Isolation, Identification, and Characterization of Microorganisms which Possess the Flocculating Activity to Food Wastewater (음식물류폐수에 특이적 응집제를 생성하는 미생물의 분리, 동정 및 응집특성)

  • Six bacteria, which showed the flocculating activity to food wastewater, were isolated from various environment. These strains were identified as Bacillus pumilus, Enterobacter sp., Pantotea agglomerans, Bacillus licheniformis, and two Bacillus sps. Among them, the flocculating activities of three strains, such as Enterobacter sp.(YK102), Bacillus sp.(YK103), and Pantotea agglomerans (YK104), were eight times or more higher than that of the control strain, Zoogloea ramigera. in the test with 0.5% kaolin. In the experiment with food wastewater, Enterobacter sp.(YK102) showed the highest flocculating activity which was 2.5 times higher than that of a control strain, Pseudomonas fluorescens.

살균 target 미생물별 사멸효과 예측 프로그램

  • 가열살균 처리시의 온도값을 이용하여 다양한 미생물들의 가열살균 효과를 D값, z값 등의 미생물 내열성 특성치와 kinetic 모델 및 수식을 이용하여 살균치 계산을 컴퓨터를 이용하여 수행함으로써 target 미생물의 살균정도를 예측하고자, Excel과 Visual Basic을 연동하여 작성된 시스템으로 Excel을 기본으로 사용한 프로그램을 개발하였다. 가열살균 target 미생물을 선택하고자 야채식품을 기반으로 하는 식품에서의 미생물을 검토하여, 내열성인 가장 큰 호기성 포자형성균인 Bacillus spp. 계통이 산을 형성하여 맛 등의 품질을 저하시켜 변패를 유발하며, 위생적인 면에서 포자수를 줄이거나 사멸할 필요가 있어서 선택하였다. 병원성 균 중에서 D121.1이 0.0065분, z값이 7.8$^{\circ}C$인 Bacillus cereus와 부패현상 관련 균 중에서 D121.1이 0.5분, z값이 12$^{\circ}C$인 Bacillus subtilis를 대상으로 해서 개발되었으며, 필요시에는 D값과 z값을 직접 대입하므로서, 여러 가지 미생물에 대해서도 적용이 가능하게 하였다.

유산균 용균 효소를 생산하는 미생물의 분리, 동정 및 배양조건

  • Isolation, identification, and culture conditions of a lytic enzyme producing microorganism against Lacto- bacillus plantarum were investigated. The selected strain was gram-positive, rod (0.7 $\times$ 2.7 $\mu$m in size), and non-motile. The strain did not have any flagella and spores. According to its cultural and physiological characteristics, the strain was identified as Bacillus sp. The optimal pH and temperature for the production of lytic enzyme were 8.0 and 30$\circ$C, respectively. The maximum enzyme activity showed 1.5 units/ml in the medium composed of 1% peptone and 0.1% NaCl.

  • PAHs의 분해를 위해 Bacillus subtilis 와 Phanerochaete chrysosporium의 두 미생물을 사용하였고, 이들을 부착시킬 담체로 코르크와 톱밥를 선정하여 미생물을 접종시켰다. Phenanthrene 분해의 경우, 반응 초기에는 분해 속도가 매우 빠르지만 반응 12일째 분해 속도가 떨어졌다. 분해 속도는 1)톱밥에 담지한 Bacillus subtilis, 2)코르크에 담지한 Bacillus subtilis, 3)톱밥에 담지한 Phanerochaete chrysosporium, 4)코르크에 담지한 Phanerochaete chrysosporium의 순으로 나타났다.

Studies on the Isolation and Identification of Bacillus sp. for the Composting of Swine Manure and the Removal of Malodorous Gases from its Liquid Compost (돈분의 퇴비화를 위한 Bacillus sp.의 분리 동정 및 그 액체 비료의 악취 제거 연구)

  • Bacillus species were isolated from swine manure to develope the microbial additive suitable for the rapid com-posting. The 3 of 4 isolated strains were identified as Bacillus cereus KD-2, B. pumilus KD-3, and B. licheni-formis KD-4. Bacillus sp. KD-1 was, however, not highly identical with any Bacillus sp. The isolated strains were analyzed their growth rates, enzyme activities, and antibacterial activities. The maximum growth tem-peratures of KD-1, KD-2, KD-3 and KD-4 were $45^{\circ}C$, $50^{\circ}C$, $53^{\circ}C$, and $55^{\circ}C$, respectively. The activities of pro-tease or amylase in mixed culture of 4 strains were similar in the range of $37^{\circ}C$ to $53^{\circ}C$ and activities of lipase in the range of $37^{\circ}C$ to $42^{\circ}C$ were twice higher than those of lipase in the range of $47^{\circ}C$ to $53^{\circ}C$. The antibacterial activity of KD-l, KD-2, or KD-3 against each other was not detected. That of KD-4 against KD-1, KD-2, or KD-3 was, however, detected. The organic compound and C/N ratio of compost fermented by the mixed culture were determined as 61.9% and 22.4%, respectively. The concentration of the ammonia gas was 12.35 mg/l in the liquid compost.

Comparison of Antifungal Activity, Plant Growth Promoting Activity, and Mineral-Solubilizing Ability of Bacillus sp. Isolated from Rhizosphere Soil and Root (근권 토양과 뿌리로부터 분리된 Bacillus sp.의 항진균 활성, 식물 생장 촉진 활성 및 미네랄 가용화능 비교)

  • The purpose of this study was to evaluate the antifungal activity, plant-growth-promoting activity, and mineral solubilization ability of 10 species of phytopathogenic fungi to select a Bacillus sp. from rhizosphere soils and roots that can be used as a microbial agent. The antifungal activity for phytopathogenic fungi varied based on the Bacillus sp. Among the selected strains, DDP4, DDP16, DDP148, SN56, and SN95 exhibited antifungal activity for nine or more species of phytopathogenic fungi. Regarding nitrogen-fixation ability, all Bacillus sp. showed similar levels of activity, and siderophore production ability was relatively high in ANG42 and DDP427. The indole-3-acetic acid production abilities were in the range of 1.83-67.91 ㎍/ml, with variations in activity based on the Bacillus sp. One strain with a high activity was selected from each species, and their mineral solubilization abilities were examined. Most Bacillus sp. could solubilize phosphoric acid and calcium carbonate, and DDP148 and SN56 could solubilize silicon and zinc, respectively. These results suggested that Bacillus sp. can be considered potential multi-purpose microbial agents for plant growth promotion and disease prevention.

  • Ten(10) strains of aerobic bacteria and two(2) strains of microaerobic bacteria were isolated out of soysauce which was brewed by the conventional process. The following bacteria were identified by studying their morphorogies and physiological characters. Bacillus pumillus-R-2; Bacillus subtilis var., aterimus-S-1; Bacillus licheriformis var-S-2; Bacillus subtilis-T-1; Sarcina maxima-T-2; Pediococcus acidi lactici-Z-2; Bacillus citreus var, . soyaB-Z-5. T-2 and Z-5 of the isolated bacteria were found good in growth even in the 24%-salted density, and Z-5 was more vigorous than T-2 though stinking. S-1 produced black-brown pigment from the medium containing various kinds of carbohydrate and the medium of soysauce which are available to S-1.

  • Beneficial bacteria, which have been used for medical purpose and for medicines for treating intestinal disorders, include strains of Bifidobacterium sp., Lactobacillus sp., Enterococcus sp., Clostridium butyricum, Lactobacillus sporogenes, Bacillus subtilis, Bacillus polyfermenticus and the like. Bacillus polyfermenticuss SCD with is commonly called as Bispan strain has been appropriately used for the treatment of long-term intestinal disorders, since the live strains in the form of active endospores can successfully reach the target intestine. In this study, the identification and characterization of Bispan strain was done using SEM observation, API 50CHB kits, isoprenoid quinone analysis, and fatty acid analysis. These results suggest that Bispan strain is very similar to Bacillus subtilis.

Isolation and Identification of Microorganisms in Korean Traditional Soybean Paste and Soybean Sauce (전통 된장 및 간장의 숙성기간별 생육 미생물의 분리 및 동정)

  • As a basic study for quality improvement of Korean soybean paste and soybean sauce, we investigated on microflora of soybean paste and soybean sauce fermentation. Major Microorganisms were isolated from the sample pastes and sauces, and identified systematically. Selected Microorganisms were identified by MIS whole cell fatty acid analysis by gas chromatography. Identification results showed that Bacillus licheniformis, bacillus pumilis and Bacillus subtilis were dominant in soybean paste and Staphylococcus vitulus, Bacillus subtilis, Bacillus pumilus, and Lactobacillus fermentum were dominant in soybean sauce. It seemed that these Microorganisms played an important role in soybean paste and soybean sauce fermentation and could be used for the further studies such as protease and amylase activities.

  • The study was performed to investigated the excellent microbial anticaries substance which is more effective than the chlorohexidine in the dental caries treatment. For the screening of alkaliphilic microorganism, more than 1200 bacterial strains were isolated from sea soil sample. A typ-ical strain which produced the most excellent antimicrobial substance was selected. The strain was identified novel alkalophilic Bacillus sp. through the results of morphological, biochemical and chemotaxonomical characteristics and 16S rDNA sequencing and designated as Bacillus alkalophilshaggy JY-827.
