The Study on Evaluating Performances of Lab Sacle-Advanced $A_{2}O$ with Changing System Using Biofilm Process (생물막 담체를 이용한 실험실 규모 $A_{2}O$공법의 시스템 변형에 따른 고도처리 성능 평가에 관한 연구)

  • Recently, as reinforced water quality standards for wastewater has been announced, more efficient and more powerful wastewater treatment processes are required rather than the existing activated sludge process. In order to meet this demands, we evaluate Task 1-4 about lab scale $A_{2}O$ process using biofilm media. Task 1, 2, and 3 use 'Module A' which has 4 partitions (Anoxic/Anerobic/Oxic/Oxic). Task 4 uses 'Module B' which has 2 partitions including a denitrification reactor with an Inclined plug flow reactor (IPFR) and a nitrification reactor with biofilm media. The denitrification reactor of Module B is designed to be upward flow using IPFR. The result of evaluating at each Task has shown that attached growth system has better capacity of removal efficiency for organic matter and nitrogen with the exception of phosphorus. Task 4 which has the most outstanding removal efficiency has 90.5% of $BOD_{5}$ removal efficiency, 97.8% of ${NH_4}^{+}-N$ removal efficiency, 65% of T-N removal efficiency and 92% of T-P removal efficiency with additional chemical phosphorus removal system operated at HRT 9hr, Qi:Qir 1:2, and BOD/T-N ratio 2.7.

Evaluation of SBBR Process Performance Focused on Nitrogen Removal with External Carbon Addition (외부탄소원을 사용한 SBBR의 공정 특성 및 질소제거)

  • A sequencing batch biofilm reactor (SBBR) operated with a cycle of anaerobic - aerobic - anoxic - aerobic has been evaluated for the nutrient removal characteristics. The sponge-like moving media was filled to about 10% of reactor volume. The sewage was the major substrate while external synthetic carbon substrate was added to the anoxic stage to enhance the nitrogen removal. The operational results indicated that maximum T-N and T-P removal efficiencies were 97% and 94%, respectively were achieved, while COD removal of 92%. The observations of significant nitrogen removal in the first aerobic stage indicated that nitrogen removal behaviour in this SBBR was different to conventional SBR. Although the reasons for aerobic nitrogen removal has speculated to either simultaneous nitrification and denitrification or anoxic denitrification inside of the media, further researches are required to confirm the observation. The specific oxygen uptake rate (SOUR) test with biofilm and suspended growth sludge indicated that biofilm in SBBR played a major role to remove substrates.

Comparison of Antimicrobial Activity of Electrolyzed Water Using Various Electrodes against Biofilm of Oral Pathogens

  • Biofilms of oral microbes can cause various diseases in the oral cavity, such as dental caries, periodontitis and mucosal disease. Electrolyzed water generated by an electric current passed via water using a metal electrode has an antimicrobial effect on pathogenic bacteria which cause food poisoning. This study investigated the antimicrobial activity of electrolyzed waters using various metal electrodes on the floatage and biofilms of oral microbes. The electrolyzed water was generated by passing electric current using copper, silver and platinum electrodes. The electrolyzed water has a neutral pH. Streptococcus mutans, Porphyromonas gingivalis and Tannerella forsythia were cultured, and were used to form a biofilm using specific media. The floatage and biofilm of the microbes were then treated with the electrolyzed water. The electrolyzed water using platinum electrode (EWP) exhibited strong antimicrobial activity against the floatage and biofilm of the oral microbes. However, the electrolyzed water using copper and silver electrodes had no effect. The EWP disrupted the biofilm of oral microbes, except the S. mutans biofilm. Comparing the different electrolyzed waters that we created the platinum electrode generated water may be an ideal candidate for prevention of dental caries and periodontitis.

Denitrification of Wastewater in a Fluidized Bed Biofilm Reactor (유동층 생물막 반응기에서의 폐수 탈질화)

  • Activated carbon particles of 1.274 mm diameter and sand particles of 0.455 mm diameter were employed as the support of the biofilm formed in fluidized bed biofilm reactors(FBBRs) for the wastewater denitrification. Ethanol was used as the electron donor in the anoxic respiration. The steady-state biofilm thickness increased as the nitrate loading rate increased, and the activated carbon particles induced thicker biofilm than the sand particles. The FBBRs with sand support showed higher efficiency and rate of the nitrate removal than those with activated carbon support, and exhibited the biomass concentration of 37 kg/㎥ and the nitrate removal rate of 21 kg N/㎥d.

The Basic Design Parameters and Effluent Characteristics for Aerobic Fixed Biofilm Reactor (호기성 고정생물막 반응기에서 기초 설계인자와 유출수의 성상)

  • A number of experiments to treat wastewater of a dye plant were conducted to investigate the basic design parameters and effluent characteristics for aerobic fixed biofilm reactor upon the variation of organic loading rate. The media used for this study were SARAN 4000 D with specific surface area $153m^2/m^3$, and 1000 D with specific surface area $307m^2/m^3$. The influent COD concentration ranged from 1250 to 4080 mg/L. Substrate removal and variation of biomass concentration were observed. At the beginning, the effluent quality was poor but improved as the time passed. However the effluent quality became seriously deteriorated when the media clogged. At influent COD of 1250mg/L, the effluent COD varied little, even if the organic loading rate increased from 0.63 to 2.5kg COD/$m^3$/day. It was also noted that the design parameters for activated sludge process could be applied to an aerobic fixed biofilm process under the experimental conditions.

Low Temperature Effects on the Nitrification in a Nitrogen Removal Fixed Biofilm Process Packed with SAC Media

  • A fixed biofilm reactor system composed of anaerobic, anoxic(1), anoxic(2), aerobic(1) and aerobic(2) reactor was packed with synthetic activated ceramic (SAC) media and adopted to reduce the inhibition effect of low temperature on nitrification activities. The changes of nitrification activity at different wastewater temperature were investigated through the evaluation of temperature coefficient, volatile attached solid (VAS), specific nitrification rate and alkalinity consumption. Operating temperature was varied from 20 to $5^{\circ}C$. In this biofilm system, the specific nitrification rates of $15^{\circ}C$, $10^{\circ}C$ and $5^{\circ}C$ were 0.972, 0.859 and 0.613 when the specific nitrification rate of $20^{\circ}C$ was assumed to 1.00. Moreover the nitrification activity was also observed at $5^{\circ}C$ which is lower temperature than the critical temperature condition for the microorganism of activated sludge system. The specific amount of volatile attached solid (VAS) on media was maintained the range of 13.6-12.5 mg VAS/g media at $20{\sim}10^{\circ}C$. As the temperature was downed to $5^{\circ}C$, VAS was rapidly decreased to 10.9 mg VAS/g media and effluent suspended solids was increased from 3.2 mg/L to 12.0 mg/L due to the detachment of microorganism from SAC media. And alkalinity consumption was lower than theoretical value with 5.23 mg as $CaCO_3$/mg ${NH_4}^+$-N removal at $20^{\circ}C$. Temperature coefficient (${\Theta}$) of nitrification rate ($20^{\circ}C{\sim}5^{\circ}C$) was 1.033. Therefore, this fixed film nitrogen removal process showed superior stability for low temperature condition than conventional suspended growth process.

Sewage Disposal by Different Structure of Fluidized Bed Biofilm Reactor (유동층 생물반응기의 구조변화에 따른 하수처리)

  • The purpose of this study is to investigate the biofilm reactors capable of doing high efficiency treatment. Vertical fluidized bed biofilm reactor(VFBBR) and spiral fluidized bed biofilm reactor(SFBBR) was used for their performence in biodegradation of artificial sewage. The factors influencing the efficiency of those reactors were compared with difference of physical condition. They had same size but different structure to gain access of its unique characteristics. When recycle solution with flow rate of 22 mL/min and artificial sewage with flow rate of 2~10 mL/min were fed into two reactors in aerobic state, the average $COD_{cr}$, removal rate for biodegradation of SFBBR was greater than VFBBR. After reactor feed sewage was constantly maintained as flow rate of 4 mL/min and the recycle solution were changed to 10~32 mL/min respectively, the average $COD_{cr}$ removal rate of artificial sewage in SFBBR was greater than VFBBR. In this experiment for addition of support media into two reactors SFBBR was 4.1% excellent than VFBBR. Above all, SFBBR excelled VFBBR in boidegradation of organic matter in sewage.

Manufacture of the Fluidizing Media Using Rice Straw and Paper Wastewater Treatment (볏짚을 이용한 유동장 여재제조 및 제지폐수처리)

  • Biological treatment using the activated sludge method and biofilm process has been developed for paper wastewater treatment. It is known that a water treatment using biofilm process has a high efficiency be-cause a great deal of microorganism could adhere to media. It is also known that various plastics such as polyurethane and polyethylene have been used as the media. In this study polyethylene was used as a media and rice straw an additive agent to improve porous and hydrophilic properties of the media for waste water treatment. Porosity and hydrophilic characteristics of polyethylene was increased as rice straw was added to polyethylene. Paper wastewater was then treated with newly developed environment materials. Rice straw showed excellent results in waste water treatment in various media. This environmentally friendly material prepared by polyethylene and rice straw could show similar results to those of a commercial porous polyurethane foam in wastewater treatment.

Simultaneous Nitrification and Denitrification in a Fluidized Biofilm Reactor with a Hollow Fiber Double Layer Biofilm Media (이중층 중공사 생물막 담체를 이용한 유동층 생물막 반응기에서의 동시 질산화와 탈질)

  • Simultaneous nitrification and denitrification of ammonia and organic compounds-containing wastewater were performed in a fluidized bed biofilm reactor with polysulfone(PS) hollow fiber as a double layer biomass carrier. The PS hollow fiber fragment has both aerobic and anoxic environments for the nitrifiaction and denitrification at the shell and lumen-side respectively. The reactor system showed about 80% nitrification efficiency stably throughout the ammonia load conditions applied in the experiment. Denitrification efficiency depended on organic load and C/N ratio. High free ammonia concentration and low dissolved oxygen resulted in nitrite accumulation which leads to enhance organic carbon efficiency in denitrification when compared to nitrate denitrification. The simultaneous nitrification and denitrification reactor system has an economic advantages in reduced chemical cost of organic carbon for denitrification as well as compact reactor configuration.

Effects of Nutritional and Environmental Conditions on Planktonic Growth and Biofilm Formation of Citrobacter werkmanii BF-6

  • Citrobacter sp. is a cause of significant opportunistic nosocomial infection and is frequently found in human and animal feces, soil, and sewage water, and even in industrial waste or putrefaction. Biofilm formation is an important virulence trait of Citrobacter sp. pathogens but the process and characteristics of this formation are unclear. Therefore, we employed in vitro assays to study the nutritional and environmental parameters that might influence biofilm formation of C. werkmanii BF-6 using 96-well microtiter plates. In addition, we detected the relative transcript levels of biofilm formation genes by RT-PCR. Our results indicated that the capacity of C. werkmanii BF-6 to form biofilms was affected by culture temperature, media, time, pH, and the osmotic agents glucose, sucrose, NaCl, and KCl. Confocal laser scanning microscopy results illustrated that the structure of biofilms and extracellular polysaccharide was influenced by 100 mM NaCl or 100 mM KCl. In addition, nine biofilm formation genes (bsmA, bssR, bssS, csgD, csgE, csgF, mrkA, mrkB, and mrkE) were found to contribute to planktonic and biofilm growth. Our data suggest that biofilm formation by C. werkmanii BF-6 is affected by nutritional and environmental factors, which could pave the way to the prevention and elimination of biofilm formation using proper strategies.
