Bioassay-directed fractionations of the seed extracts of Trichosanthes kirilowii, have resulted in the isolation of two new compounds, 2-(4-hydroxy-3-methoxyphenyl)-3-(2-hydroxy-5-methoxyphenyl)-3-oxo-1-propanol (2) and isoetin 5'-methyl ether (5,7,2',4'-tetrahydroxy-5'-methoxyflavone) (3), together with two known compounds, 7-hydroxychromone (1) and 5,7,4'-trihydroxy-3',5'-dimethoxyflavone (tricin, 4). Their structures were characterized by spectroscopic analysis such as 2D-NMR, HRTOFMS, and UV. Compound 3 showed cytotoxicity against human lung cancer cell line A549, human skin melanoma SK-Mel-2, and mouse melanoma B16F1, with IC50 of 0.92, 8.0, and 7.23 μg/mL, respectively.
본 연구는 연잎 추출물의 미백 화장품 첨가물로서 사용이 가능한지를 연구하였다. 연잎 추출물의 항산화 활성을 측정하게 위해 전자공여능 측정, xanthine oxidase 억제 효과 실험을 실시하였고, 미백활성을 알아보기 위하여 tyrosinase 저해활성을 측정하여 1,000 μg/ml의 농도에서 42.7%의 효과를 나타내었다. 또한 연잎 추출물에 대한 세포생존율을 MTT assay로 측정한 결과 1,000 μg/ml 농도에서 81.61%를 이상의 세포생존율을 확인할 수 있었다. 미백 관련 인자인 MITF, TRP-1, TRP-2 및 tyrosinase의 단백질 발현과 mRNA 발현 억제를 25, 50, 100 μg/ml 농도에서 측정하였다. MITF, TRP-1, TRP-2 및 tyrosinase의 단백질 발현은 100 μg/ml 농도에서 각각 69.6%, 27.7%, 67.3%, 67.8%의 저해 효과를 나타내었고, MITF, TRP-1, TRP-2 및 tyrosinase의 mRNA발현은 100 μg/ml 농도에서 각각 67.5%, 71.4%, 85.7%, 83.6%의 억제를 나타내었다. 이러한 연구결과를 보았을 때, 연잎 추출물이 항산화 및 미백활성에 효과를 나타내었고, 화장품 첨가물로서의 가능성을 확인할 수 있었다.
울릉도 자생 삼나물(Aruncus dioicus)은 최근 monoterpenoids 생리활성 물질이 밝혀지면서 여러 가지 항산화와 관련된 생리활성이 확인되고 있다. 삼나물 ethyl acetate 분획물에 의한 미백효과를 검증하기 위하여 흑색세포종인 B16F10을 이용하여 실험을 진행하였다. 삼나물 ethyl acetate 분획물(ADE)의 세포독성을 측정하기 위하여 MTT assay를 실시한 결과, ADE $500{\mu}g{\cdot}mL^{-1}$ 이상의 농도에서 미비한 세포독성(10% 이상)을 확인하였으며, 이후 실험에서는 5, 10, $50{\mu}g{\cdot}mL^{-1}$ 농도가 세포 내에서 tyrosinase 활성과 멜라닌 양의 변화를 측정하였다. 그 결과 ADE 농도에 따라 tyrosinase 활성이 감소하고 총 멜라닌 함량이 감소하는 것을 확인하였다. 특히 $50{\mu}g{\cdot}mL^{-1}$에서 35.6% tyrosinase 활성억제, 58.8% 멜라닌 함량이 감소하는 것을 확인하였다. 또한 ADE에 의해 미백과 관련된 tyrosinase, tyrosinase related protein 1(TRP1), TRP2, microphthalmia associated transcription factor(MITF) 및 그 상위 단계인 cAMP와 protein kinase A(PKA)의 단백질 양이 감소하여 cAMP response binding protein(CREB)의 인산화는 감소하고 extracellular signal related kinase(ERK)의 인산화는 증가하는 것을 확인하였다. 본 연구에서는 울릉도 자생 삼나물의 미백효과에 관한 효능을 확인하고 기능성화장품의 소재로서의 활용 가능성이 있음을 확인하였다.
In this study, we used the mixture made from the Rice bran 45 ㎏, Dendropanax 5 ㎏, the sugar of the 10% of the total weight, and the enzyme of the 0.1% of the total weight. After the mixture were fermented for 90 days under 20 $^{\circ}C$, we measured the cell viability and the inhibition rate of the melanin biosynthesis, the activity of tyrosinase and superoxide dismutase (SOD) in malignant melanoma, B16F10 cells, in order to survey the whitening effect and the mechanism of the effects on the sample. As a result, the samples significantly suppressed the cell viability of B16F10 in more than 500 ${{\mu}g}$/㎖ and significantly inhibited the generation of melanin induced by ${\alpha}$-MSH in more than 1,000 ${{\mu}g}$/㎖. Sample decreased the activity of tyrosinase while increased the activity of SOD in dose dependent manner. Therefore, we considered that the fermentation extract made from a Rice bran and Dendropanax will be able to produce high value-added products, if used as a commercial.
본 논문은 재배가 용이하고 민간요법에서 다양하게 이용되고 있는 곰보배추의 에탄올 추출물이 화장품 소재로서 가능성이 있는지를 알아보기 위해 항산화, 미백효능에 대하여 관찰하였다. B16F10 세포에서 곰보배추 에탄올 추출물 25, 50, $100{\mu}g/mL$ 농도에서 독성이 나타나지 않았다. DPPH radical 소거능을 관찰한 결과 모든 농도에서 소거능을 보여주었고, $50{\mu}g/mL$의 농도에서 65.2%, $100{\mu}g/mL$에서 77.6%의 강한 항산화 효능을 나타냈다. Raw 264.7 세포 내에서 ROS 생성 저해능을 관찰한 결과 농도 의존적으로 유의하게 나타냈고, $100{\mu}g/mL$ 농도에서는 39.1% 억제하는 것으로 나타났다. NO생성 억제를 관찰하기 위해 Raw 264.7 세포에 곰보배추 에탄올 추출물을 25, 50, $100{\mu}g/mL$ 농도별로 첨가하여 관찰한 결과 농도 의존적으로 NO생성을 억제하였다. 시험관 내에서 L-DOPA와 L-tyrosine을 이용하여 곰보배추 에탄올 추출물이 tyrosinase activity를 농도 의존적으로 억제하는 것을 나타냈다. 세포 내에서 MSH를 가한 B16F10 세포에 곰보배추 에탄올 추출물을 25, 50, $100{\mu}g/mL$의 melanin 함량을 관찰한 결과 농도 의존적으로 억제하는 것을 보여주며 $100{\mu}g/mL$에서 30.7%로 억제하였다. 따라서 곰보배추 에탄올 추출물이 항산화 기능이 있는 미백 기능성 화장품의 소재로서 개발 가능성이 충분히 있는 것으로 사료된다.
To develope skin-whitening or therapeutic agents against hyperpigmentation, aqueous extract from Amomum xanthioides (AEAX) was evaluated for melanogenesis inhibitory activity in B16F10 melanoma cell. The treatment with AEAX at the 0.5 and 1.0 mg/ml level significantly inhibits the biosynthesis of melanin compared with untreated control. The AEAX-treated cells at the 1.0 mg/ml level were more efficient than commercial arbutin at 0.1 mg/ml. The tyrosinase activity also significantly decreased in AEAX-treated cells at the 0.5 and 1.0 mg/ml level. The Western analyses confirmed the significantly decreased expression of tyrosinase and tyrosinase-related protein-1 by AEAX treatment. These results indicate that AEAX may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression and serve as a new candidate in the design of new skin-whitening or therapeutic agents.

Abnormal melanin synthesis results in several hyperpigmentary disorders such as freckles, melanoderma, age spots, and other related conditions. In this study, we investigated the anti-melanogenic effects of an extract from the microalgae Chlamydomonas reinhardtii (CE) and potential mechanisms responsible for its inhibitory effect in B16F10, normal human epidermal melanocyte cells, and human skin-equivalent models. The CE extract showed significant dose-dependent inhibitory effects on ${\alpha}$-melanocyte-stimulating, hormone-induced melanin synthesis in cells. Additionally, the CE extract exhibited suppressive effects on the mRNA and protein expression of microphthalmia-associated transcription factor, tyrosinase, tyrosinase-related protein-1, and tyrosinase-related protein-2. The CE extract also inhibited the phosphorylation of protein kinase A and extracellular signal-related kinase, which function as upstream regulators of melanogenesis. Using a three-dimensional, reconstructed pigmented epidermis model, the CE-mediated, anti-pigmentation effects were confirmed by Fontana-Masson staining and melanin content assays. Taken together, CE extract can be used as an anti-pigmentation agent.
Skin photoaging occurs due to chronic exposure to solar ultraviolet radiation (UV), the main factor contributing to extrinsic skin aging. Clinical signs of photoaging include the formation of deep, coarse skin wrinkles and hyperpigmentation. Although melanogenesis and skin wrinkling occur in different skin cells and have different underlying mechanisms, their initiation involves intracellular calcium signaling via calcium ion channels. The ORAI1 channel initiates melanogenesis in melanocytes, and the TRPV1 channel initiates MMP-1 production in keratinocytes in response to UV stimulation. We aimed to develop a drug that may simultaneously inhibit ORAI1 and TRPV1 activity to help prevent photoaging. We synthesized nootkatol, a chemical derivative of valencene. TRPV1 and ORAI1 activities were measured using the whole-cell patch-clamp technique. Intracellular calcium concentration [Ca2+]i was measured using calcium-sensitive fluorescent dye (Fura-2 AM). UV-induced melanin formation and MMP-1 production were quantified in B16F10 melanoma cells and HaCaT cells, respectively. Our results indicate that nootkatol (90 μM) reduced TRPV1 current by 94% ± 2% at -60 mV and ORAI1 current by 97% ± 1% at -120 mV. Intracellular calcium signaling was significantly inhibited by nootkatol in response to ORAI1 activation in human primary melanocytes (51.6% ± 0.98% at 100 μM). Additionally, UV-induced melanin synthesis was reduced by 76.38% ± 5.90% in B16F10 melanoma cells, and UV-induced MMP-1 production was reduced by 59.33% ± 1.49% in HaCaT cells. In conclusion, nootkatol inhibits both TRPV1 and ORAI1 to prevent photoaging, and targeting ion channels may be a promising strategy for preventing photoaging.
아시아에서 한방약초로 널리 알려진 당귀 추출물의 미백활성을 알아보기 위하여 tyrosinase 저해활성을 측정한 결과 1,000 ${\mu}g/ml$의 농도에서 70% 이상의 활성을 나타내었다. 또한 당귀 추출물에 대한 멜라노마 세포(B16F10)의 세포생존율을 확인한 결과 500 ${\mu}g/ml$의 농도에서 99% 이상의 세포생존율을 확인할 수 있었다. 미백 관련 인자인 MITF, TRP-1, TRP-2 및 tyrosinase의 mRNA 발현량을 측정한 결과 50 ${\mu}g/ml$의 농도에서 각각 85.7%, 123.9%, 68.8%, 208%로 당귀 추출물을 처리하지 않은 군보다 감소하였음을 확인할 수 있었다. 이러한 연구 결과에 따라 당귀 추출물이 melanin 합성과 관련이 있는 유전자 발현의 억제효과가 있음을 확인할 수 있었으며, 미백 화장품 소재로서의 가능성을 확인하였다.
