In order to select probiotics with a high survival rate in gut and the growth inhibition of virulent pathogens to human beings or animals, we have screened lactic acid bacteria and yeasts from Jeot-gal to assess resistance against the artificial gastric acid and bile juice. Lactic acid bacteria and yeasts isolated were incubated for 24 h in artificial bile juice after incubation for 2 h in artificial gastric acid. Especially, strain HW 161 and strain NK 181 showed the higher survival for 2 h incubation in artificial gastric acid. All of 3 strains of lactic acid bacteria and 2 strains of yeast were showed higher growth rate than the control in artificial bile. The antimicrobial activity of lactic acid bacteria and yeasts was also investigated to prove efficacy as probiotic organisms. Lactic acid bacteria were shown the inhibition of Gram positive and negative bacteria, but yeasts narrow inhibition.
Bacteria isolated from ultrapure water made by a high-purity water manufacturing system in a semiconductor manufacturing plant were classified into two groups which either grew in diluted nutrient broth medium (oligotrophic bacteria) or could not grow (copiotrophic bacteria). The nutritional flexibility of oligotrophic and copiotrophic bacteria was investigated. The oligotrophic bacteria were shown to be able to utilize a significantly broader range of organic substrates than the copiotrophic bateria. This finding substantiates the hypothesis that nutritional flexibility is adaptive for oligotrophic bacteria.
For more convenient and rapidly isolation of Bacillus thuringiensis subsp. israelensis(Bti), 1) heat treatment spore forming bacteria, 2) growth in enrichment culture media for Bacillus sp. and 3) selection of bacteria producing a lecithinase for Bacillus thuringiensis subsp. israelensis, were performed. Spore forming bacteria were counted 4.8 $\times $ 10$^{8}$cells/g soil on NAPGCY media, 9.2 $\times $ 10$^{7}$cells/g soil on NA media, and 3.6 $\times $ 10$^{8}$cells/g soil on NAAC media, respectively. Bacteria producing only a lecithinase were reached at 25.2% on medium contained egg york, bacteria only producing a delta-endotoxin were reached at 23.2% by phase contrast microscope, and bacteria producing a lecithinase & a delta-endotoxin simultaneously were reached at 13.7%. Bacillus thuringiensis which producing a lecithinase and a delta-endotoxin simultaneously among bacteria producing a lecithinase, were reached at 56.5%; A half of Bacillus thuringiensis was produced a delta-endotoxin, but not produced a lecithinase. Among 8 isolates of Bacillus thuringiensis, two strain of Bti which has a mosquito-cidal toxin, were detected by PCR using a specific primer of $\delta $-endotoxin gene from Bti.
Approximately 1% of intestinal bacteria of human and rats was alkalotolerant. Among these bacteria of human, bacteria producing $\beta$-glucosidase, $\beta$-glucuronidase and sulfotransferase were 40%, 4% and 0%, respectively. Among alkalotolerant intestinal bacteria of rats, bacteria producing, these enzymes were 70%, 8% and 0%, respectively. $\beta$-Glucosidase and $\beta$-glucuronidase of alkalotolerant intestinal bacteria of human and rat were induced by the medium of high pH: these enzymes activities were increased by elevating pH of the medium, but the growths were not changed. The enzyme activities at the medium of pH 7 were about ten-fold higher than those at the medium of pH 6.
To investigate the interaction of bacteria and Microcystis isolated from a hypertrophic reservoir(Seo-Nakdong River), the response of five bacteria in relation to the different treatment of Microcystis and microcystin production by addition of dominant bacteria Staphylococcus sciuri were examined. Five bacteria (S. sciuri, S. capitis, S. epidermis, Pseudomonas aeruginosa and Aeromonas aimonicida) were identified from the reservoir. In the experiment of bacterial response, two types of bacterial growth pattern were observed. All bacteria showed active growth in heated Microcystis-added media. Especially, three species of bacteria (S. sciuri, S. capitis and P. aeruginosa) among them showed active growth in live Microcystis-added media. In Microcystis response, increase of microcystin production showed when dominant bacteria, S. sciuri was added.
Tetracycline is one of the mostly used antibiotics around the world. The purpose of this study was to evaluate the fate of two different types of tetracycline resistant bacteria in biological wastewater treatment plants. Tetracycline resistant enterics and heterotrophic bacteria were monitored under two different lab-scale experimental conditions. Tetracycline resistant enteric bacteria showed the lower percentages of total enteric bacteria and net specific growth rate in the monitored activated sludge system as compared to tetracycline resistant heterotrophic bacteria. Therefore, total enterics, potentially E.coli, might not be the best indicator microorganism for evaluating the antibiotic resistant bacteria in biological wastewater treatment plant.

The seven strains, Pseudomonas paucimobilis, Pseudomonas cepacia, Staphylococcus auricularis, Staphylococcus saprophyticus, Acidovorax spp., Acinetobacter calcoaceticus, and Actinobacillus capsulatus were tested with three slimicides. Most of the tested bacteria were inhibited with slimicide K (an isothiazolin based compound), even at its low concentration, except for Actinobacillus capsulatus and Staphylococcus auricularis. Both slimicides B (an organic bromine based compound) and S (aldehydes) also couldn't prevent these two strains even at their highest concentration. Five different sizes of plasmid DNA were isolated from Actinobacillus capsulatus. Staphylococcus auricularis, a gram-positive bacteria, showed the slimy substances around its cell distinctively. The results suggest that two strains, Actinobacillus capsulatus, Staphylococcus auricularis, have presumably developed a resistance to the slimicide, by plasmid DNA or slimy substance. Our findings also suggest that not only gram-negative bacteria, but also gram-positive bacteria should not be neglected

This study was conducted to empirically analyze the effects and interactions among beneficial bacteria, commensal bacteria, and acne bacteria, which are factors in the skin microbiomes, on detrimental bacteria by 289 people, who are 20 to 49 years old among Koreans. As a result of multiple regression models using bio big data of skin microbiomes, when the difference in skin microbiomes according to the sex and age of the subjects was controlled, the beneficial bacteria showed a negative (-) effect on the detrimental bacteria, while the commensal and acne bacteria showed a positive (+) effect. Particularly, the negative (-) effect of beneficial bacteria on detrimental bacteria was different through interaction with acne bacteria according to the level of commensal bacteria. These results demonstrate that the activation of beneficial bacteria inhibits detrimental bacteria, and the effect of skin microbiomes on detrimental bacteria is balanced with skin microbiomes through interaction with independent influence. Therefore, it is suggested that when studying skin microbiomes products to help the proliferation of beneficial bacteria and to create a skin environment that inhibits detrimental bacteria in the personalized cosmetics manufacturing industry, it is necessary to consider the independent effects and interactions among skin microbiome factors together.

The encapsulation of bacteria may be used to harness them for longer periods of time in order to make them viable, whereas antibiotic treatment would result in controlled release of therapeutic molecules. Encapsulated Escherichia coli GFP (green fluorescent protein) (E. coli GFP) was used here as a model for therapeutic substance - GFP fragments release (model of bioactive substances). Our aim was to evaluate the performance of bacteria encapsulated in hollow fibers (HFs) treated with antibiotic for induction of cell death. The polypropylene-surface-modified HFs were applied for E. coli encapsulation. The encapsulated bacteria were treated with tetracycline in vitro or in vivo during subcutaneous implantation into mice. The HF content was evaluated in a flow cytometer, to assess the bacteria cell membrane permeability changes induced by tetracycline treatment. It was observed that the applied membranes prevented release of bacteria through the HF wall. The E. coli GFP culture encapsulated in HF in vitro proved the tetracycline impact on bacteria viability and allows the recognition of the sequence of events within the process of bacteria death. Treatment of the SCID mice with tetracycline for 8 h proved the tetracycline impact on bacteria viability in vivo, raising the necrotic bacteria-releasing GFP fragments. It was concluded that the bacteria may be safely enclosed within the HF at the site of implantation, and when the animal is treated with antibiotic, bacteria may act as a local source of fragments of proteins expressed in the bacteria, a hypothetical bioactive factor for the host eukaryotic organism.
To identify the treatment effect of lactic acid bacteria for diabetes, the treatment effects of a single administration of acarbose (a diabetes treatment drug) or lactic acid bacteria, and the mixture of acarbose and lactic acid bacteria on diabetes in a type 1 diabetes animal model, were studied. In this study, streptozotocin was inoculated into a Sprague-Dawley rat to induce diabetes, and sham control (Sham), diabetic control (STZ), STZ and composition with live cell, STZ and composition with heat killed cell, STZ and composition with drugs (acarbose) were orally administered. Then the treatment effect on diabetes was observed by measuring the body weight, blood glucose, and serum lipid. For the histopathological examination of the pancreas, the Langerhans islet of the pancreas was observed using hematoxylin and eosin staining, and the renal cortex, outer medullar, and inner medullar were also observed. The induced diabetes decreased the body weight, and the fasting blood glucose level decreased in the lactic-acid-bacteria-administered group and the mixture-administered group. In addition, the probiotic resulted in the greatest decrease in the serum cholesterol level, which is closely related to diabetes. Also, the hematoxylin and eosin staining of the Langerhans islet showed that the reduction in the size of the Langerhans islet slowed in the lactic-acid-bacteria-administered group. The histopathological examination confirmed that the symptoms of diabetic nephropathy decreased in the group to which viable bacteria and acarbose were administered, unlike in the group to which dead bacteria was administered. The mixture of lactic acid bacteria and acarbose and the single administration of lactic acid bacteria or acarbose had treatment effects on the size of the Langerhans islet and of the kidney histopathology. Thus, it is believed that lactic acid bacteria have treatment effects on diabetes and can be used as supplements for the treatment of diabetes.
