Population Genetic Analyses of Gibberella fujikuroi Isolated from maize in Korea

  • We analyzed 88 strains of Gibberella fujikuroi (Analmorph: Fusarium section Liseola) from maize in Korea for mating population, mating type, fumonisin production vegetative compatibility, and random amplified polymorphic DNA (RAPD) patterns. We found 50 strains that were MATA-2, 22 that were MATA-1, 1 that was MATD-1, and 15 that were not reproducibly fertile with any of the mating type testers. Of the 50 MATA-2, 15 were female fertile, while 10 of the 22 MATA-1 strains were female fertile. A total of 1,138 nitrate non-utilizing (nit) mutants were recovered from a total of 88 strains. These strains were grouped into 39 vegetative compatability groups (VCGs) by demonstrating heterokaryosis between nit mutants. A single maize ear could be infected by more than one VCG of F. moniliforme. RAPD analysis measured genetic diversity among 63 strains of F. moniliforme. Several VCGs were distinguished by RAPD fingerprinting patterns. Most strains produced significant levels of fumonisins. However, 6 MATA-2 strains from a single VCG produced higher levels of fumonisin $\textrm{B}_3$ than that of fumonisin $\textrm{B}_1$ or $\textrm{B}_2$. From these data, we concluded that most Korean strains of F. moniliforme associated with maize belonged to mating population A and produced significant levels of fumonisins. Futhermore, RAPD analysis could differentiate strains associated with different VCGs.

Mating Behavior, Mycotoxin Production, and Vegetative Compatibility of Gibberella fujikuroi Species Complex from Sorghum in Korea

  • Fusarium isolates of Gibberella fujikuroi species complex were obtained from sorghum grown in five provinces of Korea in 1996 and 1997. These isolates were characterized based on their mating behavior, mycotoxin production, and vegetative compatibility. Only three mating populations (A, D, and F) were recovered from a total of 155 isolates examined. The relative frequency of the mating populations was significantly different: F was predominant (80%), while D and A were observed at low frequencies of 9% and 3%, respectively. Female fertile isolates were more common within F (44 our of 124) than D (2 out of 14), while none of the five A isolates were female fertile. The inbreeding effective population sizes ($\textrm{N}_e$)for mating type and male/hermaphrodite ratios in mating populations A and D produced significant amounts of fumonisins, while F isolates produced none or only traces of fumonisin B$_1$. In contrast. F isolates produced higher amounts of moniliformin (average of 3,820 ppm) than A and D isolates (averages of 77 and 1,819 ppm, respectively). Fifty-one isolates were tested for vegetative compatibility using nitrogen non-utilization mutants of each isolate, and 44 vegetative compatibility groups (VCGs) were identified. A single VC type (VC1) was found in all of the five A isolates examined. Six of the D isolates examined consisted of three VC types: two for VC2, two for VC3, and the rest for VC4. All of the F isolates tested were incompatible in every combination and , thus, each constituted a unique VCG.

Mitochondrial Effects on the Physiological Characteristics of Lentinula edodes

  • In the mating of filamentous basidiomycetes, dikaryotic mycelia are generated through the reciprocal movement of nuclei to a monokaryotic cytoplasm where a nucleus of compatible mating type resides, resulting in the establishment of two different dikaryotic strains having the same nuclei but different mitochondria. To better understand the role of mitochondria in mushrooms, we created four sets of dikaryotic strains of Lentinula edodes, including B2×E13 (B2 side) and B2×E13 (E13 side), B5×E13 (B5 side) and B5×E13 (E13 side), E8×H3 (E8 side) and E8×H3 (H3 side), and K3×H3 (K3 side) and K3×H3 (H3 side). The karyotypes and mitochondrial types of the dikaryotic strains were successfully identified by the A mating type markers and the mitochondrial variable length tandem repeat markers, respectively. Comparative analyses of the dikaryotic strains on the mycelial growth, substrate browning, fruiting characteristics, and mitochondrial gene expression revealed that certain mitochondria are more effective in the mycelial growth and the production of fruiting body, possibly through the activated energy metabolism. Our findings indicate that mitochondria affect the physiology of dikaryotic strains having the same nuclear information and therefore a selection strategy aimed at mitochondrial function is needed in the development of new mushroom strain.

근연간 교배방법에 의한 팽이 탄핵균주의 교배형분석 및 균주 육성 (Development of a New Strain and Mating Type Analysis by Inbreeding Method in Flammulina velutipes)

  • 새로운 백색 팽이 품종을 육성하고자 수집균주 중 백색자실체를 생성하는 5개의 이핵계통을 선발하고 이들로부터 단핵균주를 분리하였다. 분리된 단핵균주 계통의 교배형을 검정한 결과 A1B1A2B2 이었으며, 이핵균주들에 비하여 균사생장 속도가 느리고 변이폭도 좁다는 것을 알 수 있었다. 분리된 단핵계통의 근연간 교배를 수행한 결과 육성된 모든 이핵균주는 백색이었으며 모본보다 우수한 균주들을 선발할 수 있었다. 특히 Fv 4-1균주는 가장 높은 수량성과 안정성을 나타내었다. 또한 상호교배에 의해 일부교배조합에서 세포질효과를 확인할 수 있었다. 따라서 근연간 교배법은 기존 재배품종들이 가진 백색과 같은 특성을 유지하면서 새로운 품종을 육성하기 위한 방법으로 사용할 수 있을 것으로 생각된다.

  • Twenty-five isolates of Fusarium fujikuroi acquired from rice seeds and rice plants evidencing symptoms of Bakanae disease were evaluated to determine their mating types and characterize the formation of their sexual state. The mating types of the isolates were evaluated via multiplex PCR with the diagnostic primers of the mating-type (MAT) region: GFmat1a, GFmat1b, GFmat2c, and GFmat2d. Among the 25 isolates, 11 were identified as MAT-1 (male), and 14 as MAT-2 (female). Four MAT-1 isolates and three MAT-2 isolates were mated and cultured to evaluate the optimal culture conditions for the production of their sexual states. Among four tested media, 10% V8 juice agar proved optimal for the perithecial production of the isolates. The isolates also generated the largest numbers of perithecia when incubated at 23oC in alternating cycles of 12 hr fluorescent light and NUV fluorescent light and 12 hr darkness.

사철느타리버섯 형질전환주(形質轉換珠)의 특성(特性) (Characteristics of Transformants in Pleurotus florida)

  • 사철 느타리버섯 영양요구성(營養要求性) 균주(菌株)를 팽이버섯 leu 2 유전자를 지닌 pM 301 백터를 이용(利用)하여 영양요구성(營養要求性)을 보완(補完) 시킴으로써 형질전환(形質轉換) 하였다. 형질전환균주(形質轉換菌株)의 균사생장(菌絲生長)을 버섯 완전배지(完全培地)나 버섯 최소(最小) 배지(培地)에서 비교(比較)하였다. 형질전환주(形質轉換株)는 1 핵(核) 균사(菌絲)와 교배(交配)후 자실체(子實體) 형태(形態)를 비교(比較)하고 포자 분석(分析)에 의하여 유전분석(分析)을 하였다. 자실체(子實體) 발생(發生)과 자실체(子實體) 모양이 형질전환주(形質轉換株)는 모균주와 다른 특성(特性)이 나타났다. 모균주(母菌株)는 교배형(交配型)이 $A_1B_1$으로 $A_2B_1$ 교배형(交配型)인 1핵(核) 균주(菌株)와 교배(交配)에 의하여 자실체(子實體)를 형성(形成)하지 못하나 형질전환주(形質轉換株)는 교배형(交配型)이 $A_1B_1$이고 $A_2B_1$ 1핵(核) 균주(菌株)와 교배(交配) 하였을때 자실체(子實體)를 형성(形成)하였다.

금개구리(Rana plancyi)의 소리 유형과 수온에 따른 소리변화 (The Call Patterns and the Change of Calls by Water Temperature in Rana plancyi (Amphibia, Anura))

  • The study of the Rana plancyi, was done during the calling period at a pond in Osong, Chungbuk, Korea from May to the middle of August of 1996 and 1997. Five basic types of call-A, B, C, D, E, - were identified in Rana plancyi according to the structure of call types. The structure of A type call has only one pulse, B type call has two pulses and C, D type call has one separated pulse which is called introductory call and pulse group as follow it. On the other hand, E type call has 3-6 separate pulses. The interval between introductory call and pulse group is $0.73{\pm}0.29$ s(n=159) in C type call and $0.60{\pm}0.21$ s (n=48) in D type call. The number of pulses in the pulse group is $30.08{\pm}8.69$ in C type call and $15.78{\pm}2.40$ in D type call. An increase in water temperature induces a decrease in C and D type call duration (C type call r= -0.4153, p<0.001, D type call r=-0.7064, p<0.001). In case of C and D type call, the interval between introductory call and pulse group influenced more call duration than pulse group duration. We regarded A, B type call as a territorial call and C, D type call as a mating call and we recognized that E type call has the function of threat or alarm.

Phytophthora nicotianae var. nocotianae에 의한 딸기 역병의 발생 (Occurrence of Phytophthora Rot of Strawberry Caused by Phytophthora nicotianae var. nicotianae)

  • A severe Phytophthora rot of strawberry caused by a species of Phytophthora has been widely occurred at major cultivation areas of Kimhae on August in 1997. Incidence of the disease was obtained in the range of 69.2~83.6% in surveyed 4 fields and showed an average of 75.2%. A species of Phytophthora was mostly isolated from the crown of infected strawberry plants and all the isolates were identified as P. nicotianae var. nicotianae (=P. parasitica). The fungus showed strong pathogenicity on strawberry by inoculation test. As a result of the leaf inoculation using mycelial disks of the fungus, both leaves and petioles were darkly browned, and were finally blighted. As a result of the root inoculation of zoospore suspension, both roots and crowns were rotten with dark brown. Although the fungus produced sporangia either on V-8 juice agar medium or liquid medium, the sporangia observed on the liquid medium appeared to be broadly turbinate and noncaducous. Moreover the fungus cultured on the liquid medium often produced sporangia having two papilla. The number of zoospores in sporangia was found to be ranged from 3 or 4 to as many as 20 or 25. In addition, the released zoospore from the sporangium became the cystospore during the prolonged culture of the fungus. The sporangia were measured as av. 49$\times$35 ${\mu}{\textrm}{m}$ with l/b ratio of 1.43. All isolates from crowns were heterothallic and A1 mating type since oospores were abundantly formed on clarified V-8 juice agar by dual culture with P. capsici A2 mating type. Aplerotic oospores were sized 24-26 ${\mu}{\textrm}{m}$. Antheridia were always amphigynous and recoreded an average of 12$\times$10 ${\mu}{\textrm}{m}$. Hyphal swlling were easily observed, and terminal or intercalary chlamydospores were abundantly formed on V-8 juice agar as well as in C/Z solution and sized av. 28.2 ${\mu}{\textrm}{m}$. This is the first report of Phytophthora rot of strawberry in Korea.

서울 및 강원지역 고양이의 혈액형 빈도 조사 (Prevalence of Feline Blood Types in Seoul and Kangwon Area of Korea)

  • To determine the distribution of feline blood types and then to estimate the risk of neonatal isoerythrolysis (NI) in non-pedigree cats, we typed blood of 482 cats of both genders and various breeds (336 domestic shorthair cat and 146 pedigree) from August 2005 through July 2007. Blood samples from Seoul and Kangwon province were typed within 5 days after collection by the simple tube method. High-titer anti-A antiserum and anti-B reagent, prepared with Triticum vulgaris lectin, were used to determine type A and type B blood, respectively. The majority of cats were type A (n = 465, 96.5%) and only 3.5% (n = 17) were type B. No type AB blood were detected. Blood type distributions among the non-pedigree and pedigree cats were similar: for non-pedigree cats, 96.4% were type A and 3.6% were type B, whereas for pedigree cats, 96.6% were type A and 3.4% were type B. All type B cats had a very strong agglutination reaction to anti-A antiserum: 8 sample for 3+ and 9 for 4+. Assuming 19% of estimated frequency for the type-B allele in domestic cats, the calculated proportion of random mating from this population at risk for developing NI was 3.4%. Based on this finding, it is strongly recommended that blood typing be performed prior to any blood transfusion or breeding to minimize blood type incompatibilities. Further comprehensive studies on the titer of naturally occurring antibodies in cat populations in Korea and the prevalence of possible NI in practice are clearly required.
