Effects of methyl jasmonate (MeJA) on ethylene production in mungbean (Phaseolus radiatus L.) hypocotyl and leaf segments were studied. Ethylene production in mungbean hypocotyl segments was decreased in proportion to MeJA concentrations and $450\;\mu\textrm{M}$ of MeJA showed 50% inhibitory effect. This inhibitory effect appeared after 3 h of incubation period and continued for 24 h. Inhibition of ethylene production by MeJA was due to the decrease in 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase activity. However, MeJA treatment had no effect on ACC content and ACC synthase activity. MeJA also inhibited auxin-induced ethylene production in hypocotyls. To investigate the mechanisms of the inhibitory effect of MeJA on the auxin-induced ethylene production, ACC synthase and ACC oxidase activity were examined after MeJA treatment. MeJA decreased the ACC content and ACC synthase activity as weD as ACC oxidase activity in the auxin-treated tissue. These results suggest that the inhibition of MeJA on auxin-induced ethylene production is not due to the direct inhibitory effect of MeJA on the ACC synthase, but to the inhibition of the ability of IAA to promote the synthesis of ACC synthase. In contrast, ethylene production from the detached mungbean leaves was stimulated by MeJA. The rate of ethylene production increased approximately 65% over the control after 12 h of incubation period by $4.5\;\mu\textrm{M}$ MeJA. When MeJA was applied to detached leaves along with IAA, the effect of MeJA appeared to be additive. In an effort to elucidate mechanisms of MeJA action on auxin-induced ethylene production in the leaf tissue, enzyme activities of ACC synthase and ACC oxidase were examined. MeJA stimulated ACC oxidase activity but did not affect ACC synthase activity in leaf tissue. This study was conducted to determine the effects of cutting position, media, and auxin on rooting of Artemisia keiskeana cuttings. In the cutting positions, the cutting of middle and basal positions had a better growth in the both upper and lower part than the cutting of the top position. Especially, the cutting of the middle position showed the highest rooting rate, at 68.3%. In the media, peatmoss showed the latest date on the rooting initiation. Also, the growth, survival rate and rooting rate of the upper part and lower part were low. However, the cutting in vermiculate showed the fastest date on the rooting initiation, and had a significant effect on the growth of the upper part and lower part, showing the highest rooting rate and survival rate in all tested media. When cuttings were treated by auxin, the rooting rate and growth of cuttings were higher than the control. The control showed the lowest rooting rate, at 41%, while, the auxin treatment showed the highest, rate over 80%. Whereas when treated by NAA, the lower part of cuttings had a better growth than these treated by IBA. Also, the higher the concentration of auxin was, the higher the rooting rate and growth rate of Artemisia keiskeana cuttings were.
The effects of stigma removal from floral organ and treatment of auxin on those regions were evaluated on the fruit growth and quality in strawberry (Fragaria ananassa Duch.). The removal of stigma reduced the fruit weight, fruit length, and fruit width, while fruit growth declined as the number of removed stigma increased. The removal of stigma also resulted in reduced fruit firmness, sugar content, and total anthocyanin content, and decreased value of Hunter a for red color and increased value of Hunter b for yellow. The number of seeds deceased as the regions of removed stigma increased. The treatment of auxin on the region of removed stigma improved fruit size, while it reduced fruit firmness, sugar content, and anthocyanin content. The delay in fruit ripening was observed from both stigma removal and auxin treatment. The incidence of malformed fruits increased as the regions with removed stigma increased. The treatment of auxin also promoted fruit malformation.
This study was carried out to investigate the optimal condition for multiple propagation through leaf tissue culture and to apply anther culture techniques to Pulsatilla koreana Nakai breeding. Leaf and anther of Pulsatilla koreana Nakai were cultured on MS, MT, LS and $B_5$ media supplemented with several growth regulators and nitrogen sources under various conditions. For callus induction and differentiation from the Pulsatilla koreana leaf segments were more effective in the combination of zeatin and auxin than auxin alone. The color of the callus was green when treated with IBA alone. Shoot differentiation was more effective when treated with zeatin than auxin alone, especially the best hormoal combination for shoot differentiation was zeatin 1.0mg/l +NAA 0.1mg/l, while 2,4-D inhibited shoot differentiation. The appeared rate of S pollen was 35% in vivo, while that of S pollen by low temperature$(4^{\circ}C)$ pretreatment for 4 days was increased by 53% and the optimum culture time for callus induction from anther was uni-nucleate stage. $B_5$ basal medium supplemented with NAA 0.5mg/l and zeatin 1 mg/l was the most effective on callus formation and the best results of plant regeneration were obtained from combination of NAA 0.5mg/l and zeatin 0.5mg/l in anther culture. $NH_{4}NO_3$ as more effectives as the nitrogen source than $KNO_3$ and the combination with zeatin 2.0mg /L was the best effective. The best combination for plant regeneration in callus induced from anther was $NH_{4}NO_3$ 1650mg/l + $KNO_3$ 3800mg/l + zeatin 2.0mg/l. Ploidy level of anther-derived plants appeared 28% haploid, 47% diploid and the others were triploid, tetraploid and mixploid. In compare with E.S.T, M.D.H and P.X banding patterns were distinguished among callus, haploid and diploid plants in electrophoresis.
Nineteen nitrogen-fixing bacteria were isolated from rice roots cultivated in the southern part of the Korean peninsula. Among them, three isolates - Stenotrophomonas sp. KNUC89, Pseudomonas sp. KNUC116, and Delftia sp. KNUC133 - showed antifungal activities against phytopathogenic fungi Rhizoctonia solani and Fusarium oxysporum. Besides they could produce auxin and siderophores.

Callus-inducing ability of Alnus hirsuta was examined by culturing various tissues (leaf, hypocotyl, cotyledon and seed) on NT (Nagata & Takebe) medium, supplemented with 2.5$\mu$M 2,4-D. Leaf-originated callus was cultured on media varying in auxin (IBA and NAA) and cytokinin (BAP) concentrations to examine the effects of auxin and cytokinin on callus growth. Maximum growth was obtained at 10 $\mu$M IBA+10$\mu$M BAP and 10$\mu$M NAA without cytokinin. Cell suspensions established from cotyledon-originated callus yielded viable protoplasts after incubation for 16-18 hours in an enzyme mixture (1% (w/v) Onozuka R-10 0.5% (w/v) Macerozyme, CPW salts and 13% (w/v) mannitol, pH 5.8). Protoplasts were cultured on NT medium, supplemented with glucose, hormones and coconut milk. After 6 weeks of culture, protoplasts sustained cell divisions to form microcallus, which showed various colors from red to white.
Brassinosteroids are known to promote cell elongation in a wide range of plant species but their effect on cell division has not been extensively studied. The effect of brassinolide on the kinetics and final division frequencies of regenerating petal protoplasts of Petunia hybrida Vilm v. Comanche was examined. Under optimal auxin and cytokinin conditions, 10-100 nM brassinolide not only reduced the time of first cell division by 4.5 days but also altered the final division frequencies after 10 days of culture. One micromolar brassinolide showed the same acceleration of first cell division but inhibited the final division frequency by approximately 9%. Under sub-optimal auxin conditions, 10-100 nM brassinolide accelerated the first cell division, but no significant increase in the 8-10 days final division frequencies. Isolated protoplasts may provide a useful model system for the investigation of the molecular mechanisms of brassinosteroid action on cell division and proliferation in higher plants.
