(R)-2-(4-hydroxyphenoxy)propionic acid (HPOPA) is a key intermediate for the preparation of aryloxyphenoxypropionic acid herbicides (R-isomer). In order to improve the HPOPA production from the substrate (R)-2-phenoxypropionic acid (POPA) with Beauveria bassiana CCN-A7, static cultivation and H2O2 addition were attempted and found to be conducive to the task at hand. This is the first report on HPOPA production under static cultivation and reactive oxygen species (ROS) induction. On this premise, the cultivation conditions and fermentation medium compositions were optimized. As a result, the optimal carbon source, organic nitrogen source, and inorganic nitrogen source were determined to be glucose, peptone, and ammonium sulfate, respectively. The optimal inoculum size and fermentation temperature were 13.3% and 28℃, respectively. The significant factors including glucose, peptone, and H2O2, identified based on Plackett-Burman design, were further optimized through Central Composite Design (CCD). The optimal concentrations were as follows: glucose 38.81 g/l, peptone 7.28 g/l, and H2O2 1.08 g/l/100 ml. Under the optimized conditions, HPOPA titer was improved from 9.60 g/l to 19.53 g/l, representing an increase of 2.03-fold. The results obtained in this work will provide novel strategies for improving the biosynthesis of hydroxy aromatics.
Entomopathogenic fungi are one of the microorganisms distributed worldwide, and they inhabit not only insects but also soils of various regions such as agricultural land and forest areas. In this study, entomopathogenic fungi distributed in the soil of forest areas in Korea were isolated using the insect-baiting method, and the relationship between the distribution and diversity of entomopathogenic fungi and the physical/chemical characteristics of the soil was analyzed. The soils were collected from five forest areas in Korea, and a total of 42 entomopathogenic fungal isolates were isolated from the collected soils. Among them, Beauveria bassiana (42.9%) and Metarhizium anisopliae (45.2%) were mainly isolated. In particular, it was confirmed that the distribution of M. anisopliae varies depending on the total nitrogen (g/kg) and organic matter content (%) of the soil. The soils in forest areas in Korea have a low pH of 4-5. Among all isolates, >50% of entomopathogenic fungi were isolated from silt loam. This study suggests that it will be helpful in understanding the relationship between the distribution and diversity of entomopathogenic fungi and the physical and chemical characteristics of soil.
Single ascospore isolates of Cordyceps bassiana were observed for their colony pigmentation on Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates and were inoculated in a brown rice medium for production of fruiting bodies. Colony pigmentation did not show any relationship with perithecial stromata formation. The isolates were also grown on opposite sides of SDAY agar plates and were observed for vegetative compatibility. Neither vegetative compatibility nor perithecial stromata could be found to be related to each other. It was concluded that fertile fruiting body production was independent of colony pigmentation and vegetative compatibility. Synnemata formation was found to be more common than perithecial stromata formation. This might be due to its highly conidiogenous anamorphic stage, i.e., Beauveria bassiana.
Biological control of forest insect pests, Agelastica coeruleci, Meganola n~elancholia,a nd Glyphodes perspectalis, turfgrass insect pest, Blitoperfhu orientr~lis,v egetable insect pests, Plutella xylostella and Agrotis segetun~ with entomopathogenic fungus, Beciuveria hassinna GY 1 - 17 isolated from rice paddy of Yangsan in the southern part of Korea were investigated. Mortalities of A. coeruleu and P. ~ylo.~rellluar vae were 100% at the rate of 7.0 and 2.0 x 107conidia/ml after 7 and 5 days and those of M. melancholia were 66.7 - 100% at the rate of 0.03875-3.1 X 107conidia/ml. However, G. perspectulis was not affected at the rate of 2.0 x lo7 to X I04conidia/ml. And mortalites of B. orientcilis and A. segetum larvae were 46.7% at 3.7 x 107conidia/ml and 63.3% at 2.5 X 107conidia/ml.
Selection of the optimal culture medium and evaluation of the antimicrobial activity against various phytopathogens were performed for four entomopathogenic fungal isolates with excellent insecticidal and antimicrobial activity against the two-spotted spider mite (Tetranychus urticae), green peach aphid (Myzus persicae), and gray mold (Botrytis cinerea). The optimal medium was selected by measuring the amount of blastospore production and the antifungal activity of the culture medium. On the basis of these experiments, GY medium was selected for Beauveria bassiana 2R-3-3-1 and Metarhizium anisopliae 4-2, SD3, and PDB medium for B. bassiana SD15. The antimicrobial activity test against other phytopathogens indicated that all four isolates showed high antifungal activities against Colletotrichum acutatum and Sclerotinia sclerotiorum. However, for Phytophthora capsici and C. fructicola, only M. anisopliae SD3 showed a high antifungal activity against P. capsici, and the other three isolates had little activity. Antibacterial activity against Clavibacter michiganensis subsp. michiganensis was high in two isolates of M. anisopliae but not in two isolates of B. bassiana. Thus, it was confirmed that entomopathogenic fungi effective for pest control could be effectively used as a control agent for various plant diseases.
Overwintering adults of sycamore lace bug (Corythucha ciliata) infected by an unidentified pathogenic fungus were found on the stems of street trees of sycamore in Cheongju city. The objective of this study was to describe this entomopathogenic fungus infecting overwintering sycamore lace bug adults. This unidentified fungus colonized the insect adult body and formed white colony with subglobose clusters of conidiocarps. The size of conidiocarps was 300 to $400{\mu}m$ and each conidium was 15 to $20{\mu}m$. The conidiospore was globus and 2.5 to $3.0{\mu}m$ in diameter, and the hyphae were 1 to $5{\mu}m$ thick. This fungus was successfully isolated and cultivated on potato dextrose agar medium (PDA). The fungal colony was white and then became light yellow. When conidia from this pure culture were inoculated into the overwintering adults, the fungus formed conidiocarps with the same morphology on the insect body and the lethal rate by the fungus was $88{\pm}16%$. This fungus has over 99% homology with Cordyceps bassiana (imperfect fungal name is Beauveria bassiana) in ITS-5.8s rDNA base sequence. The fungal ecology and the infection process of the fungus into its host need to be clarified before using this fungus as a biological control agent.
Beauveria bassiana infects a number of pest species and is known to produce insecticidal substances, such as the nonribosomal peptides (NRPs) beauvericin and bassianolide. However, most NRPs and their biological roles in B. bassiana remain undiscovered. To identify NRPs that potentially contribute to pathogenesis, the 21 predicted NRP synthetases (NRPSs) or NRPS-like proteins of B. bassiana ARSEF 2860 were primarily ranked into three functional groups: basic metabolism (7 NRPSs), pathogenicity (12 NRPSs), and unknown function (2 NRPSs). Based on the transcript levels during in vivo growth on diamondback moth (Plutella xylostella (Linnaeus)), half of the Group II NRPSs were likely to be involved in infection. Given that the metabolites biosynthesized by these NRPSs remain to be determined, our result underlines the importance of the NRPSome in fungal pathogenesis, and will serve as a guide for future genomic mining projects to discover functionally essential and structurally diverse NRPs in fungal genomes.

Two systemic fungicides, SF1 (Bavistin, a carbandazim fungicide 50% WP, Rallis India ltd., India) and SF2 (Bayleton 25% WP-Triadiamefon, a Triazole compound, Rallis India Ltd., India) were screened for control of muscardine disease in silkworm, Bombyx mori. One and two percent of SF1 and 0.05 and 0.1 % of SF2 in aqueous solution were found to be effective in in vivo condition for the control of the disease. These fungicides, on feeding through mulberry leaves continuously for two days to 4$^{th}$ and 5$^{th}$ instar silkworm larvae inoculated topically with conidia of Beauveria bassiana (4$\times$10$^{6}$ conidia/ml) resulted in reduction in mortality due to muscardine by over 90% as against 100% mortality in inoculated control. SF1 at 1% reduced the mortality by 90% in 4$^{th}$ instar and 91% in final instar silkworm while at 2%, the reduction was 92% and 96%, respectively. SF2 at 0.05 and 0.1 % concentration reduced the mortality by 82 and 88% during 4$^{th}$ instar and by 88 and 92% during 5$^{th}$ instar, respectively.

Entomopathogenic fungi were isolated directly from a cadaver of adult Monochamus alternatus supporting fungal sporulation, using a semi-selective medium and then screened several fungal colonies. The pathogenicity of each fungus was tested using oak longicorn beetle, Moechotypa diphysis, as substitutive insect. As the result, only one of them showed high pathogenicity against M. diphysis, with up to 100% mortality within 21 days of inoculation. Selected fungus was named as MaW1 and identified by Beauveria bassiana using microscopic examination and DNA analysis. Pathogenicity was also evaluated to M. alternatus.
The efficacy of conidia of Beauveria bassiana (Balsamo) Vuillemin strain CS-1 that showed high mortality in laboratory and net house was examined against diamondback moth larvae in the field. Conidia ($4{\times}10^{7}$ conidia/ml) were applied to larvae on chinese cabbage in a 1.5% emulsifiable oil-emulsion amended with 4% clay at a volume of 0.55 L/3.3 $m^{2}$. There were no significant differences among plants and replicates in the deposition of spray droplets on water-sensitive papers or of conidia on leaves and larvae. Weather conditions were rainy and cool during first few days, and then hot and dry. Persistence of conidia was equally short on both leaves and larvae. Nevertheless, treatment of B. bassiana potentially reduced larval populations. But the field efficacy was lower than the laboratory efficacy. This reduced efficacy was ascribed to unfavorable environmental conditions.
