Optimal conditions for pigmentation in Bacillus licheniformis SSA3 and cloning of a DNA fragment involved in pigment production

  • Bacillus licheniformis SSA3 can produce a dark-brown antimutagenic pigment. The optimal conditions for production of this pigment are reached at 0.1% tyrosine, in pH 6-8, within 7-9 days, at $30^{\circ}C$, and in aerobic condition. We cloned a DNA fragment involved in pigment synthesis from Bacillus licheniformis SSA3 using a mutant strain. The cloned DNA was 7kb in size, which can produce the same pigment even in E. coli.

Effect of Glycyrrhizia uralensis Extract Addition on the Quality of Cheonggukjang (감초추출물 첨가가 청국장의 품질에 미치는 영향)

  • This study was conducted to investigate the effects of Glycyrrhizia uralensis water extract on the quality of Cheonggukjang(fermented soybean paste). Soybeans were soaked for 24 hrs, steamed for 1 hr at 121, mixed with G. uralensis extract(0, 1, 3 and 5%) and fermented for 54 hrs at 40 after inoculation of Bacillus lichenifomis. The amounts of viscous substance and reducing sugar, and pH of the products were increased when the G. uralensis extract was added. The color of the products was changed to dark yellow by the addition of G. uralensis extract. Ammonia odor and bitter taste decreased as the G. uralensis extract content increased. The products added with 5% of G. uralensis extract showed higher acceptability scores than the others. Results suggest that the water extract of G. uralensis could be used as an additive to improve the quality of Cheonggukjang.

Effect of Phytase-Producing Bacteria on the Fermentation of Soybean and Corn Meals (Phytase 생산균주의 콩과 옥수수 가루의 발효에 미치는 영향)

  • Two Phytase-producing bacteria isolated from the fermented soybean and corn meals, Enterobacter cloacae and Bacillus lichenifomis, were used to investigate the degradation of phytic acid and changes of some nutrient contents in fermented soybean and corn meals. The pH in fermented soybean meal with E, cloacae was rapidly dropped after 48 hours, but the pH in fermented corn meal was declined gradually for 5 days. The degradation of phytic declined acid were optimized at $35^{\circ}C$.pH 8.0 for 5 days and at $30^{\circ}C$, pH 7.0 for 5 days fermented with E. cloacae and B. iichenifomis, respectively. Riboflavin and vitamin $B_{12}$ contents were greatly increased after the fermentation with these two bacteria, and also available lysine, methionine and tryptophan contents were greatly increased.

Purification and Characterization of a New Fibrinolytic Enzyme of Bacillus licheniformis KJ-31, Isolated from Korean Traditional Jeot-gal

  • Jeot-gal is a traditional Korean fermented seafood and has long been used for seasoning. We isolated 188 strains from shrimp, anchovy, and yellow corvina Jeot-gal, and screened sixteen strains that showed strong fibrinolytic activities on a fibrin plate. Among those strains, the strain that had the largest halo zone was chosen and identified as Bacillus licheniformis by using 16S rDNA sequencing and an API CHB kit. The fibrinolytic activity of Bacillus licheniformis was characterized and designated as bpKJ-31. The active component of bpKJ-31 was identified as a 37 kDa protein, designated bacillopeptidase F, by internal peptide mapping and N-terminal sequencing. The optimum activity of bpKJ-31 was shown at pH 9 and $40^{\circ}C$, with a chromogenic substrate for plasmin. It had high degrading activity for the $B{\beta}$-chain and $A{\alpha}$-chain of fibrin(ogen), and also acted on thrombin, but not skim milk and casein. The amidolytic activity of bpKJ-31 was inhibited by 1 mM phenylmethanesulfonyl fluoride, but 1 mM EDTA did not affect the enzyme activity, indicating that bpKJ-31 is an alkaline serine protease, like a plasmin. The bpKJ-31 showed approximately 14.3% higher fibrinolytic activity than the plasmin. These features of bpKJ-31 make it attractive as a health-promoting biomaterial.

Antioxidant Activity of Fermented Barley, Wormwood, Sea Tangle, and Soybean (발효 보리, 쑥, 다시마, 대두의 항산화효과)

  • Superoxide is involved in causing inflammation, cancer, and arteriosclerosis in many cases. Taking antioxidant material can be helpful in preventing the diseases. Natural food such as barley, wormwood, sea tangle, and soybean contain antioxidant ingredients. Antioxidant activity increase was determined by fermenting them with microorganism. To determine the activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) solution was used. When barley, wormwood, sea tangle, and soybean were fermented with Bacillus lichenifomis Bl, antioxidant activities of each fermented product increased 2.6, 1.6, 2.7, and 1.7 folds, respectively. Also, absorbance of fermented soybean was higher than that of soybean at the range of 250~290nm, which might be involved in differences of antioxidant activity of the two. Paraquat suppressed Esherichia coli DH5$\alpha$ growth by making superoxide inside the strain. However, when ethanol extract from fermented soybean was added into the GM (glucose-mineral) media containing the strain, its growth was recovered, suggesting that ethanol extract can move across E. coli, and can function as anti-oxidant material in vivo. Thus, it will be possible to develope antioxidant material from fermented soybean which can be taken orally.

Quality Characteristics of Kipfel Cookie Prepared with Chitosan-Chungkukjang (키토산 청국장을 첨가하여 제조한 깊펠 쿠키의 품질 특성)

  • Quality characteristic of Kipfel cookies with 0, 20, 40 and $60\%$ of freeze dried chitosan-chungkukjang(CC), which was fermented for 24 hours at $40^{\circ}C$ with steamed soybean added with $0.25\%$ of chitosan(MW 2,025 kDa) powder and inoculated $2\%$ of Bacillus lichenifomis, were investigated Bulk density was higher in the CC cookie, but there were no difference among the CC cookies. While hardness was decreased, brittleness and springiness were increased in the higher ratio of CC, but no significant difference was observed in cohesiveness and gumminess. According to increasing the CC ratio, L$\ast$ values and hue angle were decreased from 62.69 and 96.71 to 44.41 and 69.30, respectively. While a$\ast$ values were increased from -1.94 to 4.95, and no changes were observed in b$\ast$ values. Glucosamine content was $27.34 mg\%$ in the control cookie, $40{\~}93.75 mg\%$ in the CC cookies. Antioxidant activity of the CC cookies were higher than the control. The activity of the control cookie was decreased during storage, while it was maintained in CC cookies. There were no differences in the sour, sweet and savory taste. Off-flavor did not detected in the $0{\~}40\%$ CC cookies, but the chungkukjang odor was slightly detected in the $60\%$ CC cookie. The $20{\~}40\%$ CC cookies did not affect on native odor of Kipfel cookie. Color acceptability also did not affect by addition of CC $20{\~}40\%$ but it was lower in $60\%$ CC cookie. Overall acceptability was the best in the $20\%$ CC cookie.

Isolation, Identification and Growth Characteristics of Main Strain Related to Meju Fermentation (메주발효에 관여하는 우량균주의 분리, 동정 및 생육특성)

  • This study was carried out to select Meju of a good quality through general composition analysis, organoleptic evaluation, and to conduct isolation, identification, and growth characteristics of main strain related to fermentation from selected Meju. Moisture and crude protein of Meju were 7.2∼28.8% and 32.7∼42.3%, respectively. The amino nitrogen contents of Kyongbuk and Chonbuk Mejus were 770.8 mg% and 239.9 mg%, respectively. And also, free amino acid and glutamic acid contents of Doenjangs made from Chonbuk and Kyongbuk Mejus were 4,169.6 mg% and 499.4 mg%, respectively. The result of sensory evaluation of Mejus collected from several regions showed Kyongbuk was the most suitable Meju in items of color, flavor, appearance and overall (p<0.05). The typical properties of B. lichenifomis NH20 strain isolated from Kyongbuk Meju showed gram positive, aerobic rod cell and motility. As major component among its cellular fatty acid composition, $C_{15:0}$ anteiso fatty acid, $C_{15:1}$ iso fatty acid, $C_{17:0}$ anteiso fatty acid, and $C_{17:0}$ iso fatty acid were 30.7, 28.9, 13.3 and 11.2%, respectively. It showed the same identification coefficient (0.653) compared to the standard strain. Therefore, it was identified to be B. licheniformis NH20 according to Bergey's Manual of Systematic Bacteriology and its fatty acid profiles. The optimum pH, temperature, salt content, and culture time of B. licheniformis NH20 were 7.0, 32$^{\circ}C$, 2%, and 9 hours, respectively.ctively.
