Effect on Melanogenic Protein Expression of Acanthoic Acid isolated from Acanthopanax koreanum in Murine B16 Melanoma

  • Melanogenesis is a well-known physiological response of human skin that may occur because of exposure to ultraviolet light, for genetic reasons, or due to other causes. In our effectors to find new skin lightening agents, acanthoic acid (AA) was investigated for its ability to inhibit melanogenesis. The effects of AA isolated from A.koreanumun the expression of $\alpha$-MSH-induced melanogenic factors (tyrosinase, tyrosinase related protein (TRP)-1, TRP-2 and MITF (microphthalmla-associated transcriptional factor)) were investigated in murine B16F10 melanoma cells. The results indicate that AA was an effective inhibitor of melanogenesis in B16F10 cells. To elucidate the mechanism of the effect of AA on melanogenesis, we performed Western blotting for melanogenic proteins. AA inhibited melanogenic factors (tyrosinase, TRP-1, TRP-2) expressions. In this study, we also confirmed that AA decreased the protein level of MITF proteins, which would lead to a decrease of tyrosinase and related genes in B16F10 melanoma cells. In order to apply AA to the human skin, the cytotoxic effects of the AA were determined by MTT assays using human keratinocyte HaCaT cells. Based on these results, we suggest that AA be considered possible anti-melanogenic agent and might be effective against hyperpigmentation disorders for the topical application.

Seed Transmission of Bipolaris coicis, B. cynodontis, B. maydis and Curvularia lunata causing Leaf Blight of Job's tears (율무에 잎마름 증상을 일으키는 Bipolaris coicis, B. cynodontis, B. maydis 및 Curvularis lunata의 종자전염)

  • Bipolaris coicis, B. cynodontis, B. maydis and Curvularia lunata were leaf blight fungi detected from 45 seed samples of Job's tears and B. coicis was the predominant species in seed samples followed by C. lunata, B. cynodontis and B. maydis. When the seed components were plated on test tube agar, B. coicis and C. lunata were highly detected from invelucre, glume, endosperm and stamen, but not detected from plumule. Seed infection with B. coicis casued seed rot, coleoptile blight and seedling blight of Job's tears. Conidial characteristics of leaf blight fungi were as follows; B. cynodontis was fusiform, brown, slightly curved, 0~5 distoseptate, and 16.8-48$\times$7.2-16.8${\mu}{\textrm}{m}$ in size; B. cynodontis was fusiform, brown, slightly curved, 0~8 distoseptate, and 16.8-72$\times$9.6-19.2 ${\mu}{\textrm}{m}$; B. maydis was fusiform, brown, distintly curved, 0~10 distoseptate, and 28.8-110.4$\times$12-21.6 ${\mu}{\textrm}{m}$. C. Lunata was fusiform, brown, typically curved, 0~3 distoseptate, and 7.2-24$\times$4.8-12 ${\mu}{\textrm}{m}$. B. coicis was highly pathogenic to Job's tears and corn, weakly pathogenic to rice, but not pathogenic to wheat and barley. c. lunata was highly pathogenic to Job's tears (No. 2), corn, wheat and barley, weakly pathogenic to Job's tears (No. 1), but not pathogenic to rice. All treatments were effective to inhibition of leaf blight fungi when carboram, benoram, fludioxonil, prochloraz, thioram, and tap water treated to infected seeds.

Inhibitory effect of glyceollin isolated from soybean against melanogenesis in B16 melanoma cells

  • Natural products with non-toxic and environmentally friendly properties are good resources for skin-whitening cosmetic agents when compared to artificial synthetic chemicals. Here, we investigated the effect of glyceollin produced to induce disease resistance responses of soybean to specific races of an incompatible pathogen, phytophthora sojae, on melanogenesis and discussed their mechanisms in melanin biosynthesis. We found that glyceollin inhibits melanin synthesis and tyrosinase activity in B16 melanoma cells without cytotoxicity. To elucidate the mechanism of the effect of glyceollin on melanogenesis, we conducted western blot analysis for melanogenic enzymes such as tyrosinase, tyrosinase-related protein-1 (TRP-1), and TRP-2. Glyceollin inhibited tyrosinase and TRP-1 protein expression. Additionally, glyceollin effectively inhibited intracellular cAMP levels in B16 melanoma cells stimulated by $\alpha$-melanocyte stimulating hormone ($\alpha$-MSH). These results suggest that the whitening activity of glyceollin may be due to the inhibition of cAMP involved in the signal pathway of $\alpha$-MSH in B16 melanoma cells.

Toxin Gene and Antibiotic Resistance of Staphylococcus aureus and Bacillus cereus Isolated from Indoor Air in Cafeteria (급식실 실내공기에서 분리된 황색포도상구균과 바실러스 세레우스의 독소 유전자 및 항생제 내성)

  • In this study, toxin gene and antibiotic resistance of food poisoning strains isolated from indoor air in the cafeteria were analyzed to prevent food poisoning. Staphylococcus aureus (16 strains) and Bacillus cereus (37 strains) isolated from indoor air in child care center were tested. The toxin genes of S. aureus and B. cereus were detected by PCR assay. The antimicrobial susceptibility test followed the disc diffusion method described by the Clinical and Laboratory Standard Institute. The seg and sei toxin genes were detected in 11 of 16 S. aureus strains (68.6%). The nheA and nheB toxin genes were detected in 37 B. cereus strains. In this study, a total of 12 toxin gene patterns of B. cereus were found, among which the nheA-nheB-nheC toxin gene was found to be the most frequent pattern. The result of the antimicrobial susceptibility test of S. aureus revealed 93.8% and 87.5% resistance to ampicillin and penicillin antibiotics, but methicillin resistance S. aureus and vancomycin resistance S. aureus were not detected. All 37 B. cereus tested in this study were resistant to ampicillin and penicillin antibiotics. Based on the result of this study, it was judged that regular ventilation and air quality management were necessary to prevent food poisoning caused by S. aureus and B. cereus contaminated in the indoor air of child care centers.

Chronology and environment of the Palaeolithic and Neolithic cultures on the southern Russian Far East

  • The results of geoarchaeological studies of the prehistoric cultural complexes on the Russian Far East (Primorye, or Maritime Province; the Amur River basin; and Sakhalin Island) are presented. Upper Palaeolithic sites are dated to ca. 40,000-10,500 B.P. They existed during the mild climate of the Chernoruchie interstadial (ca. 40,000-21,000 B.P.); during harsh climate at the Last Glacial Maximum, ca. 20,000-18,000 B.P., in several places on the Russian Far East (Primorye, Amur River basin, and Sakhalin); and during climatic amelioration in the Late Glacial time, ca. 16,000-10,500 B.P. The earliest Neolithic sites, represented by Osipovka and Gromatukha cultures, existed at ca. 13,000-10,000 B.P. in the environment of coniferous forests with admixture of broadleaved taxa. Since ca. 8000 B.P., Neolithic cultures appeared in all of the Russian Far East. They existed until ca. 3000 B.P., first during the Holocene Climatic Optimum, ca. 8000-5000 B.P., in the environment of coniferous-broadleaved forests, and later, at ca. 5000-3000 B.P., in the environment of birch-oak and coniferous forests.

Overexpression of Cuphea viscosissima CvFatB4 enhances 16:0 fatty acid accumulation in Arabidopsis

  • Cuphea viscosissima plants accumulate medium-chain fatty acids (MCFAs), i.e., those containing 8 ~ 14 carbons, in their seeds, in addition to the longer carbon chain fatty acids (≥16 carbons) found in a variety of plant species. Previous studies have reported the existence of three C. viscosissima MCFA-producing acyl-acyl carrier protein (ACP) thioesterases with different substrate specificities. In this study, CvFatB4, a novel cDNA clone encoding an acyl-ACP thioesterase (EC 3.1.2.14), was isolated from developing C. viscosissima seeds. Sequence alignment of the deduced amino acid sequence revealed that four catalytic residues for thioesterase activity are conserved and a putative N-terminal chloroplast transit peptide is present. Overexpression of CvFatB4 cDNA, which was under the control of the cauliflower mosaic virus 35S promoter, in Arabidopsis thaliana led to an increase in 16:0 fatty acid (palmitate) levels in the seed oil at the expense of 18:1 and other non-MCFAs.

Inhibitory Effects of Latilactobacillus curvatus BYB3 Cell-Free Extract on Human Melanoma B16F10 Cells and Tumorigenic Mice

  • Latilactobacillus curvatus BYB3 (BYB3) is a species of lactic acid bacteria, formerly named Lactobacillus curvatus, which is isolated from kimchi. In this study, the effect of BYB3, Lactobacillus rhamnosus GG, and Lactobacillus acidophilus GP1B strain extracts at various concentrations was examined on B16F10, a mouse melanoma cell line. Cell viability was examined via MTT assay, and the results indicated that compared to the other two probiotics, BYB3 significantly decreased the total percentages of viable cells. The effects of BYB3 on cell migration and proliferation in B16F10 cells were evaluated using wound healing mobility and proliferation assays, respectively; the results indicated that BYB3 inhibits cell migration and proliferation in a concentration-dependent manner. Using human dermal fibroblast cells to investigate BYB3 extract in vivo had no effect on skin-related cells. Nonetheless, the BYB3 extract inhibited tumor growth in a mouse model, as demonstrated by liver slices. Therefore, this suggests that using BYB3 extract to inhibit melanoma may be a novel approach.

Tracing of Some Root Colonizing Pseudomonas in the Rhizosphere Using lux Gene Introduced Bacteria (lux Gene을 도입한 생물적 방제 미생물의 근권 정착과 식물 생장 촉진 효과)

  • The use of bioluminescence as a sensitive marker for the detection of Pseudomnas sp. in the rhizosphere was investigated. Transposon Tn4431 which contains a promoterless luciferase operon and tetracycline resistant gene was used. This transposon, present on a suicide vector (pUCD623) in E. coli HB101, was mated with spontaneous rifampicin mutant of Pseudomonas fluorescens B16, a plant growth promoting rhizobacteria (PGPR), and then rifampicin and tetracycline resistant survivors were isolated. Twenty tow mutants wer isolated from the conjugants between E. coli HB101 and P. fluorescens B16. One of these, B16::Tn4431 (L22) recombinant which glowed brightly in the dark was selected for analysis. The cucumber seeds inoculated with L22 were grown in moisten two layers of filter paper and nonsterile soil contained in half cut PVC pipe. The roots were removed from the filter paper and PVC pipe, then placed on the 1/2 LB media plates. The plates were incubated at room temperature for 16 hr. L22 could successfully be detected in the rhizoplane by using the ordinary negative camera film (ASA100-400) with 30 minutes exposure under dark condition. The root colonizing ability and the plant growth promoting effect of L22 were not reduced compared to the untreated bacteria and wild type. L22 was superior to will type.

A Study on the Performance Improvement of Turbo Coded OFDM Systems Considering Frequency Offset (주파수 오프셋을 고려한 Tued OFDM 시스템의 성능 개선에 관한 연구)

  • In this paper, it is analyzed theoretically that the performance degradation, caused by carrier frequency offset, in an OFDM/M-ary PSK system. Then, when Turbo coding is adopted to an OFDM/M-ary PSK system, the degree of performance enhancement is evaluated. Finally, the maximum frequency offset is calculated to satisfy the BER performance required in a Turbo coded OFDM/M-ary PSK system. As results of analysis, it is shown that the more the number of M-ary is, the worse the BER performance is. Moreover, 7dB, 9dB, and 17dB of $E_b/N_o$ are required in QPSK, 8PSK and 16PSK systems, respectively in order to satisfy the error performance, $BER=10^{-3}$ for voice communication. If $E_b/N_o$ are 10㏈ and 15㏈, the frequency offset should be below 0.05 and 0.075, respectively, for voice communication. When Turbo coding is adopted to an OFDM/M-ary PSK system, the less the number of M-ary is, the greater the performance enhancement of Turbo coding is. If the number of a M-ary system of the system is below 16, it is found that required $E_b/N_o$ is about 8dB to satisfy $BER=10^{-5}$ Moreover, in the system the Turbo coding scheme, voice communication is available with greatly low$E_b/N_o$, and 8dB of $E_b/N_o$ is enough for data communication regardless of the permission range of frequency offset.
