)

  • Alcoholic liver disease (ALD) is a significant risk factor in the global disease burden. The stem bark of the Betulaceae plant Alnus japonica, which is indigenous to Korea, has been used as a popular folk medicine for hepatitis and cancer. However, the preventive effect of Alnus japonica leaf extracts on alcohol-related liver damage has not been investigated. The objective of this study was to investigate the hepatoprotective effects of the extracts of Alnus japonica leaf (AJL) against ethanol-induced liver damage in HepG2/2E1 cells. Treatment with AJL significantly prevented ethanol-induced cytotoxicity in HepG2/2E1 cells by reducing the levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). This protective effect was likely associated with antioxidant potential of AJL, as evidenced by the attenuation of reactive oxygen species (ROS) and malondialdehyde (MDA) production and restoration of the depleted glutathione (GSH) levels in ethanol-induced HepG2/2E1 cells. Our findings suggest that FCC might be considered as a useful agent in the prevention of liver damage induced by oxidative stress by increasing the antioxidant defense mechanism.

Single Dose Oral Toxicity Test of Water Extracts of Stachys sieboldii and Acorus gramineus, and their Mixture in ICR Mice (ICR 마우스를 이용한 초석잠, 석창포 단독추출물 및 복합추출물의 단회경구투여 독성시험)

  • Stachys sieboldii Miq. (SSM) and Acorus gramineus Soland. (AGS) have been used as traditional medicines for thousands of years in parts of Asia, including Korea, China, and Japan. Recent researches on SSM and AGS have documented a wide spectrum of therapeutic properties, including anti-inflammatory, anti-oxidative, neurodegenerative disease effects. However, the toxicity and safety of SSM and AGS, and their mixture (medicinal herber mixture, MHMIX) were not confirmed. Therefore, this study was performed to evaluate the acute toxicity and safety of SSM, AGS and MHMIX. SSM, AGS and MHMIX were orally administered at a dose of 5,000 mg/kg in ICR mice. Animals were monitored for the mortality and changes in the body weight, clinical signs and gross observation during the 14 days after dosing, upon necropsy. We also measured parameters of organ weight, clinical chemistry, and hematology. No dead and no clinical signs were found during the experiment period after administration of a single oral dose of SSM, AGS and MHMIX. There were no adverse effects on clinical signs, body weight, or organ weight and no gross pathological findings in any treatment group. Therefore, LD50 value of SSM, AGS and MHMIX may be over 5,000 mg/kg and it may have no side toxic effect to ICR mice. The results on the single-dose toxicity of SSM, AGS and MHMIX indicate that it is not possible to reach oral dose levels related to death or dose levels with any harmful side effects.

  • Non-alcoholic fatty liver disease (NAFLD), especially including non-alcoholic steatohepatitis (NASH) is one of the common diseases with 25% of prevalence globally, but there is no thera-peutic access available. Amomum villosum var. xanthioides (Wall. ex Baker) T.L.Wu & S.J.Chen (AX), which is a medicinal herb and traditionally used for treating digestive tract disorders in Asia countries. We aimed to examine pharmacological effects of ethyl acetate fraction of AX (AXEF) against ER stress-induced NASH mice model using C57/BL6J male mice by tunicamycin (TM, 2 mg/kg) injection focusing on the oxidative stress. Mice were orally administrated AXEF (12.5, 25, or 50 mg/kg), silymarin (50 mg/kg) or distilled water daily for 5 days, and outcomes for fatty liver, inflammation, and oxidative stress were measured in serum or liver tissue levels. AXEF drastically attenuated hepatic ER stress-induced NASH which were evidenced by decreases of li-pid droplet accumulations, serum liver enzymes, hepatic inflammations, and cell death signals in the hepatic tissue or serum levels. Interestingly, AXEF showed potent antioxidant effects by quenching of reactive oxidative stress and its final product of lipid peroxide in the hepatic tissue, specifically increase of metallothionein (MT). To confirm underlying actions of AXEF, we ob-served that AXEF increase MT1gene promoter activities in the physiological levels. Collectively, AXEF showed antioxidant properties on TM-induced ER stress of NASH by enhancement of MTs.

Ecological Characteristics and Distribution of Plant Resources of Pyrus and Malus sp. in Jindong Valley, Gangwon Province

  • Four major naturally populated areas around the Jindong valley in Inje-Gun, Gangwon province for wild Pyrus ussuriensis, Malus sieboldii, and Malus baccata are mostly located on the southeast side of the mountains at 630-745m in altitude at sea level and are in very good sunny areas. The total of 77 taxa dividing into 32 families, 58 genera, 65 species, and 12 varieties has been inhabited in these areas. Most of these plants are heliophytes, which love sunshine, but some of rhizo-plants such as Sasa borealis, climbing plants such as Actinidia arguta and Celastrus orbiculatus, or naturalized plants like Bidens frondosa are also shown. These areas have been continually destroyed, so it is possibly thought that the second transition has been progressed. The index of species diversity of plant groups in these areas is 0.672∼1.465 based on the Shannon-Wiener's method, but its index for the area that rhizo-plants like Sasa borealis are growing as an under planting decreases relatively. In this study, the oldest plant of Pyrus ussuriensis in Korea, which is 120cm in diameter at breast height (DBH), 19m in tree height and 25m in width, has been found.

New Records of Three Aquatic Fungi Isolated from Freshwater in Samcheok and Yeongju, Korea (강원 삼척 및 경북 영주의 담수지역에서 발굴된 수생균류 3종의 국내 최초 보고)

  • Three aquatic fungi were isolated from samples of freshwater-deposited plant litter and foam collected in Samcheok, Gangwon-do, and Yeongju, Gyeongsangbuk-do, Korea. Based on their morphological characteristics and a phylogenetic analysis of the internal transcribed spacer (ITS) rDNA region, the three isolates NNIBRFG329, NNIBRFG339, and NNIBRFG19 were confirmed as aquatic fungi: Articulospora tetracladia, Margaritispora aquatica, and Aquanectria penicillioides. These species were known as aquatic fungi but neither species has been previously reported in Korea.

  • Three endophytic fungal strains were isolated from aquatic plants collected in Jeju. The fungi were identified by sequencing and phylogenetically analyzing their internal transcribed spacer (ITS) regions. Consequently, the three strains NNIBRFG2982, NNIBRFG2984 and NNIBRFG 3040 were confirmed as the fungal species Acremonium tubakii, Colletotrichum cliviae and Cylindrocarpon pauciseptatum, respectively, all of which are new to Korea.

A Floristic Study on the Indigenous Habitat of Donghae-si and Samcheok-si in Gangwon-do, Korea (강원도 동해시·삼척시 일대 고유생육지의 식물상)

  • This study was carried out to investigate the flora of indigenous habitat of Donghae-si and Samcheok-si in Gangwon-do, Korea. The vascular plants were collected 18 times (from March to October 2011), and were identified as 804 taxa; 127 families, 445 genera, 720 species, 8 subspecies, 69 varieties, 5 forms and 2 hybrids. In the flora of this area, the calcicolous plants were 55 taxa; Cheilanthes argentea, Clematis serratifoliam, Quercus variabilis. Epipactis papillosa, etc. The endangered plants designated and protected by the wildlife protection law, the Ministry of Environment, Korean were counted to one taxon; Cymbidium macrorhizum. The red list of vascular plants according to IUCN valuation basis were examined, endangered species (EN); Prunus yedoensis and Cymbidium macrorhizum, Vulnerable species (VU); Juniperus chinensis, Pulsatilla tongkangensis, Diarthron linifolium and Swertia wilfordii, Near Threatened species (NT); Paeonia japonica, Allium senescens, Epipactis papillosa and Pogonia minor, Least Concern species (LC); Asplenium ruta-muraria, Platycladus orientalis, Monotropa hypopithys and Eleutherococcus divaricatus var. chiisanensis, etc., Data Deficient species (DD); Paeonia lactiflora var. trichocarpa, and Not Evaluate species (NE); Prunus choreiana, Panax ginseng and Polygonatum infundiflorum etc. Korean endemic plants of this area were 26 taxa; Aconitum pseudolaeve, Corydalis hirtipes, Vicia chosenensis, Salvia chanroenica and Hemerocallis hakuunensis, etc. The floristic regional indicator plants found in this area were 130 taxa comprising 11 taxa of grade V, 15 taxa of grade IV, 33 taxa of grade III, 15 taxa of grade II, 56 taxa of grade I. The naturalized plants were identified as 67 taxa and the percent of naturalized index (NI) was 8.3% of total 804 taxa vascular plants.
