Effects of Plant Growth Regulators on in vitro Propagation of Echinosophora koreensis Nakai

  • To establish in vitro nodal culture conditions of Echinosophora koreensis Nakai, one of rare and endangered species famous for beautiful flowers in the Korean Peninsula, the influence of plant growth regulators (PGRs) on shooting and rooting from in vitro shoots was investigated. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the half-strength Driver and Kuniyuki's media in the range of 2.22 to 8.88 ${\mu}M $induced 2.5 to 2.7 shoots per axillary bud; and addition of 2.27 ${\mu}M $ thidiazuron (TDZ) produced 3.2 shoots, during 4 weeks of culture, while zeatin and isopentenyl adenine (2ip) were not effective on shoot multiplication as observed from several combination treatments of BA with other PGRs. Shoots established were smaller than 2 cm in length, in most of the treatments. while in BA 8.88 ${\mu}M $ treatment more than 30% of shoots were longer than 2 cm and shorter than 4 cm. In rooting, naphthalene acetic acid (NAA) from 5.37 to 21.48 ${\mu}M $ showed the rooting rate from 40.0 to 62.5%. Indole butyric acid (IBA) addition had little effect on rooting (<10%), although some roots in IBA-containing media were longer than those in NAA. Micropropagation from axillary buds of nodular explants was applicable and promising to multiplication and conservation of Echinosophora koreensis Nakai.

바이오리액터 배양기에 의한 석곡 유식물체 대량 증식 (Mass production of the seedlings of Dendrobium moniliforme using bioreactor culture)

  • Protocorms were newly formed from the culture of axillary buds, obtained in the seedlings of Dendrobium moniliforme in vitro. Its formation ratio was calculated to 43.7% on MS medium containing 1.0 mg/L BA. To test their survival ratio, we gradually increased the inoculation of transplant populations from single to more than three, and then found that the ratio in three populations went up as high as 95.2% rather than those of one or two. In bioreactor, explant obtained from the axillary bud grew well in lower concentration as 1/4 MS medium, while clearly grew slow in a little bit high concentration as 1/2 MS medium. We found that the explant of axillary bud, obtained from the Dendrobium moniliforme seedlings, would grow five times after culturing in a bioreactor for six weeks in 1/4 MS medium.

  • 본 실험은 여름딸기 무병묘 대량증식을 위해 bioreactor배양의 증식 및 경제성 효과를 비교하고자 실시하였다. 배양 방법은 반고체, 고체, 현탁배양 및 bioreactor 배양 등 4가지 방법을 이용하였다. 배양 6주 후, 식물체의 초장은 고체배양이 3.6cm로 가장 짧았으며, bioreactor 배양이 8.3cm로 가장 길었다. 생체중과 건물중은 bioreactor 배양이 2,261mg과 525mg으로 다른 배양방법에 비하여 가장 무거웠다. 액아는 반고체, 고체 및 현탁배양은 거의 발생하지 않았으나, bioreactor 배양은 주당 7개의 액아가 발생하였다. 경제성 분석 결과 기본식물 생산 시 bioreactor배양이 303원/주으로 고체배양의 845원/주보다 542원/주 적었다. 따라서 딸기 무병묘 생산 시 bioreactor배양이 대량증식 및 경제적인 면에서 효율적이었다.

  • The effect of plant growth regulators was investigated on in vitro shoot proliferation from axillary bud explants of Hypericum erectum. To determine the optimal cytokinin for proliferation of axillay buds, we carried out screening four cytokinins (BA, kinetin, 2iP, TDZ). When nodal segments were cultured on MS medium supplemented with $4.5\;{\mu}M$ TDZ (thidiazuron), a number of shoots were induced. Our results indicated that the addition of TDZ to culture medium resulted in the induction of significantly more axillary buds than in the addition of other cytokinins. The optimal concentration of TDZ for proliferation of axillary buds was $10\;{\mu}M$. 92% of shoots spontaneously rooted without any plant growth regulator (PGR) and formed whole plantlets within one month. More than 95% of these regenerants survived and they did not show any detectable variation in morphology or growth characteristics compared to their donor plants.

홍경천(Rhodiola rosea L.)의 액아배양을 통한 다신초 유도 및 기내 대량증식 (In vitro propagation and multiple shoot induction of Rhodiola rosea L. by axillary bud culture)

  • 본 실험은 약리적 효능이 뛰어난 홍경천의 기관 (액아가 포함된 줄기 조직)배양을 통한 대량증식 방법의 일환으로 액아배양을 통한 다신초를 유도, 증식조건에 관해 BA와 GA3의 영향을 조사하였고 신초증식에 필요한 적정 sucrose의 농도를 확인하였다. 그 후 적절한 기내 발근 및 토양순화 조건을 탐색하여 최적의 홍경천의 생산 조건을 확립하였다. 다신초의 유도 및 증식은 $GA_3$와 BA를 혼합 처리가 효과적이었다. 신초증식에 효과적인 sucrose의 농도는 50 g/L가 첨가된 배지였으며, 평균 7 cm 이상의 신장을 보였다. 기내 발근은 옥신류 식물생장조절물질 무첨가 배지에서 가장 높은 발근길이와 개수를 나타냈다. 기내발근된 유식물체의 토양순화는 모래상을 제외하고는 모두 양호한 생장을 보였고, 특히 모래, 피트모스, 펄라이트가 모두 첨가된 배합토에서 가장 양호한 생장을 보였다. 이상의 결과는 수년전 국내에 도입하려다 실패한 약용식물인 홍경천의 기내배양을 통한 재배의 가능성을 제시하는 것이고 더 나아가 다양한 생물소재의 활용 방안의 기초재료로 활용될 것으로 보여진다.

액아배양(腋芽培養)에 의한 신품종(新品種) 대추나무의 실용적(實用的)인 대량증식(大量增殖)에 관(關)한 연구(硏究) (Study on Practical Micropropagation of Juiube Cultivars through Axillary Bud Culture)

  • 조직배양(組織培養)에 의(依)한 대추나무 신품종(新品種) 실용적(實用的)인 대량증식방법(大量增殖方法)을 구명(究明)하기 위(爲)하여 액아(腋芽)를 이용(利用), 기내생장(器內生長) 및 야외순화(野外馴化) 시험(試驗)을 실시(實施)한 결과(結果)는 다음과 같다. 1. BAP 0.5mg/l가 들어있는 1/2MS배지(培地)에 charcoal을 첨가(添加)하여 기내생장(器內生長)을 조사(調査)한 결과(結果), '금성(錦城)'의 경우(境遇) charcoal 500mg/l 처리(處理)하였을 때 줄기의 신장(伸張) 및 발생(發生)된 줄기의 수(數)가 각각(各各) 6.4cm, 10개(個)로 가장 높았고 '복조(福棗)'에서는 1,000mg/l 처리시(處理時) 7.5cm, 12.4개(個)로 가장 좋았다. 2. 조사(調査)한 IBA 농도중(濃度中), 그 농도(濃度)가 증가(增加)할수록 발근율(發根率) 및 callus 생장(生長)이 양호(良好)하였으며, IBA 1.0mg/l 처리구(處理區)에서 줄기의 생장(生長)이 가장 좋았다. 3. 조직배양(組織培養)에 따른 생장반응(生長反應)의 차(差)를 조사(調査)한 바, '금성(錦城)'의 경우(境遇) 발생(發生)되는 줄기수에 있어 액아(腋芽)가 정경(頂莖)보다 2배(倍)정도 많았으나 '복조(福棗)'에서는 정경(頂莖)이 줄기신장(伸張) 및 발생(發生)되는 줄기수(數)에 있어 액아(腋芽)보다 좋은 결과를 보였다. 4. 기내(器內) 배양묘(培養苗) 순화시험(馴化試驗)에서는 근원기(根原基)만 형성(形成)된 묘(苗)가 활차율(活差率)이 97.8%로 발근묘(發根苗)보다 월등(越等)히 높았다. 5. 온실내(溫室內) 삽목시험(揷木試驗)에서는 paclobutrazol 처리(處理)가 발근율(發根率) 및 뿌리생장(生長)에 있어 IBA, NAA, $Rooton^{(R)}$ 처리구(處理區)보다 효과적(效果的)이었다.

Multiple shoot induction and plant regeneration from axillary buds of Magnolia 'Vulcan'

  • An efficient protocol for multiple shoot induction and plant regeneration from axillary bud culture of Magnolia 'Vulcan' was developed in the present study. Primary shoots were obtained from axillary bud explants cultured on Murashige and Skoog (MS) medium containing 1.0 mg/L 6-benzylaminopurine (BA). To induce multiple shoots effectively, primary shoot tips were cultured on MS medium supplemented with different concentrations of BA and zeatin at 0, 0.2, 0.5, and 1.0 mg/L. Of these treatments, the MS medium with 0.5 mg/L BA resulted in the highest number of shoots per explant with an average value of 5.9, and it produced the greatest shoot height at 4.8 cm after 12 weeks of culturing. In the rooting of in vitro produced shoots, the greatest percentage of explants forming roots (91.3%), number of roots per explant (9.7), and root length (2.8 cm) were obtained in half-strength MS medium supplemented with 6.0 mg/L indole-3-butyric acid (IBA). Regenerated plantlets were successfully acclimatized and hardened off inside the culture room with 87.5% survival rate. Plants were transferred to a greenhouse with a 97.2% survival rate. The highly efficient shoot multiplication and plant regeneration system reported herein can be used for large-scale clonal propagation of valuable Magnolia species or cultivars.

자작나무 성숙목의 근주맹아를 이용한 기내증식 (In Vitro Propagation Using Stool Shoots of Mature Betula platyphylla var. japonica)

  • 15년생 자작나무의 근주 맹아지 액아를 이식절편체로하여 기내배양시켜 효과적으로 증식시킬 수 있었다. 줄기증식에는 WPM에 BAP 0.5와 1.0mg/l 첨가한 배지가 효과적이었다. 기내발근에는 GD배치를 이용하였고 0.2mg/l IBA 첨가시 100% 발근되었다. 이렇게 증식된 식물체는 토양에 이식하여 95% 이상 활착되었으며 활착후 정상적인 생장을 보였다. 이상의 결과는 자작나무 성숙목의 기내 대량 증식 가능성을 시사한다.

In vitro plant regeneration from axillary buds of Hibiscus syriacus L.

  • Presently, we report a simple, reproducible and high frequency plant regeneration in Hibiscus syriacus L. using axillary buds. H. syriacus was regenerated from axillary buds directly or through a callus phase. Regenerated shoots were directly induced from young and fresh axillary buds cultured on Murashige and Skoog medium (MS) supplemented with 0.01 mg/L of the growth regulator thidiazuron (TDZ) after 2 weeks of culture. Directly induced shoots were transferred to hormone-free MS medium and root development was observed after 6 weeks. On the other hand, old and stale axillary buds were regenerated to shoots via callus induction on MS medium containing 0.01–2 mg/L TDZ after 4 weeks. A TDZ concentration of 0.01 mg/L was most effective in callus formation. Green callus was transferred to MS medium containing 0.01 mg/L α-naphthalene acetic acid (NAA) and 0.5 mg/L benzylaminopurine (BA). After 4 weeks, callus had developed into multiple shoots. Plantlets were formed from 10 week cultures of single shoots on hormone-free MS medium. Regenerated plantlets were cultured on MS medium for one month and then transferred to pots containing garden soil. Potted plants were acclimatized for one month and grown to maturity under greenhouse conditions. The present study has shown that various concentrations of plant growth regulator can be effective for in vitro plant regeneration of H. syriacus. The direct and indirect regeneration protocol presented here will be useful for understanding the manipulation and propagation of H. syriacus.

Cryopreservation of in vitro-cultured Axillary Shoot Tips of Japanese Bead Tree (Melia azedarach) using Vitrification Technique

  • In vitro-grown axillary buds of Melia aredarach were successfully cryopreserved by vitrification. On the MS medium supplemented with BA 1 mg/L, multiple shoots were developed within $4{\sim}5$ weeks. Plantlets of Melia azedarach were cold-hardened at $10^{\circ}C$ for a 16-hr photo-period for 6 weeks. Excised axillary shoot-tips from hardened plantlets were precultured on a solidified Murashige & Skoog agar medium (MS) supplemented with 0.7 M sucrose for 1 day at $25^{\circ}C$. Axillary shoot-tip meristems wert dehydrated using a highly concentrated vitrification solution (PVS2) for 60 min at $0^{\circ}C$ prior to a direct plunge into liquid nitrogen (LN). The PVS2 vitrification solution consisted of 30% glycerol (w/v), 15% ethylene glycol (w/v), 15% DMSO (w/v) in MS medium containing 0.4M sucrose. After short-term warming in a water bath at $40^{\circ}C$, the meristems were transferred into 2 ml of MS medium containing 1.2M sucrose for 15 min and then planted on solidified MS culture medium. Successfully vitrified and warmed meristems resumed growth within 2 weeks and directly developed shoots without intermediary callus formation. The survival rate of cold-hardened plantlets for 3 and 4 weeks was 90%. We did not find any difference in PCR-band patterns between control and cryopreserved plants. This method appears to be a promising technique for cryopreserving axillary shoot-tips from in vitro-grown plantlets of Medicinal plants.
