In vertebrates, the vitamin D receptor (VDR), a member of the nuclear receptor superfamily, binds the biologically active ligand $1{\alpha},25-(OH)_2$-vitamin $D_3$ (1,25 $D_3$). Nearly all vertebrates, including Agnatha, possess a VDR with high ligand selectivity for 1,25 $D_3$ and related metabolites. Although a putative ancestral VDR gene is present in the genome of the chordate invertebrate Ciona intestinalis, the functional characteristics of marine invertebrate VDR are still obscure. To elucidate the ascidian Halocynthia roretzi VDR (HrVDR), we cloned full-length HrVDR cDNA and investigated the transcriptional activity of HrVDR in HEK293 cells. HrVDR consists of 1,680 nucleotides (559 amino acids [aa]), including a short N-terminal region (A/B domain; 26 aa), DNA-binding domain (C domain; 72 aa), hinge region (D domain; 272 aa), and C-terminal ligand-binding domain (E domain; 161 aa). The amino acid sequence identity of HrVDR was greatest to that of C. intestinalis VDR (56%). In the luciferase reporter assays, the transcriptional activity of HrVDR was not significantly increased by 1,25 $D_3$, whereas the farnesoid X receptor agonist GW4064 increased the transactivation of HrVDR. These results suggest the presence of a novel ligand for and a distinct ligand-binding domain in ascidian VDR.
Type strain, Methylosinus trichosporium OB3b, was used to convert methane to methanol. To prevent further oxidation of methanol, NaCl and EDTA were used as inhibitors of methanol dehydrogenase. The reaction temperature was $25^{\circ}C$, and the concentrations of cell and sodium formate added to the reaction mixture were 0.6 mg dry cell wt/ml and 20 mM, respectively. During 12hr reaction, 8 mM methanol was accumulated in the reaction mixture. In this reaction $K_m$ and $V_{max}$ values were found to be 532.6 mM and 1.749 mmol/hr, respectively, and the conversion rate was approximately 37%. To increase the concentration of methanol in the medium, a repeated batch reaction was carried out. In this process, methane was injected every eight hours, and the produced methanol concentration was 18 mM.

Two high resolution compressive schemes, CICSAM(Ubbink, 1997) and HRIC(Muzaferija & Peric, 1999), in interface capturing method are reviewed briefly with respect to the extended forms suitable for unstructured meshes. And then those are applied to three typical test cases of translation test, shearing flow test and collapsing water problem with an obstacle. It is accomplished by implementing the high resolution schemes in the in-house CFD code(PowerCFD) for computing 3-D flow with an unstructured cell-centered method, which is based on the finite-volume technique and fully conservative. The calculated results show that CICSAM is better than HRIC with respect to accuracy and robustness, although either scheme can be used as a good choice for free surface or two-phase flow simulation.
In this study, we show a convenient MTT assay for detect the susceptibility of yeast-like form of Trichosporon beigelii against antifungal agents. This assay was developed based on mitocondrial respiration by determining reduction of 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) to formazan. Cells of T beigelii are seeded into 96-well microtiter plates, and antifungal agents, amphotericin B, magainin and CA-ME hybrid peptide were added with various concentrations. After 24 hr incubation, MTT was added, then incubations were continued for 4 hr. Formazan formation was quantified photometrically after extraction of the formazan with acid sodium dodesyl sulfate (SDS). From this assay, we could obtained MICs of antifungal agents against T. beigelii. The presented method can easily be used as an effective methods to assess the antiftingal action of various agents on yeasts with minimal amounts of antifungal agents.
DNA damage by UV and environmental agents are the major cause of genomic instability that needs to be repaired, otherwise it give rise to cancer. Accordingly, mammalian cells operate several DNA repair pathways that are not only responsible for identifying various types of DNA damage but also involved in removing DNA damage. In mammals, nucleotide excision repair (NER) machinery is responsible for most, if not all, of the bulky adducts caused by UV and chemical agents. Although most of the proteins involved in NER pathway have been identified, only recently have we begun to gain some insight into the mechanism by which proteins recognize damaged DNA. Binding of Xeroderma pigmentosum group C protein (XPC)-hHR23B complex to damaged DNA is the initial damage recognition step in NER, which leads to the recruitment of XPA and RPA to form a damage recognition complex. Formation of damage recognition complex not only stabilizes low affinity binding of XPA to the damaged DNA, but also induces structural distortion, both of which are likely necessary for the recruitment of TFIIH and two structure-specific endonucleases for dual incision.
The water extract of Seongpungtang(SPT) has commonly been used for treatment of ischemic brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extract of SPT rescues brain cells from ischemic damage. To elucidate the protective mechanism of ischemic induced cytotoxicity, the regulation of Lipopolysaccharide (LPS) and PMA (phobol-12-myristate-13-acetate) induced iNOS expression in microglial cells was investigated. LPS and PMA treatment for 48 hr in microglial cells markedly induced nitric oxide (NO), but treatment of the cells with the water extract of SPT decreased nitrite formation. In addition, LPS and PMA treatment for 48 hr induced severe cell death in microglial cells. However treatment of the cells with the water extract of SPT did not induce significant changes compared to the control cells. Furthermore, NO production was markedly decreased by treatment of nuclear factor kappa B(NF-kB) inhibitor, pyrrolidine dithiocarbamate(PDTC). According to the above results, it is suggested that the protective effects of the water extract of SPT against ischemic brain damage may be mediated by regulation of iNOS during ischemic condition.
Background : An acceleration or deceleration of the heart rate (HR), which reflects autonomic effects, is observed before the onset of paroxysmal atrial fibrillation (PAF). The purpose of this study was to assess the discrepancy in the autonomic interactions before the onset of PAF for different patterns of change in the HR. Materials and Methods : From 105 Holter tapes with the PAF recorded, 55 episodes (42 patients, 34 men, $58{\pm}12$ years) of PAF (>5 min), preceded by a sinus rhythm for more than 1 hour, were selected and submitted to time-domain and frequency-domain HR variability analyses. Fifty-five episodes were divided into 2 groups: group A PAF (n=30) with acceleration of the HR during the last 2 minutes before the PAF and group B (n=25) with deceleration of the HR. Results : A significant linear decrease in the mean R-R interval was observed in group A ($924{\pm}30$ to $835{\pm}28ms$, $P$=0.001) and an increase from $831{\pm}32$ to $866{\pm}31ms$ in group B PAF episodes ($P$=0.046). In the frequency-domain analyses, the LF/HF ratio exhibited a progressive linear increase before the PAF in group A ($P$=0.005). The HF normalized units (HFnu) and natural logarithm-transformed HF ($In$HF) values decreased from $30.8{\pm}4.0$ to $16.1{\pm}1.8$ ($P$=0.003) and $4.49{\pm}0.25$ to $4.07{\pm}0.22$ ($P$=0.001), respectively. Contrary to the results in group A, a significant increase in the HF components (HFnu and $In$HF) (from $22.6{\pm}3.2$ to $30.2{\pm}4.0$, $P$=0.005, and $4.27{\pm}0.27$ to 4.75 0.33, $P$=0.001, respectively) and a resultant decrease in the LF/HF ratio were observed in group B PAF episodes. No significant changes were observed in the LF components in either PAF group. Conclusion : Autonomic stimuli leading to an acceleration or deceleration of the HR before the onset of AF are due to parasympathetic modulation. Parasympathetic modulation plays a key role in the initiation of PAF.

Boron Phosphide films were deposited on the glass substrate at the low temperature, 55$0^{\circ}C$, by the reaction of B$_2$H$_{6}$ with PH$_3$ using CVD. $N_2$ was employed as carrier gas. The optimal gas rates were 50 $m\ell$/min for B$_2$H$_{6}$, 50 $m\ell$/min for PH$_3$$m\ell$/min and 1.5 $\ell$/min for $N_2$. To investigate the annealing effect, the films were annealed for 1hour, 3hours in $N_2$ambient at 55$0^{\circ}C$ and tested. The deposition rate was 1000$\AA$/min and the refractive index of film was 2.6. The measurement of X-RD shows that the films have the preferred orientation of (1 0 1) and the intensity of the peak for (1 0 1) orientation decreases according to the annealing time. The data of VIS spectrophotometer proved that the films are transparent in the visible range and the maximal transmittance increases according to the annealing time; 75.49% for as-deposited, 76.71% for 1hr-annealed and 86.4 % for 3hrs-annealed. The measurement of AFM shows that the average surface roughness increases according to the annealing time; 73$\AA$ for as-deposited, 88.9$\AA$ for 1hr-annealed and 220$\AA$ for 3hrs-annealed. Also, The data of the secondary electron emission rate(Υ) shows that the secondary electron emission rate increases according to the annealing time; 0.317 for 1hr-deposited, 0.357 for 1hr-annealed and 0.537 for 3hrs-annealed. And, The measurement of FT-IR that the characteristic of transmittance in the infrared range was stabilized through annealing.ing.
To investigate the re-treatment time of Microcotyle sebastis by oral administration of praziquantel, the residue levels of praziquantel in plasma of rockfish Sebastes schlegeli administered orally at a dose of 200 mg/kg B.W. were analyzed by reversed-phase HPLC, and the concentrations of praziquantel in the plasma were correlated with the extermination of M. sebastis. The absorption and depletion of praziquantel in the blood of rockfish were fast and the residual concentrations of praziquantel declined below $4\mu g/mL$ within 24 hr post treatment. Most of worms were exterminated within 3 hr after oral administration of praziquantel, however, a small number of M. sebastis were not killed by the treatment until end of the experiment. Considering fast drop of praziquantel in blood and extermination pattern of M. sebastis in the present results, retreatment at an interval of 9-12 hr would be effective for eradication of M. sebastis.

Merging of two supermassive black holes would generate gravitational waves that can be detected by the Pulsar Timing Array (PTA) in the nHz band. In order to assess the plausibility of GW detection with PTA and to develop the data analysis scheme, it is important to understand the underlying properties of black holes and black hole binaries. In this work, we present mass and redshift distributions of black hole mergers using the Horizon Run 5 (HR5) data and discuss their implications for GW detection. We find a general conjecture about the black hole merger tree is true with the Horizon Run 5. For example, a) relatively lighter black holes merge at higher redshifts and b) binary mergers do contribute to the formation of more massive black holes toward low redshifts. We also present our plan to use the black hole properties extracted from the HR5 data in order to generate simulated GW signals to be injected into actual PTA data analysis pipelines. Mass and distance obtained from the HR5 would be key ingredients to generate a more realistic PTA source data set.
