The possibility of substituting potato or sweet potato for the wheat, one of the raw materials for soy sauce, was studied by measuring the amylase and proteolytic activities of Koji. Also optimum conditions of Koji making were determined. It was found that substitution of up to 30% (starch content) of wheat content (15% of the total bean and wheat content) with potato yielded good qulity of soy sauce. Use of more than 30% potato yielded a Koji of low enzymatic activity. This was attributed to the high moisture content of potato. It ws also found that substitution of up to 50% (starch content) of wheat content(25% of the total bean and wheat content) with sweet potato yieldede a good quality of soy sauce. But the taste was inferior to the control (the soy sauce which was made with 50% of bean and 50% of wheat).

A laboratory study was made for developing an fermentative method of producing conventional Korean soy sauce type of product from sandfish and defatted soy bean Koji of Aspergillus oryzae as raw material and enzme source, respectively. As an attempt to shorten the fermentation period, the admixture, consisting of pre-chopped sandfish slurry and the Koji of Asp. oryzae (100:15, wet fish weight/dry Koji weight) with added water equal to the fish weight (v/w), was first allowed, while being agitated at 450rpm, to undergo digestion for a 5 hour period at $50^{\circ}C$ with no added salt and then then, after adding 20% salt (w/v), the hydrolysate mixture was ripened for up to 13 weeks at $30^{\circ}C$ and $45^{\circ}C,$ At intervals, an aliquot was withdrawn for determining microbiological, chemical and organoleptic changes taking place in the sandfish-defatted soy bean koji mixture during both digestion and ripening period.
The superior materials for substitution of bean and red pepper paste were selected through the examination of chemical compositions and the function. The following results were obtained. (1) The enzyme activities were higher in the process of koji manufacturing for brewing bean paste and red pepper paste in barley koji and in corn powder koji, but those of wheat koji were inferior (2) Corn powder was proved as the most excellent substitute, because its components and functioning were the best. (3) In the production of the paste the use of corn powder and bean with equal amount was proved as good as those product of the traditional way of the brewing. (4) By using the corn powder, the production cost can be cut down to 25% in the production of bean paste, and to 23% in the production of red pepper paste, comparing with those of traditional brewing products
The various conditions of substituting wheat gluten for the bean, one of the raw materials for soy sauce manufacture, was studied by measuring the activities of the amylase and proteolytic enzyme of koji. It was found that substitute wheat gluten for up to 60% of bean content (30% of the total bran and wheat content) yielded good quality of soy sauce. By using more than 30% of wheat gluten the availability of nitrogen of raw materials was decreased. This was attributed to the low enzyme activity in koji containing more than 30% wheat gluten.
The possibility of substituting raw materials for soy sauce by corn gluten was studied by measuring the amylase and proteolytic activities of koji. Also optimum conditions of koji making were determined. It was found that substitution of up to 30% of the defatted bean content (or 15% of the total bean and wheat content) with corn gluten yielded a good quality of soy sauce. Use of more than 15% corn gluten (based on total bean and wheat content) yielded a soy sauce of poor taste and low nitrogen content even though corn gluten has a high nitrogen content. This drop in nitrogen was attributed to the low enzyme activity in koji containing more than 15% corn gluten and the difference in availability of nitrogen in bean compared to corn gluten.

The purpose of this study was to compare the quality characteristics of each brown rice glutinous rice red pepper paste prepared. In this study, brown rice, glutinous rice, and red pepper paste were prepared with various enzyme preparations. A 20-day study of the physicochemical and microbiological properties of each enzyme through the results process showed significant differences. In particular, brown rice red pepper paste manufactured using cozies purchased from Sunchang was functionally superior to rice cozies and bean cozies, with probiotics of 7.1-7.5×107 CFU/mL, lactobacillus of 1.9-2.2×103 CFU/mL, 48.67-52.30%, and 58.57-63.90%. When the results were combined, the composition of microorganisms and enzymes of fermented soybean paste purchased from Sunchang was not confirmed, but brown rice chili paste with nutritional and functional properties could be prepared compared to A.oryzae and A. kawachii.
Ke Ho Lee and Kun Hyung Chang (Army Research and Testing Lab.): Studies on koji for optimum condition of growth and identification of Aspergillus spp. Cpart 2). Kor. J. Microbiol. Vol. 2, No. 1, p. 17-18 (1964). The experiments reported in this paper are concerned with the identifications, the growth conditions and lethal temperatures of the three strains of Aspergillus spp. which have been isolated from the soils and Meju (fermented soy bean in Korea). The results obtained in the light of the manual of Raper and Thom for identification of the Genus, Aspergillus, have been shown that the three strains of Aspergillus spp. are pertained to Aspergillus flavusoryzae group. The optimum temperature, pH and the lethal temperatures of the Aspergillus spp. have been measured.
As a series of fundamental research projects to produce doenjang (Korean fermented soy paste) of better quality, two kinds of doenjang were manufactured from a traditional meju (Korean soy bean koji) and the mixed with Aspergillus oryzae and Bacillus natto, and histological changes in the cell structures of soy bean of the two were reported doenjang samples were observed and compared during the entire period of fermentation processes. Cell walls of the soy bean were ruptured by pressure and heat during the pressure cooking process and some of them were observed to have the ghost-like shapes. Remarkable differences in the plasmolysis of the cytoplasms were observed between the seed coat and the inner part of soy bean. Small vacuoles resulting from the fusion of the glycoprotein globules by protease and from the hydrolysis of the starch granules by amylase were also observed. Penetration of microorganisms was transferred from the seed coat to the inside of soy bean as the fermentation proceeded. Slimy substances were observed on the seed coat and the parenchyma cells of soy bean fermented with the mixed with Aspergillus oryzae and Bacillus natto. Cell walls of soy bean became difficult to stain and they showed unusual, polygonal shapes as the fermentation proceeded. Samples fermented with the mixed with Aspergillus oryzae and Bacillus natto showed more remarkable tendencies than traditional meju.
Changes of Enzymatic activities and chemical components during Koji preparation of soy sauce with various ratio of soy bean and wheat were studied as first step for checking the current ratio of raw materials for improved soy sauce and determination of proper ratio of them in the respect to its quality and taste, and following results could be obtained. 1. The Protease in the dryed Koji were mainly conmposed of a part which active at the neutral (about pH 6.0) range, while parts which active at acid and alkaline side were inferior. The more amount of wheat increases as raw materials of Koji, the stronger Protease activities of acid and neural side were, while the weaker alkaline side were. 2. Activity of Enzymes were increased rapidly in earlier stage then gradually in later stage or Protease and ${\beta}-Amylase$ rapidly throughout except drying of ${\beta}-Amylase$ during the course of Koji preparation. The more amount of wheat as raw material increases, the stronger Protease and ${\beta}-Amylase$ activity except. ${\beta}-Amylase$ were. 3. Reducing sugar, amino nitrogen, total nitrogen were increased, while total sugar were decreased during the course of Koji preparation. 4. The more amount of wheat increases as raw materials, the more increase reducing sugar, total sugar were, while the total nitrogen were decreased, no noticeable differences were observed in the amino nitrogen among the dryed Kojies.
This study was carried out to investigate effects of iron content on the quality of soy sauce, bean paste and red pepper paste, and to elucidate the origin of iron and change of the contents during production processes. For the first step, the iron contents in commercial soy sauce and changes of the contents during brewing process were determined. The results obtained were as follows. 1, Iron contents of raw materials were 108 ppm in soy bean, 133ppm in defatted soy bean, 79 ppm in wheat, 5 ppm in sodium chloride, 58 ppm in seed koji, 300-2000 ppm in spore of Aspergillus oryzae, 240 ppm in wheat gluten, 20 ppm in sodium carbonate (above figures were of dry weight basis), 6 ppm in hydrochloric acid, 18 ppm in caramel and 0.3ppm in brewing water respectively. 2, Iron contents in koji were 200-240 ppm (as dry weight basis) and increased, more or less, in progress of koji-making period. 3. Iron contents in the mashes during fermentation were 40 rpm after 1 month, 43-47 ppm after 3 months and 49-62ppm after 6 months. 4. In chemical soy sauce, the iron content was 159 ppm after hydrolysis of wheat gluten with hydrochloric acid, and 184 ppm after neutralization. 5. Higher iron contents were detected both in fermented and chemical soy sauce when the concentration of total nitrogen increased, but the levels were higher in chemical soy sauce than in fermented one at the same concentration of total nitrogen. 6. In the case of fermented soy sauce, the iron content in the filtrate was decreased by press-filtration, but no significant change was found between before and after heat-sterilization. 7. Iron contents in commercial soy sauce were varied with the producers, however, the average value was 62.7 ppm as calculated as 1.0 percent of total nitrogen. And the average level of iron in home-made soy sauce produced by conventional method was 37.68 ppm.
