0.1% Iodoacetamide에 의해 유도된 흰쥐 위염 모델에서 한약처방(JAUN-1)의 유익한 효능규명 (Beneficial Effects of Herbal Mixture (JAUN-1) on 0.1% Iodoacetamide-induced Gastritis Rat Model)

  • To verify the effects of JAUN-1, which is a water-extracted herbal mixture, on gastroenteric disorders induced by 0.1 percent of iodoacetamide (IA) in rats. We divided four groups, $Na{\"{\i}}ve$ + Distilled Water (DW), 0.1% IA + DW, 0.1% IA + Proton pump inhibitor (Lansoprazole, 5 mg/kg) and 0.1% IA + Herbal mixture (JAUN-1, 50mg/kg) and performed following experimental methods to confirm its advantageous effects against ulcerogenic stomach in rats induced by 0.1% IA; cell cytotoxicity, analysis of lesions score, Hematoxylin & Eosin (H&E) stain, RT-PCR for ${\beta}-actin$, COX-1 and COX-2 and evaluation of intestinal prokinetic activity. No cytotoxicity was elucidated at the concentration of 1, 5, 10, 50, 100, $500\;{\mu}g/ml$ and 1mg/ml JAUN-1 through MTT Assay using by human stomach epithelial AGS cells, respectively. In addition, the JAUN-1 treated group and the lansoprazole treated group significantly decreased in lesions score compared to the DW treated group in the gastritis induced rat model, and results of immunohistochemistry by H&E staining showed that histological recovery in Proton Pump Inhibitor (PPI) and JAUN-1 treated groups rather than the DW administrated group. Another outcome was that ${\beta}-actin$ relative COX-2 expression level was significantly promoted in the DW treated group while ${\beta}-actin$ relative COX-1 expression level was no meaningful change in this rat model. Finally, intestinal prokinetic activity was recovered from low level of prokinetic activity due to 0.1% IA induced gastritis to the similar level of Normal group. These results suggested that JAUN-1 may have beneficial effects against 0.1% IA-induced gastritis rat model through decreasing lesions score, histological recovery, ${\beta}-actin$ relative COX-1, 2 expression level and prokinetic activity.

  • $eta$-Sialon synthesis was investigated via the simulataneous reduction and nitriding of Wando pyrophyllite. When Wando pyrophyllite-graphite-$Si_3N_4$ seed mixture was heated at 135$0^{\circ}C$ for as long as 10 hours in 80% $N_2$-20%$H_2$ atomsphere $eta$-$Si_3N_4$ solid solution was mainly formed together with a small amount of $\alpha$-$Si_3N_4$ The value z of the forming $Si_{6-x}Al_2O_2N{8-z}$ was decreased with heating time.

Antioxidants and Anti-inflammatory Effects of Fermented Houttuynia cordata Thunb and Scoria Mixture Extract

  • In this study, antioxidant and anti-inflammatory of fermented Houttuynia cordata Thunb and scoria mixture extract were investigated in vitro. Radical-scavenging activities of the ethanol extracts were examined by using ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS)) radicals assay. Consequently, we confirmed that fermented Houttuynia cordata Thunb and scoria mixture extract dependent removed DPPH and ABTS radical. Also, to confirm anti-inflammatory activity of ethanol extract, we treated fermented Houttuynia cordata Thunb and scoria mixture extract on BV-2 cell with LPS. The result showed that fermented Houttuynia cordata Thunb and scoria mixture extract concentration-dependent inhibited NO production. Therefore, fermented Houttuynia cordata Thunb and scoria mixture extract showed inhibition radical oxygen activities and inflammatory and have available for a pharmacological composition on neuritis-protection and treatment.

Tree Size Distribution Modelling: Moving from Complexity to Finite Mixture

  • Tree size distribution modelling is an integral part of forest management. Most distribution yield systems rely on some flexible probability models. In this study, a simple finite mixture of two components two-parameter Weibull distribution was compared with complex four-parameter distributions in terms of their fitness to predict tree size distribution of teak (Tectona grandis Linn f) plantations. Also, a system of equation was developed using Seemingly Unrelated Regression wherein the size distributions of the stand were predicted. Generalized beta, Johnson's SB, Logit-Logistic and generalized Weibull distributions were the four-parameter distributions considered. The Kolmogorov-Smirnov test and negative log-likelihood value were used to assess the distributions. The results show that the simple finite mixture outperformed the four-parameter distributions especially in stands that are bimodal and heavily skewed. Twelve models were developed in the system of equation-one for predicting mean diameter, seven for predicting percentiles and four for predicting the parameters of the finite mixture distribution. Predictions from the system of equation are reasonable and compare well with observed distributions of the stand. This simplified mixture would allow for wider application in distribution modelling and can also be integrated as component model in stand density management diagram.

사료 내 β-Glucanase 활성이 강화된 복합효소제 급여가 육계의 생산성과 혈청성분 및 육질에 미치는 영향 (The Effects of Dietary Enzyme Mixture Fortified with β-Glucanase Activity on the Growth Performance, Serum Components, and Meat Quality of Broiler Chicks)

  • 본 연구에서는 Ross 208 종의 broiler 31,800수를 대조구와 복합 효소제 첨가구로 나누어 5주간에 걸쳐 대조구는 옥수수-대두박을 기초로 한 시판 육계사료를 급여하고, 시험구는 ${\beta}-glucanase$를 강화한 복합 효소제를 0.3% 첨가 급여하여 생산성과 혈청성분과 육질에 미치는 영향을 조사하였다. 시험기간 동안의 각 시험구의 증체량 및 사료 효율, 생산지수, 육성율을 측정하였고 무작위로 각 시험구당 20수를 선발하여 혈청 콜레스테롤 및 glucose 농도를 측정하였다. 또 ND와 IBD 항원을 음용 투여한 브로일러의 ND와 IBD에 대한 항체역가를 조사하였으며, 육질검사로 도체의 pH 및 육색, 가열감량, 보수력, 전단력을 측정하였다. 사육시험 결과, 0.3% 복합 효소제 시험구가 대조구에 비해 평균체중 19%, 생산지수 6.8%, 사료효율 5.5%, 육성율 5%의 증가를 나타내었다. 복합 효소제 시험구는 대조구보다 혈청내 HDL-콜레스테롤 양을 증가시키고 glucose의 양을 감소시켰다. 육계 혈청 내 ND 및 IBD 항원에 대한 항체역가도 복합 효소제 시험구가 대조구보다 높았다. 육질검사에 있어서는 복합 효소제 시험구가 대조구보다 육색과 전단력은 차이가 없었으나 보수력(p<0.05)과 가열감량이 개선되는 결과를 나타내었다. 이상의 결과로부터 ${\beta}-glucanase$ 활성을 강화한 복합 효소제의 첨가 급여는, 육계의 생산성과 혈청성분과 면역기능과 육질을 개선시키는 효과가 있는 것으로 사료된다.

Purification and Characterization of $Ginsenoside-{\beta}-Glucosidase$

  • In this paper, the saponin enzymatic hydrolysis of ginsenoside Rg3 was studied. The $ginsenoside-{\beta}-glucosidase$ from FFCDL-48 strain mainly hydrolyzed the ginsenoside Rg3 to Rh2, the enzyme from FFCDL-00 strain hydrolyzed Rg3 to the mixture of Rh2 and protopanaxadiol (aglycon). The $ginsenoside-{\beta}-glucosidase$ from FFCDL-48 strain was purified with a column of DEAE-Cellulose to one spot in the SDS polyacrylamide gel electrophoresis. During the purification, the enzyme specific acitvity was increased about 10 times. The purified $ginsenoside-{\beta}-glucosidase$ can hydrolyze the Rg3 to Rh2, but do not hydrolyze the $p-nitrophenyl-{\beta}-glucoside$ which is a substrate of original exocellulase such as ${\beta}-glucosidase$ of cellulose. The molecular weight of $ginsenoside-{\beta}-glucosidase$ was 34,000, the optimal temperature of enzyme reaction was $50^{\circ}C,$ and the optimal pH was 5.0.

Regulation of $\beta$-Xylosidase (XylA) Synthesis in Bacillus stearothermophilus

  • Syntheses of the B. stearothermophilus xylanolytic enzymes such as xylanases, ${\beta}$-xylosidases, ${\alpha}$-arabinofurano-sidases, and esterases, were observed to be regulated by the carbon source present in the culture media. Xylan induced synthesis of ${\beta}$-xylosidase at the highest level while xylose gave about 30% of the ${\beta}$-xylosidase activity induced by xylan. The lowest syntheses of the xylanolytic enzymes above mentioned were detected in the basal medium containing glucose as a sole carbon source. When a mixture of xylan and glucose was used as a carbon source, we could observe glucose repression of xylanase (about 70-fold) and ${\beta}$-xylosidase (about 40-fold) syntheses. Whereas, the level of the glucose repression of the expression of the xylA gene encoding the major ${\beta}$-xylosidase of B. stearothermophilus was assessed to be about l0-fold when the relative amounts of the xylA transcript were determined. From the sequence of the xylA gene, we could find two CRE-like sequences (CRE-l: nucleotides +124 to +136 and CRE-2:+247 to +259) within the reading frame of the xylA gene, either or both of which were suspected to be involved in catabolite repression of the xylA gene.

  • A mixture of aluminum isopropoxide (Al(OC3H7i)3) solution and sodium hydroxide (NaOH) solution was hydrolyzed in the range between pH 1~14. the powder obtained from sol-gel process was calcined at several temperatures and crystallization behaviors of various samples were investigated. The hydrolyzed sols of pH 1~6 wre clear, or near clear. On the other hand, powder precipitated from sols of pH 7~14. The sample obtained from pH 3 solution crystallized via complicated route, and $\beta$-Al2O3 and $\beta$"-Al2O3 phases appeared at lower temperature than samples from other pH conditions. And the quantity of remained $\beta$"-Al2O3 phase after heat treatment at 150$0^{\circ}C$ was more than samples from other pH conditions. After sintering, ionic conductivities were 1.3$\times$10-4S.cm-1 to 0.76$\times$10-4S.cm-1.0-4S.cm-1.
