The bacterial effector protein RavZ is secreted by the intracellular pathogen Legionella pneumophila and inhibits host autophagy through an irreversible deconjugation of mammalian ATG8 (mATG8) proteins from autophagosome membranes. However, the roles of the LC3 interacting region (LIR) motifs in RavZ function remain unclear. In this study, we show that a membrane-targeting (MT) domain or the LIR motifs of RavZ play major or minor roles in RavZ function. A RavZ mutant that does not bind to mATG8 delipidated all forms of mATG8-phosphatidylethanolamine (PE) as efficiently as did wild-type RavZ. However, a RavZ mutant with a deletion of the MT domain selectively delipidated mATG8-PE less efficiently than did wild-type RavZ. Taken together, our results suggest that the effects of LIR motifs and the MT domain on RavZ activity are complementary and work through independent pathways.
Autophagy, which is a process of self eating, has gained interest in the past decade due to its both beneficial and controversial roles in various biological phenomena. The discovery of autophagy genes (ATG) in yeast has led to focused research designed to elucidate the mechanism and regulation of this process. The role of autophagy in a variety of biological phenomena, including human disease, is still the subject of debate. However, recent findings suggest that autophagy is a highly regulated process with both beneficial and negative effects. Indeed, studies conducted using various model organisms have demonstrated that increased autophagy leads to an extended lifespan. Despite these findings, it is still unknown if all pathways leading to extended lifespan converge at the process of autophagy or not. Here, an overview of modern developments related to the process of autophagy, its regulation and the molecular machinery involved is presented. In addition, this review focuses on one of the beneficial aspects of autophagy, its role in lifespan regulation.
Autophagy is an evolutionally conserved cytoplasmic degradation system in which varieties of materials are sequestered by a double membrane structure, autophagosome, and delivered to the lysosomes for the degradation. Due to the wide varieties of targets, autophagic activity is essential for cellular homeostasis. Recent genetic evidence indicates that autophagy has a crucial role in the regulation of animal lifespan. Basal level of autophagic activity is elevated in many longevity paradigms and the activity is required for lifespan extension. In most cases, genes involved in autophagy and lysosomal function are induced by several transcription factors including HLH-30/TFEB, PHA-4/FOXA and MML-1/Mondo in long-lived animals. Pharmacological treatments have been shown to extend lifespan through activation of autophagy, indicating autophagy could be a potential and promising target to modulate animal lifespan. Here we summarize recent progress regarding the role of autophagy in lifespan regulation.
The present experiment was performed to investigate the acute effects of cadmium on ultrastructural and biochemical changes in mouse kidney and compare these changes with liver damage. Mouse were injected with cadmium chloride at a dose of 5 mg/kg body weight. After treatment, mouse were sacrificed at time intervals of 6, 12, 24 and 48 hours. It was observed that ultrastructural changes in mouse kidney were composed of swelling of mitochondria, dilation in endoplasmic reticulum, wrinkling at basal infolded membrane, formation of autophagosome and partial loss of microvilli in brush. border, and that ultrastructural changes in liver were mitochondrial change, dilation and deterioration of rough endoplasmic reticulum and proliferation of smooth endoplasmic reticulum. Biochemical effects of cadmium were more severe on liver than kidney. Therefore, acutely injected cadmium caused not only liver damage, but also kidney damage.
Autophagy is the sequential process whereby cell components are degraded, which can occur due to nutrient deprivation. Its regulation has an essential role in many diseases, functioning in both cell survival and cell death. Autophagy starts when mTORC1 is inhibited, resulting in the activation of several complexes to form a cargo that fuses with a lysosome, where it undergoes degradation. In this review, we describe a plant extract that is well known in Korea, namely Korean ginseng extract; we studied how its derivatives and metabolites can regulate autophagy and thus mediate the pathogenesis of certain diseases.

Ataxia-telangiectasia mutated (ATM), a master kinase of the DNA damage response (DDR), phosphorylates a multitude of substrates to activate signaling pathways after DNA double-strand breaks (DSBs). ATM inhibitors have been evaluated as anticancer drugs to potentiate the cytotoxicity of DNA damage-based cancer therapy. ATM is also involved in autophagy, a conserved cellular process that maintains homeostasis by degrading unnecessary proteins and dysfunctional organelles. In this study, we report that ATM inhibitors (KU-55933 and KU-60019) provoked accumulation of autophagosomes and p62 and restrained autolysosome formation. Under autophagy-inducing conditions, the ATM inhibitors caused excessive autophagosome accumulation and cell death. This new function of ATM in autophagy was also observed in numerous cell lines. Repression of ATM expression using an siRNA inhibited autophagic flux at the autolysosome formation step and induced cell death under autophagy-inducing conditions. Taken together, our results suggest that ATM is involved in autolysosome formation and that the use of ATM inhibitors in cancer therapy may be expanded.
자가소화작용은 세포 내 물질들을 이중막 구조의 자가포식체를 형성하여 라이소좀(lysosome)과 융합한 후 이들을 분해시키는 과정이다. 자가소화작용은 유전적, 세포적, 생화학 수준에서 연구가 진행되어왔지만, 자가포식체의 미세구조 분석 및 정량적 분석은 체계적으로 이루어지지 않았다. 본 연구에서는 먼저, 기초 자가소화작용이 진행되는 환경과 기아상태로 자가소화작용을 유도한 환경의 세포에서 각각 자가포식체 막 형성에 중요한 역할을 하는 ATG5가 결핍된 섬유아세포와 정상 섬유아세포에서의 자가포식체의 미세구조를 비교 관찰 하였다. 정상 섬유아세포에서는 초기 패고포어, 초기 및 후기 자가포식체, 오토라이소좀과 같은 자가포식체의 단계별 미세구조를 확인한 반면, ATG5가 제거된 섬유아세포에서는 이중막 구조가 완성되지 않은 자가포식체 구조가 증가한 것을 확인하여 ATG5가 자가포식체 성숙에 중요한 역할을 함을 다시 한번 검증할 수 있었다. 본 연구를 통해 구축한 자가포식체 미세구조 분석법을 활용하여 확장된 폴리글루타민을 가지는 N-말단 헌팅틴 유전자를 발현시키는 헌팅턴병의 세포학적 모델에서 단계별 자가포식체를 분석하였다. 그 결과, 흥미롭게도 헌팅틴 단백질이 발현된 세포에서 후기 자가포식체가 증가하는 것을 확인할 수 있었으며, 이를 통해 헌팅틴 돌연변이 단백질 응집체 제거에 필요한 자가소화작용 활성화를 위해서 자가포식체와 라이소좀의 결합 단계를 촉진하는 것이 효과적일 가능성에 대해서 유추할 수 있었다. 결론적으로 전자현미경을 활용한 자가포식체의 단계별 미세구조 관찰 및 정량 분석이 다양한 인간 질병모델에 적용되고, 자가소화작용과 연관된 병리 메커니즘 연구에 기여할 수 있을 것으로 생각된다.
이 연구는 비만이 심장 조직에서 자가포식 관련 단백질 발현에 미치는 영향을 확인하기 위해 고지방 식이(20주)를 통해 비만을 유도한 후 8주간의 트레드밀 운동을 실시하고, 자가포식의 유도, 형성 그리고 자가포식포와 라이소좀 융합단계를 조절하는 단백질의 발현을 확인하였다. 실험동물(SD rat)은 20주간의 고지방식이(탄수화물: 20%, 지방: 60%, 단백질: 20%)를 통해 비만을 유도하였으며, 8주간의 트레드밀 운동(주 5일, 하루 30분, 5분; 8m/min, 5분; 11m/min, 20분; 14m/min)을 실시하였다. 집단 구분은 정상식이 비교군(n=10), 고지방식이 비교군(n=10), 고지방식이 운동군(n=10)으로 구분하였다. 8주간의 트레드밀 운동 실시 전과 후에 경구당부하검사를 실시하여 곡선 하 면적(area under the curve; AUC)을 산출하였으며, 공복시 인슐린 농도와 포도당 농도를 통해 인슐린 저항성 지표인 HOMA-IR과 체중 당 복부지방량(abdominal visceral fat/Body weight; AVF/BW)를 산출하여 비교하였다. 또한 심장 조직에서 자가포식 관련 단백질을 분석하여 운동 트레이닝의 효과를 검증하였다. 장기간의 고지방식이를 통해 HFD-CON 그룹에서는 비만이 유도되었으며, ND-CON 그룹에 비해 체중, AUC, HOMA-IR, AVF/BW가 증가되는 것으로 나타났다. 하지만 8주간의 트레드밀 운동을 실시한 HFD-TE 그룹에서는 AUC, HOMA-IR, AVF/BW가 개선되는 것으로 나타났다. 체중의 경우, 감소되는 경향은 있었지만, 통계적으로 유의한 차이는 없었다. 자가포식 유도에 관여하는 mTOR와 AMPK는 비만상황에서 모두 감소되었지만, 운동을 통해 증가되는 것으로 나타났다. 자가포식 형성에 관련된 Beclin-1, BNIP3, ATG-7, p62, LC3는 비만상황에서 모두 증가하는 것으로 나타났으며, 운동을 통해 감소되는 것으로 나타났다. 자기포식포와 라이소좀 융합단계 조절하는 Cathepsin L과 LAMP2는 비만상황에서 모두 감소되었으며, 운동을 통해 증가하는 것으로 나타났다. 트레드밀 운동과 같은 신체활동은 대사성 질환에서 나타나는 병리학적 현상을 개선하고 자가포식 과정을 정상적으로 유도하는 것으로 나타났다. 따라서 트레드밀 운동이 심장 관련 질환의 예방 및 치료에 있어 일차적으로 고려해야할 필요성이 있다고 제안한다.
본 연구에서는 레스베라트롤이 당뇨병 및 비알코올성 지방간 질환 개선 효과를 가지는지를 규명하기 위해 고지방 식이 유도 비만 쥐를 대상으로 레스베라트롤을 4주간 osmotic pump를 사용하여 공급한 후 정상대조군과 고지방식이 제공 비만군과 비교 분석하였고 그 결과는 다음과 같다. 1) 고지방식이 유도 비만 쥐를 대상으로 8 mg/kg/day의 레스베라트롤을 4주간 처리한 결과 체중 변화, 간 조직 중량, 식이 섭취량에 영향을 미치지 않았다. 2) 레스베라트롤은 공복 혈당, 혈청 내 인슐린, 중성지방, 총 콜레스테롤 농도를 낮추었고, 인슐린 작용을 촉진시키는 혈청 아디포넥틴 수준을 개선시켰다. 또한, 고지방식이에 의해 높아진 간 조직 내 중성지방과 총콜레스테롤 농도를 낮추어 레스베라트롤이 지방간 개선 효과를 가질 수 있음을 제안하였다. 3) 자가포식의 표지인자인 autophagosome 생성과 LC3-II 형성 분석 결과, 고지방식이에 의해 과도한 자가포식이 유도되었음을 확인하였다. 레스베라트롤 처리는 이중막을 가지는 autophagosome 생성과 LC3-II 형성을 감소시켜 고지방식이에 의해 유도된 과도한 자가포식을 억제시킴을 보여주었다. 결론적으로 고지방식이와 함께 레스베라트롤을 제공하는 것은 당뇨병과 비알코올성 지방간 질환 관련 대사 인자들을 개선시키고, 이는 간에서의 자가포식 조절과 관련이 있다고 제안한다.
Background: Ginsenoside Rg3, a primary bioactive component of red ginseng, has anti-cancer effects. However, the effects of Rg3-enriched ginseng extract (Rg3RGE) on apoptosis and autophagy in breast cancer have not yet been investigated. In the present study, we explored the anti-tumor effects of Rg3RGE on breast cancer cells stimulated CoCl2, a mimetic of the chronic hypoxic response, and determined the operative mechanisms of action. Methods: The inhibitory mechanisms of Rg3RGE on breast cancer cells, such as apoptosis, autophagy and ROS levels, were detected both in vitro. To determine the anti-cancer effects of Rg3RGE in vivo, the cancer xenograft model was used. Results: Rg3RGE suppressed CoCl2-induced spheroid formation and cell viability in 3D culture of breast cancer cells. Rg3RGE promoted apoptosis by increasing cleaved caspase 3 and cleaved PARP and decreasing Bcl2 under the hypoxia mimetic conditions. Further, we identified that Rg3RGE promoted apoptosis by inhibiting lysosomal degradation of autophagosome contents in CoCl2-induced autophagy. We further identified that Rg3RGE-induced apoptotic cell death and autophagy inhibition was mediated by increased intracellular ROS levels. Similarly, in the in vivo xenograft model, Rg3RGE induced apoptosis and inhibited cell proliferation and autophagy. Conclusion: Rg3RGE-stimulated ROS production promotes apoptosis and inhibits protective autophagy under hypoxic conditions. Autophagosome accumulation is critical to the apoptotic effects of Rg3RGE. The in vivo findings also demonstrate that Rg3RGE inhibits breast cancer cell growth, suggesting that Rg3RGE has potential as potential as a putative breast cancer therapeutic.
