  • Salmonella is one of the most common bacteria that causes heavy losses in swine industry and major causative pathogen of food poisoning in public health. Various methods for the identification of Salmonella such as Gram staining, agglutination test, enzyme-linked immunosorbent assay (ELISA), polymerase chain reaction (PCR) have been used. Several studies have demonstrated that Matrix Assisted Laser Desorption Ionization Time of Flight (MALDI TOF) Mass Spectrometry (MS) identification is an efficient and inexpensive method for the rapid and routine identification of isolated bacteria. In this study, MALDI TOF MS could provide rapid, accurate identification of Salmonella spp. from swine compared with end point PCR and real time PCR.

Subgingival calculus at Aerococcus viridans, Leuconostoc spp. bacteria 5, 1 species detection.

장기 보존 한약 파우더의 미생물 모니터링 (Monitoring on Microbial flora of Herbal Powder in Long Term Preservation)

  • Objectives : This study was carried out to moniter microbial flora on freeze-dried herbal powder and identify isolated bacteria. Methods : We measured the total number of bacteria and fungi in 29 herbal powder which had made according to the guideline of KFDA. For the identification, we observed microscopic properties and carried out polymerase chain reaction(PCR). The purified DNA was analyzed by DNA sequencer. Results : Among the 29 herbal powders, the fungi were detected only one sample as unacceptable range of total aerobic bacteria. Isolated bacteria were identified as Bacillus cereus, B. subtilis, B. megaterium, B. licheniformis, Erwinia tasmaniensis, E. amylovora, and Pantoea agglomerans by 16S rDNA analysis. E. tasmaniensis was observed 20 herbal samples. Conclusions : According to above results, further studies for the effective sterilization of low herbal materials should be needed.

사과 푸른곰팡이병의 길항미생물의 분리 및 동정 (Isolation and Identification of Antifungal Bacteria on Blue Mold in Apple)

  • In order to screen the antagonistic bacteria which inhibit the growth of the apple pathogen, Penicillum expansum, we isolated an effective bacterial strain and investigated into the antifungal activity of the antagonist and it's identification. The eleven strains of bacteria which strongly inhibited P. expansum were isolated from the nature, and the best antagonistic bacterial strain designated as CH142, was selected. The antagonistic strain CH142 was identified to be the genus Bacillus subtilis based on morphological and biochemical characterization. The CH142 showed 55.9% of antifungal activity against the growth of P. expansum. By the treatment of the culture broth and the heat treated culture filtrate of it, the B. subtilis CH142 showed 90% and 15% of antifungal activity, respectively.

  • Isolation and identification of mesophilic and psychrotrophic bacteria distributed in Korean refrigerated beef were attempted. Total isolated colonies were 192, and identified as 5 genera and 10 species. Among them, mesophilic bacteria were Enterobacter aerogenes, E. agglomerans, Serratia liquefaciens, Proteus mirabilis, and "psychrotrophic" bacteria were Pseudomons fluorescens, P. putida, P. pickettii, P. mendocina, P. stutzeri, Alcaligenes faecalis. Dominant species was Serratia liquefaciens as mesophiles, and Pseudomonas putida as psychrotroph.chrotroph.

서울시역 한강수계에서의 합성세제 분해세균의 분리 및 동정 (Isolation and Identification of Detergents Degrading Bacteria in the Han River Downstream)

  • Detergent-degrading bacteria isolated from the Han River downstream running through the center of Seoul area were identified. Samples were monthly taken from the surface waters at four designated sites from October, 1980 to August, 1981. Isolated strains were characteristically Gram-negative bacteria in all, and they were classified within 18 genera, 43 spceies. The dominant strains of detergent -degrading bacteria were within geneus Pseudomonas. On the regional distribution of detergent-degrading bacteria, Euterobacteriaceae and genus Aeromonas were suddenly increased at the Third Bridge of site 2 and the First Bridge of site 3, and others were almost the same regardless of sites.

  • The purpose of this paper is to study on the morphological and physiological nature generally observed for the identifications, and the four strains of thermal resistanting aerobic bacteria isolated from swelled cans and the different soils collected from the wide area in Korea. The results obtained in the light of the Bergey's Manual for the identification of the bacteria, have been shown that the four strains of bacteria are pertained to Bacillus subtilis. The optimum temperature, pH and the thermal resistance (2.5 min. at $121^{\circ}C$) of the bacteria have been measured.

  • Objectives: The objective of this study is to analyze the predominant microorganisms in the industry treating MWFs(Metal working fluids). Methods: The bacteria and fungi were collected by agar plate impaction and bulk MWFs in storage tank at 54 sites in 9 shops in South Korea. The dominant bacteria and fungi isolated from agar media were identified by fatty acid analysis and morphological analysis, respectively. Results: Totally 111 dominant bacteria were identified in the process, outdoor, and bulk MWFs. The predominant bacterial genus was Micrococcus and Bacillus in the process and outdoor, Pseudomonas in bulk MWF. Among the identified 119 strains of fungi, Cladosporium and Penicillium genus were dominated. The ratios of bacteria designated biosafety level 2 and 1 were 30% and 21%, respectively. Conclusions: This study has investigated the dominant microorganisms in soluble MWF using industry. And it was useful that the qualitative evaluation method along with quantitative analysis for better understanding of the biological factors in the work environment.

  • 본 실험은 한국산 잠견에 기생하는 세균을 분리하여 생리학적 형태학적 및 배양적인 특성을 파악하여 그 미생물을 동정하고 저장중인 잠견에 미생물 침식을 방지하는 과학적인 근거를 모색했다. 그 실험결과는 다음과 같다. 1. 잠견으로부터 분리된 12 strain의 Bacteria의 형태학적인 실험결과 colony의 형태는 table II와 같다. 종합적인 형태적 특성은 table III과 같으며 gram stain과 spore stain은 Fig, I, II와 같다. 2. 분리된 12 strain의 배양적인 특성은 table IV, V, Ⅵ와 같다. 3. 분리된 12 strain의 생리적인 특성은 table Ⅶ와 같다. 4. 이상, 형태학적 배양적 생리적인 특성에 의거하여 분리균주의 유연관계를 추정하면 다음과 같다.   • The presence of slime in paper mills is practically universal. Many researches have been performed for many years to resolve the problem caused by the slime in pulp and paper mill. Many papers have been published to show the bacteria is a major cause of paper mill slime. Now that the recycling of the water has been increased and the regulations of a toxic chemical dosage have become more strengthen, the importance of the control of slime in pulp and paper mill recently has been more recognized. Therefore, to produce quality products at the lowest economic and environmental costs, a through study of the microbial ecology and the indentification of troublesome slime-forming bacteria is a quite necessary. The purpose of this paper is to indentify slime~forming bacteria isolated from the papermaking process. The samples were taken from four parts of making fine paper : machine chest, head box, wire part, white water tank. Machine chest showed the most numbers of bacteria, numbering $2.55{\times}10^7$. The different colony types were taken from the 105 dilution plate. Nine bacteria were identified u sing the Biolog system and the vitek system: 6 gram-negative bacteria, 3 gram-positive bacteria. They are Pseudomonas paucimobilis B., Staphylococcus sp., Acinetobacter calcoaceticus., Pseudomonas cepacia, Actinobaci1lus capsulatus, Acidovorax sp., Flavobacterium sp., and Staphylococcus auricularis in addition to one unidentified sp., Among them. Pseudomonas paucimobillis was found in all places where the samples were taken. And, each parts had the different predominant bacteria in it : Pseudomonas paucimobilis B. in machine chest, Acinetobactor calcoaceticus. in Wire Part and Staphylococcus sp. in head box.
