Isolation and Purification of Polyhydroxylated Alkaloids from Silkworm (Bombyx mori L.)

  • Several polyhydroxylated alkaloids were isolated from the extracts of freeze-dried silkworm powder, and purified by ion exchange chromatographic analysis. Through the HPLC analysis, we could identify 1-deoxynojirimycin (DNJ) and a kind of calystegin $B_2$ (HS-58) as well as a noble compound (HS-74) from the purified polyhydroxylated alkaloids. In order to know the characteristics of these isolated alkaloids as enzyme inhibitors, glycosidase inhibition activities of these identified alkaloids including other two non-purified alkaloids (SWP 3-1 and SWP 3-2) were investigated.

  • Borrelina virus was inoculated into Hyphantrea cunea DRURY in the labolatory and in the field. The pathogenecity of Borrelina virus upon Bompyx mori L. and Dendrolinus spectabilis BUTLER, too, was examined with following results. 1) $10^8$/ml, $10^7$/ ml, $10^6$/ml concentration of nuclear-polyhedrosis virus was inoculated into the larvae of H.cunea at various ages. The corrected mortality of the larvae were 97.4%, 95.2%, 94.7% in the 3rd instar, and 88.6%, 73.6%, 62.5% in the 6th instar, respectively, with three different concentration of NPV. 2) The symptom of disease of the larvae appeared on 4days after inoculation and most of the larvae were dead within 18 days. 3) The youngest larvae treated with the highest concentration of NPV showed the highest mortality. With older larvae and lower concentriton treated, it appeared that the time needed for death grew longer, marking slower death curve. 4) When we sprayed NPV of $10^6$/ml concentration to H. cunea in the field, the mortality was 94.8% in the first year, 84.6% in the second year and 78.3% in third year. By this, we could admit the continuous effects of the pathogens for several years. 5) About the larvae of B. mori of 3rd and 5th instar and D.spectabilis of 3rd instar inoculated with $10^8$/ml concentration of inoclum, we could not see any pathogenic effects.

Mathematical modelling of the stability of carbon nanotube-reinforced panels

  • The present paper studies the stability analysis of the continuously graded CNT-Reinforced Composite (CNTRC) panel stiffened by rings and stringers. The Stiffened Panel (SP) subjected to axial and lateral loads is reinforced by agglomerated CNTs smoothly graded through the thickness. A two-parameter Eshelby-Mori-Tanaka (EMT) model is adopted to derive the effective material moduli of the CNTRC. The stability equations of the CNRTC SP are obtained by means of the adjacent equilibrium criterion. Notwithstanding most available literature in which the stiffener effects were smeared out over the respective stiffener spacing, in the present work, the stiffeners are modeled as Euler-Bernoulli beams. The Generalized Differential Quadrature Method (GDQM) is employed to discretize the stability equations. A numerical study is performed to investigate the influences of different types of parameters involved on the critical buckling of the SP reinforced by agglomerated CNTs. The results achieved reveal that continuously distributing of CNTs adjacent to the inner and outer panel's surface results in improving the stiffness of the SP and, as a consequence, inclining the critical buckling load. Furthermore, it has been concluded that the decline rate of buckling load intensity factor owing to the increase of the panel angle is significantly more sensible for the smaller values of panel angle.

Effect of Pesticide Residue in Soil on Silkworm, Bombyx Mori L- Survey Analysis

  • Silkworm larval mortality specifically during spinning stage leading to non-spinning with specific morphological symptoms was frequently complained by sericulturists in Karnataka, India during 2015. Survey was conducted and information collected through questionnaire from the identified farmers who faced the problem of non-spinning in both traditional and non-traditional areas of Karnataka. Survey results indicate that the problem is specific to the silkworm crop of those farmers' who shifted from other crops of agriculture/ horticulture/olericulture/ floriculture to Sericulture. Silkworm rearing performance of the batches fed with these leaves confirmed that the pesticide sprayed to the crops previous to mulberry, remain in the soil and when mulberry plantation is taken up in these gardens, the pesticide is absorbed by the roots of mulberry plants and transported to the leaves. Silkworms that feed on these mulberry leaves, grow and ripen normally but during spinning stage, larvae die with external symptoms like regurgitation, body shrinkage, rectal protrusion later become hook shaped leading to non-spinning or partial metamorphosis into pupa and death or spin flimsy cocoons. Larval mortality ranged from a minimum of twenty five percent to a maximum of hundred percent. The problem was noticed from the first harvest of leaves and lasted for a maximum period of 36 months. Cocoon crop loss depends on the concentration, duration and type of pesticides used previously for other crops.

Exact third-order static and free vibration analyses of functionally graded porous curved beam

  • An exact solution based on refined third-order theory (TOT) has been presented for functionally graded porous curved beams having deep curvature. The displacement field of the refined TOT is derived by imposing the shear free conditions at the outer and inner surfaces of curved beams. The properties of the two phase composite are tailored according the power law rule and the effective properties are computed using Mori-Tanaka homogenization scheme. The equations of motion as well as consistent boundary conditions are derived using the Hamilton's principle. The curved beam stiffness coefficients (A, B, D) are obtained numerically using six-point Gauss integration scheme without compromising the accuracy due to deepness (1 + z/R) terms. The porosity has been modeled assuming symmetric (even) as well as asymmetric (uneven) distributions across the cross section of curved beam. The programming has been performed in MATLAB and is validated with the results available in the literature as well as 2D finite element model developed in ABAQUS. The effect of inclusion of 1 + z/R terms is studied for deflection, stresses and natural frequencies for FG curved beams of different radii of curvature. Results presented in this work will be useful for comparison of future studies.

  • 1. Ascorbic acid는 병B 배자까지는 점차적으로 증가하고 그 후에는 감소함을 알 수 있었다. 2. Triglyceride 및 total cholesterol은 병A 배자까지 증가하고 그 후는 감소함을 알 수 있었다. 3. Phospholipid는 병A 배자에서 감소하고 병B 배자에서 증가 다시 감소하였다가 점청기배자에서 부화직전까지 증가함을 알 수 있었다. 4. Free Fatty acid는 병A 배자에서 감소하고 병B 배자와 기A 배자에서 증가를 나타내고 다시 감소하였다가 부화직전에 증가하는 것을 알 수 있었다.

  • Mulberry, Mori folium leaves (ML) and fruits (MF), Lycii fructus (LF), and Panax ginseng have been known to maintain normal blood sugar levels (BSL) in folk medicine. Here we investigated the effects of coated rice with these herb extract to diabetic patients. Alloxan-induced diabetes mellitus (DB) mice (80 mg/kg BW., i.v.) separated into 3 groups. One group received the extract in high dose (6.5 mg/kg B.W.), and another one group received in low dose (1.3 mg/kg B.W.) perorally for 30 days. (+) control received no extracts. (-) control received neither of them. BSL, volume of water consumed (VWC), body weights (BW) of each group were measured and compared. BSL in both of the extracts-received groups got close to normal levels on 4th week after starting the experiments. VWC also showed the same pattern. BSL in (+) control remained high through the experiment periods. BW in (+) control remained high, but those in the extracts-received groups recovered to normal levels. The clinical trials for both of healthy volunteers and DB patients, conducted in a University Hospital, also showed that the rice coated with the extracts could control the BSL. From these results, we suggest that the functional rice with herb extracts could be used as a new alimentotherapy for DB patients.

Insect Ornithine Decarboxylase (ODC) Complements SPE1 Knock-Out of Yeast Saccharomyces cerevisiae

  • Ornithine decarboxylase (ODC) is a rate-limiting enzyme in the biosynthesis of polyamines, which are essential for cell growth, differentiation, and proliferation. This report presents the characterization of an ODC-encoding cDNA (SlitODC) isolated from a moth species, the tobacco cutworm, Spodoptera litura (Lepidoptera); its expression in a polyamine-deficient strain of yeast, S. cerevisiae; and the recovery in polyamine levels and proliferation rate with the introduction of the insect enzyme. SlitODC encodes 448 amino acid residues, 4 amino acids longer than B. mori ODC that has 71% identity, and has a longer C-terminus, consistent with B. mori ODC, than the reported dipteran enzymes. The null mutant yeast strain in the ODC gene, SPE1, showed remarkably depleted polyamine levels; in putrescine, spermidine, and spermine, the levels were > 7, > 1, and > 4%, respectively, of the levels in the wild-type strain. This consequently caused a significant arrest in cell proliferation of > 4% of the wild-type strain in polyamine-free media. The transformed strain, with the substituted SlitODC for the deleted endogenous ODC, grew and proliferated rapidly at even a higher rate than the wild-type strain. Furthermore, its polyamine content was significantly higher than even that in the wild-type strain as well as the spe1-null mutant, particularly with a very continuously enhanced putrescine level, reflecting no inhibition mechanism operating in the putrescine synthesis step by any corresponding insect ODC antizymes to SlitODC in this yeast system.

Evaluation of F$_1$ Hybrids Between RD$_1$ and Bivoltine Breeds of the Silkworm (Bombyx mori L.) for Exploitation in Dry Zones

  • With the main objective to know the feasibility of exploitation of F$_1$ hybrids in semi arid and arid climatic conditions of India, the present study was carried out by utilising RD$_1$ as female parent and six bivoltine silkworm breeds viz., CSR$_2$, CSR$_4$, CSR$_{5}$, CSR$_{17}$, CSR$_{18}$ and CSR$_{19}$ along with NB$_4$D$_2$ as male parents. Different hybrids exhibited their superiority for various economic characters during different seasons. Among F$_1$ hybrids, RD$_1$${\times}$CSR$_{5}$ was adjudicated as the best hybrid in terms of expression of significant positive hybrid vigour over mid parental value for five economic characters namely hatching %, cocoon yield, cocoon weight, cocoon shell weight and filament size, highest multiple trait average evaluation index value of 56.77 and comparatively uniform cocoon size with coefficient of variation (CV%) of 3.80 and Standard Deviation (SD) of 7.99 during September - October 2003. Results of the present study revealed that the F$_1$ hybrid RD$_1$${\times}$CSR$_{5}$ can be successfully exploited on commercial in semi arid and arid climatic conditions in India. India.dia.

Vibration analysis of damaged core laminated curved panels with functionally graded sheets and finite length

  • The main objective of this paper is to study vibration of sandwich open cylindrical panel with damaged core and FG face sheets based on three-dimensional theory of elasticity. The structures are made of a damaged isotropic core and two external face sheets. These skins are strengthened at the nanoscale level by randomly oriented Carbon nanotubes (CNTs) and are reinforced at the microscale stage by oriented straight fibers. These reinforcing phases are included in a polymer matrix and a three-phase approach based on the Eshelby-Mori-Tanaka scheme and on the Halpin-Tsai approach, which is developed to compute the overall mechanical properties of the composite material. Three complicated equations of motion for the panel under consideration are semi-analytically solved by using 2-D differential quadrature method. Several parametric analyses are carried out to investigate the mechanical behavior of these multi-layered structures depending on the damage features, through-the-thickness distribution and boundary conditions. It is seen that for the large amount of power-law index "P", increasing this parameter does not have significant effect on the non-dimensional natural frequency parameters of the FG sandwich curved panel. Results indicate that by increasing the value of isotropic damage parameter "D" up to the unity (fully damaged core) the frequency would tend to become zero. One can dictate the fiber variation profile through the radial direction of the sandwich panel via the amount of "P", "b" and "c" parameters. It should be noticed that with increase of volume fraction of fibers, the frequency parameter of the panels does not increase necessarily, so by considering suitable amounts of power-law index "P" and the parameters "b" and "c", one can get dynamic characteristics similar or better than the isotropic limit case for laminated FG curved panels.
