6-Methylquinolinium Dichromate를 이용한 치환 벤질 알코올류의 산화반응 속도 (Kinetics of the Oxidation of Substituted Benzyl Alcohols using 6-Methylquinolinium Dichromate)

  • $H_2O$ 용매 하에서 6-methylquinoline 과 chromium trioxide의 반응을 통하여 6-methylquinolinium dichromate [$(C_{10}H_9NH)_2Cr_2O_7$]를 합성하여, 적외선분광광도법(IR), 유도결합 플라즈마(ICP) 등으로 구조를 확인하였다. 여러 가지 용매 하에서 6-methylquinolinium dichromate를 이용하여 벤질 알코올의 산화반응을 측정한 결과 유전상수(${\varepsilon}$) 값이 큰 용매 순서인 시클로헥센 < 클로로포름 < 아세톤 < N,N-디메틸포름아미드 용매 하에서 높은 산화반응성을 보였다. 산촉매($H_2SO_4$)를 이용한 DMF 용매 하에서 6-methylquinolinium dichromat는 벤질 알코올과 그의 유도체들(p-$OCH_3$, m-$CH_3$, H, m-$OCH_3$, m-Cl, m-$NO_2$)을 효과적으로 산화시켰다. 그리고 전자받개 그룹들은 반응속도가 감소한 반면에 전자주개 치환체들은 반응속도를 증가시켰다. 또한 Hammett 반응상수(${\rho}$) 값은 -0.67(303K) 이였다. 그러므로 본 실험에서 알코올의 산화반응 과정은 속도결정단계에서 수소화 전이가 일어나는 메카니즘임을 알 수 있었다.

  • We have developed the novel one-pot synthetic method for regioselective and stereoselective N-protected amines through the reaction of various ethers with chlorosulfonyl isocyanate (CSI). Also, we found a novel technique to compare directly the stability of carbocations in the solution phase and established the stability order of the various carbocations. And we reported the cleavage of benzyl and p-methoxybenzyl protecting groups of alcohols and phenols in the presence of other functional groups using CSI. (omitted)

  • 2-브로모프로피오닐화된 수지를 이용해 insulin A (1-21) 사슬을 합성하였다. 시스테인의 결사슬은 각각 acetamidomethyl, benzyl, 그리고 benzhydryl기로 보호하였으며 그루타민과 아스파라긴은 p-nitrophenyl기로 활성화하여 합성에 이용하였다. 매 짝지음단계마다 DCC/HOBT coupling agent로 각 아미노산을 축합하였으며 반응의 완결여부는 닌히드린시험으로 측정하였다. 생성물은 NH$_3$/MeOH-Dioxane(v/v 1:1)로 수지로부터 분리하여 DEAE Sephadex A-25와 Sephadex LH-20으로 정제하였으며 최종생산물은 HPLC, electrophoresis로 확인한 결과 순수한 것(>99.9%)으로 나타났으며 총수득율은 6%이었다.

  • ABA type block copolymers composed of poly($\gamma-benzyl$ L-glutamate) (PBLG) as the A component and poly (ethylene glycol) as the B component were obtained by polymerization of $\gamma-benzyl$ L-gletamate N -carboxyanhydride, initiated by amino groups at both ends of poly(ethylene glycol) . From circular dichroism measurements in ethylene dichloride solution as well as from infrared spectTa measurements in solid state, it was found that the polypep- tide block exists in the a-helical conformation, as in PBLG homopolymer. $Poly-N^5$ (3-hydroxypropyl glutamine) (PHPG)/poly(ethylene glycol)block copolymer hydrogel was obtained by the treatment of PBLG/PBG block copolymer with the mixture of 3-ammine-1-propanol and diamlnooctane. The water content of PHPG/PEG block copolymer hydrogel was about 80wt% when the concentration of crosslinking agent was below 5 mole % per polymer.

Substituent Effects on the Binding Energies of Benzyl Alcohol-H2O Clusters: Ab initio Study

  • Computations are presented for the ortho- and para-substituted benzyl alcohol-$H_2O$ clusters. A variety of conformers are predicted, and their relative energies are compared. Binding energies of the clusters are computed, and detailed analysis is presented on the effects of substitution on the strength of the hydrogen bond in the clusters. F- and $NH_2-$ substituted clusters are studied to analyze the effects of electron-withdrawing and electron-pushing groups. In para-substituted clusters, the inductive effects are dominant, affecting the binding energies in opposite way depending on whether the hydroxyl group is proton-donating or -accepting. For ortho-substituted clusters, more direct involvement of the substituting group and the resulting geometry change of the hydrogen bond should be invoked to elucidate complicated pattern of the binding energy of the clusters.

Free Radical-mediated Ring Expansion Reactions:Endocyclic Cleavage of Cyclopropylcarbinyl Radicals

  • Ring expansion reactions via endocyclic cleavage of cyclopropylcarbinyl radicals derived from the reaction of [1-benzyloxycarbonylbicyclo[n. 1.O]alk-(n+l)-yl] -1-imidazolethiocarboxylates with tributyltin hydride/AIBN proceeded to produce 3-cycloalkenecarboxylates in good yields. Benzyl (5'-phenoxypentyl) -3-cyclohepten-1 -carboxylate was obtained in 33% yield from the reaction of benzyl 5-methylenebicyclo [4. 1.0]- 1-carboxylates with 4-phenoxybutyl iodide under radical conditions. Selective cleavage of endocyclic bond in cyclopropane to the cyclohexane, results from stabilization of the resultant radical by the carbonyl groups, such as the benzyloxycarbonyl group, which lower the transition state energy for the final cyclopropane cleavage in the ring expansion.

Octapeptide (Alanine Angiotensin) 의 合成 (Synthesis of an Octapeptide (Alanine Angiotensin))

  • We have shown that carboxy-peptidase destroys the biological activity of angiotensin octa-and deca-peptides. Since Proline occurs as the seventh amino acid from the amino end of the chain and since carboxypeptidase does not cleave proline from a peptid chain, it is evident that the heptapeptid H.asp-arg-val-tyr-ileu-his-pro.OH is formed by this hydrolysis. This peptide must then be biologically inactive. In order to determine whether the phenyl group of the C-terminal amino acid was the necessary requirement for biological activity of the octapeptide, $ala^8$ angiotensin octapeptide(amino acids of peptides numbered from amino end) was synthesized. For this synthesis the four dipeptides were prepared: carbobenzoxy-L-prolyl-L-alanine-P-nitrobenzyl-ester, m.p. $134-135^{\circ}C,$ carbobenzoxy-L-isoleucyl-imidazole benzyl-L-histidine methyl ester, m.p. $114-116^{\circ}C,$ carbobenzoxy-L-valyl-L-tyrosine hydrazide and carbobenzoxy B-benzyl-L-aspartyl-nitro-L-arginine. The first three dipeptides were obtained as crystalline compounds. Imidazole-benzyl-L-histidine was used in the hope that it would block the histidine imidazole against side reactions in steps subsequent to the formation of the C-terminal tetrapeptide. Also, it was through that the imidazole benzylated peptides would be easier to crystallize. This, however, was not the case. The tetrapeptide, carbobenzoxy-L-isoleucyl-L-im, benzyl-histidyl, L-prolyl-L-alanine-nitrobenzyl ester was not obtained in a crystalline form. Neither could the mono-or dihydrobromide of the tetrapeptide free base be induced to crystallize. Carbobenzoxy-L-valyl-L-tyrosine azide was condensed with the tetrapeptide free base to yield the protected hexapeptide; carbobenzoxy-L-valyl-L-tyrosyl-L-isoleucyl-L-im, benzyl, histidyl-L-Prolyl-L-alanine-nitrobenzyl ester. Upon removal of the carbobenzoxy group with hydrogen bromide in acetic acid an amorphous free base hexapeptide ester was obtained. This compound gave the correct C, H, N analysis and contained the six amino acids in the correct ratio. The octapeptide was obtained by condensing this hexapeptide with carbobenzoxy-B-benzyl-L-aspartyl-nitro, L-arginine using the mixed anhydride method of condensation. This amorphous product was proven to be homogenous by chromatography in two solvent systems and upon hydrolysis yielded the eight amino acids in correct ratio. The five protecting groups were removed from the octapeptide by hydrogenolysis over palladium black catalyst. Biological assay of the free peptide indicated that it possessed less than 0.1 per cent of both pressor and oxytocic activity of the phenylalanine8 angiotensin. This suggests that the phenyl group is a point of attachment between angiotensin and its biological receptor site.

Anion Exchange Membrane Having Permselectivity specific Anion in Electrodialysis

  • To change permselectivity between anions through the anion exchange membrane in electrodialysis, the various modified anion exchange membranes were prepared: highly crosslinked anion exchange membranes, anion exchange membranes having benzyl trialkylammonium groups with different carbon number of alkyl chain as anion exchange groups and anion exchange membranes having pyridinium groups with a hydrophilic or hydrophobic substituent at a different position as anion exchange groups. It became clear from the evaluation of these membranes that the degree of the hydrophilicity of the anion exchange membranes greatly affects the permselectivity between two artions. To increase the hydrophiticity of the anion exchange membranes further, electrodialysis was carried out in the presence of ethylene glycols and the permeation of strongly hydrated anions increased and that of less-hydrated anions decreased. It became clear that the change in the permselectivity between two artions is due to the change in the affinity of anions to the membranes, not the change in mobility ratio of the anions in the membranes phase.
