Monitoring microbiological contamination, pre-decontamination, and irradiation status of commercial dried laver (Porphyra sp.) products (시판 유통 김의 미생물 오염도, 사전 살균처리 및 전자선 조사 여부 확인)

  • Eighteen commercial laver (Porphyra sp.) products were purchased from Korean market and were monitored for their microbial contamination, pre-decontamination, and luminescence properties. The laver samples showed considerable variation in their microbial contamination, from $10-10^7CFU/g$ of total aerobic counts, <$10-10^2CFU/g$ of coliforms in 4 dried laver samples, and <$10-10^6CFU/g$ of yeasts and molds except in 3 samples. In addition, $10^2CFU/g$ of Bacillus cereus was found in one sample. DEFT/APC analysis was suitable for demonstrating whether the samples were pre-decontaminated or not, with DEFT/APC values lower than 2.0 log for non-heated samples and 1.0-8.5 log for heatprocessed samples. In photostimulated luminescence (PSL) calibration, 15 samples irradiated at 1 kGy showed positive (irradiated) values more than 5000 PCs. Furthermore, thermoluminescence (TL) analysis by separating the marker minerals from samples revealed the potential to be employed in identifying irradiation status by determining $1^{st}$ TL glow at $125-175^{\circ}C$ and TL ratio ($TL_1/TL_2$) of all the samples.

Comparison of Neural Cell Differentiation of Human Adipose Mesenchymal Stem Cells Derived from Young and Old Ages (연령별 지방 중간엽 유래 줄기세포의 신경세포로의 분화 능력 비교)

  • Recently, adipose mesenchymal stem cells (AdMSC) that are similar to bone marrow MSC and blood derived MSC are thought to be another source for stem cell therapy. However, the diseases that can be applied for stem cells therapy are age-dependent degenerative diseases. Accordingly, the present study investigated the growth and differentiation potential to neural cells of human AdMSC (hAdMSC) obtained from aged thirty, forty and fifty. The growth of cells and cell viability were measured by passage and neural differentiation of hAdMSC was induced in neural differentiation condition for 10 days. Our results demonstrated that cell number, viability and morphology were not different from hAdMSC by age and passage. Immunofluorescence analysis of neural cell marker (TuJ1, NSE, Sox2, GFAP or MAP2) demonstrated no significant differences in neural cell differentiation by age and passage. As the number of passage was increased, the mRNA level of MAP2 and Sox2 was decreased in hAdMSC from age of 50 compared to hAdMSC from age of 30. In conclusion, the present study demonstrated that ability of neural cell differentiation of hAdMSC was maintained with ages, suggesting that autologous stem cells from aged people can be applied for stem cell therapy with age-dependent neural disease with the same stem cell quality and ability as stem cell derived from young age.

Analysis of Microsatellite Patterns in the Genome of Red Sea Cucumber (홍해삼 유전체 분석에 의한 microsatellite의 분포도 연구)

  • This study was conducted to analyze genome of red sea cucumber and to use it as basic data for the development of genetic markers for red sea cucumber. Microsatellite marker analysis of Ulleungdo_normal and Ulleungdo_native red sea cucumbers revealed that dinucleotide simple sequence repeats (SSRs) had the highest ratio, at 81.3~81.4%, and the number of the detected SSRs tended to decrease as the number of repeating sequence units in SSRs increased. In general, microsatellites with between 5 and 10 iterations were most common. As the size of the SSR repeating sequence units increased, the SSR iterations gradually decreased. The di-, tri-, and tetra-nucleotides in SSRs were detected in the highest numbers as (AT)5, (AAT)5, and (AAAT)5, respectively. (CG) and (CCG) had very low frequencies compared to the numbers of other repeating SSR units. The numbers of di-and tri-nucleotide repeats were up to 35 and 32, respectively, and then increased discontinuously up to 44 and 43 repeats, respectively. Tetra-, penta-, and hexa-nucleotides in SSRs occurred in numbers up to 25, 21 and 14, respectively. This analysis of red sea cucumber indicated that it maintains its own repetition sequence and repetition number; therefore, we suggest that using it as basic data for molecular marker will be possible in future research.

A Study of the Relationships between the Ratio of $2^{nd}$ to $4^{th}$ Digit Length and Cerebral Laterality (제2수지-제4수지 길이 비율과 대뇌 편측화 관계 연구)

  • Objectives : Cerebral laterality is thought to be an important marker for neurodevelopment. Prenatal testosterone could influence both cerebral laterality and 2nd to 4th finger length ratio(2D : 4D). EEG coherence and 2D : 4D were examined to investigate the relationship between prenatal testosterone level and cerebral laterality. Methods : EEG was recorded in 24 healthy subjects in the eyes closed resting state. Differences in 2D : 4D finger ratio were used to discriminate "masculine finger type" and "feminine finger type" groups. The 2D : 4D ratio was lower and greater than one for the "masculine finger type" group and "feminine finger type" group, respectively. We used coherence analysis to estimate the cortical functional connectivity. Results : There were statistically meaningful relationships among cerebral functional connectivity, sex and finger ratio. Man and masculine finger type group showed higher intra-hemispheric coherence than those of woman and feminine finger type group. Woman and feminine finger type group showed higher inter-hemispheric coherence than those of man and masculine finger type group. Conclusions : These results imply that prenatal testosterone might act as important determinants of cerebral laterality. Further examination of the relationship between 2D : 4D and EEG coherence in schizophrenia could give some clues for the neurodevelopmental hypothesis of schizophrenia genesis.

Simultaneous Determination of the Flavonoids and Limonoids in Citrus junos Seed Shells Using a UPLC-DAD-ESI/MS

  • Citrus junos seeds (CS) have been traditionally used for the treatment of cancer and neuralgia. They are also used to manufacture edible oil and cosmetic perfume. A large amount of CS shells without oil (CSS) are discarded after the oil in CS is used as foods or herbal remedy. To efficiently utilize CSS as a by-products, it needs to be studied through chemical analysis. Therefore, we developed an ultra-performance liquid chromatography (UPLC)-diode array detection (DAD) method for simultaneous determination and quantitative analysis of five components (two flavonoids and threes limonoids) in CSS. A Waters Acquity UPLC HSS T3 column C18 (2.1 × 100 mm, 1.8 ㎛) was used for this separation. It was maintained at 40 ℃. The mobile phase used for the analysis was distilled water and acetonitrile with gradient elution. To identify the quantity of the five components, a mass spectrometer (MS) with an electrospray ionization (ESI) source was used. The regression equation showed great linearity, with correlation coefficient ≥ 0.9912. Limits of detection (LOD) and limits of quantification (LOQ) of the five compounds were 0.09 - 0.13 and 0.26 - 0.38 ㎍/mL, respectively. Recoveries of extraction ranged from 97.45% to 101.91%. Relative standard deviation (RSD) values of intra- and inter-day precision were 0.06 - 1.15% and 0.19 - 0.25%, respectively. This UPLC-DAD method can be validated to simultaneously analyze quantities of marker flavonoids and limonoids in CSS.

  • Vitellogenin (Vtg), a phospholipoglycoprotein precursor of egg yolk is synthesized and secreted from the liver in response to estrogens in female fish. Vtg is normally undetectable in the blood of male fish, but can be induced by exposure to chemicals possessing estrogenic activity. Thus, the presence of Vtg in blood of male fish can serve as a useful biomarker for assessing previous exposure to estrogenic compounds. In the present study, Vtg was abnormally expressed in Rhynchocypris oxycephalus using estradiol benzoate ($E_2$). As the result, it was found that the level of Vtg in blood from R. oxycephalus was increased by treated quantity of $E_2$ with dose-effect manner. Monoclonal antibodies were generated against Vtg of R. oxycephalus. The hybridoma were screened with an enzyme immunoassay for the production of specific anti-Vtg antibodies. Five positive cell lines with a high specificity were selected. Monoclonal antibodies against vtg of R. oxycephalus that was developed in this study, may be a useful bio-indicator for the detection of estrogenic contamination in the aquatic ecosystem.

Development of Simple Sequence Repeat Markers from Adenophora triphylla var. japonica (Regel) H. Hara using Next Generation Sequencing (차세대염기서열분석법을 이용한 잔대의 SSR 마커 개발)

  • Background: Adenophora triphylla var. japonica (Regel) H. Hara shows vegetative growth with radical leaves during the first year and shows reproductive growth with cauline leaves and bolting during the second year. In addition, the shape of the plant varies within the same species. For this reason, there are limitations to classifying the species by visual examination. However, there is not sufficient genetic information or molecular tools to analyze the genetic diversity of the plant. Methods and Results: Approximately 34.59 Gbp of raw data containing 342,487,502 reads was obtained from next generation sequencing (NGS) and these reads were assembled into 357,211 scaffolds. A total of 84,106 simple sequence repeat (SSR) regions were identified and 14,133 primer sets were designed. From the designed primer sets, 95 were randomly selected and were applied to the genomic DNA which was extracted from five plants and pooled. Thirty-nine primer sets showing more than two bands were finally selected as SSR markers, and were used for the genetic relationship analysis. Conclusions: The 39 novel SSR markers developed in this study could be used for the genetic diversity analysis, variety identification, new variety development and molecular breeding of A. triphylla.

Evaluation of the acute toxicity of theoredoxin (TRX) transgenic soybean to Daphnia magna

  • Theoredoxin (TRX) transgenic soybeans were developed using the human Theoredoxin gene under the control of the ��-conglycinin promoter with a selection marker, the phosphinothricin acetyltransferase (PAT) gene. This study was done to assess the acute toxicity of a genetically modified (GM) soybean using the fresh water planktonic crustacean Daphnia magna. The acute toxicity effect of the TRX soybean and non-GM soybean (Gwangan) on D. magna was investigated at different concentrations (0, 156, 313, 625, 1,250, 2,500, and 5,000 mg·L-1). The TRX soybean used for the test was confirmed to express the TRX/PAT genes by PCR and enzyme-linked immunosorbent assay (ELISA). D. magna feeding tests showed no significant differences in the cumulative immobility or an abnormal response with either the TRX soybean or non-GM soybean. The feeding study showed a similar abnormal response and cumulative immobility of the D. magna between the TRX soybean and Gwangan treatments. Additionally, the 48 h-EC50 values for the TRX and Gwangan soybeans were 755.6 and 778 mg·L-1, respectively. The soybean NOEC (no observed effect concentration) value for D. magna was suggested to be 156 mg·L-1. These results suggest that there is no significant difference in toxicity to Daphnia magna between the TRX soybean and its non-GM counterpart.

Risk assessment and evaluation of epidermal growth factor (EGF) transgenic soybean: responses of Cyprinus carpio fed on EGF transgenic soybean

  • The epidermal growth factor (EGF) transgenic soybean was developed and biosynthesis of human epidermal growth factor (hEGF) in soybean seeds was confirmed. Also, EGF transgenic soybean were found to contain a herbicide resistance selectable marker by introduction of phosphinothricin acetyltransferase (PAT) gene from the Streptomyces hygroscopicus. For biosafety assessment, the EGF transgenic soybean expressing the EGF biosynthesis gene EGF and herbicide resistant gene PAT was tested to determine effects on survival of Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. C. carpio was fed 100% ground soybean suspension, EGF soybean or non-genetically modified (GM) counterpart soybean (Gwangan). Gene expression of EGF soybean was confirmed by PCR and ELISA to have EGF/PAT. Feeding test showed that no significant differences in cumulative immobility or abnormal response between C. carpio samples fed on EGF soybean and non-GM counterpart soybean. The 48 h-EC50 values of the EGF and non-GM soybean were 1,688 mg·L-1 (95% confidence limits: 1,585 - 1,798 mg·L-1) and 1,575 mg·L-1 (95% confidence limits: 1,433 - 1,731 mg·L-1), respectively. The soybean NOEC (no observed effect concentration) value for C. carpio was suggested to be 625 mg·L-1. We concluded that there was no significant difference in toxicity for non-target organisms (C. carpio) between the EGF soybean and non-GM counterparts.

CALCAREOUS NANNOPLANKTON FROM THE SEOGUIPO FORMATIN OF CHEJU ISLAND, KOREA AND ITS PALEOCEANOGRAPHIC IMPLICATIONS

  • Twenty species of calcareous nannofossils belonging to 11 genera are identified from the Seoguipo Formation in Cheju island, Korea. On the basis of the marker species, the Seoguipo Formation is biostratigraphically assigned to the Pseudoemiliania lacunosa Zone (NNl9), which corresponds to the combined zones of Emiliania annula - Emiliania ovata (CN13a-CN14a) of latest Pliocene and Early Pleistocene. Generally, cold-water species is dominant in the lower part, whereas warm-water one in the upper part. This is interpreted the palaeoceanographic condition has changed from cooling to warm phase. The change in floral composition and abundance of specific species allows the recognition of 4 ecostaratigraphic units in the Seoguipo Formation and the migration of oceanographic frontal boundary. According to nannofossil distribution in the study area, the position of an oceanographic boundary between warmer water and cooler water appeared to have oscillated north-south over the Korea Strait and Cheju island in response to glacial and interglacial cycles. The geologic time of the interpreted paleoceanographical changes determined by nannofossil biochronology is well agreed with the results obtained from the Japan Sea (East Sea) and Japan-Sea side of Japan.
