Colon Delivery of Prednisolone Based on Chitosan Coated Polysaccharide Tablets

  • Colon drug delivery is advantageous in the treatment of colonic disease and oral delivery of drugs unstable or suceptible to enzymatic degradation in upper GI tract. In this study, multilayer coated system that is resistant to gastric and small intestinal conditions but can be easily degraded by colonic bacterial enzymes was designed to achieve effective colon delivery of prednisolone. Variously coated tablets containing prednisolone were fabricated using chitosan and cellulose acetate phthalate (CAP) as coating materials. Release aspects of prednisolone in simulated gastrointestinal fluid and rat colonic extracts (CERM) were investigated. Also, colonic bacterial degradation study of chitosan was performed in CERM. From these results, a three layer (CAP/Chitosan/CAP) coated system exhibited gastric and small intestinal resistance to the release of prednisolone in vitro most effectively. The rapid increase of prednisolone in CERM was revealed as due to the degradation of the chitosan membrane by bacterial enzymes. The designed system could be used potentially used as a carrier for colon delivery of prednisolone by regulating drug release in stomach and the small intestine.

급수관내 염소 주입이 미생물의 증식과 부식에 미치는 영향 (The Influence of Chlorine Application on Corrosion and Bacterial Growth in Home Plumbing Systems)

  • This research was conducted using a model home plumbing system composed of copper, stainless steel, galvanized iron, carbon steel, and PVC (polyvinyl chloride) pipe. The number of bacteria present in stainless steel pipe and PVC was higher than other pipes. High turbidity and zinc release were found in galvanized iron pipe material and detected during the first 6 months. Conversely, there was a decrease in turbidity and zinc release after 6 months resulting in levels similar to other pipes. Copper concentration decreased as operation times increased. In this experiment, the number of bacteria detected in biofilm for a copper pipe continued to increase. Pipe material influenced bacterial numbers in biofilm and water. This showed that elevated chlorine could not control bacterial growth in biofilm for galvanized iron and stainless steel systems. It also suggested that the dosing of chlorine might not be available for all kinds of pipes. Therefore, another complementary method should be introduced to manage biofilm effectively in water distribution systems.

Antibacterial Activity of Ciprofloxacin-incorporated Central Venous Catheters and its Mechanism Against Planktonic Bacterial Cells

  • Bloodstream infections (BSI) are caused by planktonic microorganisms, sometimes leading to serious infections such as bacteremia and sepsis. BSI occurs more frequently to the patients wearing the central venous catheter (CVC). The ciprofloxacin-incorporated CVC (CFX-CVC) has been reported previously to possess antimicrobial activity. In this study, the antibacterial activity of CFX-CVC and its mechanism against planktonic BSI cells were explored by using the shake flask test and by examining the release rate of 260 nm-absorbing substances from the bacterial cells indicative of the membrane damage of the bacterial cells. CFX-CVC reduced more than 99.9% of the viable planktonic BSI cells demonstrating its potent antibacterial activity. It provoked bacteriolysis causing leakage of a large amount of 260 nm-absorbing materials from the planktonic bacterial cells like S. aureus and E. coli. These results provide evidence that the antibacterial activity of CFX-CVC came from the inhibition of the stability of the planktonic bacterial cells.

석영 및 철피복 모래에서 박테리아 부착.탈착: 이온강도의 영향 (Adhesion and Release of Bacteria in Quartz and Iron-coated Sands: Effect of Ionic Strength)

  • 본 연구에서는 칼럼실험을 이용하여 석영모래와 철피복 모래에서 박테리아(Escherichia coli, Bacillus subtilis, 그리고 Staphylococcus aureus)의 부착 및 탈착에 미치는 이온강도의 영향을 조사하였다. 실험결과, 철피복 모래에서는 이온강도가 1 mM에서 100 mM로 증가함에 따라 질량회수율이 일정(E. coli = 13.7${\pm}$0.5%, B. subtilis = 9.8${\pm}$1.3%, S. aureus = 13.0${\pm}$2.1%)한 반면, 석영모래에서는 80.7%에서 45.3%로 감소하였다(S. aureus). 용출용액의 이온강도가 100 mM에서 0.1 mM로 감소함에 따라, 석영모래에서는 평균 39.1%의 박테리아 탈착이 일어났지만, 철피복 모래에서는 탈착이 관찰되지 않았다. 철피복 모래에서 관찰된 이러한 현상은 박테리아와 철피복 모래사이의 inner-sphere complexes(이온강도의 영향을 받지 않는)에 의한 결합 때문이다. 본 연구는 박테리아와 표면변형 다공성 여재의 상호작용에 대한 지식을 증진시킨다.

Development and Characterization of Membrane for Local Delivery of Cephalexin

  • Laminated films composed of drug-containing reservoir layer and drug-free membrane were prepared. Zero-order drug release with lag time was achieved by laminating drug-free film onto the reservoir layer, while burst effect was observed on cast-on film. The rate controlling membrane was either attached to or cast directly into the reservoir. The release rate was independent on the reservoir composition but dependent on the composition of rate-controlling membrane. In growth inhibitory test of cephalexin from Eudragit RS film to Streptococcus Mutans, the disk even after release test for 72 hours showed more bacterial growth inhibition than that of control. Permeation of drug through rat skin was proportional to the HPC fraction in the film. We could control the release of cephalexin from the film by changing the fraction of Eudragit RS, HPC and DEP content. Consequently, Eudragit RS/HPC film was found to be very effective system for local delivery of drugs.

PC12 세포에서 신경전달물질 방출을 저해하는 물질 S9940 물질의 탐색 (Screeing of S9940 as an Inhibitor of Neurotransmitter Release from PC12 Cells)

  • We established an in vitro experimental system using the following procedure. We first introduced tritium-labelled norepinephrine ([3H]-NE)into PC12 cells. The [3H]-NE incorporated into PC12 cells were then stimulated by a high concentration (60 mM) of $K^+$ during 12 minutes. Then, we counted the amount of [3H]-NE release from PC12 cells with the scintillation counter. After screening fungal, Streptomyces or bacterial product using this experimental system, we obtained S9940 from Streptomyces spp. which inhibited [3H]-NE release from PC12 cells. S9940 also inhibits the release of ATP as a neurotransmitter of PC12 cells and rat cortical neurons. The inhibitory effect was seen even when the PC12 cells were treated with low $K^+$ buffer containing ionomycin $(1\muM)$ as an ionopore. This result suggests that the inhibitory action of S9940 on neurotransmitter release appeared after the influx of $Ca^{2+}$.

Invisible Signals from the Underground: Bacterial Volatiles Elicit Plant Growth Promotion and Induce Systemic Resistance

  • Plant growth-promoting rhizobacteria (PGPR) are a wide range of root-colonizing bacteria with the capacity to enhance plant growth and control plant pathogens. Here we review recent progress that indicate some PGPR strains release a blend of volatile organic compounds (VOCs) that promote growth in Arabidopsis seedlings and induce resistance against Erwinia carotovora subsp. carotovora. In particular, the volatile components 2,3-butanediol and acetoin released exclusively from the PGPR strains triggered the greatest level of growth promotion and induced systemic resistance. Pharmacological applications of 2,3-butanediol promoted the plant growth and induced resistance, while bacterial mutants blocked in 2,3-butanediol and acetoin synthesis was devoid of growth-promotion and induced resistance capacities. The results suggested that the bacterial VOCs play a critical role in the plant growth promotion and induced resistance by PGPR. Using transgenic and mutant lines of Arabidopsis, we provide evidences that the signal pathway activated by volatiles from one PGPR strain is dependent on cyto-kinin activation for growth promotion and dependent on an ethylene-signaling pathway for induced pathogen resistance. This discovery provides new insight into the role of bacterial VOCs as initiators of both plant growth promotion and defense responses in plants.

폴리카프로락톤 매트릭스로부터 세파드록실의 방출에 미치는 BSA의 영향 (The Effect of BSA on the Release of Cefadroxil from a Polycaprolactone Matrix)

  • In order to investigate the effect of bovine serum albumin (BSA), as a pore former, on the controlled release of an antibiotic from a biodegradable polymeric device, polycaprolactone (PCL)-cefadroxil matrices were prepared by the solvent casting method. The amount of cefadroxil released from various formulations at $37^{\circ}C$ was measured by HPLC. The duration of antimicrobial activity of matrices against S. aureus was evaluated by measuring the diameters of the inhibition zone. The morphology of the matrices was investigated by scanning electron microscopy (SEM). The release rate and extent of cefadroxil from PCL matrix increased as the loading dose and particle size of BSA/cefadroxil mixture powder increased. Cefadroxil released from the matrix exhibited antibacterial activity for up to 4 days. SEM of the cross-section of matrix showed the typical channel formation after 3 days of release study. Thus, a biodegradable polymeric matrix loaded with antibiotic/BSA mixture can effectively prevent bacterial infection on its surface, thereby bringing about an enhancement of biocompatibility of biomaterials.

Production of Phagocyte Activating Supernatants by Olive Flounder (Paralichthys olivaceus) Leucocytes Stimulated with Genomic DNA of Escherichia coli

  • Effects of Escherichia coli genomic DNA on the production of phagocyte activating supernatants by the head kidney leucocytes isolated from olive flounder (Paralichthys olivaceus) were investigated. Phagocyte activating activity of the supernatants was estimated by. measuring reactive oxygen species (ROS) production in target head kidney phagocytes. All supernatants from olive flounder head kidney leucocytes-stimulated with E. coli DNA induced significantly (P<0.01) higher ROS production from target pagocytes than the unstimulated control supernatant. Maximum enhancement of chemiluminescent response was observed $5.0-10.0{\mu}g\;mL^{-1}$ of bacterial DNA while the increment ability was decreased significantly (P<0.01) at the concentration of $20.0{\mu}mL^{-1}$. The results demonstrate that olive flounder head-kidney leucocytes stimulated with bacterial DNA release a soluble phagocyte activating cytokines capable of enhancing the respiratory burst activity from target phagocytes.
