(9) 본 세균의 배양 상등액은 초파리에 대하여 높은 살충률을 보여 열에 안정한 exotoxin의 존재를 확인하였다.
We report the case of a 46-year-old Chinese male patient who visited our clinic complaining of infertility. Semen analysis revealed azoospermia, and azoospermia factor c region partial deletion (b1/b3) was detected using Y chromosome microdeletion analysis. Testicular sperm extraction was performed after genetic counseling. The bilateral ductus deferens and a portion of the epididymis were absent, whereas the remaining epididymis was expanded. Motile intratesticular spermatozoa were successfully extracted from the seminiferous tubule. On histopathology, nearly complete spermatogenesis was confirmed in almost every seminiferous tubule. To our knowledge, this is the first case report of b1/b3 deletion with a congenital bilateral absence of the vas deferens and almost normal spermatogenesis.

To evolve silkworm hybrids with higher survival and productivity and easy and effective seed cocoon production, three new evolved bivoltine hybrids of silkworm (Bombyx mori L.), created with participation of sex-limited lines at eggs and larva stage were evaluated with control Super$_1$${\times}$Hessa$_2$ hybrid at Plovdiv Agrarian University during 2000 - 2002. ANOVA analysis was employed and the performance of hybrids was observed in respect of main quantitative traits. The results obtained show significant superiority of new hybrids T$_{15}$ 4/${\times}$TBV$_{2}$24/, XT$_{215}$38/${\times}$TV$_{3}$2/, XT$_{215}$38/${\times}$B$_{2}$6/ and their reciprocal crosses over the control. They were characterized with 99% hatchability of eggs, 93-95% silkworm survival, 2.32- 2.42 g cocoon weight, 53.5-55.7 cg shell weight, 22.9-23.1% shell ratio, 1,307-1,326 m filament length, 3.08-3.17 denier, 95-96% reelability, 44.4-45.1% silk ratio, 569-593 number of normal eggs per lying and 347-364 mg weight of normal eggs per lying. Newly evolved hybrids were manifested high productivity, 41.0-43.6 kg cocoon yield and 7.33-7.78 kg raw silk yield per one box (20,000$\pm$200 viable eggs), which significant surpass the control Super$_1$${\times}$Hessa$_2$ hybrid with 7.6-14.4% and 10.8-17.6%, respectively. T$_{15}$ 4/${\times}$TBV$_{2}$24/ and XT$_{215}$38/${\times}$TV$_{3}$2/ were considered as highly productive hybrids to local conditions and found suitable to rear in spring season.n.

We describe the generation of transgenic silkworm that carrying the chimeric fibroin light chain (L-chain) gene. Previously, we have cloned the complete fibroin L-chain gene from the silkworm Baekok-Jam, Bombyx mori, and the complete fibroin gene from the oak silkworm, Antheraea yamamai. The 444 bp repetitive sequence of A. yamamai fibroin gene was inserted into the exon 6 of B. mori fibroin L-chain gene to produce chimeric fibroin L-chain gene. The chimeric fibroin L-chain gene was cloned into the polyhedrin gene site of Autographa californica nuclear polyhedrosis virus (AcNPV) to yield a recombinant baculovirus as a fibroin gene targeting vector, One-day-old fifth instar female silkworm larvae were injected with the recombinant baculovirus and then mated with normal male moths. Genomic DNA from their progenies was extracted and screened for the desired targeting event by using PCR and Southern blot analysis. The analysis showed that the chimeric fibroin gene had intergrated into the L-chain gene on the genome by homologous recombination and was transmitted through generations. The transgenic silkworm carrying the chimeric fibroin gene were approximately 43.2% in $F_2$ generation, and the silkworms synthesized the fusion protein in cocoons layer.

The general and specific combining abilities and hybrid vigour of seven polyvoltine breeds [BL24, BL62, BL6S, BL67, BL68, BL69, and Pure Mysore (PM)] with five bivoltine breeds (NB4D2, CSR2, CSR5, CSR18 and CSR19) were evaluated in a line ${times}$ tester design by analysing six quantitative traits viz., fecundity, yield 110,000 larvae both by number and weight, cocoon weight, shell weight and cocoon shell ratio. ANOVA estimates showed significant effects for all the traits indicating the role of both additive and non- additive gene actions. Estimation of GCA revealed that the breed BL67 exhibited significant positive effects toy all the six traits followed by BL68 for four important economic traits viz., fecundity, cocoon weight, shell weight and cocoon shell ratio. Among testers, CSR5 showed significant GCA effects for five traits whereas CSR2 exhibited for shell weight and shell ratio. Three hybrids viz., BL67${times}$ NB4D2, BL67 ${times}$CSR5 and BL67${times}$ CSR19 excelled in four quantitative traits for SCA. Majority of the hybrids expressed positive hybrid vigour for cocoon yield/10,000 larvae by weight, cocoon weight and shell weight. Four hybrids namely PM ${times}$ NB4D2, BL67 ${times}$NB4D2, BL67${times}$ CSR5 and BL67${times}$ CSR19 manifesting positive hybrid vigour for five economic traits were adjudicated as best Heterotic hybrids and recommended for commercial exploitation to increase the silk yield.

Three newly authorized bivoltine silkworm hybrids namely, $CSR2{\times}CSR4$ (productive single hybrid), $(CSR6{\times}CSR26){\times}(CSR2{\times}CSR27)$ (productive double hybrid) and $CSR18{\times}CSR19$ (robust single hybrid) were chosen for the present study. These hybrids were subjected to different temperature and humidity treatments i.e., $25{\pm}$1^{\circ}C and RH $65{\pm}5%$ (control), $30{\pm}1^{\circ}C$, with combinations of low relative humidity (RH $65{\pm}5%$) and high RH ($85{\pm}5%$) at different stages during rearing and spinning of silkworm larvae. The larvae of after 3rd moult were subjected to different thermal and humidity stress till the assessment of cocoon traits. The comparative rearing and reeling performance clearly indicated that the deleterious effect of high temperature and high RH was more pronounced for the majority of traits such as cocoon uniformity, cocoon weight, shell weight, shell percentage, reelability, filament length, raw silk percentage raw silk recovery denier and waste percentage on silk weight than other temperature and RH treatments and this effect was almost similar for all three silkworm hybrids studied. The present investigation clearly indicate that the deleterious effect of high temperature and high RH was more pronounced on rearing and spinning of silkworm larvae than other temperature and RH treatments and similar effect was noticed for all the three silkworm hybrids studied. The cocoon characters can be improved by providing ideal environmental conditions even during spinning stage of larvae affected with high temperature and RH. The study also suggest that high temperature and low humidity has greater effect during rearing stage than spinning stage.

In the present study, hybrid vigour in terms of heterosis and over dominance of newly evolved bivoltine hybrids and their reciprocals raised from the promising newly evolved bivoltine breeds were calculated for the important parameters like pupation, cocoon yield, cocoon weight, shell weight, shell ratio, raw silk percentage, filament length and neatness. Through there was no significant difference among the hybrids for the characters pupation, cocoon yield and cocoon weight, the magnitude of heterosis and over dominance were varied significantly for these characters. However, significant differences were observed between new hybrids and control hybrid for the characters like cocoon shell weight, cocoon shell ratio, filament length and raw silk percentage. The results indicate that majority of new hybrids have shown higher heterosis and over dominance than control hybrid. The data also indicate that highest mean heterosis of all new hybrids was recorded for cocoon yield followed by filament length. Whereas highest mean over dominance was recorded for cocoon yield followed by cocoon weight. Negative mean heterosis and over dominance were recorded for the trait cocoon shell ratio. Based on the results, the importance of three hybrids viz., CSR2 ${\times}$ CSR4, CSR2 ${\times}$ CSR5 and CSR3 ${\times}$ CSR6 and their reciprocals (CSR4 ${\times}$ CSR2, CSR5 ${\times}$ CSR2 and CSR6 ${\times}$ CSR3) which exhibited significant heterosis and over dominance, for commercial exploitation was discussed.

The present investigation was carried out to study the possible cause for reciprocal difference in silkworm hybrids. By utilising the polyvoltine race Pure Mysore (PM) and newly evolved breeds (CSR2, CSR5, CSR16 and CSR17), the direct and reciprocal crosses of polyvoltine ${\times}$ bivoltine and also bivoltine hybrids were studied. The hybrids of polyvoltine ${\times}$ bivoltine (direct) are superior to their reciprocal crosses in respect of cocoon yield, cocoon weight and filament length. The reciprocal crosses of polyvoltine ${\times}$ bivoltine are superior to their direct crosses in respect of fecundity and short larval duration. No significant differences were observed in the characters like cocoon shell ratio, raw silk percentage, denier, reelability and neatness in both polyvoltine ${\times}$ bivoltine direct crosses and their reciprocals. The expression of cocoon characters as a function of sex revealed that direct crosses (polyvoltine ${\times}$ bivoltine) showed higher cocoon weight, pupal weight, shell weight and longer filament length in females than the reciprocal crosses (bivoltine ${\times}$ polyvoltine), where as these characters in males were almost the same in both direct and reciprocal crosses, indicating that the sex-linked genetic factor played a more important role. it was clear that difference in cocoon yield observed in reciprocal crosses of polyvoltine ${\times}$ bivoltine was due to the low cocoon and shell weight in females which was turn due to presence of early maturity genes (Lme) linked with sex-chromosome (X) which effect on larvae period of the silkworm. In bivoltine hybrids, i.e., both direct and their reciprocals crosses, all the characters viz., hatching percentage, larval duration, survival, cocoon weight, cocoon shell weight, cocoon shell ratio, raw silk percentage, filament length, denier, reelability and neatness did not show any significant difference (except number of eggs laid by moth) which could account for presence of same maturity genes (Lm) in both direct and reciprocal crosses. it was clear that reciprocal differences occur when the hybrids are prepared from the parental strains with different voltinism.

The reproductive performance of multivoltine breed 'Nistari' and bivoltine breeds as NB18, P5 and a bivoltine hybrid (NB18 ${\times}$ P5) of B. mori were studied in different seasons in terms of total number of eggs laid, unlaid and hatching efficiency of the laid eggs to critically assess the breeds / hybrid potentiality. The bivoltine hybrid showed better performance in all the seasons as compared to the breeds investigated, as expected due to possibility of hybrid vigour. Among the breeds, bivoltine P5 had a better egg laid performance in S1 (February - March), S2 (May - June) and S3 (September - October) seasons but with an increase in the number of unlaid eggs. The hatching percentage, although didn't show any definitive trend, still suggested a marginal better performance in the hybrid. Most importantly, egg laying rhythmicity was studied by mating the female moth at 6.00 a.m. for $3{\frac{1}{2}}$ hours, and subsequently allowing the laying of eggs both under BOD condition ($25{\pm}1^{\circ}C$ and $85{\pm}5%$ RH) and at ambient temperature ($23-28^{\circ}C$ and $74.7{\pm}5%$) separately. Maximum (90%) egg laid occurred from 3.30 to 7.30 pm (i.e. 4 - 6 h after decoupling). Similar laying patterns were observed when females where mated at 12 noon and decoupled at 3.30 pm, revealing that late photoperiod and early scotoperiod were favourable for egg laying in case of multivoltine breed and bivoltine prefers late photoperiod for egg laying.

Cells respond to an accumulation of unfolded proteins in the endoplasmic reticulum (ER) by increasing transcription of genes encoding molecular chaperones and folding enzymes. The information is transmitted from the ER lumen to the nucleus by intracellular signaling pathway, called the unfolded protein response (UPR). In Saccharomyces cerevisiae, such induction is mediated by the cis-acting unfolded response element (UPRE) which has been thought to be recognized by Hac1p transcription factor. We cloned the ATFC gene showing similarity with Hac1p, and then examined to determine whether ATFC gene product specifically binds to UPRE by electrophoretic mobility shift assays. ATFC gene product displayed appreciable binding ${to ^{32}}P-labelled$ UPRE. Therefore, we concluded that ATFC represents a major component of the putative transcription factor responsible for the UPR leading to the induction of ER-localized stress proteins.
