Biodiversity and Enzyme Activity of Marine Fungi with 28 New Records from the Tropical Coastal Ecosystems in Vietnam

  • The coastal marine ecosystems of Vietnam are one of the global biodiversity hotspots, but the biodiversity of marine fungi is not well known. To fill this major gap of knowledge, we assessed the genetic diversity (ITS sequence) of 75 fungal strains isolated from 11 surface coastal marine and deeper waters in Nha Trang Bay and Van Phong Bay using a culture-dependent approach and 5 OTUs (Operational Taxonomic Units) of fungi in three representative sampling sites using next-generation sequencing. The results from both approaches shared similar fungal taxonomy to the most abundant phylum (Ascomycota), genera (Candida and Aspergillus) and species (Candida blankii) but were different at less common taxa. Culturable fungal strains in this study belong to 3 phyla, 5 subdivisions, 7 classes, 12 orders, 17 families, 22 genera and at least 40 species, of which 29 species have been identified and several species are likely novel. Among identified species, 12 and 28 are new records in global and Vietnamese marine areas, respectively. The analysis of enzyme activity and the checklist of trophic mode and guild assignment provided valuable additional biological information and suggested the ecological function of planktonic fungi in the marine food web. This is the largest dataset of marine fungal biodiversity on morphology, phylogeny and enzyme activity in the tropical coastal ecosystems of Vietnam and Southeast Asia. Biogeographic aspects, ecological factors and human impact may structure mycoplankton communities in such aquatic habitats.

The first record of the rare fern Pteris griffithii (Polypodiales: Pteridaceae: Pteridoideae) in the Bhutan Himalayas

  • Pteris griffithii Hook., one of the rarest fern species on the Indian subcontinent, is reported from Bhutan for the first time. The identity of this species was confirmed through morphological determination at the National Herbarium (THIM) of the National Biodiversity Centre (NBC) of Bhutan. It was found only in one location, in Gyelpozhing in eastern Bhutan, at an elevation of 521 m a.s.l. on 10 January 2016. Given that a very limited study of this species was conducted, the knowledge baseline with regard to its distribution is poor. It is also reported that this species has not been found for several years. The species is also considered to be very rare or critically endangered in some countries; however, there are no assessments on the International Union for Conservation of Nature (IUCN) Red List for this particular species. This paper attempts to provide baseline information considering its rarity and data deficiency. This species is also reported from the adjacent neighboring Indian state of Arunachal Pradesh as very rare, and also from Myanmar; however, confirmation of its presence in China is not clear at this time. Therefore, considering its data deficient status, we attempt to document it scientifically to create a knowledgebase pertaining to this particular species. Concurrently, this species merits further research to understand its distribution patterns in Bhutan and any related anthropogenic threats.

  • To distinguish manchurian clematis, Clematis terniflora var. mandshurica and three-leaf clematis, C. apiifolia, we collected 9 nuclear ITS sequences and two sequences of trnQ-trnH intergenic spacer and trnH region in GenBank database. Those sequences were aligned to find differences between those of C. terniflora var. mandshurica and C. apiifolia. Two primer pairs were newly designed base on the differences between two species and conducted multiplex PCR. The size of amplified fragments using generated primers were 380 base pairs (only three-leaf clematis) and 189 base pairs (both species). This genetic marker based on PCR is useful to discrimination of C. terniflora var. mandshurica and C. apiifolia

Taxonomic Review of L. makinoana Complex (Orchidaceae; Epidendroideae; Malaxidae) in Korea

  • 한국에서 자생하고 있는 Liparis 속의 11분류군 중 Liparis koreana (Nakai) Nakai을 제외하고 대부분은 경기도와 강원도부터 제주도 및 도서지방을 포함한 전국에 분포하고 있다. 최근 국내 Liparis japonica(Miq.) Maxim. 로 알려진 개체군에 대해 학명이 잘못 적용된 결과가 보고되었고, 본 개체군을 포함하여 Liparis makinoana complex에 대한 국내 개체군에 대해 재검토를 하였다. 그 결과 L. makinoana complex은 제주 Type (Type 1)과 내륙의 2 Types (Type 2, 3)으로 3 Types가 확인되었다. Type 1-3은 형태학적으로 유사하지만, 종의 형태학적 형질과 개화기, 그리고 분포지역에서 뚜렷한 차이를 보였다. Type 1은 꽃이 가장 크며, 순판은 넓은 난형이고, 개화기는 5-6월이며, 제주도에서 분포하는 것으로 확인되었다. Type 2는 꽃이 중간크기이며, 순판이 난형이고, 개화기는 6월 중순-7월 중순이며, 중북부지방에 주로 분포한다. 반면에, Type 3는 꽃이 가장 작고, 순판이 좁은 난형이고, 개화기가 6월 초-7월 초이며, 백두대간을 따라 분포하는데, Type 3는 Type 1과 Type 2에 비해 화서가 길다.

Tyrosinase inhibition effects of Korean edible brown, green, and red seaweed extracts

  • The tyrosinase inhibition effects of 23 marine-derived seaweeds harvested in Korea were screened to determine their potential as skin-whitening agents. Of the 23 species initially screened, the total phenolic (TP) content of brown, green, and red seaweeds were 7.62-280.11, 5.24-62.37, and 0.63-28.76 phloroglucinol equivalents (PGE) mg/g, respectively. Brown seaweed extracts exhibited much stronger inhibitory activities than green and red seaweed extracts. Among the brown seaweeds, Ecklonia cava had the highest TP content (280.11 PGE mg/g) and the strongest tyrosinase inhibitory effect with a half maximal inhibitory concentration (IC50) value of 4.38 ㎍/mL. The kinetics of tyrosinase inhibition, analyzed by Lineweaver-Burk plots, found E. cava extract to be a non-competitive inhibitor. This study's results indicated that E. cava's inhibition of tyrosinase may have potential applications in the cosmetic industry.

The Current status of the cooperative research on the biodiversity in North-East Asia

  • In this paper, the cooperative research on the biodiversity conservation in North-East Asia is introduced. This research was initiated by National Institute of Biological Resources (NIBR) in 2012. The bilateral MOU and following MOA had been signed between NIBR and National University of Mongolia (NUM) in 2012, whereas those had been signed between NIBR and Institute of Zoology, Chinese Academy of Science (IOZ, CAS) in 2013. Details of the cooperative research based on those were presented.

  • Fungi commonly encountered on monocotyledonous substrates were evaluated for their in vitro ability to produce enzymes involved in lignocellulose breakdown. Most were capable of structural polysac-charide utilization, but few produced enzymes associated with lignin breakdown. None of the mono-cotyledon-inhabiting fungi produced reactions as strongly as wood decay fungi.

Development of DNA Barcode Database and Identification System of Forest Mushrooms in Korea

  • Over five thousand higher fungal specimens were collected from 32 forest areas of Chungcheong and Gyeongsang province from 2012 to 2013. We obtained 513 strains and 3,120 ITS sequences. Mushrooms were first identified with macro- and micro-scopic characters, and their identification was confirmed on the basis of ITS sequences. Voucher specimens were designated for each species found in Korea. Construction of DNA barcoding Database is currently underway with sequences of 409 species. During the development of the database, some new species were recognized, along with several Korean new records. When the system has been completed, it will provide essential molecular information for metagenomic and phylogenetic researches for higher fungi.
