Antimutagenic Potential of Phellinus igniarius

  • Mutagenic activities of extracts from the filtrate of the cultured broth (PI-I), mycelia (pI-II), and the fruiting bodies (PI-III) of Phellinus igniarius were examined by Ames/Salmonella tests. No mutagenic activity was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PI-I, PI-II, and PI-III showed inhibitory effects on the mutagenic activities by the directly-acting mutagens, 4-nitro-ο-phenylenediamine(NPD) and sodium azide ($NaN_3$), and also by the indirectly-acting mutagens, 2-aminofluorene (2-AF) and benzo[a]pyrene (B[a]P). These results suggest that P. igniarius possesses some antimutagenic activity and may contain some chemopreventive agents.

In Vivo Antitumor Activity of Hydrophilic Arginine-Conjugated Linoleic Acid Complex

  • Although conjugated linoleic acid (CLA) exerted potent antitumor activities in several animal models, application of CLA as a bioactive ingredient has been limited due to its hydrophobicity. This study was designed to determine the antitumor activity of arginine-CLA complex (Arg-CLA), a hydrophilic form of CLA. Mouse forestomach cancer was induced by gavage with benzo(a)pyrene (B(a)p) for 4-weeks prior to Arg-CLA (0.2 and 0.5%) feeding. Complete necropsies were performed to determine the number, size and locations of all the forestomach tumors at 20 weeks post-B(a)P administration. All mice in the B(a)P group developed tumors, and tumor incidences were decreased by 31 % and 44% in 0.2% and 0.5% Arg-CLA-fed groups, respectively, whereas no decrease was observed when Arg or com oil was given alone. Our results suggest that Arg-CLA suppresses mouse forestomach cancer.

Assessing the Formation of Polycyclic Aromatic Hydrocarbons in Grilled Beef Steak and Beef Patty with Different Charcoals by the Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) Method with Gas Chromatography-Mass Spectrometry

  • This study investigated the effects of different charcoals on the occurrence of 16 polycyclic aromatic hydrocarbons (PAHs) in grilled beef steaks and beef patties. Seven different charcoals were used as follows: from oak wood (C1), from orange wood (C2), from Valonia oak wood (C3), from Marabu wood (C4), extruded charcoal from beech wood (C5), from coconut shells (C6), and from hazelnut shells (C7). The grilling times for each charcoal type were 6 min for the beef patties and 7 min for the beef steaks, until the internal temperature reached at least 74℃. The total concentration of 16 PAHs (PAH16) in beef steaks grilled with C1 (35.75 ㎍/kg) and C7 (36.39 ㎍/kg) was higher than that of C3 (23.80 ㎍/kg) and C6 (24.48 ㎍/kg; p<0.05). The highest amounts of PAH16 (216.40 ㎍/kg) were determined in the beef patty samples grilled using C5 (p<0.05). The summation of benzo[a]anthracene, chrysene, benzo[b]fluoranthene and benzo[a]pyrene, referred to as PAH4, was not detected in any of the beef steaks, whereas it was determined in the beef patties grilled with C2 (7.72 ㎍/kg) and C5 (22.95 ㎍/kg; p<0.05). The PAH16 concentrations of the beef patty samples in each charcoal group were significantly higher compared to the beef steaks (p<0.05). To avoid the formation of high PAH levels, the use of extruded charcoal and hazelnut shell charcoal should therefore be avoided when charcoal grilling beef steaks and beef patties, and low-fat meat products should be preferred.

Roasting 온도에 따른 오미자, 황기, 길경 및 맥문동을 첨가한 한방차의 벤조피렌 함량 변화 (The Changes of Benzo[${\alpha}$]pyrene Content in Herbal Tea Containing Schizandra chinensis, Astragalus membranaceus, Platycodon grandiflorum and Liriope platyphylla Affected by Roasting Temperature)

  • Roasting 온도를 $80{\sim}140^{\circ}C$로 달리한 한방차의 성분 변화를 분석한 결과는 다음과 같다. 처리온도의 상승에 따라 수분함량이 감소하고, 일부 탄화가 발생하며 조회분 함량이 상대적으로 상승하는 소폭 변화가 있었고, 조단백질 및 조지방 함량은 거의 변화가 없는 것으로 나타났다. 벤조피렌 함량은 0.17~0.35ppb으로 처리온도가 상승할수록 $B({\alpha})P$ 함량이 증가하는 경향을 보였다. 이러한 결과를 볼 때, 처리온도와 원재료에 따라 $B({\alpha})P$ 함량에 차이가 발생한 것으로 나타났는데, roasting의 경우 실제 내부온도는 약 $200^{\circ}C$내외에 그치지만 roaster 표면의 온도는 약 $2,000^{\circ}C$에 이르기 때문에 이 표면과 직접 접촉한 부분에서 일부의 $B({\alpha})P$가 생성된 것으로 판단된다. 한방차의 고형분용출율을 0.18~0.35%(w/w)를 나타내었는데, roasting 온도가 상승할수록 고형분용출율이 감소하는 경향을 나타내었다. 처리온도가 $80{\sim}110^{\circ}C$처리구간에서는 큰 변화를 나타내지 않은 반면 $110{\sim}140^{\circ}C$처리구간에서는 고형분용출율이 급격히 감소하는 경향을 보였다. 처리온도가 상승할수록 고형분용출율이 감소하는 것은 내부 조직이 치밀하여 상대적으로 용출이 어렵기 때문으로 생각된다.

  • An analysis of changes in herbal tea composition according to the difference in processing conditions showed slightly reduced crude protein content and increased, moisture, crude fat and solid elution rate after treatment using the ash puffing process compared to roasting. Benzopyrene content was significantly reduced to 0.18 ppb from 0.51 ppb. This result indicated that, the $B({\alpha})P$ content differed depending on the processing condition and raw materials. Generation of food $B({\alpha})P$ is mainly include the thermal decomposition of food cooking, when the processing which is a main component of food carbohydrate, protein, fat reason despite severe heat treatment as a whole is to be detected even though the $B({\alpha})P$ in this way is considered to be. The taste, aroma and color did not show a large difference, but the strong bitters taste decreased.

STUDY CYTOCHROME P450IA1 GENE EXPRESSION BY RTPCR.

  • To investigate the mechanism of the regulation of cytochrome P450IA1 gene expression, ethoxyresorufin deethylase(EROD) and benzo(a)pyrene hydroxylase in B6 mouse liver, in isolated perfused rat liver system. and in B6 mouse hepatocyte Hepa-I cells were examined. In C57BL/6N mouse, 3-methylcholan- throne( 3MC ) treatment have resulted in the stimulation of EROD activity based on fluorometry by 2.79 fold comparirng with that of control. Measurement of mRNA of cytochrome P450 was carried out by either nothern blot or dot blot analysis. Findings are similar to that of studies with enzymes. Furhtermore, when RTPCR method was applied to detect mRNA in Hepa I cell and liver tissues the results were more clear. Cytochrome P450IA1 upstream DNA containing CAT construct was transfected into Hepa-1 cells. After transfection of CAT construct, 3MC and flavonoids, such as, chrysin, hesperetin, kaempferol, morin, myricetin and aminoyrine were treated. 48 Hours after treatments, cells were harvested and assayed for CAT mRNA by RTPCR. 3MC treatment to hepa I cells transfected with trout P450IA1-CAT construct increased CAT mRNA by 2.81 fold when it was compared with that of control. This increase CAT mRNA was decreased by concomitantly treated flavonoids and aminopyrine. The level of CAT protein was 29.2-58.0% of 3MC stimulated CAT protein. Results of this study suggested that RTPCR seems to be a very good method to study regulation of gene expression in liver tissue or Hepa cells.

Biodegradation and Removal of PAHs by Bacillus velezensis Isolated from Fermented Food

  • Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous in the environment. They are highly toxigenic and carcinogenic. Probiotic bacteria isolated from fermented foods were tested to check their ability to degrade and/or detoxify PAHs. Five probiotic bacteria with distinct morphologies were isolated from a mixture of 26 fermented foods co-cultured with benzo(a)pyrene (BaP) containing Bushnell Haas minimal broth. Among them, B. velezensis (PMC10) significantly reduced the abundance of BaP in the broth. PMC10 completely degraded BaP presented at a lower concentration in broth culture. B. velezensis also showed a clear zone of degradation on a BaP-coated Bushnell Haas agar plate. Gene expression profiling showed significant increases of PAH ring-hydroxylating dioxygenases and 4-hydroxybenzoate 3-monooxygenase genes in B. velezensis in response to BaP treatment. In addtion, both live and heat-killed B. velezensis removed BaP and naphthalene (Nap) from phosphate buffer solution. Live B. velezensis did not show any cytotoxicity to macrophage or human dermal fibroblast cells. Live-cell and cell-free supernatant of B. velezensis showed potential anti-inflammatory effects. Cell-free supernatant and extract of B. velezensis also showed free radical scavenging effects. These results highlight the prospective ability of B. velezensis to biodegrade and remove toxic PAHs from the human body and suggest that the biodegradation of BaP might be regulated by ring-hydroxylating dioxygenase-initiated metabolic pathway.

먹물버섯 자실체 및 균사체 추출물의 돌연변이 억제효과 (Antimutagenic Effect of the fruiting Body and the Mycelia Extracts of Coprinus comatus)

  • The inhibitory effect of Coprinus comatus on the mutagenicity in Salmonella assay system and SOS chromotest were studied. In Ames test, the ethanol and water extracts and the cultured mycelia fractions of Coprinus comatus did not show any mutagenicity, but the Coprinus comatus ethanol extracts showed inhibitory effects of 8 0∼90% on the mutagenicity induced by indirect mutagen of benzo(a)pyrene(B(a)P) and aflatoxin B1(AFB1) in Salmonella typhimurium TA98 and TA100. The antimutagenic effect increased with increasing concentration of the ethanol extract toward N methyl N' nitro N nitrosoguanidine(MNNG). However, the water extracts inhibited about 40∼50% against direct and indirect mutagen. The cultured mycelial filtrate of Coprinus comatus, the fractionⅡ, showed antimutagenic effect of 90% on MNNG and 25∼50% on B(a)P and AFB1. In SOS chromotest, the ethanol extracts of Coprinus comatus showed antimutagenic effect of 65∼81% on SOS function induced by 4 NQO, and the cultured mycelia fractionⅡ showed low inhibitory effect of 20∼50%.

  • Human exposure to environmental carcinogens can be detected by a number of methods including immunoassay, $^{32}P-postlabeling$ assay, and fluorescence technique. These assays have been applied to measure biological markers of carcinogen-adducts formed with macromolecules such as DNA, RNA and protein. In an attempt to investigate causal relationships between carcinogen exposure and tumor formation, specific carcinogen-adducts have been quantitated from human tissues and body fluids of cancer patients, occupational workers heavily exposed to certain carcinogens, smokers and controls. Carcinogens studied for biological human monitoring include benzo(a)pyrene, aflatoxin B1, UV light, ethylene oxide, 8-methoxypsoralen, 4-aminobiphenyl, vinyl choride, N-nitrosamine, cisplatin and other chemotherapeutic agents. Relevance of human monitoring for cancer research, progress in this field, methods to detect carcinogen-adducts are reviewed here. It is hoped that these approaches will be used for the risk assessment of carcinogen exposure, cancer etiology study and cancer prevention in humans.
