Some Observations on the Fine Structure of Korean Bat Livers (한국산 박쥐 간장의 미세구조적 고찰)

  • To understand the fine structure of Korean bat livers , the authors investigated the liver of four Korean bats; Rhinolephus ferrumequinum korai, Myotis macrodactylus, Myotis dauhentonii ussrinesis, and Murina leucogaster intermedia by transmission electron microscopy. The hepatocytes of Rhinolophus ferrumequinum korai had large-sized mitochondria and many peroxisomes. In the Myotis macrodactylus, juctional completes, especially desmosomes, were well developed. The Myotis daubentonii ussrinesis had many glycogen particles in the cytoplasm. Also, the space of Disse and sinusoidal spare was filled with amorphous materials. In the Murina leucogaster intemedia, basement membrane was prominent in the sinusoid, and no Kupffer and Ito cells were observed These results suggest the characteristic differences in the liver ultrastructure among Korean bats.

Study on the Pathogenesis of Hantaan Virus with Monoclonal Antibodies (단일항체를 이용한 한국형출혈열의 병인성 연구)

  • Hantaan virus(HV) 76-118 strain was inoculated into suckling ICR mice by intra-nasal route with an inoculum of $10LD_{50}$. Mortality was 65% at the 3rd week after inoculation, but declined to 35% at the 4th week. Infectivity was determined by the measuring immuno-fluorescent antibody in sera. The peak of infectivity was 80% at the 4'th week after inoculation. Viremia was reached peak level of $1.7{\times}10^4\;PFU/ml$ by day 10. Immunofluorescent antibody and neutralizing antibody appeared by 2 weeks and 15-17 days respectively, but achieved similar titer by 35 days. By using a monoclonal antibody to HV 76-118, viral antigens were initially detected in inguinal and axillary lymph node by 2 days. Viral antigens in bone marrow and lung were delayed much more than in those of lymph node. These were similar with those of intra-peritoneal and intra-muscular route. Immune complex against IgG, IgM and C3 appeared by 16 days, 14 days, and 18 days respectively. The pattern of immunofluorescence in the basement membrane of glomeruli was diffuse membranous. Spotted pattern was also observed in the tissue stained with anti-mouse C3 antibody. By 20 days, control tissue was also shown immune complex in the glomeruli.

Differential Expression of miR-34c and Its Predicted Target Genes in Testicular Tissue at Different Development Stages of Swine

  • To verified the target genes of miR-34c, bioinformatics software was used to predict the targets of miR-34c. Three possible target genes of miR-34c related to spermatogenesis and male reproductive development: zinc finger protein 148 (ZNF148), kruppel-like factor 4 (KLF4), and platelet-derived growth factor receptor alpha (PDGFRA) were predicted. Then, the expression of miR-34c and its target genes were detected in swine testicular tissue at different developmental stages by quantitative polymerase chain reaction. The results suggested that the expression of PDGFRA has the highest negative correlation with miR-34c. Then immunohistochemical staining was done to observe the morphology of swine testicular tissue at 2-days and 3, 4, 5-months of age, which indicated that PDGFRA was mainly expressed in the support cells near the basement membrane during the early development stages of testicular tissue, but that the expression of PDGFRA was gradually reduced in later stages. Therefore, western blot analyzed that the highest expression of PDGFRA was generated in 2-days old testicular tissues and the expression levels reduced at 3 and 4-months old, which correlated with the results of immunohistochemical staining. In conclusion, PDGFRA is a target gene of miR-34c.

Resveratrol blunts tumor necrosis factor-${\alpha}$-induced monocyte adhesion and transmigration

  • The leukocyte recruitment and transmigration across the endothelial barrier into the vessel wall are crucial steps in atherosclerosis. Leukocyte trafficking on the endothelium is elicited by induction of endothelial adhesion molecules, and its transmigration is mediated by degradation of basement membrane proteins through enzymatic activity of matrix metalloproteinases (MMP). The current study investigated whether resveratrol, a polyphenol present in grapes and red wine, was capable of inhibiting leukocyte adhesion to tumor necrosis factor (TNF)-${\alpha}$-activated endothelium. It was found that resveratrol inhibited the TNF-${\alpha}$-activated endothelial expression of vascular cell adhesion molecule-1 in a dose-dependent manner. In addition, resveratrol hampered THP-1 monocyte adhesion to activated endothelial cells. This study further examined whether resveratrol interfered with transendothelial migration of leukocytes. The MMP-2 gelatinolytic activity of endothelial cells was enhanced by TNF-${\alpha}$, which was attenuated by an addition of ${\geq}25{\mu}M$ resveratrol. In addition, 25 ${\mu}M$ resveratrol mitigated the MMP-9 activity of THP-1 cells, followed by a marked inhibition of transendothelial migration. These results demonstrated that resveratrol suppressed monocyte adhesion and migration induced by TNF-${\alpha}$ through modulating expression of adhesion molecules and gelatinolytic activity of MMP. These findings suggest that dietary resveratrol may be therapeutic agent for inhibiting leukocyte recruitment into the subendothelium during inflammatory atherosclerosis.

A case of transitional carcinoma in the nasal cavity of a dog (개의 비강 내 이행암종 증례)

  • A 10-year-old female Yorkshire terrier with the clinical signs of nasal swelling, epistaxis and nasal discharge was presented to the Veterinary Teaching Hospital in the Cheju National University. Abnormal nasal mass was detected in physical examination and radiographic findings. After surgical excision, the sample of nasal mass was referred to Pathology Department of Veterinary Medicine. Grossly, the mass was soft, friable, and $2.5{\times}4cm$ cm in size. Histopathologically, the mass was composed of mediumsized non-keratinizing columnar to polyhedral cells arranged in anastomosing ribbon and large nest. It has complex in-folding of thick epithelial layers separated by fibrovascular septa. Tumor cells showed characteristic palisading arrangement of columnar cells, and perpendicularly distributed to the basement membrane. The cells had pale basophilic cytoplasm, oval nucleus and one or more nucleoli, and indistinct cellular border. Many tumor cell emboli were presented in lymphatics. Immunohistochemistry revealed that tumor cells were cytokeratin (CK) 19 and CK clone MNF116 positive and but CK7 and CK high molecular weight negative. Based on the gross, histopathologic, and immunohistochemical findings, the mass was diagnosed as transitional carcinoma in nasal cavity. In our best knowledge, this is the first report of transitional carcinoma originated from transitional zone of canine nasal cavity in Korea.

AN IMMUNOHISTOCHEMICAL STUDY ON PROLIFERATING CELL NUCLEAR ANTIGEN, ${\alpha}$-1-ANTICHYMOTRYPSIN, FIBRONECTIN, TRANSGLUTAMINASE IN INFLAMMED GINGIVA (염증성 치은에서 Proliferating Cell Nuclear Antigen(PCNA),${\alpha}$-1-antichymotrypsin, Fibronectin, Transqlutarninase의 분포에 관한 면역조직화학적 연구)

  • Recently, available interests concerning the biologic significance of the extracellular matrix and proliferating cells associated with periodontal disease has been increased. The distribution or expression of cellular proliferation by PCNA, macrophage detection by ${\alpha}$-l-antichymotrypsin, fibronectin playing a important role in host defence mechanisms indirectly, and transglutaminase that cross linked to fibronectin and stimulate fibrin stabilization were studied in inflammed and healthy gingiva. The excised tissue samples were fixed neutral formalin for 24 hours, embedded with paraffin, sectioned at 4-61lffi in thickness, and immunohistochemically processed by LSAB method. The positive reaction to PCNA was localized in the suprabasal and basal layer of inflammed gingiva and an increasing reactivity was observed than healthy gingiva. ${\alpha}$-I-antichymotrypsin positive cells were localized in the basal layer of inflammed gingiva, and there was no or rare positive cells in healthy gingiva. The positive reaction to fibronectin in inflammed gingiva was more than healthy gingiva,"and shown in the connective tissue subjacent to basement membrane of epithelium and in the periphery of the collagen fiber bundles. The positive cells by transglutaminase in inflammed gingiva were noted in suprabasal, spinous, and keratin layer of epithelium, and slightly increased in the capillaries of connective tissues. But the results of this study demonstrated in vitro reaction. Therefore, the role of PCNA,${\alpha}$-l-antichyrnotrypsin, transglutaminase, fibronectin and coefficient with other growth factor and extracellular matrix were further investigated in vivo.

Podocytopathy and Morphologic Changes in Focal Segmental Glomerulosclerosis (초점분절사구체경화증에서 발세포병증과 형태 변화)

  • Podocytopathy is glomerular lesions characterized by podocyte injury. It is observed in various glomerular diseases, but minimal change disease and focal segmental glomerulosclerosis (FSGS) are the prototypes. In this review, morphologic features of podocyte injury and subtypes of FSGS will be reviewed briefly. Effacement of podocyte foot processes is the most common feature of podocyte injury. As podocytic injury progresses, intracytoplasmic vacuoles, subpodocytic cyst, detachment of podocytes from the glomerular basement membrane and apoptosis develop. Glomerular capillary loops in epithelium-denuded area undergo capillary collapse. Synechia and hyalinosis may accompany this lesion. To manifest segmental sclerosis, podocyte loss above a threshold level may be required. Injured podocytes can injure neighboring intact podocytes, and thereby spread injury within the same lobule. FSGS can be categorized into five subtypes by morphologic characteristics; not otherwise specified (NOS), perihilar, cellular, tip, and collapsing types. Each subtype has been reported to show different clinical courses and associated conditions, but there are controversies on its significance. With recent progress in the discovery of genetic abnormalities causing FSGS and plasma permeability factors, we expect to unravel pathophysiology of FSGS and to understand histological sequences leading to FSGS in near future.

The Effect of Silk Fibroin-Alginic Acid Sponge Treatment as an Artificial Skin on Wound (인공피부용 실크 피브로인-알긴산 해면체의 창상치유 효과)

  • Purpose: This study was designed to compare the wound healing effect of silk fibroin, alginate and fibroin/ alginate blend sponge with clinically used Nu gauze in a rat skin defect model. Methods: Two full thickness excisions were made on the back of Sprague-Dawley rat. The excised wound was covered with either of the silk fibroin(SF), alginate (SA), or fibroin/alginate blend sponge(SF/SA). On the postoperative days of 3, 7, 10 and 14, the wound area was calculated by image analysis software. At the same time, a skin wound tissue was biopsied. Results: Healing time 50% ($HT_{50}$) of SF/SA sponge treated group was dramatically reduced as compared with that of control treatment. We also found that the $HT_{50}$ of SF/SA sponge was significantly decreased as compared with either those of SF or SA treatment. Furthermore, SF/SA treatment significantly increased the size of epithelialization and collagen deposition as well as the number of PCNA positive cells on epidermal basement membrane as comapred with those of control treatment. Conclusion: Our results suggest that the wound healing effect of SF/SA blend sponge is the best among other treatments including SF and SA during the whole wound healing period.

Functional characterization of the distal long arm of laminin: Characterization of Cell- and heparin binding activities

  • Basement membrane laminin is a multidomain glycoprotein that interacts with itself, heparin and cells. The distal long arm plays major cell and heparin interactive roles. The long arm consists of three subunits (A, B1, B2) joined in a coiled-coil rod attached to a terminal A chain globule (G). The globule is in turn subdivided into five subdomains (Gl-5). In order to analyze the functions of this region, recombinant G domains (rG, rAiG, rG5, rGΔ2980-3028) were expressed in Sf9 insect cells using a baculovirus expression vector. A hybrid molecule (B-rAiG), consisting of recombinant A chain(rAiG) and the authentic B chains (E8-B)was assembled in vitro. The intercalation of rAiG into E8-B chains suppressed a heparin binding activity identified in subdomain Gl-2. By the peptide napping and ligand blotting, the relative affinity of each subeomain to heparin was assigned as Gl> G2= G4> G5> G3, such that G1 bound strongly and G3 not at all. The active heparin binding site of G domain in intact laminin appears to be located in G4 and proximal G5. Cell binding was examined using fibrosarcoma Cells. Cells adhered to E8, B-rAiG, rAiG and rG, did not bind on denatured substrates, poorly bound to the mixture of E8-B and rG. Anti-${\alpha}$6 and anti-${\beta}$1 integrin subunit separately blocked cell adhesion on E8 and B-rAiG, but not on rAiG. Heparin inhibited cell adhesion on rAiG, partially on B-rAiG, and not on E8. In conclusion, 1) There are active and cryptic cell and heparin binding activities in G domain. 2) Triple-helix assembly inactivates cell and heparin binding activities and restores u6131 dependent cell binding activities.

Development of the rumen of fetuses and neonates in Korean native goats (한국재래산양의 태아 및 신생아의 제1위 발달에 관하여)

  • The development of rumen in fetuses between 60, 90, 120 days of gestation and neonates of Korean native goats was investigated by light, scanning electron microscopy. The results were summarized as follows; 1. In the 60-day-old fetuses, the stomach was developed and differentiated into four compartments of rumen, reticulum, omasum, and abomasum. The ruminal epithelial layers were differentiated into two zones; a small dark basal and a large light luminar zones. 2. In the 90-day-old fetuses, the light luminar zone of the rumen was 6-12 times thicker than the dark zone. 3. In the 120-day-old fetuses, the wall of the rumen had increased in thickness, and its the mucosa was still smooth. Several undulations of various depths involving the basal zone, basement membrane and lamina propria were observed at irregular within the light zone. 4. In the neonate, the wall of the rumen had increased in thickness and the luminar surface had become wavy due to the appearance of several shallow furrows in the luminar surface of the epithelium especially between the tips of the relatively more developed undulations. 5. Scanning electron microscopic studies: In the 90-day-old fetuses, numerous microvilli and microridges were observed on the superficial epithelial layer of the rumen. The ruminal papillae were already formed at 120 days of gestation. In the 120-day-old fetuses, the ruminal papillae formed short dome-shape. In the neonate, the ruminal papillae formed pebble-shape or short finger like.
