A series of in vitro incubation studies with washed rumen bacteria were conducted to determine the influence of incubation time and concentrations of peptides, alanine, ammonia nitrogen and carbohydrate on the rate of peptide disappearance and on bacterial growth. Disappearance rate of tri-alanine (ala3) under various conditions was between 30.6 and $58.2mg\;hr^-$ per gram bacterial dry matter. Ala3 was removed from the incubation medium in an almost linear fashion as incubation time and ala3 concentration was increased. Washed rumen bacteria utilized ala3 faster than di-l-alanine (ala2) at all concentrations. Adding 9mM carbohydrate significantly increased ala3 disappearance, but level of ammonia nitrogen had no influence on ala3 disappearance. The presence of alanine in the medium significantly lowered ala3 utilization by rumen bacteria. Bacterial dry matter and nitrogen growth yield were not influenced by alanine and peptides when incubation medium already contained a sufficient level of ammonia nitrogen. Increased ammonia nitrogen in the presence of ala3 did not stimulate bacterial growth. Carbohydrate significantly increased bacterial dry matter and nitrogen growth as expected. Results indicate that the rate of peptide utilization by rumen bacteria may be altered by type and concentration of peptides, and energy supply, and this may be mediated through changes in numbers and type of bacteria.
Effect of nitrogen-load condition on hydrogen ($H_2$) production and bacterial community in a continuous anaerobic hydrogen fermentation were investigated. The slight $H_2$ production on extremely low nitrogen-load condition (C/N ratio: 180) at the start-up period. The highest $H_2$ production was obtained when the C/N ratio was 36, the $H_2$ production yield ($mol-H_2/mol-glucose$) reached to 1.7, and it was indicated that Clostridium pasteurianum mainly contributed to the $H_2$ production. The $H_2$ production was decreased on both the lower (C/N: 72) and higher (C/N: 18) nitrogen-load conditions. The excess nitrogen-load was not always suitable for the hydrogen production. The fluctuation of $H_2$ production seemed to be caused by a change in the bacterial community according to the nitrogen-load condition, while a recovery of $H_2$ productivity was possible by a control of nitrogen-load condition through the bacterial community change. When the nitrogen-load condition was not suitable for hydrogen production, the lactic acid concentration was increased and also lactic acid bacteria were definitely detected, which suggested that the competition between hydrogen fermentator and lactic acid producer was occurred. These results demonstrated that the nitrogen-load condition affect on the $H_2$ productivity through the change of bacterial community in anaerobic hydrogen fermentation.

The bacterial growth was most vigorous when the temperature and pH of the culture broth was 30~$35^{\circ}$C, and 7, respectively. As for carbon source, dextrose was best followed in order by dextrose(monosaccharides)>sucrose(disaccharides)$\geq$saccharose(di-saccharides)>starch (polysaccharides). Among different sugars, bacterial growth was favored by in the order of brown, black and white sugars, indicating that the bacterial growth might be promoted by the minor elements presented as impurities in the less purified sugars. As for nitrogen source, organic forms were better to bacterial growth than inorganic forms, that is polypeptone was best followed in order by soy sauce, soybeen milk and inoganic nitrogens. Differences in bacterial growth among different forms of inorganic nitrogen were negligible.

The effects of algal inoculation on chemical oxygen demand (COD) and total nitrogen (TN) removal, and indigenous bacterial dynamics were investigated in municipal wastewater. Experiments were conducted with municipal wastewater inoculated with either Chlorella vulgaris AG10032, Selenastrum gracile UTEX 325, or Scenedesmus quadricauda AG 10308. C. vulgaris and S. gracile as fast growing algae in municipal wastewater, performed high COD and TN removal in contrast to Sc. quadricauda. The indigenous bacterial dynamics revealed by 16S rRNA gene amplification showed different bacterial shifts in response to different algal inoculations. The dominant bacterial genera of either algal case were characterized as heterotrophic nitrifying bacteria. Our results suggest that selection of indigenous bacteria that symbiotically interact with algal species is important for better performance of wastewater treatment.
To assess the relative contribution of bacterial and phytoplankton biomasses to particulate organic matter (POM) in the water column, microbial abundance and biomass were from two transects in the western channel of the Korea Strait in 1996. Bacterial abundance had a mean value of $5.9{\times}10^5$ cells/ml and chlorophyll-a averaged 0.14 ${\mu}g/l$. Bacterial abundance in the Korea Strait showed a positive relationship with chlorophyll-a concentration, while the distribution of POM did not covary with chlorophyll-a. Particulate organic carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon and nitrogen biomasses were 7.29 ${\mu}gC/l$ and 1.24 ${\mu}gN/l$, respectively, during the study periods. Bacterial biomass was larger in October than in August due to the autumn phytoplankton bloom. Phytoplankton biomass based on chlorophyll-a was 7.67 ${\mu}gC/l$ for carbon and 1.10${\mu}gN/l$l for nitrogen. The ratio of bacterial carbon (BC) to phytoplankton carbon (Cp) averaged 0.95 in the Korea Strait in 1996. Bacteria may play a more significant role in the dynamics of POM than phytoplankton do in August, with BC/Cp ratio of 1.26. The ratio of BC to Cp increased with a decrease in chlorophyll-a concentration. Averaged over all the samples in both cruises, the contribution of microbial biomass to POC and PON was about 43% and 51%, respectively. Bacterial assemblage constituted a significant fraction of POC (21%) and PON (27%). Phytoplankton accounted for 22% of POC and 24% of PON. Microbial biomass played a more important role in the dynamics of POC and PON in October than in August due to a significant increase in microbial biomass in the southern transect (transect-B) in October by the autumn phytoplankton bloom. This study showed that marine microbes may constitute a significant part in the reservoir of POM in the Korea Strait.
In order to investigate the relationship among the occurrence of bacterial leaf blight, amount of nitrogen application and planting density, a field experiment was carried out with factorial combinations of N levels (7.5, 15, 30kg/10a) and planting densities (10\times20,\;15\times30,\;20\times40cm). The results obtained in this study were; (1) The incidence of bacterial leaf blight was increased with increase of nitrogen application; (2) In double amount of nitrogen application (30kg/10a), the incidence of bacterial leaf blight was higher, regardless of planting density; (3) The incidence of bacterial leaf blight in Standard nitrogen application was significantly higher $(R=0.94^{**})$ than at $15\times30cm$ density and $20\times40cm$ planting density.
Microbial community plays important roles in the culture environment of mud crab Scylla serrata. One of the environmental management efforts for the cultivation of S.serrata is by stabilizing microorganisms involved in nitrogen cycle process. The availability of dissolved inorganic nitrogen in its culture environment under a recirculating system closely relates to the nitrogen cycle, which involves both anaerobic and aerobic bacterial activities. Anaerobically, there are two major nitrogen compound degradation processes, i.e., denitrification and dissimilatory nitrate reduction to ammonium (DNRA). This study aimed to identify denitrifying and DNRA bacteria isolated from the recirculating cultivation of S. serrata. The water samples were collected from anaerobic filters called close filter system, which is anaerobically conditioned with the addition of varying physical filter materials in the recirculating mud crab cultures. The results showed that three denitrifying bacterial isolates and seven DNRA bacterial isolates were successfully identified. The phylogenetic analysis based on 16S rRNA gene of the denitrifying bacteria revealed that HIB_7a had the closest similarity to Stenotrophomonas daejeonensis strain MJ03. Meanwhile, DNRA bacterial isolate of HIB_92 showed a 100% similarity to Bacillus sonorensis strain N3, Bacillus vallismortis strain VITS-17, Bacillus tequlensis strain TY5, Geobacillus sp. strain DB24, Bacillus subtilis strain A1, and Bacillus mojavensis strain SSRAI21. This study provides basic information denitrifying and DNRA bacterial isolates identity which might have the potential to be applied as probiotics in aquaculture systems in order to maintain optimal environmental conditions.

Two successive in vitro experiments were carried out to examine the effect of MPS bacterial inoculation on growth, and nitrogen and phosphorus accumulation of maize plants under greenhouse condition in the same soil. There were four treatments, uninoculated control and three phosphate solubilizing bacterial inoculations, viz., Pseudomonas striata, Burkholderia cepacia and Serratia marcescens. The inoculated plants showed the higher plant height, total dry mass, nitrogen and phosphorus accumulation when compared to uninoculated control plants in both experiments. In the combined data analysis from two experiments, the plants inoculated with P. striata and B. cepacia showed significantly higher plant height, total dry mass and P accumulation when compared to S. marcescens inoculated plant and uninoculated control plants. The P. striata and B. cepacia inoculation enhanced total dry matter accumulation by 14% and phosphorus accumulation by 25% over the uninoculated control plants. The nitrogen and phosphorus concentration of maize plants were also increased due to MPS bacterial inoculation, however, the effect was not significant.
Recently, an autotrophic single-stage nitrogen removal (ASSNR) process based on the anaerobic ammonium oxidation (ANAMMOX) reaction has been proven as an economical ammonia treatment. It is highly evident that double-layered gel beads are a promising alternative to the natural biofilm for ASSNR because of the high mechanical strength of poly(vinyl alcohol) (PVA)/alginate structure and efficient protection of ANAMMOX bacteria from dissolved oxygen (DO) due to the thick outer layer. However, the thick outer layer results in severe mass transport limitation and consequent lowered bacterial activity. Therefore, the effects of the thickness of the outer layer on the overall reaction rate were tested in the biofilm model using AQUASIM for ammonia-oxidizing bacteria (AOB), nitrite-oxidizing bacteria (NOB) and ANAMMOX bacteria. A thickness of 0.5~1.0 mm is preferred for the maximum total nitrogen (TN) removal. In addition, a DO of 0.5 mg/L resulted in the best total nitrogen removal. A higher DO induces NOB activity and consequent lower TN removal efficiency. The optimal density of AO B and NO B density was 1~10% for a 10% ANAMMOX bacterial in the double-layered PVA/alginate gel beads. The real effects of operating parameters of the thickness of the outer layer, DO and concentrations of biomass balance should be intensively investigated in the controlled experiments in batch and continuous modes.
