Development of a Unit Module for the Process of Large Scale Dialysis for Silk fibroin (실크 피브로인의 대규모 투석 공정의 단위 모듈의 개발)

  • For the development of a large-scale dialysis process for silk fibroin solution, a batch and continuous dialysis chamber was designed and built, which are to be used as a unit module for the process. A series of dialysis experiments were carried out to estimate the kinetic parameters for dialysis using the solutions of salt-solubilized silk fibroin. The solution was dialyzed by distilled water either batch-wise or continuously. Dialysis kinetics was monitored by measuring refractometery at high salt concentration and conductivity at lower salt concentration. The apparent dialysis coefficients were $5.4g/m^2-hr$ and $1.8g/m^2-hr$, for well mixed batch and continuous dialysis, respectively. The alteration of molecular weight distribution of dialyzed silk fibroin solution was monitored.

Effects and Batch Kinetics of Agitation and Aeration on Submerged Cultivation of Ganoderma Iucidum (영지의 액체배양에 미치는 통기.교반의 효과와 동력학적 특성)

  • The effects of agitaion and aeration on mycelial growth, exo-polysaccharide (EPS) production, and substrate consumption upon the submerged cultivation of G. lucidum were investigated, and the batch kinetics of the EPS fermentation of G. lucidum were interpreted as function of agitation speed and aeration rate. In a 2.6 L jar fermenter system, the optimum agitation speed and aeration rate for EPS production were determined to be 400 rpm and 1.0 vvm, respectively. The maximum production of EPS obtained was 15 g/L. The logistic model for mycelial growth fitted the experimental data better than that determined by the Monod and the two-thirds power models. The Luedeking-Piret equation adequately modelled the kinetic data obtained for product and substrate.

  • Combustion performance of an internally cycloned circulating fluidized bed for paper sludge was discussed through a series of batch type experiments. Operation parameters such as water content, feeding mass of sludge and secondary air injection rate were varied to find out the effect on the combustion performance, which was examined with carbon conversion rate and pollutant emission such as CO and NOx. A conventional solid fuel reaction was observed in the experiments of varying water content and feeding mass of the sludge, which is characterized with kinetic limited reaction zone, diffusion limited reaction zone and transition zone. Secondary air injection with swirl enhances the mixing of the gas phase as well as the solid phase, and improves combustion efficiency accompanied with higher carbon conversion rate and lower pollutant emission rate.

  • To optimize the culture conditions for Beauveria bassiana 726, the effects of culture medium, pH, and temperature on mycelium and spore production were investigated. The optimum temperature and pH for the cultivation of B. bassiana 726 were 28 $^{\circ}C$ and 5.0, respectively. The optimized medium was composed of 1.0~2.0% total sugar from molasses, 0.5% corn steep liquor and 0.05% KH$_2$PO$_4$. In the cultivation of B. bassiana 726 with the optimum medium, the specific growth rate and substrate utilization were well-fitted with the proposed kinetic model in the shake flask and stirred tank reactor. When the fed-batch cultivation using carbon suorce, nitrogen source, and mineral salt as a feeding medium was compared with batch cultivation in stirred tank reactor, mycelium (12.7 g/L) and spore production (5.4$\times$$10^8/mL$) were enhanced up to 110% and 85%, respectively.

  • This study proposed adsorption of Pb(II) ions from aqueous solution using activated carbon prepared from areca catechu shell (ACS AC) using Timphan Method. The effects of independent variables on adsorption kinetic and isotherm have been investigated by conducting experiments in batch mode at neutral pH. The structural characterization of adsorbent was done by FT-IR and SEM analysis. The Pb(II) adsorption was correlated very well with the pseudo second-order kinetic (PSOKM) and Langmuir isotherm models (LIM). Increasing NaOH mass for activation and adsorption temperature increased weakly all the parameters of adsorption kinetic and isotherm. The Pb(II) ions adsorption capacity of the ACS AC at 27 and $45^{\circ}C$ was 50.51 and 55.25 mg/g, respectively. Thermodynamic parameters were determined, and the results confirmed the Pb(II) ions adsorption should be endothermic and spontaneous process, and both physical and chemical adsorption should be taken place.

Applicability of adsorption kinetic model for cation/anion for chitosan hydrogel bead (키토산비드를 이용한 양이온/음이온의 흡착모델 적용)

  • Batch adsorption tests were performed to evaluate the applicability of adsorption kinetic model by using hydrogel chitosan bead crosslinked with glutaraldehyde (HCB-G) for Cu(II) as cation and/or phosphate as anion. Pseudo first and second order model were applied to determine the sorption kinetic property and intraparticle and Boyd equation were used to predict the diffusion of Cu(II) and phosphate at pore and boundary-layer, respectively. According to the value of theoretical and experimental uptake of Cu(II) and phosphate, pseudo second order is more suitable. On comparison with the value of adsorption rate constant (k), phosphate kinetic was 2-4 times faster than that of Cu(II) at any experimental condition indicating the electrostatic interaction between ${NH_3}^+$ and phosphate is dominated at the presence of single component. However, when Cu(II) and phosphate simultaneously exist, the value of k for phosphate was sharply decreased and then the difference was not significant. Both diffusion models confirmed that the sorption rate was controlled by film mass transfer at the beginning time (t < 3 hr) and pore diffusion at next time section (t > 6 hr).

Kinetic and Equilibrium Study of Lead (II) Removal by Functionalized Multiwalled Carbon Nanotubes with Isatin Derivative from Aqueous Solutions

  • The carboxylated multiwall carbon nanotubes (MWCNT-COOH) and functionalized with isatin derivative (MWCNT-isatin) have been used as efficient adsorbents for the removal of lead (Pb) from aqueous solutions. The influence of variables including pH, concentration of the lead, amount of adsorbents and contact time was investigated by the batch method. The adsorption of the lead ions from aqueous solution by modified MWCNTs was studied kinetically using different kinetic models. The kinetic data were fitted with pseudo-first-order, pseudo-second-order, and intra-particle diffusion models. The sorption process with MWCNT-COOH and MWCNT-isatin was well described by pseudo-second-order and pseudo-first-order kinetics, respectively which it was agreed well with the experimental data. Also, it involved the particle-diffusion mechanism. The values of regression coefficient of various adsorption isotherm models like Langmuir, Freundlich and Tempkin to obtain the characteristic parameters of each model have been carried out. The Langmuir isotherm was found to best represent the measured sorption data for both adsorbent.

  • The microbial organisms named "Pseudomonas sp. LK-14" were isolated from farm land and shallow river sediment, activated, augmented and identified; which were using 2,4-D (2,4-Dichlorophenoxyacetic acid) as a sole carbon source and energy source. 2,4-D removal efficiency of LK-14 with 2,4-D sole carbon source (reactor S) were higher than that of Activated Sludge with 2,4-D sole carbon source (reactor A). Dynamic bioligical reaction kinetic parameters (sole carbon source was 2,4-D) obtained from batch reactor experiments were ${\mu}_{max}$ $0.105hr^{-1}$, $K_{s,24D}$ 15.64mg/L, $K_{i,24D}$ $1.94h^{r-1}$, $Y_{24D}$ 0.39 for LK-14 and ${\mu}_{max}$ $0.008hr^{-1}$, $K_{s,24D}$ 26.95mg/L, $K_{i,24D}$ $1.75hr^{-1}$, $Y_{24D}$ 0.10 for Activated Sludge. Using these parameters, we could predict the behaviors of 2,4-D substrate utilized by LK-14 and Activated Sludge in batch reactors. The kinetic parameters are enable to predict the 2,4-D substrate and microbial population behavior entering into wastewater treatment plants by using unsteady states dynamic simulation modeling technique.

  • Yarn dyed wastewater has to be treated prior to disposal into the water bodies due to its high content of harmful organic compounds. In this study, the performance of Chemical Oxygen Demand (COD) removal and kinetic rate constant are investigated via hybrid electrocoagulation-Fenton in a continuous system using wastewater discharged from a yarn dyed industry in Surabaya city. The wastewater was treated in a batch mode using electrocoagulation to reduce Total Suspended Solid, followed by Fenton method in a continuous system to reduce COD level. Various Fe(II) feeding modes, molar ratio of $Fe(II)/H_2O_2$, initial pH of wastewater, and flow rate are used in this study. The results show that COD removal process obeys a pseudo-first order kinetics. At $Fe(II)/H_2O_2$ ratio of 1:10, initial wastewater pH of 3.0, and feed flow rate of 30 mL/min, the COD removal efficiency was observed to be 80%, and the kinetic rate constant is $0.07046min^{-1}$. The chemical cost for the treatment estimated to be IDR 160 per L wastewater, which is cheaper than the previously reported batch system of IDR 256/L.

Numerical Model for SBR Aerobic Digestion Combined with Ultrasonication and Parameter Calibration (초음파 결합형 SBR 호기성 소화의 모델과 매개변수의 보정)

  • Based on the activated sludge model(ASM), a mathematical model which represents the aerobic sludge digestion by sequencing batch reactor(SBR) combined with ultrasonic treatment was composed and performed in this study. Aerobic digestion using sequencing batch reactor(SBR) equipped with ultrasound treatment was also experimented for the purpose of parameter calibration. Most of the presented kinetic parameters in ASM or ASM2 could be used for the aerobic digestion of sludge but the parameters related in hydrolysis and decay rate needed modification. Hydrolysis rate constant of organic matter in aerobic condition was estimated at $0.3day^{-1}$ and the maximum growth rate for autotrophs in aerobic condition was $0.618day^{-1}$. Solubilization reactions of particulate organics and nitrogen by ultrasonication was added in this kinetic model. The solubilization rate is considered to be proportional to the specific energy which is defined by specific ultrasound power and sonication time. The solubilization rate constant by ultrasonication was estimated at $0.202(W/L)^{-1}day^{-1}$ in this study. Autotrophs as well as heterotrophs also decomposed by ultrasonic treatment and the nitrification reaction was limited by the lack of autotrophs accumulation in the digester.
