Evaluation of Bacillus subtilis Native Strains for Plant Growth Promotion and Induced Systemic Resistance in Tomato and Red-pepper (토마토, 고추의 생육촉진 및 병 저항성 의 농업적 활용을 위한 토착 Bacillus subtilis의 생물활성 평가)

  • Bacillus subtilis strains isolated from different regions of Korea were screened for their plant growth promotion and induced systemic resistance (ISR) in tomato and red-pepper. The plant growth promotion on red-pepper and tomato revealed maximum plant height (22.73 cm) on red pepper treated with B. subtilis strain JE 21-1 and 30.18cm in case of tomato treated with B. subtilis strain JE 8-1. There was also significant improvement in root and shoot dry weight in both the plants. The strain JE 21-1 showed better promise for all growth parameters in red-pepper and tomato when compared to other strains and positive check BTH. Different strains screened in square plate method also revealed maximum plant height and leaf width, and suppressed anthracnose on red pepper in case of strain JE 21-1 at $10^6$ and $10^7$ cells/ml when compared to other strains. In all the bacterial inoculations the population was significantly high when compared to untreated check. In plant growth promotion with respect to fruit length and weight, fruit length was maximal in treating with JE 9-4 and ES 2-2, while fruit weight was maximal in treating with JE 3-6, ES4-2, ES2-2 and JE 21-2 on red pepper. In case of tomato, comparatively better fruit weight was in JE 21-1, ES 3-3 and JE 10-2 when compared to BTH and untreated control. The soft rot disease caused by Pectobacterium carotovorum SCCI was completely suppressed in case of transgenic tobacco harboring GUS gene related to PR1a and increased the level of salicylic acid significantly in combined application of JE 9-4 on par with BTH. Thus, this study clarified some potential Bacillus subtilis strains for plant growth promotion and ISR in red-pepper and tomato.

Clinical Manifestations of Invasive Infections due to Streptococcus pyogenes in Children (소아에서 발생한 A군 연쇄구균에 의한 침습성 질환의 임상적 특성 분석)

  • Purpose: Streptococcus pyogenes is an important cause of invasive diseases in children. We aimed to describe the clinical characteristics of invasive infections due to S. pyogenes in children in Korea. Methods: A retrospective study of children under 18 years of age with invasive infections due to S. pyogenes at Seoul National University Children's Hospital between March 1992 and December 2012, and Seoul National University Bundang Hospital between March 2003 and December 2012 was conducted. Demographic factors, clinical characteristics, laboratory findings, treatment, mortality and morbidity of all patients were reviewed. Results: A total of 30 among 36 cases identified as invasive disease due to S. pyogenes were available for review. There was a predominance for male subjects (male:female=2.75:1). The median age was 50 months (range 12 days to 15 years) and 53.3% were under 5 years of age. Skin and soft tissue infections (9/30, 30.0%), bacteremia without identified focus (4/30, 13.3%) and bone and joint infections (6/30, 20.0%) were the most frequent clinical presentations. Streptococcal toxic shock syndrome (3/30, 10.0%) pulmonary, abdomen and central nervous system infections (2/30, 6.7%) were also seen. There was a peak in number of patients in year 2012 (9/30, 30.0%). There were no cases of mortality. Erythromycin and clindamycin resistance rates were low by 3.8% and 7.5%, respectively. Conclusion: We studied the clinical presentations of invasive infections due to S. pyogenes during the past 20 years in Korean children. The findings of this study help us understand the characteristics of the disease, enhancing early recognition and prompting adequate antibiotic therapy which is important in reducing morbidity and mortality.

Relationship between the Oral Cavity and the Stomach of Helicobacter pylori (구강과 위내 Helicobacter pylori의 상호관련성)

  • Helicobacter pylori(H. pylori) is bacterial infection, with more than half of the world population infected and oral cavity is considered second reservoir of H. pylori infection. The purpose of this study was to evaluate role of oral cavity in H. pylori infection by comparison of the mode H. pylori infection in oral cavity and stomach. We recruited 100 subjects without systemic disease including gastrointestinal disease. Samples in oral cavity taken on gingival sulcus fluid(GSF) of lower left central incisor and 1st molar, area of buccal mucosa, dorsum of the tongue, palatal and saliva. We analyzed by Nested polymerase chain reaction(PCR) for oral infection and Urea Breath Test(UBT) for gastric infection. The results were as follows : 1. Among these 100 subjects, 36(36%) were positive by Nested PCR and 33(33%) were positive by UBT(p>0.05). 2. In detection rate of H. pylori in sites taken sample, 11(11%), 8(8%), 9(9%), 3(3%), 9(9%), 7(7%) were positive on GSF of lower left central incisor and 1st molar, area of buccal mucosa, dorsum of the tongue, palatal and saliva, respectively. Statical significance was observed in samples of GSF of lower left central incisor and area of dorsum of the tongue(p<0.05). 3. In comparison of the mode of H. pylori infection in oral cavity and stomach by analytic method, positive in oral cavity and stomach was 10(10%), negative in oral cavity and positive in stomach was 23(23%), positive in oral cavity and negative in stomach was 26(26%) and negative in oral cavity and stomach was 41(41%)(p>0.05). Conclusively, we can guess that oral H. pylori is not associated with gastric H. pylori infection and normal flora.

Overexpression and Activity Analysis of Cystathionine γ-Lyase Responsible for the Biogenesis of H2S Neurotransmitter (새로운 신경전달물질 H2S 발생 효소, cystathionine γ-lyase의 대량발현 조건과 활성측정)

  • There is a growing recognition of the significance of $H_2S$ as a biological signaling molecule involved in vascular and nervous system functions. In mammals, two enzymes in the transsulfuration pathway, cystathionine ${\beta}$-synthase (CBS) and cystathionine ${\gamma}$-lyase (CGL), are believed to be chiefly responsible for $H_2S$ biogenesis. Genetic inborn error of CGL leads to human genetic disease, cystathioninuria, by accumulating cystathionine in the body. This disease is secondarily associated with a wide range of diseases including diabetes insipidus and Down's syndrome. Although the human CGL (hCGL) overexpression is essential for the investigation of its function, structure, reaction specificity, substrate specificity, and protein-protein interactions, there is no clear report concerning optimum overexpression conditions. In this study, we report a detailed analysis of the overexpression conditions of the hCGL using a bacterial system. Maximum overexpression was obtained in conditions of low culture temperature after inducer addition, performing low aeration during overexpression, and using a low concentration inducer (0.1 mM, IPTG) for induction. Expressed hCGL was purified by His-tag affinity column chromatography and confirmed by Western blot using hCGL antibody and enzyme activity analysis. We also report that the His tag with TEV site attached protein exhibits 76% activity for ${\alpha}-{\gamma}$ elimination reaction with L-cystathionine and 88% for ${\alpha}-{\beta}$ elimination reaction with L-cysteine compared to those of wild type hCGL, respectively. His tag with TEV site attached protein also exhibits a 420 nm absorption maximum, which is attributed to the binding cofactor, pyridoxal 5'-phosphate (PLP).

Operative Treatment of Mitral Valve Regurgitation Due to Chordal Rupture and/or Papillary Muscle Rupture (건삭 파열 및 유두근 파열로 인한 승모판 판막 폐쇄부전의 외과적 치험)

  • Background: As the rupture of chordae and/or papillary muscle became the main cause of mitral valve regurgitation, mitral reconstructive surgery has a very important role. In this regard, we analyzed the clinical result and postoperative early result of operative treatment performed in our hospital, Material and Method: For this analysis, forty nine patients (male 26, female 23, mean age 49.0$\pm$16.5) who underwent mitral valve operation caused by the rupture of chordae and/or papillary muscle from August 1991 to April 2002 were reviewed. Among forty nine patients, twenty two (44.9%) received mital valve reconstruction and twenty seven (59.2%) received mitral valve replacement. Result: As to the pathological etiology of rupture of mitral and papillary muscle, twenty five cases (51.0%) were nonspecific degeneration, eleven cases (22.4%) were myxomatous degeneration, seven cases (14.3%) were subacute bacterial endocarditis. Three patients suffered mortality after operation (6.1%) and valve replacement was performed again on one patient because of remnant mitral insufficiency after valve reconstruction. The 5-year survival rate after operation for the entire mitral valve regurgitation patients was 81 .4%. We have also compared and analyzed the operation results of a group of patients who underwent valve reconstruction and the other group of patients who underwent valve replacement from thirty six patients who had suffered from mitral valve regurgitation caused by degenerative disease. The mortalities were 0% and 14.3%, respectively and the 5-year survival rates were 90.2% and 64.3%, respectively, but there were no statistical significance. Conclusion: The most common pathological etiology of mitral valve regurgitation caused by rupture of chordae and/or papillary muscle was nonspecific degeneration, In case of degenerative disease is the cause of mitral valve regurgitation, valve reconstruction showed better long-term effects in many respects and better operation results compared to valve replacement.

Selection and Quality Evaluation of Sprout Soybean [Glycine max (L.) Merrill] Variety for Environment-Friendly Cultivation in Southern Paddy Field (남부지역 친환경 논 재배를 위한 나물콩 품종 선발 및 품질 평가)

  • We carried out the experiment to select the suitable sprout soybean varieties for environment-friendly cultivation in paddy field of southern part area, compares of excess moisture injury degree and yield ability among 29 sprout soybean varieties. Plant growth of sprout soybean was generally low in beginning and recovered after flowering due to rainfall. In paddy field cultivation, number of pod per individual and number of seed per individual were less in difference than upland cultivation, and maturing date was delayed 5-14 days than upland cultivation in most species. When environment-friendly cultivation, pest injury was not caused major problem for the growth during the vegetative period of soybean due to ground spider as natural enemy to insect pest. However, damage of stink bugs showed severe during grain filling period, and Dawonkong, Anpyeongkong, Dachaekong and Wonhwangkong showed susceptible to sting bug. SMV infection was weak and showed some necrosis symptoms in Sokangkong, but black root rot was not infected at all. Bacterial pustule began to be infected slowly from pod enlargement stage in most species, displayed severe symptoms in Dawonkong, Pungsannamulkong, Seonamkong and Sobaeknamulkong. The symptoms of pod anthracnose, pod blight and purple spot were greatly appeared after flowering. Disease resistance varieties was Paldokong, Kwangankong, Doremikong, Somyeongkong, Pungsannamulkong, Iksa-namulkong, Seonamkong, Sojinkong, Pureunkong, Bosugkong, Namhaekong and Sorokkong. Lodging index showed 3 in Saebyeolkong, and other species displayed slight lodging in 0-3 degree. 100-seed weight is 9.8-17.2g extent and increased 0.1-3.7g than upland cultivation in most species, but decreased in some species. Government purchase standard, species correspond to small-seed-size namulkong (Sizing screen diameter 4.0-5.6 mm) was Dawonkong, Dachaekong, Bosugkong, Seonamkong, Sokangkong, Hannamkong, Somyeongkong and Wonhwangkong. Species which seed yield was higher than Pungsannamulkong (266kg/10a) were Sorokkong, Hannamkong, Bosugkong and Sowonkong. Considering sprout soybean species, disease endurance, insect resistance, lodging resistance, 100-seed weight, yield ability and excess moisture tolerances synthetically, Seonamkong, Hannamkong, Doremikong, Bosugkong, Pungwonkong, Kwangankong, Sowonkong, Dagikong, Paldokong, Eunhakong and Pungsannamulkong were promising for environment-friendly cultivation in paddy field.

A New Rice Cultivar 'Manjong', Suitable to the Rice-Cash Crop Double Cropping System (중생 소득작물 후작지 적응 벼 품종 '만종')

  • 'Manjong' is a new japonica rice cultivar developed in 2009 derived from a cross between 'Yeongdeog34' and 'Nampyeongbyeo' at the Department of Functional Crop, NICS, RDA. This cultivar is suitable for the double cropping system after the cash crop cultivation. Heading date of 'Manjong' is later than 'Keumobyeo' under the late transplanting cultivation on July 10. It has high grain fertility under cold conditions and low premature heading. One of the distinguishing characteristics of this variety is its resistance to major diseases like leaf blast and rice stripe virus disease. However, it shows susceptibility to major insect pests and bacterial blight disease. Milled rice kernels are translucent with non-glutinous endosperm and have 7.2% protein and 18.7% amylose contents. Ripening ratio and head rice ratio is very higher than those of 'Keumobyeo'. The palatability of cooked rice is also better than 'Keumobyeo'. The milled rice yield of 'Manjong' in local adaptability tests after harvest of the cash crop is $4.81MT\;ha^{-1}$. This cultivar is suitable for planting in the plain paddy fields of Honam and Yeonnam regions in Korea.

A New Early Maturing Rice Cultivar "Junamjosaeng" with Multiple Disease Resistance and High Grain Quality Traits (고품질 복합내병성 조생종 벼 신품종 "주남조생")

  • Junamjosaeng is a new japonica rice cultivar developed in 2006 from a cross between Milyang165*3 and Koshihikari at the Department of Functional Crop Science, NICS, RDA. This cultivar is suitable for the double cropping system (i. e., before and after the cash crop). Heading date of Junamjosaeng is 6 days earlier than Keumobyeo under the late transplanting cultivation on July 10. It has a high grain fertility under cold conditions and low premature heading. One of the distinguishing characteristics of this variety is its resistance to major diseases like leaf blast, bacterial blight races ($K_1$, $K_2$, $K_3$) and rice stripe virus disease. However, it showed susceptibility to major insect pests. Milled rice kernels are translucent with non glutinous endosperm and have 6.7% protein and 19.8% amylose contents. Milling recovery of head rice is 75.7%. The palatability of cooked rice is better than Keumobyeo. The milled rice yield of Junamjosaeng in local adaptability tests after harvest of the cash crop was $4.43\;tons\;ha^{-1}$. This cultivar is suitable for planting in the plain paddy fields of Honam and Yeonnam regions in Korea.

A New Soy-paste Soybean Cultivar, 'Daeyang' with Disease Resistance, Large Seed and High Yielding (장류용 내병 대립 다수성 신품종 '대양')

  • A new soybean cultivar for soy-paste, 'Daeyang', was developed from the cross among 'Jangyeobkong', 'Hwaeomputkong' and 'Suwon192' by the soybean breeding team at the National Institute of Crop Science (NICS) in 2007. A promising line, SS97214-80-1, was selected and named this line 'Milyang163'. It was prominent and had good result from regional adaptation yield trials (RYT) for three years from 2005 to 2007 and released as the name of 'Daeyang'. It has a determinate growth habit, purple flower, grey pubescence, yellow seed coat, yellow hilum, large spherical seed (25.2 g per 100 seeds). 'Daeyang' is resistant to soybean mosaic virus and moderately resistant to bacterial pustule, the major soybean disease in Korea. The average yield of 'Daeyang' was 2.58 ton per hectare in the regional yield trials (RYT) carried out for three years from 2005 to 2007 which was 3 percent higher than the check variety, 'Taekwang'.

A New High Yielding Rice Variety with Multi-Disease Resistance, 'Keunseom' (중생 복합내병충성 초다수성 벼 '큰섬')

  • 'Keunseom', a new second generation Tongil-type rice variety (Oryza sativa L.), is a mid-maturing ecotype developed by the rice breeding team of Department of Functional Crop, NICS, RDA in 2006. This variety was originated from a cross between 'Dasanbyeo' and 'Namyeongbyeo' in 1996's summer season, which developed by pedigree breeding method. The pedigree of 'Keunseom' was YR18234-B-B-98-3-5-1, and it was designated 'Milyang202' in 2002. 'Keunseom' has tolerance to lodging, because it has short culm length as 77 cm. This variety is resistance to bacterial blight K1 race, rice stripe virus, rice dwarf virus, and leaf blast disease. Milled rice kernel of 'Keunseom' is a clean translucent with non-glutinous endosperm, and has good quality as it was clear in chalkness. The milled rice yield potential of 'Keunseom' was about 719 kg/10a at ordinary fertilizer level of local adaptability test. This cultivar would be adaptable to the mid and southern plain of Republic of Korea.
