Protoplast Culture by Electrofusion of Protoplasts from Solanum sisymbriifolium and Other Solanum Species

  • This research was conducted to get the basic materials necessary to obtain the somatic hybrid plant between Solanum sisymbriifolium and other Solanum species (S. integrifolium and S. toxicarium). Regarding the formation of colony from the protoplast in S. sisymbriifolium, S. integrifolium and the fused protoplast mixture; for the S. sisymbriifolium, a colony was observed in F medium(Kao medium containing $5.0mg{\cdot}L^{-1}\;NAA,\;1.0mg{\cdot}L^{-1}$ 2,4-D and $1.0mg{\cdot}L^{-1}$ BA); and for the S. integrifolium, in G medium (a half strength MS medium containing 0.03 M sucrose, 0.4 M mannitol, $1.0mg{\cdot}L^{-1}\;NAA,\;1.0mg{\cdot}L^{-1}$ kinetin) respectively. In mixed cultured protoplast after electriofusion treatment, the cell division and colony formation were observed in both media F and G. For the shoot and root formation rate, there was no difference between the parent of each breed and mixed protoplast regardless of the medium. In the fused protoplast mixture of S. sisymbriifolium and S. toxicarium, a colony formation was also observed in both media F and H(a half strength MS medium containing 0.03 M sucrose, 0.4 M mannitol, $1.0mg{\cdot}L^{-1}\;NAA,\;1.0mg{\cdot}L^{-1}$ kinetin); and there was no difference in the shoot and root formation rate between the parent and the mixed protoplast.

정보검색기반 결함위치식별 기술의 성능 향상을 위한 버그리포트 품질 예측 (Bug Report Quality Prediction for Enhancing Performance of Information Retrieval-based Bug Localization)

  • 버그리포트는 소프트웨어의 유지보수 단계에서 발생한 결함 정보를 담고 있는 문서로서 개발자가 해당 결함을 수정하기 위해 필수적인 정보이다. 이 때 개발자가 버그리포트를 해결하기 위해 결함을 추적하는 시간을 단축시키기 위한 정보검색기반 결함위치식별 기술들이 제안되었다. 그러나 정보검색에 유용하지 못한 내용들로 작성된 낮은 품질의 버그리포트가 등록 될 경우 결함위치식별 성능이 크게 저하된다. 본 논문에서는 낮은 품질의 버그리포트를 선별하기 위한 품질 예측 방법을 제안한다. 이 과정에서 버그리포트의 쿼리로써의 품질 요소를 정의하고, 기계학습을 사용하여 품질을 예측한다. 제안 방법을 오픈 소스 프로젝트에 적용하여 기존 품질 예측 기술 대비 평균 6.62% 더 정확하게 예측하였다. 또한 기존 결함위치식별 기술에 제안 예측 기술과 자동 쿼리 재구성 기술을 함께 적용한 경우 결함위치식별 정확도를 1.3% 향상시켜, 제안 품질 예측 기술이 정보검색기반 결함위치식별 기술의 성능 향상을 도울 수 있음을 확인하였다.

  • 토양에서 분리, 선별한 Streptomyces 속의 한 균주가 생산하는 Penicillinase의 최적생산조건을 조사한 결과를 요약하면 다음과 같다. 탄소원으로서는 glucose, 실소원으로는 L-as-paragine이 가장 우수했으며 금속감중에서 Mn C $l_2$를 첨가했을때 효소생성을 37% 증가시켰다. Amino 산중에서 L-leucine이 본 효소생산을 약간 증가시켰으며 L-histidine L-methionine은 크게 저하시켰다. Riboflavine, i-inositol, hesperidine, niacinamide, biotin, DL-$\alpha$- lipoic acid, folic acid가 효소생산을 증가시켰으며 항생물질중에서 cephradine, cephalexin, ampicillin, cloxacillin의 첨가가 효소생성을 크게 증가시켰다. 본 효소생산의 최고 pH는 7.0이며 최적온도는 28$^{\circ}C$였다. 또 500$m\ell$ Erlenmeyer flask에서 175$m\ell$의 배지를 가하여 3 일간 배양했을 때 효소생성이 최고에 도달했다.

당근의 다량증식을 위한 순원기 배양 (Shoot Primordium Culture for Multiplication of Carrot)

  • 당근의 기내 증식에 있어서 계대배양을 반복하더라도 분열능과 재분화능에 차이가 없고 변이가 생기지 않는 순 원기배양법을 확립코자 하였다. 엽 원기 2개가 붙은 경정조직을 BA 2.0 mg/L 와 NAA 0.2 mg/L 를 첨가한 MS 기본 액체배지에 치상하고 24시간 강한 광선 아래 2rpm으로 수직 회전배양하였을 때 순 원기가 유기되고 덩어리로 자라났다. 순 원기 유기는 공여친의 유전자형과 재배 계절 및 발육단계의 영향을 거의 받지 않는 것으로 판단되었다. 이 순 원기 덩어리는 사방 2~3 mm정도의 크기로 순 원기가 유기된 배지에 계대배양 하였을때 잘 증식되었는데 표면에 작은 신초를 가진 것과 그렇지 않은 것, 그리고 캘러스의 3가지 형태로 분화하였다. 표면에 작은 신초를 가진 것이 역시 표면에 작은 신초를 가진 순 원기 덩어리로 증식되었다. 이 표면에 작은 신초를 가진 것이 1/2MS 기본배지에 KN 0.2-1.0 mg/L 과 IAA 0.2~1.0 mg/L 를 첨가한 고체배지에서 주로 식물체로 잘 분화하였다. 일단 순 원기가 유기되어 덩어리를 형성한 후에는 1년 동안에 8.7$\times$$10^{12}$개의 비율로 증식될 수 있을 것으로 계산되었다. 그리고 13번의 계대배양 후에도 증식능에 차이가 없었으며 염색체상의 이상도 없었다.

Characterization of BST films for high tunable thin film capacitor

  • This is for the electrical characterization by IDC pattern using BST$(Ba_{0.5}Sr_{0.5}TiO_3)$ thin film. BST materials had been chosen for high frequency applications due to it's high permitivity and tunability. The BST thin films have been deposited on $Al_2O_3$ Substrates by Nd-YAG pulsed laser deposition with a 355nm wavelength at $700\;^{\circ}C$. The post deposition annealing at $750^{\circ}C$ in flowing $O_2$ atmosphere for 1 hours. The capacitance of IDC patterns have been measured from 1 to 10 GHz as a function of electric field ($\pm40$ KV/cm) at room temperature using inter-digital Au electrodes deposited on top of BST. The IDC patterns have three type of fingers number. For the 10 pairs finger was the best capacitance onto $Al_2O_3$ substrate. The capacitance was 0.9pF. Also Dielectric constant was been 351 at 100 mTorr and annealing temperature $750^{\circ}C$ for 1 hour. The loss tangent was been 0.00531.

HPLC analysis of Polycyclic Aromatic Hydrocarbons(PAHs) in Mussels(Mytilus edulis) living in the Intertidal Zone of Kori, Korea

  • Polycyclic aromatic hydrocarbons(PAHs) are ubiquitous contaminants in coastal marine environment. PAHs enter estuarine and nearshore marine environment via several routes such as combustion of fossil fuels, domestic and industrial effluents and oil spills. In August of 1997, mussels(Mytilus edulis) were collected at 6 sites near Kori nuclear power plant in order to analyze the PAH content by HPLC with uv/vis detection. Unfortunately, I could not find any living oysters in which I firstly intended to measure the PAH content in the study area. NPTHL and ANCPL were the major dominant PAH compounds in mussels living in the intertidal zone of Kori, Korea, and DahA, BbF, BaP were the next dominant PAH group in mussels in the study area. The mean concentrations of 15 PAH in mussels ranged from 3.2 to 1,680 ppb(mean 105$\pm$60.5 ppb). Compared with other studies world over, the concentrations of carcinogenic PAHs were relatively low in mussels in the study area, even though total PAH content was rather high. According to N/P(Naphthalene/Phenanthrene) ratio(147) and the ratio of 2∼3 ring to 3∼5 ring PAHs(58∼90 %) in mussels in the study area, I expect that the major source of PAHs in this study area is rather fresh petroleum-derived. This study presents preliminary data for the PAH levels in mussels from the intertidal zone of Kori, and the data will hopefully be utilized for the assessment of oil pollution in the East Sea, Korea.

화학적으로 변형된 전구용액을 이용한 YBCO 박막 제조 (Fabrication of YBCO films in MOD processing via chemically modified precursor solution)

  • Superconducting YBCO films were successfully fabricated by MOD process using chemically modified precursor solution. In this study, a chemically modified precursor solution for MOD processing was synthesized using metal-organic salts and organic additives. It was shown that crack-free and uniform precursor films were formed after calcination in humidified Oxygen atmosphere. Less than 3 hours are required to finish the calcination process. XRD measurement shows that $BaF_2,\;CuO,\;Y_2O_3$ are major constituent of precursor films. Furthermore, YBCO films without any secondary phases were successfully fabricated after annealing in wet $Ar/O_2$ atmosphere. The YBCO film prepared on a $LaAlO_3$ single crystal substrate ($10mm{\times}10mm$) gives transport $I_c$ of 10A at 77K. This chemical modification approach is a possible candidate for improving MOD-processing of YBCO coated conductor.

PMN 계 유전체 적용 EL 소자의 광전특성 연구 (The Study of Opto-electric Properties in EL Device with PMN Dielectric Layer)

  • In this study, the opto-electric properties of EL devices with PMN dielectric layer with variation of firing tempereature were investigated. For the PMN dielectric layer process, the paste was prepared by optimization of quantitative mixing of PMN powder, $BaTiO_3$, Glass Frit, $\alpha$-Terpineol and ethyl cellulose. The EL device stack consists of Alumina substrate ($Al_2O_3$), metallic electrode (Au), insulating layer (manufactured PMN paste), phosphor layer (ELPP- 030, ELK) and transparent electrode (ITO), which is well structure as a thick film EL device. The phase transformation properties of PMN dielectric with various firing temperatures of $150^{\circ}C$ to $850^{\circ}C$ was characterized by XRD. Also the opto-electric properties of EL devices with different firing temperature were investigated by LCR meter and spectrometer. We found the best opto-electric property was obtained at the condition of $550^{\circ}C$ firing which is 3432.96 $cd/m^2$ at 1948.3 pF Capacitance, 40 kHz Frequency, 40% Duty, Vth+330 V voltage.

PMF 모델을 이용한 대기 중 PM-10 오염원의 확인 (Source Identification of Ambient PM-10 Using the PMF Model)

  • The objective of this study was to extensively estimate the air quality trends of the study area by surveying con-centration trends in months or seasons, after analyzing the mass concentration of PM-10 samples and the inorganic lements, ion, and total carbon in PM-10. Also, the study introduced to apply the PMF (Positive Matrix Factoriza-tion) model that is useful when absence of the source profile. Thus the model was thought to be suitable in Korea that often has few information about pollution sources. After obtaining results from the PMF modeling, the existing sources at the study area were qualitatively identified The PM-10 particles collected on quartz fiber filters by a PM-10 high-vol air sampler for 3 years (Mar. 1999∼Dec.2001) in Kyung Hee University. The 25 chemical species (Al, Mn, Ti, V, Cr, Fe, Ni, Cu, Zn, As, Se, Cd, Ba, Ce, Pb, Si, N $a^{#}$, N $H_4$$^{+}$, $K^{+}$, $Mg^{2+}$, $Ca^{2+}$, C $l^{[-10]}$ , N $O_3$$^{[-10]}$ , S $O_4$$^{2-}$, TC) were analyzed by ICP-AES, IC, and EA after executing proper pre - treatments of each sample filter. The PMF model was intensively applied to estimate the quantitative contribution of air pollution sources based on the chemical information (128 samples and 25 chemical species). Through a case study of the PMF modeling for the PM-10 aerosols. the total of 11 factors were determined. The multiple linear regression analysis between the observed PM-10 mass concentration and the estimated G matrix had been performed following the FPEAK test. Finally the regression analysis provided source profiles (scaled F matrix). So, 11 sources were qualitatively identified, such as secondary aerosol related source, soil related source, waste incineration source, field burning source, fossil fuel combustion source, industry related source, motor vehicle source, oil/coal combustion source, non-ferrous metal source, and aged sea- salt source, respectively.ively.y.

Studies on magneto-electro-elastic cantilever beam under thermal environment

  • A smart beam made of magneto-electro-elastic (MEE) material having piezoelectric phase and piezomagnetic phase, shows the coupling between magnetic, electric, thermal and mechanical under thermal environment. Product properties such as pyroelectric and pyromagnetic are generated in this MEE material under thermal environment. Recently studies have been published on the product properties (pyroelectric and pyromagnetic) for magneto-electro-thermo-elastic smart composite. Hence, the magneto-electro-elastic beam with different volume fractions, investigated under uniform temperature rise is the main aim of this paper, to study the influence of product properties on clamped-free boundary condition, using finite element procedures. The finite element beam is modeled using eight node 3D brick element with five nodal degrees of freedom viz. displacements in the x, y and z directions and electric and magnetic potentials. It is found that a significant increase in electric potential observed at volume fraction of $BaTiO_3$, $v_f$ = 0.2 due to pyroelectric effect. In-contrast, the displacements and stresses are not much affected.
