Regeneration and Acclimatization of Regenerants in Long-term in vitro Culture of Japanese Blood Grass (Imperata cylindrica 'Rubra')

  • Long-term culture of cell lines is an important issue in in vitro culture and in plant science. In this study, the regeneration ability and ex vitro acclimatization of regenerants were evaluated. The ploidy level of regenerants derived from long-term cultured cell lines was measured in Imperata cylindrica 'Rubra', Poaceae. Adventitious buds (shoots) were successfully induced from five-year-cultured calli on MS medium containing 0.1 mg/L BA or 2.0 mg/L TDZ, combined with 0.01 mg/L auxins (IAA, IBA, NAA and 2,4-D), respectively. Adventitious roots were also induced on MS medium containing 0.01 mg/L auxins (IBA, NAA and 2,4-D), respectively. Interestingly, regenerants with both red and green leaf were successfully obtained when regenerants were cultured on MS medium with 9% sucrose. Regenerants derived from long-term cultured calli were transferred to pots using an optimal acclimatization process and successfully adapted to both pot and soil conditions. Moreover, the ploidy level was measured using calli and regenerants that had been kept on MS medium containing various kinds of plant growth regulators (PGRs).

  • Shoot proliferation was obtained from shoot tips and nodal explants of Vitex negundo L. on MS medium supplemented with either BAP or KIN (0.1-2.0 mg/L) alone or in combination with NAA (0.1 mg/L). The concentrations of cytokinins combined with NAA produced multiple shoots from shoot tips and nodal explants. The highest mean percentage (84.3$\pm$8.0) of shoot multiplication's were observed on nodal explants in the presence of BAP (1.5 mg/L) and NAA (0.1 mg/L) followed by shoot tips (65.0$\pm$5.0). The regenerated shootlets were rooted on MS basal medium IAA, IBA, NAA (0.1-1.5 mg/L). The maximum number of roots (51.0$\pm$2.6) was achieved on the medium containing IBA (1.0 mg/L) followed by other auxins (NAA, IAA). The regenerated plants were successfully transferred to a mixture of vermiculate and soil. About 95% of the plantlets survived when transferred to the field.

  • Effects of growth regulators(2,4-D, IAA, NAA, BA and kinetin) and chilling treatment on callus induction. embryogenesis and regeneration through anther cultures of B. falcatum L. were examined. Frequency of callus induction and embryogenesis was effectively increased by the treatment of chilling at 5$^{\circ}C$ for 10 days before anther inoculation. Optimal level of growth regulator for callus induction and embryogenesis from anther was 2,4-D 1.0 mg/L in Murashige and Skoog(MS) basal medium supplemented with 30 g/L sucrose, 8 g/L agar. Frequency of embryogenesis from anther derived callus was increased up to 48% or 45% by addition of IAA 0.1+ kinetin 1.0 mg/L of IAA 0.1+ BA 1.0 mg/L in MS medium, respectively, Optimal medium for obtaining green callus was MS basal supplemented with BA 1.0mg/L. Addition of auxins(IAA or NAA) inhibited the formation of green callus from anther derived callus.

  • This study was conducted to development of rapid propagation method by leaf cuttings in Hibiscus hamabo native to southern seaside of Korea, and special object of this study was to determine the effects of IAA, IBA, and NAA on rooting in leaf cuttings of H. hamabo. Rooting was promoted by dipping treated with IAA and IBA. And rooting percentage was greatest at 1,000 and 2,000$mg{\cdot}L^{-1}$ IBA. At higher concentrations of IAA and IBA, more adventitious roots were developed. Also, IAA at high concentrations (above 2,000$mg{\cdot}L^{-1}$) and IBA ranged from 500~2,000$mg{\cdot}L^{-1}$ promoted on root number and rootingratio. However, root formation in Hibiscus leaf cuttings inhibited by NAA application.

  • The effects of various plant growth regulators, both auxins and cytokinins, on cell growth and berberine production were investigated in cell suspension cultures of Thafictrum rugosum. Indole-%-acetic acid (IAA) was found to be the best for berberine production among five examined plant growth regulators and the optimum concentration of IAA was 1 $\mu \textrm M$. The enhancement compared to control 2, 4-dichlorophenoxyacetic acid (2, 4-D) was more than 60%. Simultaneous addition of cytokinins such as kinetin and 6-benzylamiroyurine (BA) was inhibitory.

  • The callus formation and shoot growth of germinating rice seedlings in the aseptic culture media treated with 2.4-D and N.A.A. were studied with 3 rice cultivars. Callus formation and shoot growth were the best in the media including $10^-4$M N.A.A.. Varietal differences also were significant in the formation of callus and shoot growth from the callus.

Seedling Propagation by Stem Cuttings in Yacon

  • Four experiments were conducted to determine growth regulators, their concentrations and treatment time, rooting medium, and plant parts for optimum seedling propagation by stem cuttings in yacon (Polymnia sonchifolia Poepp. & Endl.). NAA was the most effective on rooting, and followed by IBA 2,4-D, and IAA. Dipping the base of cuttings in 50 or 100 ppm solutions of IBA increased rooting ratios. The 100 ppm solution of IBA was 14% higher than the 50 ppm solution for rooting ratios. Rooting medium with equal volumes of clay loam and sand was the most suitable for rooting of cuttings. Cuttings with both terminal bud and the first node or cuttings with both the first and second nodes were higher in rooting ratios than cuttings with only terminal bud or the first or second nodes. Days to rooting was not affected by plant parts.

  • This experiment was aimed to develop possible methods to control the maturity of grape berries through the application of exogenous plant growth substances and physical treatments such as defoliation or girdling. 1. Chlormequat and paclobutrazol increased anthocyanin but did not affect soluble solids contents and maturity. Girdling enhanced maturation and solids accumulation whereas defoliation delayed maturity. Solids content of berries in defoliation treatment did not reach to the level of other treatments even when fully ripened. 2. Ripening of grapes is greatly delayed for 20 to 30 days by the application of auxins (2,4-D and fenoprop) compared to the untreated control. Uneven ripening of berries in those clusters was observed when the concentration of auxin was over 50 ppm. Thus, about 30% of berries remained green until the normal berries were overripened. 3. Gibberellin did not affect the maturity of grape berries but maturity was greatly delayed when GA was applied with auxins. Also, uneven coloration between berries was observed such as in the application of auxin alone. 4. Ethephon application combinded with calcium at veraison showed no effect on berry ripening but increased anthocaynin contents. It can be concluded that harvest time of 'Campbell Early' grapes can be effectively extended by delaying the maturity through the application of auxin.

  • Immature ovule of intergeneric $F_1$ hybrid between Platycodon grandiflorum x Codonopsis lanceolata for producing embryogenic callus. somatic embryos and plant regeneration were cultured in vitro on various medium as well as MT(Murashige Tucker)medium treated with different concentration of plant growth regulators. Embryogenic callus induction was highest in the treatment of NAA 0.5 $mg/{\ell}$ and zeatin 0.01 $mg/{\ell}$ added on MT medium, whereas it was lower in treatments with auxins alone. MT medium were more effective in production of somatic embryos from incubated embryogenic callus. Most favorable plant growgh regulator for producing somatic embryos was 2. 4-D 0.5 $mg/{\ell}$ and zeation. BAP 0.01$mg/{\ell}$, but hormone-free and auxins alone were less effective. NAA 0.01$mg/{\ell}$ added with zeation 0.5 $mg/{\ell}$ was effective as high as NAA 0.01 $mg/{\ell}$ alone in normal plant regeneration from somatic embryo.
