Polyclonal antiserum against Manduca sexta allatotropin has been utilized to investigate the localization of allatotropin-immunoreactivity in the brain of the si1k moth Bombyx mori. Manduca sexta allatotropin-immunoreactive (Mas-AT-IR) neurons were found in all larval brains investigated, but not in prepupal, pupal and adult brains. In the larval stages, first appearance of Mas-AT-immunoreactivity w8s shown in the brain of first instar larvae, which contains four pairs of bilateral Mas-AT-IR cell bodies. Labeled neurons increased to six pairs in the second instar larval brain, including two pairs of median neurosecretory cells in the pars intercerebralis. In the third and fourth instar larvae, five pairs of labeled cell bodies were distributed throughout each brain. In the fifth instar, there were about ten pairs of bilateral cell bodies in the day-1 brain, about seven pairs in the day-3 brains, and five pairs in the day-5 brains, respectively. Mas-AT-labeling was observed in both axons within nervi corpora cavdiaci (NCC) 1+11 and corpora allata. This suggests that the Mas-AT produced from the brain neurons is transported via some axons of the NCC 1+11 and nervi corpora allati I to the corpora allata, which appears to be a main accumulation site for the Mas-AT neuropeptide in some brain neurons produced in B. mori.

DNJ concentration in silkworm, Bombyx mori was traced through the life. DNJ absent in egg existed over the concentration of 5 mg gdw-1 from 1st instar to 4th day of 5th instar during larva period. In previous studies we have shown that a sw17255 gene was expressed in hemocyte-specific tissues of the silkworm, Bombyx mori L. (Lepidoptera: Bombycidae). It was verified that the sw17255 core promoter region contains elements that regulate the expression of this gene in hemocyte tissue; the selected promoter region spans nucleotides -1 to -2,112 upstream of the start codon. Each of the luciferase reporter gene expression vectors under the control of 4 different kinds of promoter candidates, (-2,112/-1), (-1,640/-1), (-1,169/-1) and (-579/-1), and the control reporter plasmid DNA, were introduced into B. mori larval coelom by direct injection using a syringe. The promoter candidate [E] (-579/-1) showed more than 1.67 fold transcriptional activity compared to control promoter activity. Higher productivity of an expressed gene in the transgenic silkworm by this promoter combination could be achieved in the near future. The foreign recombinant protein could be easily harvested from the blood of the transgenic silkworm.

We investigated some biochemical properties of acetylcholinesterase (AChE) in the Bombyx mori larval head. 1% Triton X-100 (v/v) was suitable for extracting AChE from the silkworm larval head but 1 M NaCl was not suitable. PAGE analysis showed a single band of AChE that was detected by histochemical staining using acetylthiocholine as a substrate. AChE was also partially purified with Sepharose 6B and DEAE-cellulose column. Finally, the specific activity of partially purified enzyme solution was 7.6. The study on inhibitor specificity indicated that the enzyme under study was a true cholinesterase (ChE) or AChE. AChE activity was maximum at the substrate concentration of $5{\times}10^{-4}$ M and the excess substrate inhibited the AChE activity. The optimal pH and temperature were pH 7.0-9.0 and 30-35$^{\circ}C$.
In this study, various antioxidant propertieswere evaluated by analyzing the proximate composition and free-sugar and DNJ contents of the Bombyx mori L., Mori fructus, Morus ramulus, Mori folium, Dioscoreae rhizome and Inonotus obliquus extracts, and the antidiabetic effect was evaluated through an in-vivo experiment. Product evaluation was conducted after preparing a beverage for the easy use of the mixed extract for biological activity, as a functional resource. The biochemical composition of the extracts was 0.31% crude protein, 0.114 g/100 mL free sugar and 161.02 mg/gdw DNJ, all of which showed excellent results in all the antioxidant ability and ${\alpha}$-glucosidase inhibition ability experiments. The beverage showed the following functionalities. The total-polyphenol content was 71.93%, but the electron-donating ability was highest in the 5% extract concentration. Moreover, when the TBARS values were experimented on, KO2 showed an especially high scavenging ability. During the five-week beverage supply after inducing diabetes with streptozotocin (STZ), the change in the blood sugar was measured, and the STZ-induced diabetes+oral-beverage group (C) showed a lower blood sugar level than the diabetes comparative group (B) in the second week. In the STZ-induced diabetes+free-diet/beverage group (D), the blood glucose level also slowly decreased in the second week. The lowest blood glucose level among the STZ-induced diabetes groups was shown in the fifth week.

The aim of this study was to investigate biological activity and biochemical properties of extracts from Bacillus subtilis-fermented silkworm (Bombyx mori L., SP) powder of different origin (Buan, Namwon, and Boeun). An additional aim was to determine the inhibition of cancer cell (B16-F10, HT-29, LNcaP, and MCF-7) proliferation and nitric oxide (NO) production from lipopolysaccharide (LPS)-induced RAW264.7 cells. Biological activities (${\alpha},{\alpha}^{\prime}$-diphenyl-${\beta}$-picrylhydrazyl [DPPH], free radical scavenging activity, fibrinolytic activity, antiproliferation activity, and anti-inflammatory activity) and biochemical properties (compositional amino acid contents, and mineral contents) were examined in water extracts from silkworm powder and B. subtilis-fermented silkworm powder. The highest amino acid contents were detected in Buan silkworm powder (BU). After fermented, the highest contents were found in B. subtilis-fermented Buan silkworm powder (BBO). The major minerals detected were K, Ca, and Mg. Rates of these minerals, especially those of Na increased after fermented. DPPH radical scavenging activity and fibrinolytic activity were stronger in the fermented group than non-fermented group. DPPH radical scavenging activity and fibrinolytic activity were highest in the extract from BBO. The inhibition activities of LNcaP and MCF-7 cells viability were significantly decreased in the BBO, and there was no inhibition activity in other cancer cells (B16-F10 and HT-29). An SRB assay of the cell viability of RAW 264.7 cells exposed to extracts of silkworm powder and B. subtilis-fermented silkworm powder revealed no toxicity in any of the groups. Compared with the LPS-treated group, the biggest reduction in NO production was detected in the BBO group. Based on these results, extracts from Boeun silkworm powder fermented with B. subtilis could be a candidate material as a dietary supplement for use in healthy functional foods.
Among many microbial pesticides, Bacillus thuringiensis is one of the most hopeful pesticide and some commercial products have been appearing on the market. Because these commercial products contain living spores and toxins of the organism, there is a danger that living spores of B. thuringiensis may be scattered by wind and cause a great damage in the sericulture areas. In order to avoide these risks it is desirable to select the strain which has low pathogenicity to the silkworm, and at the sometime being highly pathogenic to the pest insects. Thus this study has been carried out to acquire some basic informations about the procedure of desicable strain selection. Three strains of B. thuringiensis var. kurstaki, var. dendrolimus and var. aizawai were used for the pathogenicity test on the silkworm, Bombux mori and the fall webwarm, Hyphantria cunea. Those strains were investigated by the agarose gel electrophoresis patterns of plasmid DNA determine whether mutation had occured. Pathogenicity tests were carried out of using isolated crystal proteins and spore-crystal protein to mixtures of each strain, seperatively. In case of using spore-crystal protein mixture, the order of pathogenicity in varities of B. thuringiensis against B.mopri and H.cunea were kurstaki, aizawai, dendrolimus and kurstaki, dendrolimus, aizawai, respectively. But using isolated crystal proteins, dendrolimus had the highest toxicity to H. cunea and the lowest toxicity to B. mori among tested three strains. From the above results, dendrolimus was presumed the most desirable straing for using microbial pesticide.
In our country there are five species of Morus genus including Morus alba L.. Also their varieties and hybrids are distributed so much. In sucession of previous report we collected control and marketing specimens of Mori Cortex Radicis, comparative experiments were pharmacognostically carried out to identify the control specimen by the differences of external and internal morphology. It was difficult to identify marketing specimens by external morphology, because they are similar in spite of conparating with control specimen which the origin is definite. In internal morphology, medullary ray is developed near the cambium to primary bark in control specimen A(Morus alba series) and C(M. Lhou series), but less developed in B(M. bombycis series). The difference of these three series was observed. The thickness of cork layer is almost the same($7{\sim}12$ layers) in A and C series, but B is thin layer and sample E(that on the market) is generally more thick and has a stick cork cell. The kinds of starch, Ca-oxalate and latex, cell centents were same, but it was easy to identify them by the differences of their distribution. The bast fibre of D(wild specimen) and E were light lignified, latex tube of A and C series was richer distributed than others. These results show that the origin of Mori Cortex Radicis on the market can be appreciated in four groups of Korean Morus genus which are M. alba, M. bambycis, M. Lhou series and the others.
The water extract from the root bark of Cortex Mori (CM, Morus alba L.: Sangbaikpi), a mulberry tree, has been known in Chinese traditional medicine to have antiphlogistic, diuretic, and expectorant properties. In this study, the cytotoxicity of CM against tumor cells and its mechanism was examined . CM exhibited cytotoxic activity on K-562, B38O human leukemia cells and B16 mouse melanoma cells at concentrations of > 1 mg/ml. A DNA fragmentation, PARP cleavage, and nuclear condensation assay showed that those cells exposed to CM underwent apoptosis. The water extract of Scutellarie Radix (SR) was used as a negative control and showed no cytotoxicity in those cells. The flow cytometric profiles of the CM-treated cells were also indicative of apoptosis. However, they did not appear to exert the G1 arrest, which is observed in other tubulin inhibitor agents such as vincristine, taxol. The protein-binding test using Biacore and a microtubule assembly-disassembly assay provided evidence showing that CM bound to the tubulins resulting in 3 markets inhibition of the assembly, but not the disassembly of microtubules. The possible nonspecific effect of the CM extract could be excluded due to the results using SR, which did not affect the assembly process. Overall, the water extract of CM induces apoptosis of tumor cells by inhibiting microtubule assembly.

A breeding programme was initiated during 2001 utilizing two polyvoltine silkworm breeds viz. $BL_{69}$, an evolved breed tolerant to high temperature and MAR, comparatively resistant to Bombyx mori nuclear polyhedrosis virus (BmNPV) with the objective to develop robust polyvoltine breeds and hybrids. The breed $NP_1$ was developed by exposing the fifth instar larvae to high temperature $(36{\pm}1^{\circ}C)$, high Relative Humidity ($85{\pm}5%$ R.H.) and inoculating third instar larvae with BmNPV inoculum. At $F_{12}$, the breed was tested for hybrid forming ability utilizing six bivoltine silkworm breeds viz. $CSR_2,\;CSR_4,\;CSR_{17},\;CSR_{18},\;CSR_{19}\;and\;NB_4D_2$. The hybrid $'NP_1{\times}CSR_{17}'$ exhibited its superiority by recording 97.2% survival, 1.892 g cocoon weight, 0.406 g cocoon shell weight, 21.5% cocoon shell ratio, 16.6% raw silk percentage and 890 m filament length whereas the control $(PM{\times}CSR_2)$ has recorded 90.2% survival, 1.599 g cocoon weight, 0.304 g cocoon shell weight, 18.9% cocoon shell ratio, 13.1 % raw silk percentage and 768 m filament length. Commercial exploitation of the new $polyvoltine{\times}bivoltine$ hybrid in sericulture industry has been discussed.
