Effect of Dietary Supplementation of Fresh Water Algae Euglena on the Performance and Egg Quality and Fatty Acid Composition of Egg Yolk in Laying Hens (산란계에서 담수녹조류 Euglena의 첨가사료가 생산성 및 계란의 품질과 지방산 조성에 미치는 영향)

  • Feeding trials were conducted with Euglena strains grown under different media. The effect of supplementation of Euglena on the laying performance, egg quality and fatty acid composition of egg yolk was studied. In experiment I, two hundred eighty 32-wk-old ISA Brown layers were randomly assigned to seven dietary treatments for 4 wks. Each treatment consisted of 4 replications with 10 birds each housed in two birds cages. Control diet was formulated to have $17\%$ CP and 2,750 kcal ME/kg. Euglena gracilis Z. (EG) was added to control diet at the level of 0.25, 0.5, $1.0\%$ and Euglena gracilis Z. bleached and DHA enriched (EGBD; a strain mutated by streptomycin and cultivated in DHA enriched medium) at the level of 0.5, 1.0, $2.0\%$ in the diet. In experiment 2, three hundred 84-wk-old ISA brown layers were randomly assigned to five dietary treatments: T1; Control, T2; T1 + EGBD $0.5\%$, T3; T1 + Euglena gracilis Z. DHA enriched (EGD; cultivated in DHA enriched medium) $0.5\%$, T4; T1 + EGD $1.0\%$, T5; T1 + EGD $2.0\%$. Each treatment had 5 replication of 12 birds each housed in two birds cages. In experiments 1 and 2, Euglena suppplementation did not significantly affect egg production but increased egg weight and feed intake. In experiment 1, EG was more effective in increasing egg yolk color score than EGBD. Egg yolk color of EG $1\%$ treatment showed the highest score. EGBD supplementation increased DHA concentration of egg yolk. EGBD $2\%$ treatment showed the highest DHA and the lowest palmitic and stearic acids concentration in the egg yolk. In experiment 2, EGBD $0.5\%$ treatment showed highest DHA level in egg yolk (P<0.05). It was conducted that EGBD is a single cell protein source rich in DHA, that can be used to produce DHA enriched eggs.

Carbofuran Cytotoxicity on Rat Fibroblast Cells and Compensation Effects of Phenobarbital Sodium and 3-Methycholanthrene on Its Toxicity (살충제 Carbofuran이 쥐의 NIH3T3 섬유모세포에 끼치는 독성 및 Phenobarbital Sodium과 3-Methylcholanthrene에 의한 보상효과)

  • This study was carried out to investigate the effects of phenobarbital sodium(PB) and 3-methylcholanthrene(3-MC) on carbofuran cytotoxicity and to develop antitoxic agents based on the effectivness. Experimental groups for carbofuran cytotoxicity were divided into five groups ; medium alone and four treatments of carbofuran (1, 25, 50 and $100{\mu}M)$, and those for compensation effects were divided into six groups ; medium alone, $IC_{50}$ carbofuran and four combinations of carbofuran and PB or 3-MC($IC_{50}$ carbofuran plus 1, 25, 50, $100{\mu}M$ of PB and 3-MC, respectively). After incubation for 48 hrs under the same conditions, MTT(Tetrazolium MTT), NR(Neutral red) and SRB(Sulforhodamine B protein) assay were performed. Fifty percentage inhibition of MTT, NR, and SRB against carbofuran in rat fibroblast cell were 60.7, 82.5 and $87.0{\mu}M$, respectively. At the combination treatments of $IC_{50}$ of carbofuran and $100{\mu}M$ of PB, the significant compensation effects were observed from the results of MTT and NR but not from that of SRB absorbance. And at the combination treatments of $IC_{50}$ of carbofuran and 3-MC, the relatively significant compensation effects were found at $50{\mu}M$ 3-MC from the results of MTT and at $100{\mu}M$ 3-MC from that of NR and SRB absorbances, respectively. From the results of light microscopy, combination treatments of $carbofuran(IC_{50})$ and PB or 3-MC showed good regeneration in carbofuran toxicity of rat fibroblast cells. These results suggest that PB or 3-MC can compensate the cytotoxity of carbofuran insecticide in rat NIH3T3 fibroblast cells.

The Characteristics on the Spatio-temporal Distributions of Phytoplankton Communities in Deukryang Bay, Southwestern Korea (득량만 식물플랑크톤 군집의 시.공간적 분포특성.)

  • The observations on the spatio-temporal distribution and seasonal fluctuations of phytoplankton community were carried out in Deukryang Bay of the Korean Southwestern Sea from June 1992 to April 1993. A total of 75 species of phytoplankton belonged to 47 genera was identified. In Deukryang Bay seasonal succession in dominant species; P. alata, G. flaccida, S. costatum, L. danicus and N. longissima in summer, St. palmeriana, Ch. curvisetus and B. paxillifera in autunm, S. costatum, Ch. curvisetus, E. zodiacus and Pn. pungens in winter, and As. glacialis, As. kariana, N. pelagica, Th. nitzschioides and S. costatum in spring, were very marked, that is to say, the communities structure of phytoplankton in Deukryang Bay appeared to be various species composition and it was occupied with diatoms all the year round. Phytoplankton standing crops fluctuated with an annual mean of $1.4{\times}10^5 cells/1 between the lowest value of 2.6{\times}10^3 cells/1 in July and the highest value of 1.0{\times}10^6 cells/1$ by S. costatum in January. Densities of the phytoplankton cell number by the samples of Deukryang Bay ranged from $2.6{\times}10^3cells/1 to 1.2{\times}10^5 cells/1 with the mean value of 3.6{\times}10^4cells/1 in summer, from 6.0{\times}10^3cells/1 to 2.6{\times}10^5 cells/1 with mean of 1.5{\times}10^5 cells/1 in autumn, from 1.3{\times}10^4cells/1 to 1.0{\times}10^6 cells/1 with mean 3.5{times}10^5 cells/1 in winter, and from 4.8{\times}10^3cells/1 to 6.0{\times}10^5 cells/1 with mean of 1.6{\times}10^5 cells/1$ in autumn. That is to say, phytoplankton standing crops was large in low temperature seasons, on the other hand small in high temperature seasons. Chlorophyll $\alpha$ concentration fluctuated between 0.l9 $\mu$g/l and 12.3 $\mu$g/l in March. in Deukryang Bay seasonal flucturation in chi-$\alpha$ concentration was not marked. Especially, chl-$\alpha$ concentration in the water around Deukryang Island located in the middle part of Deukryang Bay showed patchy distributions with a very high concentration. And chl-$\alpha$ concentration was high during a year. Therefore, phytoplankton production in Deukryang Bay could be very high year-round.

  • The change of external/internal factors of the cell rquires specific biological functions to maintain life. Such functions encourage particular genes to jnteract/regulate each other in multiple ways. Accordingly, we applied a linear decomposition model IFSA, which derives hidden variables, called the 'expression mode' that corresponds to the functions. To interpret gene interaction/regulation, we used a cross-correlation method given an expression mode. Linear decomposition models such as principal component analysis (PCA) and independent component analysis (ICA) were shown to be useful in analyzing high dimensional DNA microarray data, compared to clustering methods. These methods assume that gene expression is controlled by a linear combination of uncorrelated/indepdendent latent variables. However these methods have some difficulty in grouping similar patterns which are slightly time-delayed or asymmetric since only exactly matched Patterns are considered. In order to overcome this, we employ the (IFSA) method of [1] to locate phase- and shut-invariant features. Membership scoring functions play an important role to classify genes since linear decomposition models basically aim at data reduction not but at grouping data. We address a new function essential to the IFSA method. In this paper we stress that IFSA is useful in grouping functionally-related genes in the presence of time-shift and expression phase variance. Ultimately, we propose a new approach to investigate the multiple interaction information of genes.

Effect of Fermented Brown Seaweed Waste (FBSW) on Milk Production, Composition and Physiological Responses in Holstein Dairy Cows (발효미역부산물의 첨가가 Holstein 비유우의 비유성적 및 생리반응에 미치는 영향)

  • This study was conducted to determine effects of fermented brown seaweed waste (FBSW) on milk production, composition and physiological responses as functional feed for 60 days in Holstein dairy cows. A total of 24 Holstein dairy cows (average age 49.33 months, average lactation days 175, Reproduction 2.0) were randomly allocated into control(basal diet), 1% FBSW (180g in basal diet) and 2% FBSW (360g in basal diet) groups with 8 replications for 60 days. Daily milk yield and composition (fat, protein, SNF, MUN) were not affected by FBSW supplementation, but Ca level in milk was significantly increased 4.29 mg/dl and 2.91 mg/dl in 1% and 2% groups compared to control group (p<0.05) at the end of the experiment, respectively. The somatic cell count (SCC) in milk was not significant. The plasma $T_4$ level (concentration) were increased in 1% and 2% FBSW compared to control group at the end of the experiment (p<0.05), but between triiodothyronine ($T_3$) and thyroxin ($T_4$) levels were not significant. Concentrations of plasma glucose in control, 1% FBSW and 2% FBSW groups were 64. 37mg/dl, 66.15mg/dl and 73.02 mg/dl and plasma NEFA level was 0.30~0.32 mEq/dl. Concentrations of BUN tended to be higher for FBSW group than control group. Although WBC, RBC, Hb, Hct, T-B, ALP, and GPT levels were not affected by FBSW supplementation, GOT level was significantly decreased in cows fed 1% FBSE diet compared to control group (P<0.05). Therefore we strongly suggest that the 1% FBSW supplementation in basal diet increases the milk yield and Ca level in Holstein dairy cows.

Active Polysaccharide and Immune Enhancement of Ganoderma lucidum Mycelium Cultured in Mushroom Complete Medium Supplemented with Ginseng Extract (수삼추출물 첨가 mushroom complete medium에서 배양된 영지버섯 균사체의 면역증진 효과 및 활성다당류)

  • After Ganoderma lucidum was cultured in mushroom complete medium (MCM) supplemented with ginseng extract (GE), crude polysaccharide (GL-GE-CP) was fractionated from mycelium. Among GL-GE-CP from mycelium in MCM supplemented with 5, 10, and 15% GE (v/v ratio of MCM to GE), GL-GE-15-CP (15% GE) most significantly enhanced macrophage stimulation and intestinal immune system modulating activity compared with GL-CP in MCM without GE. When GL-GE-15-CP was further fractionated on DEAE-Sepharose CL-6B, GL-GE-15-CP-II displayed more potent activity than subfractions from GL-CP on macrophage stimulation, interleukin-12 production, and intestinal immune system modulation (1.75-, 5.68-, and 1.76-fold, respectively). Anti-metastasis effect against colon 26-M3.1 carcinoma cells was also enhanced by GL-GE-15-CP-II (72.8% inhibition). In addition, GL-GE-15-CP-II contained neutral sugar (83.00%) and uronic acid (9.11%), and consisted of Ara, Man, Gal and Glc (molar ratio of 0.39:0.50:0.75:1.00). Furthermore, GE supplementation helped to enhance the immunomodulation in G. lucidum, and it is assumed that neutral polysaccharides play an important role.

The Effect of Hyperthermia Combined with Radiation on Crypts of the Mouse Jejunum (마우스공장 소낭선의 방사선 효과에 온열요법의 병용이 미치는 영향에 관한 실험적 연구)

  • The effect of local hyperthermia of 41 to $43^{\circ}C$ for 30 minutes on radiosensitivity of normal tissue was studied utilizing jejunal crypt microcolony assay. Hyperthermia of this range enhanced the radiation effect and the effect was mainly additive without significant effect on the slopes of cell survival curves. At the isoeffect level of 20 microcolony formation, the thermal enhancement ratio was 1.02, 1.10 and 1.39 for $41^{\circ},\;42^{\circ}\;and\;43^{\circ}C$, respectively. The distribution of microcolony formation along the circumference of jejunum was not uniform, having more colonies around the mesenteric border, and this suggests the effect of uneven cooling by blood circulation.

Evaluation and Reduction of Microbiological Hazard of Spoon and Spoon Case Carried by Nursery School Children (어린이집 유아 휴대 수저 및 수저집의 미생물학적 위해 분석 및 저감화)

  • This study was conducted to investigate the microbiological hazard of spoons and their cases carried by nursery school children and to evaluate the reduction effects of washing methods and ultraviolet (UV) treatments against Escherichia coli on the spoon and spoon case. A total of 78 spoons and their cases were sampled to test about total aerobic bacteria, coliform bacteria, Staphylococcus aureus, Bacillus cereus and Salmonella spp. Total aerobic bacteria were detected over 2.7 log CFU/100 $cm^2$ in 20 out of 36 spoons (55.6%), 9 out of 20 zipper-type spoon cases (45.0%) and 13 out of 22 plastic-type spoon cases (59.1%). Coliform bacteria were also detected in 19 out of 36 spoons (52.8%), 14 out of 20 zipper-type spoon cases (70.0%) and 14 out of 22 plastic-type spoon cases (63.6%). The pathogens tested in this study were not found in all samples except for the zipper-type spoon cases which were contaminated with Staph. aureus (2 samples) and B. cereus (3 samples). The results indicated that the sanitary conditions of spoons and their cases should be improved promptly. To evaluate the reduction effects of washing methods and UV treatments against E. coli, the spoons and their cases were treated at different cleaning times with and without soap, and different UV exposure times, respectively. E. coli with initial cell number of 4 log CFU on the spoons and their cases was not detected when they were cleaned at running water for 30 sec after dish sponging with soap for 30 sec. In UV treatments, E. coli with the same level of washing method was not detected after UV exposure for 15 minute in the spoons and their cases. From the results, the washing and UV treatment should be used to control the microbial contamination of spoons and their cases for more than 1 and 15 minutes, respectively.

Mass Screening of Lovastatin High-yielding Mutants through Statistical Optimization of Sporulation Medium and Application of Miniaturized Fungal Cell Cultures (Lovastatin 고생산성 변이주의 신속 선별을 위해 통계적 방법을 적용한 Sporulation 배지 개발 및 Miniature 배양 방법 개발)

  • For large and rapid screening of high-yielding mutants of lovastatin produced by filamentous fungal cells of Aspergillus terreus, one of the most important stage is to test as large amounts of mutated strains as possible. For this purpose, we intended to develop a miniaturized cultivation method using $7m{\ell}$ culture tube instead of traditional $250m{\ell}$ flask (working volume $50m{\ell}$). For obtaining large amounts of conidiospores to be used as inoculums for miniaturized cultures, 4 components i.e., glucose, sucrose, yeast extract and $KH_2PO_4$ were intensively investigated, which had been observed to show positive effect on enhancement of spore production through Plackett-Burman design experimet. When optimum concentrations of these components that were determined through application of response surface method (RSM) based on central composite design (CCD) were used, maximum spore numbers amounting to $1.9\times10^{10}$ spores/plate were obtained, resulting in approximately 190 fold increase as compared to the commonly used PDA sporulation medium. Using the miniaturized cultures, intensive strain development programs were carried out for screening of lovastatin high-yielding as well as highly reproducible mutants. It was observed that, for maximum production of lovastatin, the producers should be activated through 'PaB' adaptation process during the early solid culture stage. In addition, they should be proliferated in condensed filamentous forms in miniaturized growth cultures, so that optimum amounts of highly active cells could be transferred to the production culture-tube as reproducible inoculums. Under these highly controlled fermentation conditions, compact-pelleted morphology of optimum size (less than 1 mm in diameter) was successfully induced in the miniaturized production cultures, which proved essential for maximal utilization of the producers' physiology leading to significantly enhanced production of lovastatin. As a result of continuous screening in the miniaturized cultures, lovastatin production levels of the 81% of the daughter cells derived from the high-yielding producers turned out to be in the range of 80%$\sim$120% of the lovastatin production level of the parallel flask cultures. These results demonstrate that the miniaturized cultivation method developed in this study is efficient high throughput system for large and rapid screening of highly stable and productive strains.

Experimental Study on Autothermal Reformation of Methanol with Various Oxygen to Methanol Ratios for Fuel Cell Applications (연료전지용 메탄올 자열 개질기의 산소-메탄올 비율에 따른 성능 실험)

  • The use of Hydrogen as a fuel is receiving considerable attention and as a result, research on novel methods of hydrogen production is necessary so that the hydrogen demands in the future can be satisfied. This study presents experimental data on methanol Autothermal Reformation that quantifies the relationship between the oxygen-to-methanol ratio ($O_2/CH_3OH$) and reformer efficiency. For each catalyst configuration, the $O_2/CH_3OH$ was varied from 0.1 to 0.4, with an increment of 0.05, to investigate the effects of $O_2/CH_3OH$ on the reactor performance, including temperature profile, conversion, and efficiency. $O_2/CH_3OH$ was increased from 0.15 to 0.20, and the catalyst bed temperature increased by $235^{\circ}C$ to approximately $550^{\circ}C$. The catalyst bed temperature increased with increasing $O_2/CH_3OH$ as the reaction shifted from endothermic to exothermic reaction and as a result, excess heat, which raised the reactor temperature, was generated. The reactor performance was shown to be highly dependent on $O_2/CH_3OH$. The optimum $O_2/CH_3OH$ = 0.30 found in the experimental tests is 30% higher than the theoretical optimum of 0.23. This is attributed to a combination of factors such as the concentrations of the $O_2$ and $CH_3OH$ gas, reaction rate, catalyst effects, heat loss from the reactor, and the difference between the actual amounts of reaction products formed and the theoretical amounts of the reaction products.
