Canavanine Effects on the Amylase Activity and Protein Content in Barley Half Seeds (Canavanine에 의한 보리 무배부 종자의 Amylase 활성과 단백질 함량의 변화)

  • L-canavanine was added to GAs treated barley seeds, and induced amylase activity, soluble protein content, and arginine content were mesured. Canavanine, added at the beginning of the incubation period, inhibited amylase activity and protein accumulation. Amylase activity decreased markedly by addition of canavanine at 6 hr after incubation, where soluble protein content was not affected. The addition of canavanine after 12 hr incubation did not show serioud inhibited effect on the amylase activity and protein accumulation. GAs incubation caused decrement in arginine content per mg protein, but it was somewhat recovered by canavanine treatment. The longer the time between GAs and canavanine addition was, the less the recovery ration was. Arginine content in the $\alpha$-amylase fraction (ammonium sulfate 20~50% saturation) was lower than in 0~20% fraction, but higher than in 50~80% fraction. These results and control expreiments, using cordycepin and cycloheximide, support the idea that canavanine might incorporate into protein.

History of Korean brewing industry (식품산업 발전사 - 국내 맥주 양조사)

  • During the Japanese colonial period, Japanese brewing companies established 2 breweries in Korea, and brought in malting barley seeds, began trial cultivation in near Daejeon area and Jeju Island, and cultivated hops in Hyesan, Hamgyeong-do. In 1933, Chosun Brewery Co. Ltd. and Showa-Kirin Brewery Co. were established in Yeongdeungpo by Japan and these 2 breweries became the birthplace of today's Hite Brewery and Oriental Brewery. Oriental Brewery tried to cultivate hops and malting barley in Korea, and once self-sufficient but now hops depend on imports, and production of domestic malting barley is about 5% of demand. Beer was only about 5% of domestic alcohol consumption in the early 1970s, but now occupies more than 50% and became the most popular alcoholic drink in Korea. Recently various beers are imported from the world, and many craft breweries serving variety of beers to consumers, thus Korean enjoy heyday of the beer culture.

Physicochemical Properties of Starch Granules Isolated from Naked Barley Seeds during Germination (쌀보리의 발아과정중 분리전분의 이화학적 특성)

  • Physicochemical properties of starch isolated from the naked barley seeds during germination were investigated, The germinated barley seeds showed an apparent increase in amylose component of the starches, and water binding capacities of the starches decreased initially and then increased with longer germination time. The size frequency distribution of starch granules showed that the granules larger than $20.2{\mu}$ in diameter were preferentially degraded, and moreover the rate of degradation was higher than that of granules of any other size. It was apparent that the mean diameter of granules was $12.7{\sim}20.2{\mu}$ at all stages of germination. The results of differential scanning calorimetry, swelling power, solubility and X-ray diffraction patterns of starch granules isolated at various stages of germination showed little changes during germination. Scanning electron microscopy revealed that the starch granules have been damaged extensively by degradative enzymes during germination, but all the starch granules were not degraded uniformly.

Effects of Sprouting Degree and Threshing Methods on Germination and Seedling Vigor in Barley (보리의 수발아정도 및 탈곡방법이 종자활력 관련 형질에 미치는 영향)

  • In order for investigating the factors related to decrease of seedling vigor in the naked barley lines, two naked lines and a covered variety were sprouted with 20 mm rainfall in the natural condition, dried and the sprouted seeds were threshed by hand and small machine thresher, Chiyota. In the case of hand-threshing of the sprouted seeds, the germination rate index (GRI), germination rate, and TTC test value of the different varieties were not different due to the different sprouting degrees, but emergence rate index (ERI) became decreased with stronger sprouting damage. In comparison with hand-threshing, the embryos of sprouted seeds from machine threshing with 700 rpm were partially damaged, and the traits related to seedling vigor were decreased highly significantly. However, the diastatic activity was sharply increased by stronger sprouting and machine threshing. The sprouting degree in the hand-threshing plot was significantly correlated with the diastatic activity(r=0.931**, 0.951**) and GRI(r=0.461), respectively.

Eco-Friendly Control of Water Foxtail (Alopecurus aequalis) on Cultivation Fields of Forage Barley Sowed before Rice Harvest (벼 수확전 청보리 파종 재배시 친환경적 뚝새풀 관리)

  • This study was conducted to develop an eco-friendly control method for water foxtail in the field sowing barley seeds before rice harvesting. When sea water was applied pre-emergent, 1 and 2 leaves, little injury was observed on barley. Percent of water foxtail control with sea water (100%) was more than 50% at 1 leaf application timing. When sodium chloride was applied 400, 800 and $1,600kg\;ha^{-1}$, little injury was observed on barley. Percent of water foxtail control with sodium chloride was 36-44% at 30 days after sowing. When ferrous sulfate was applied at 400, 800 and $1,600kg\;ha^{-1}$, emergent injury was observed 35-50% on barley. Percent of water foxtail control with ferrous sulfate was 48-79% at 30 days after sowing. When barley was sowed at 200, 300 and $400kg\;ha^{-1}$, seeding rate standing plants have many of the more crops, water foxtail occurrence was low. And, Forage yield of 300 kg ha-1 seeding in the highest. Based on the results, it is possible to control water foxtail in the field sowing barley seeds before rice harvesting more than 50%.

Serological detection of barley stripe mosaic virus infection in the seeds of barley and wheat cultivars grown in Korea (혈청학적방법에 의한 보리와 밀종자의 보리${\cdot}$줄무늬 모자이크 바이러스 감염상조사)

  • An attempt was made to find out whether barley stripe mosaic virus (BSMV) occurs in barley and wheat cultivars in Korea. Modified Hamilton's technique for serological detection of barley stripe mosaic virus was used to assay for BSMV in mature seed embryos. Of the 51 barley and wheat seed lots assayed, BSMV was detected in 34 of them. BSMV was detected in covered, naked, and matting barley as well as in wheat. The level of BSMV infection in the infected cultivars varied from $2\~35\%$. This is the first reported occurrence of barley stripe mosaic virus in Korea.

Inhibition of Seed Germination and Induction of Systemic Disease Resistance by Pseudomonas chlororaphis O6 Requires Phenazine Production Regulated by the Global Regulator, GacS

  • Seed coating by a phenazine-producing bacterium, Pseudomonas chlororaphis O6, induced dose-dependent inhibition of germination in wheat and barley seeds, but did not inhibit germination of rice or cucumber seeds. In wheat seedlings grown from inoculated seeds, phenazine production levels near the seed were higher than in the roots. Deletion of the gacS gene reduced transcription from the genes required for phenazine synthesis, the regulatory phzI gene and the biosynthetic phzA gene. The inhibition of seed germination and the induction of systemic disease resistance against a bacterial soft-rot pathogen, Erwinia carotovora subsp. carotovora, were impaired in the gacS and phzA mutants of P chlororaphis O6. Culture filtrates of the gacS and phzA mutants of P. chlororaphis O6 did not inhibit seed germination of wheat, whereas that of the wild-type was inhibitory. Our results showed that the production of phenazines by P. chlororaphis O6 was correlated with reduced germination of barley and wheat seeds, and the level of systemic resistance in tobacco against E. carotovora.

Vigor Determination in Barley Seeds by the Multiple Criteria (보리 종자세 검정방법 비교)

  • The seeds of three barley varieties of which initial seed vigor were different were used to measure seed vigor of accelerated aging techniques. A vigor index derived from the product of percent germination and plumule length was used to estimate seed vigor. The index was compared with the results of other tests. The results of warm germination test well suited to the measurements of seed vigor at the advanced stages of seed deterioration. Other vigor measurements except ATP and GADA values, showed close relationships with the vigor index. The measurements of plumule length in cold test and tetrazolium test were found to be appropriate for predicting seed quality.

Spatiotemporal Patterns of Starch Deposition in Amaranth Grains (Amaranthus cruentus L.)

  • In this study, we investigated whether there is another amaranth GBSS isoform in an attempt to characterize the synthesis of amylose in the pericarp. We used I2/KI staining to analyze the temporal and spatial starch accumulation patterns during seed development. The spatiotemporal starch accumulation patterns in developing seeds were observed by staining with I2/KI. Starch granules were observed in the pericarp in the initial developmental stage (3 DAP). A few starch granules were detected in the perisperm in the early-late developmental stage (8 DAP), during which the pericarp starch contents rapidly decreased. Starch granules were distributed throughout the perisperm in the mid-late developmental stage (15 DAP). Similar results were reported for other cereal crops, including barley, rice, and sorghum. Starch granules in the pericarp are synthesized during the early seed developmental stages but are absent in mature seeds. We recently reported that starch deposits in the perisperm of developing amaranth seeds are detectable only after the initial developmental stage. Prior to this stage, the pericarp is the major site of starch deposition. A recent study suggested that GBSSII isoforms are responsible for amylose synthesis in pericarps.
